Over-expression of chloroplastic glycerol-3-phosphate acyltransferase gene (LeGPAT) increased unsaturated fatty acid contents in phosphatidylglycerol (PG) of thylakoid membrane in tomato. The effect of this increase on the xanthophyll cycle and chloroplast antioxidant enzymes was examined by comparing wild type (WT) tomato with the transgenic (TG) lines at chilling temperature (4 °C) under low irradiance (100 µmol m-2 s-1). Net photosynthetic rate and the maximal photochemical efficiency of photosystem (PS) 2 (Fv/Fm) in TG plants decreased more slowly during chilling stress and Fv/Fm recovered faster than that in WT plants under optimal conditions. The oxidizable P700 in both WT and TG plants decreased during chilling stress under low irradiance, but recovered faster in TG plants than in the WT ones. During chilling stress, non-photochemical quenching (NPQ) and the de-epoxidized ratio of xanthophyll cycle in WT plants were lower than those of TG tomatoes. The higher activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in TG plants resulted in the reduction of O2-. and H2O2 contents during chilling stress. Hence the increase in content of unsaturated fatty acids in PG by the over-expression of LeGPAT could alleviate photoinhibition of PS2 and PS1 by improving the de-epoxidized ratio of xanthophyll cycle and activities of SOD and APX in chloroplast. and N. The tested tree species included pioneer species Acacia mangium, early succession stage species Schima superba, mesophyte intermediate-succession species Machilus chinensis, and shade-tolerant plant or late-succession species Cryptocarya concinna which occur in the lower subtropical forest community. A comparison with the current ambient level of UV-B radiation (UV-B) showed the leaf net photosynthetic rate (PN), transpiration rate (E), and stomatal conductance (g s) of the four species ranged from significantly decreased to no significant change. Additionally, the thickness of palisade and mesophyll in leaves of four tree species were decreased sharply by enhanced UV-B. The thickness of spongy parenchyma in leaves was also decreased except for M. chinensis. UV-B increased the leaf width of A. mangium but its leaf length, leaf thickness, and dry mass per unit area were not affected. Significantly increased stomata width was observed in A. mangium leaf epidermis in response to UV-B. Significantly decreased stomata width and significantly increased stomata density of leaf abaxial epidermis in M. chinensis were also observed. The stomata density of abaxial epidermis of C. concinna was remarkably increased by enhanced UV-B. The height and branch biomass of A. mangium and the height of S. superba were reduced visibly by enhanced UV-B. The four plant species could be classified into three groups of UV-B sensitiveness by hierarchical cluster analysis. The effect of high irradiance (HI) during desiccation and subsequent rehydration of the homoiochlorophyllous desiccation-tolerant shade plant Haberlea rhodopensis was investigated. Plants were irradiated with a high quantum fluence rate (HI; 350 µmol m-2 s-1 compared to ca. 30 µmol m-2 s-1 at the natural rock habitat below trees) and subjected either to fast desiccation (tufts dehydrated with naturally occurring thin soil layers) or slow desiccation (tufts planted in pots in peat-soil dehydrated by withholding irrigation). Leaf water content was 5 % of the control after 4 d of fast and 19 d of slow desiccation. Haberlea was very sensitive to HI under all conditions. After 19 d at HI, even in well-watered plants there was a strong reduction of rates of net photosynthesis and transpiration, contents of chlorophyll (Chl) and carotenoids, as well as photosystem 2 activity (detected by the Chl fluorescence ratio RFd). Simultaneously, the blue/red and green/red fluorescence ratios increased considerably suggesting increased synthesis of polyphenolic compounds. Desiccation of plants in HI induced irreversible changes in the photosynthetic apparatus and leaves did not recover after rehydration regardless of fast or slow desiccation. Chlorophyll a fluorescence kinetics, net photosynthetic rate (PN), water relations, and photosynthetic pigment contents were studied during acclimation of in vitro grown tobacco to higher irradiance (HL; 700 μmol m-2 s-1). Plantlets were grown on medium containing sucrose in glass vessels (G-plants) or in Magenta boxes (M-plants) with better CO2 supply in the latter ones. The effect of HL was studied either (1) in plantlets grown under original in vitro conditions (closed vessels), (2) in in vitro plantlets exposed to ambient CO2 concentration (covers removed), or (3) in plantlets transplanted to ex vitro into pots with sand and nutrient solution. Higher PN, and fraction of closed photosystem 2 (PS2) centres (1 - qP), and lower content of xanthophyll cycle pigments were found in M-plants compared to G-plants. HL treatment caused photoinhibition particularly in plants kept in closed vessels. This was indicated by the decrease in the ratio of Fv/Fm and by the increase in non-photochemical quenching, 1 - qp, and content of xanthophyll cycle pigments. Better CO2 supply ensured by the removal of closure lead to the moderate reduction of symptoms of photoinhibition, although stomatal conductance (gs), transpiration rate (E), and PN were negatively affected. The main reason was the decrease in relative air humidity, which caused similar reduction of PN, E, and gs after the transfer of plantlets to ex vitro. Nevertheless, plant response to HL seemed not to be affected by any possible root injury caused by transfer to ex vitro. Five-year-old plants of two olive cultivars (Frantoio and Moraiolo) grown in large pots were exposed for 7 to 8 months to ambient (AC) or elevated (EC) CO2 concentration in a free-air CO2 enrichment (FACE) facility. Exposure to EC enhanced net photosynthetic rate (PN) and decreased stomatal conductance, leading to greater instantaneous transpiration efficiency. Stomata density also decreased under EC, while the ratio of intercellular (Ci) to atmospheric CO2 concentration and chlorophyll content did not differ, except for the cv. Moraiolo after seven months of exposure to EC. Analysis of the relationship between photosynthesis