Strong tolerance of freezing is an important strategy for insects living in extremely cold regions. They produce highly effective cryoprotectant systems consisting of ice-nucleating proteins and polyols, which enables tolerable freezing of the body fluid. Therefore, the measurement of the concentrations of polyols and the activity of ice nucleators in the haemolymph is an essential tool for describing tolerance to ice formation in insects occurring in particularly cold places. This study evaluates three parameters: insect body supercooling point (SCP), haemolymph glycerol content and the profile of haemolymph ice nucleating activity that characterize the strategies of cold adaptation and cold hardiness in two previously unstudied beetles, Chrysolina graminis graminis L. and Galerucella nymphaea L., inhabiting Yakutia (Russian Far East, latitude 62°N). The high SCP values, ice nucleating activity and survival of the chrysomelids after freezing indicate that both species are tolerant of freezing. According to the profiles of ice-nucleating activity, the haemolymph from C. graminis graminis is characterized by a higher nucleating potential than that from G. nymphaea. The glycerol level is also higher in C. graminis graminis. The results indicate that both species develop tolerance to low temperatures, but the cold hardiness potential of C. graminis graminis is greater than that of G. nymphaea. This was revealed by the survival test, in which beetles were frozen to a temperature of -22°C for 30 min; 86% of C. graminis graminis and 72% of G. nymphaea survived the test. Thus, the freeze-tolerance of these beetles seems to be based on the production of an integrated cryoprotectant system, the quality of which apparently influences the range of their cold resistance., Natalia G. In some regions of Argentina and Brazil, the South American fruit fly Anastrepha fraterculus (Wied.) (Diptera: Tephritidae) causes significant damage to crops. An efficient integrated management program requires knowledge of pest population dynamics, dispersion patterns, sexual and oviposition behaviour, and adaptive landscape. The present study combined simple sequence repeat (SSR) molecular markers and morphometric datasets in order to analyse the population structure and infer the oviposition resource use strategy of the females. Infested guava fruits were collected from nine wild trees in Tucumán, Argentina, and a total of 140 adult A. fraterculus were recovered. These were then measured for six morphometric traits and 89 of them were genotyped for eight SSR loci. Genetic variability estimates were high (expected heterozygosity = 0.71, allelic richness = 12.5), with 8 to 20 alleles per locus. According to Wright's F-statistics estimates, the highest proportion (83%) of genetic variation occurred within individuals while variance between and within fruits were similar (≈ 8.5%). Analysis of the cryptic genetic structure based on SSR using different approaches, namely discriminant analysis of principal components (DAPC) and sparse non-negative matrix factorization (SNMF), yielded results consistent with the occurrence of two clusters with virtually no admixture. Average kinship between individuals which had emerged from the same fruit (0.07) was lower than that expected for full-sib families. Univariate and multivariate analyses of phenotypic data showed 54-66% of variance among individuals within fruits and 34-46% among fruits. The comparison between phenotypic (PST) and molecular (FST) differentiation identified wing width and length as possible target of positive selection. The average kinship and high genetic variation within fruits, together with the highly significant genetic differentiation among fruits, supports the hypothesis that each fruit was colonised by about three ovipositing females. The results also indicate that females were able to disperse widely from the emergence site before mating and starting oviposition activity.
