Ascorbic acid (Asc) is a major plant antioxidant. L-galactono-1,4-lactone dehydrogenase (GLDH) is an enzyme that catalyzes the last step of Asc biosynthesis in higher plants. Effects of endogenous Asc on resistance to high-temperature stress were studied by using GLDH-overexpressed (GO-2) and GLDH-suppressed transgenic rice (GI-2) as experimental materials. After high-temperature treatment, the maximal quantum yield of PSII was significantly lower in GI-2, and higher in GO-2 compared to wild type rice. The content of reactive oxygen species (ROS) was the highest in GI-2. The higher Asc content resulted in lower lipid peroxidation in GO-2. The contents of chlorophyll, soluble proteins, and Rubisco large and small subunit were positively correlated to the Asc content. These results show that the higher Asc content reduced the accumulation of ROS and maintained the function of rice leaves. Periodic flooding of trees in tropical floodplains and reservoirs where water levels fluctuate is a common phenomenon. The effects of flooding and subsequent recovery on gas exchange, chlorophyll fluorescence and growth responses of Melaleuca alternifolia seedlings, a tall shrub species used in floodplain and reservoir forest restoration in southern China, were studied during a grow season (from March to December in 2007). M. alternifolia seedlings were flooded for 180 days, drained and left to recover for another 60 days. Survival rates of the seedlings were 100% during the 180-day flooding period. Chlorophyll (Chl) content, net photosynthetic rate
(PN), stomatal conductance (gs), and transpiration rate (E) of the flooded seedlings were all significantly lower than those of the control. Significant reductions of photochemical quenching coefficient (qp) and increases of nonphotochemical quenching (NPQ) in the flooded seedlings were observed. However, there were no significant differences in the maximal quantum yield of PSII photochemistry (Fv/Fm) between treatments. All seedlings survived during the two-month recovery period after the flooded treatment was drained, and the biomass and height of the recovered seedlings approached those of the control at the end of the experiment. During the first-month recovery period, Chl content, PN, gs and E in the recovered seedlings were all obviously low, then increased gradually and rose to the levels similar to the control by the end of the experiment. Quenching analysis revealed significant reductions of qp and increments of NPQ in the recovered seedlings at the beginning of draining, and a nearly complete recovery for both parameters by the end of the experiment. However, Fv/Fm of the recovered seedlings did not differ significantly from the control during the recovery period. Our study demonstrated that M. alternifolia seedlings can survive and grow through 180 days of flooding with a subsequent 60-day recovery period in drained conditions, indicating that seedlings of this species would be suitable for afforestation in areas exposed to intermittent flooding. and Y. Calmodulin (CaM) is a highly conserved calcium sensor protein associated with chilling tolerance in living organisms. It has four EF-hand domains for binding of four Ca2+, two of them located in the N-terminus, and the other two in the C-terminus. A notothenioid CaM gene fragment (CaMm), which only codes for N-terminus of CaM (with two EF-hand domains), was introduced into Nicotiana benthamiana. Effects of its overexpression on chilling tolerance in plants were explored. During 4◦C or 0◦C chilling treatment, both CaMm and CaM transgenic plants showed higher PSII maximum quantum yield, actual quantum yield, and soluble protein content, lower electrolyte leakage and malondialdehyde content than that of the control. The changes in these physiological indices were comparable between the CaMm and CaM transgenic plants during the treatments. Low temperature (LT) is one of the major factors that limit crop production and reduce yield. To better understand the cold-tolerance mechanism in the plantains, a sensitive cultivar Williams (Musa acuminata AAA cv. Williams) and a tolerant cultivar Cachaco (Musa paradisiaca ABB cv. Dajiao) were used. LT resulted in increased malondialdehyde (MDA) content, elevated contents of hydrogen peroxide (H2O2) and superoxide radical (O2.-), and decreased photochemical efficiency (Fv/Fm) and net photosynthetic rate (PN), but cv. Cachaco showed better LT tolerance than cv. Williams. After LT treatment for 120 h, total scavenging capability (DPPH. scavenging capability) in Williams showed a significant decrease but no significant alternations was found in Cachaco. Ascorbate peroxidase (APX) and peroxidase (POD) displayed a significant increase but superoxide dismutase (SOD) showed no significant alternations and catalase (CAT) showed a significant decrease in Cachaco after 120 h of LT treatment. All the four antioxidant enzymes above showed a significant decrease in Williams after 120 h of LT treatment. Our results suggest that higher activities of APX, POD, SOD, and DPPH. scavenging capability to a certain extent can be used to explain the higher cold tolerance in the plantain, which would provide a theoretical guidance for bananas production and screening cold-resistant variety. and Q.