and Ci indicated no significant change in carboxylation efficiency of ribulose-1,5-bisphosphate carboxylase/oxygenase after five months of exposure to EC. Based on estimates derived from the PN-Ci relationship, there were no apparent treatment differences in daytime respiration, CO2 compensation concentration, CO2-saturated photosynthetic rate, or photosynthetic rate at the mean Ci, but there was a reduction in stomata limitation to PN at EC. Wheat provides a unique genetic system in which variable sink size is available across the ploidies. We characterized monocarpic senescence in diploid, tetraploid, and hexaploid wheat species in flag leaf from anthesis up to full grain maturity at regular intervals. Triticum tauschii Acc. cv. EC-331751 showed the fastest rate of senescence among the species studied and the rate of loss per day was highest in terms of photosynthesis rate, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) content, and flag leaf N content coupled with a higher rate of gain in grain N content. Cultivars Kundan and HD 4530 maintained high flag leaf N content throughout grain filling as compared to the diploids and showed a slower rate of senescence. RuBPCO content was higher in the diploids as compared to Kundan and HD 4530 at anthesis. However, the rate of decline in RuBPCO content per day was also higher in the diploids. This degradation in RuBPCO was mediated by high endoproteolytic activities in the diploids which in turn supported its higher rate of N mobilization as compared to the tetraploid and hexaploid wheat. In the frame of the foreseen climate global changes we analysed the physiological responses of Arbutus unedo L. to the variations of carbon dioxide concentration, leaf temperature, and irradiance by measurements of leaf gas exchange and leaf water potential performed both in field and in the laboratory. Stomatal conductance was not affected by increase of leaf temperature. The growth conditions of potted plants likely made stomata more sensitive to the variation of external parameters than naturally growing plants. The interaction between high CO2 concentration and temperature involved important down-regulation mechanisms in the metabolic pathway of the carbon fixation. From an ecological point of view, the ability of A. unedo to adapt to the field stress makes it highly competitive in the Mediterranean plant community. and M. Vitale, F. Manes.
We analyzed the response of potted strawberry tree (Arbutus unedo L.) seedlings exposed to water stress by withholding water for 10 d (WS). Leaf water potential, net CO2 assimilation, and stomatal conductance decreased with increasing water deficit. A 30 % reduction of chlorophyll (Chl) content in the antenna complexes was observed in WS-plants. Simultaneously, a decline of photochemical efficiency (Fv/Fm) occurred as a result of an excess of solar radiation energy when carbon assimilation was limited by stomata closure due to soil water deficit. The non-photochemical quenching of Chl fluorescence (ΦNPQ) significantly increased, as well as the leaf contents of zeaxanthin (Z) and antheraxanthin (A) at the expense of violaxanthin during the WS-period. Elevated predawn contents of de-epoxidized xanthophyll cycle components were associated with a sustained lowering of predawn photosystem 2 efficiency; this suggested an engagement of Z+A in a state primed for energy dissipation. Thus, the ability of strawberry trees to maintain the functionality of the xanthophyll cycle during the Mediterranean summer is an efficient mechanism to prevent irreversible damages to the photosynthetic machinery through thermal energy dissipation in the antenna and the reduction in photochemical efficiency. and R. Sixty seven-days-old plants of Ammi majus L. were subjected for 46 d to sand culture at varying concentrations of NaCl, i.e. 0 (control), 40, 80, 120, and 160 mM. Increasing salt concentrations caused a significant reduction in fresh and dry masses of both shoots and roots as well as seed yield. However, the adverse effect of salt was more pronounced on seed yield than biomass production at the vegetative stage. Calculated 50 % reduction in shoot dry mass occurred at 156 mM (ca.15.6 mS cm-1), whereas that in seed yield was at 104 mM (ca.10.4 mS cm-1). As in most glycophytes, Na+ and Cl- in both shoots and roots increased, whereas K+ and Ca2+ decreased consistently with the successive increase in salt level of the growth medium. Plants of A. majusmaintained markedly higher K+/Na+ ratios in the shoots than those in the roots, and the ratio remained more than 1 even at the highest external salt level (160 mM). Net photosynthetic (PN) and transpiration (E) rates remained unaffected at increasing NaCl, and thus these attributes had a negative association with salt tolerance of A. majus. Proline content in the shoots increased markedly at the higher concentrations of salt. Essential oil content in the seed decreased consistently with increase in external salt level. Overall, A. majusis a moderately salt tolerant crop whose response to salinity is associated with maintenance of high shoot K+/Na+ ratio and accumulation of proline in shoots, but PN had a negative association with the salt tolerance of this crop. and M. The mangroves Rhizophora lamarkii, Ceriops roxburghiana, Bruguiera gymnorrhiza, Aegiceras corniculatum, and Lumnitzera racemosa were screened for their carbon metabolic pathways by measuring net photosynthetic rate (PN), 13C discrimination rate, leaf anatomy, titratable acidity, and activities of phosphoenolpyruvate carboxylase, NADH-malate dehydrogenase, alanine aminotransferase, aspartate aminotransferase, and pyruvate phosphate dikinase. The tested mangroves had a well developed succulence, opening of stomata during day time and closure in the night hours, and absence of diurnal fluctuation of organic acids in their leaves which excludes the possibility of these species being CAM plants. Moreover, the leaf anatomy had not exhibited Kranz syndrome. The high values of discrimination against 13C, low PN, high CO2 compensation concentration, and the activities of aminotransferases in the direction of alanine formation suggest that the species may follow C3 mode of carbon metabolic pathway. and V.