A total of 345 faecal samples were collected from domestic, captive and wild birds in rural areas, urban areas and a Zoo in Algeria. Samples were screened for the presence of parasites belonging to the genus Cryptosporidium Tyzzer, 1910 by microscopy and PCR analysis of the small-subunit rRNA (SSU), actin and 60-kDa glycoprotein (gp60) genes. Cryptosporidium spp. were detected in 31 samples. Sequence analysis of SSU and actin genes revealed the presence of C. baileyi Current, Upton et Haynes, 1986 in domestic chicken broilers (n = 12), captive ostriches (n = 4) and a wild mallard (n = 1), and C. meleagridis Slavin, 1955 in a graylag goose (n = 1), chickens (n = 11) and turkeys (n = 2). Twenty-three chicken and two turkey broilers from five farms were positive for cryptosporidia, with an overall prevalence of 2% and 6%, respectively. Both C. meleagridis and C. baileyi were detected in farmed chicken broilers, with a prevalence ranging from 9% to 69%. Farmed turkeys broilers were positive only for C. meleagridis, with a 13% prevalence at the animal level. Subtyping of C. meleagridis isolates at the gp60 locus showed the presence of subtype IIIgA22G3R1 in graylag goose and chicken broilers and IIIgA23G2R1 in chicken and turkey broilers. Infection with cryptosporidia was not associated with any clinical diseases. a1_Understanding of the diversity of species of Cryptosporidium Tyzzer, 1910 in tortoises remains incomplete due to the limited number of studies on these hosts. The aim of the present study was to characterise the genetic diversity and biology of cryptosporidia in tortoises of the family Testudinidae Batsch. Faecal samples were individually collected immediately after defecation and were screened for presence of cryptosporidia by microscopy using aniline-carbol-methyl violet staining, and by PCR amplification and sequence analysis targeting the small subunit rRNA (SSU), Cryptosporidium oocyst wall protein (COWP) and actin genes. Out of 387 faecal samples from 16 tortoise species belonging to 11 genera, 10 and 46 were positive for cryptosporidia by microscopy and PCR, respectively. All samples positive by microscopy were also PCR positive. Sequence analysis of amplified genes revealed the presence of the Cryptosporidium tortoise genotype I (n = 22), C. ducismarci Traversa, 2010 (n = 23) and tortoise genotype III (n = 1). Phylogenetic analyses of SSU, COWP and actin gene sequences revealed that Cryptosporidium tortoise genotype I and C. ducismarci are genetically distinct from previously described species of Cryptosporidium. Oocysts of Cryptosporidium tortoise genotype I, measuring 5.8-6.9 µm × 5.3-6.5 µm, are morphologically distinguishable from C. ducismarci, measuring 4.4-5.4 µm × 4.3-5.3 µm. Oocysts of Cryptosporidium tortoise genotype I and C. ducismarci obtained from naturally infected Russian tortoises (Testudo horsfieldii Gray) were infectious for the same tortoise but not for Reeve's turtles (Mauremys reevesii [Gray]), common garter snake (Thamnophis sirtalis [Linnaeus]), zebra finches (Taeniopygia guttata [Vieillot]) and SCID mice (Mus musculus Linnaeus)., a2_The prepatent period was 11 and 6 days post infection (DPI) for Cryptosporidium tortoise genotype I and C. ducismarci, respectively; the patent period was longer than 200 days for both cryptosporidia. Sarcopenia is an independent risk factor for morbidity and mortality in patients suffering from small cell lung cancer (SCLC), however, a universal indicator of sarcopenia usable in clinical practice is still missing. A novel indicator for describing the severity of cancer could be helpful in tailoring the anti-tumor therapy. The aim of this study was to evaluate the computed tomography (CT) scans of total muscle area and radiation attenuation in patients suffering from small cell lung cancer. We used staging CT scans performed at the time of diagnosis to measure total muscle area (TMA) and average psoas density (PD) at level of the 3rd lumbar vertebra. TMA and PD were statistically evaluated in association with overall survival and disease staging. We used Mann-Whitney test and Spearman´s correlation coefficient for statistical testing and p-value under 0.05 was considered statistically significant. Retrospectively we examined 47 patients suffering from SCLC (mean age 65.05±7.3 years, BMI 23.97±4.4 kg/m2 , BSA 1.77±0.2 m2 , 30-day mortality was 4.3 % with 10 months median survival). As sarcopenia was pointed TMA under 55 and 39 cm2 /m2 for men and women respectively. The sarcopenic patients had significantly shorter median survival (7 vs. 11 months, p=0.05). We observed a significant relationship between survival and performance status (Spearman´s correlation, R=-0.39, p=0.05). The patients were divided into two groups according to the extensive (ED, n=34) or limited (LD, n=13) form of the disease. We observed significant difference in PD (42.49±6.1 vs. 47.67±4.5 HU, p=0.006) between ED vs. LD groups.
Bolbelasmus (Bolbelasmus) unicornis (Schrank, 1789) (Coleoptera: Geotrupidae) is a rare and threatened beetle distributed mostly in Central, Eastern and Southeast Europe. As a species of special conservation significance it is included in Annexes II and IV of the Habitat Directive of the European Union. Several new records of this species documented using light traps and soil sampling were recently reported in Serbia (the central part of the Balkan Peninsula). In this paper we present and discuss the current distribution of this species in the region studied based on GIS occurrence data. The distribution is mapped and values of environmental variables within this beetle's range are quantified using GIS technology. In addition, we predict its potential range in Serbia using a model based on the current distribution of this beetle, environmental variables and distribution of its food source.
Mitochondrial disorders manifest enormous genetic and clinical heterogeneity - they can appear at any age, present with various phenotypes affecting any organ, and display any mode of inheritance. What mitochondrial diseases do have in common, is impairment of respiratory chain activity, which is responsible for more than 90 % of energy production within cells. While diagnostics of mitochondrial disorders has been accelerated by introducing Next-Generation Sequencing techniques in recent years, the treatment options are still very limited. For many patients only a supportive or symptomatic therapy is available at the moment. However, decades of basic and preclinical research have uncovered potential target points and numerous compounds or interventions are now subjects of clinical trials. In this review, we focus on current and emerging therapeutic approaches towards the treatment of mitochondrial disorders. We focus on small compounds, metabolic interference, such as endurance training or ketogenic diet and also on genomic approaches.
The mite family Ascouracaridae Gaud et Atyeo, 1976 contains large-sized mites (mostly > 1 mm) which live inside the quills of birds of several orders. To date, no representative of this family has been found associated with the order Strigiformes (owls). In this paper, a new species of this family, Cystoidosoma hermaphroditus sp. n., is described from the tropical screech owl, Megascops choliba (Vieillot) (Aves: Strigiformes) from Brazil. This species is unique in having an external spermaduct, a primary duct and a rudimentary bursa copulatrix present in males. This is the first astigmatan feather mite described from the order Strigiformes in this country. Kawasaki disease (KD) is a medium vessel systemic vasculitis that predominantly occurs in children below five years of age. It is an acute febrile condition in which coronary artery aneurysms and myocarditis are the most common cardiovascular complications. It is most often characterized by hypercytokinemia. The etiopathogenesis of KD is not fully understood. The present review synthesizes the recent advances in the pathophysiology and treatment options of KD. According to different studies, the genetic, infections and autoimmunity factors play a major role in pathogenesis. Several susceptibility genes (e.g. caspase 3) and cytokines (e.g. IL-2, IL-4, IL-6, IL-10, IFN-γ and TNF-α) have been identified in KD. Patients with high cytokine levels are predisposed to KD shock syndrome. The importance of respiratory viruses in the pathogenesis of the disease is unclear. Coronavirus disease 2019 (COVID-19) caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) may induce in children and adults an abnormal systemic inflammatory response. This syndrome shares characteristics with KD. It has been called by many terms like MIS-C (Multisystem Inflammatory Syndrome in Children), PIMS-TS (pediatric inflammatory multisystem syndrome temporally associated with SARS-CoV-2), hyperinflammatory shock syndrome, cytokine storm (cytokine release syndrome) or simply, Kawasaki-like syndrome. The cytokine’s role in the development of KD or Kawasaki-like syndrome being triggered by COVID-19 is controversial. The presences of the antiendothelial cell autoantibodies (AECAs) together with the newly developed hypothesis of immunothrombosis are considered potential pathogenic mechanisms for KD. In consequence, the diagnosis and treatment of KD and Kawasaki-like syndrome, one of the most common causes of acquired heart disease in developed countries, are challenging without a clearly defined protocol.
Daily rhythmicity in the sexual behaviour of Monema flavescens Walker (Lepidoptera: Limacodidae) was studied under laboratory conditions. There was a distinct diel periodicity in female calling, male responsiveness and mating behaviour of M. flavescens. As females aged there was an advance in the onset and more time spent calling. One day old females started calling 4 h after the onset of the scotophase, and 5 to 6 day old females called during the first hour of the scotophase. About 34.5% of females called on the night they emerged (1 day old) and the peak in calling of 97.8% was recorded for 2 day old females, after which the incidence of calling decreased rapidly with advancing age. Wind tunnel and copulation tests showed that males were sexually mature on the third night and female moths on the second night. The highest value for the percentage mating was recorded for 3 day old virgin females 4 to 6 h after the onset of the scotophase. In field tests, traps baited with 2 day and 3 day old virgin females captured more males than any other trap and most males were captured 4 to 6 h (1 to 2 day old), 3 to 5 h (3 to 4 day old) and 2 to 4 h (5 to 6 day old) after the onset of the scotophase. These results indicate that there is a daily rhythm in the reproductive behaviour of M. flavescens and provides a better understanding of its sexual behaviour.
