Etiolated sunflower cotyledons developed in complete darkness and lacking photosystem (PS) 2 were exposed to continuous 200 µmol(photon) m-2 s-1 "white light" for 1, 3, 6, 12, and 18 h prior to evaluations of excitation-energy dissipation using modulated chlorophyll a fluorescence. Photochemical potential of PS2, measured as the dark-adapted quantum efficiency of PS2 (FV(M)/FM), and thermal dissipation from the antenna pigment-protein complex, measured as the Stern-Volmer non-photochemical quenching coefficient (NPQ), increased to 12 h of irradiation. Following 12 h of irradiation, thermal dissipation from the antennae pigment-protein complex decreased while the efficiency of excitation capture by PS2 centers (F'V/F'M) and light-adapted quantum efficiency of PS2 (ΦPS2) continued to increase to 18 h of irradiation. The fraction of the oxidized state of QA, measured by the photochemical quenching coefficient (qP), remained near optimal and was not changed significantly by irradiation time. Hence during the development of maximum photochemical potential of PS2 in sunflower etioplasts, which initially lacked PS2, enhanced thermal dissipation helps limit excitation energy reaching PS2 centers. The partial shading effect on the photosynthetic apparatus of the sunflower (Helianthus annuus L.) was examined by monitoring oxygen evolution, maximum quantum yield of PSII photochemistry in dark-adapted leaves (Fv/Fm), the chlorophyll (Chl) concentrations and the Rubisco contents, and leaf mass per area (LMA) at the leaf level and by determining the concentrations of cytochrome (Cyt) f and the reaction centres of photosystem (PS) I and PSII at the thylakoid level. In this experiment, partial shading was defined as the shading of 2nd leaves with shade cloths, and the whole treatment was defined as the covering of the whole individuals with shade cloths. In the leaf level responses, oxygen evolution, LMA, Chl concentrations and Rubisco contents decreased in all shade treatments administered for six days. Fv/Fm remained constant irrespective of the shade treatments. On the other hand, in the thylakoid-level responses, the concentrations of the thylakoid components per unit Chl and the stoichiometry of the two photosystems showed no statistical difference among the shade treatments. The data obtained from the present study indicate that the partial shading affected the leaf-level responses rather than the thylakoid-level responses. The light received at the lower leaves might serve as a factor in the regulation of the leaf properties of the upper leaves due to the whole plant photosynthesis, while this factor did not have an effect at the thylakoid level., J. Effects of high-temperature stress (HTS) and PEG-induced water stress (WS), applied separately or in combination, on the functional activity and ultrastructure of the photosynthetic apparatus (PSA) of maize (Zea mays L.) and sunflower (Helianthus annuus L.) plants were investigated. In maize plant tissues WS provoked the decrease in RWC by 10.9 %, HTS by 7.0 %, and after simultaneous application of the both treatments the decrease was 32.7 % in comparison with control plants. Similar but more expressed changes were observed in sunflower plants. Sunflower was more sensitive to these stresses. Net photosynthetic rate decreased significantly after all treatments, more in sunflower. In mesophyll chloroplasts after separately applied WS and HTS the number of grana and thylakoids was reduced and electron-transparent spaces appeared. At combined stress (WS+HTS) granal and stromal thylakoids were considerably affected and chloroplast envelope in many of them was partially disrupted. and I. Dekov, T. Tsonev, I. Yordanov.
Environmental factors that induce spatial heterogeneity of stomatal conductance, gs, called stomatal patchiness, also reduce the photochemical capacity of CO2 fixation, yet current methods cannot distinguish between the relative effect of stomatal patchiness and biochemical limitations on photosynthetic capacity. We evaluate effects of stomatal patchiness and the biochemical capacity of CO2 fixation on the sensitivity of net photosynthetic rate (PN) to stomatal conductance (gs), θ (θ = δP N/gs). A qualitative model shows that stomatal patchiness increases the sensitivity θ while reduced biochemical capacity of CO2 fixation lowers θ. We used this feature to distinguish between stomatal patchiness and mesophyll impairments in the photochemistry of CO2 fixation. We compared gas exchange of sunflower (Helianthus annuus L.) plants grown in a growth chamber and fed abscisic acid, ABA (10-5 M), for 10 d with control plants (-ABA). PN and gs oscillated more frequently in ABA-treated than in control plants when the leaves were placed into the leaf chamber and exposed to a dry atmosphere. When compared with the initial CO2 response measured at the beginning of the treatment (day zero), both ABA and control leaves showed reduced PN at particular sub-stomatal CO2 concentration (ci) during the oscillations. A lower reduction of P N at particular g s indicated overestimation of ci due to stomatal patchiness and/or omitted cuticular conductance, gc. The initial period of damp oscillation was characterised by inhibition of chloroplast processes while stomatal patchiness prevailed at the steady state of gas exchange. The sensitivity θ remained at the original pre-treatment values at high gs in both ABA and control plants. At low gs, θ decreased in ABA-treated plants indicating an ABA-induced impairment of chloroplast processes. a1_Different parameters that vary during leaf development may be affected by light intensity. To study the influence of different light intensities on primary leaf senescence, sunflower (Helianthus annuus L.) plants were grown for 50 days under two photon flux density (PFD) conditions, namely high irradiance (HI) at 350 μmol(photon) m-2 s-1 and low irradiance (LI) at 125 μmol(photon) m-2 s-1. Plants grown under HI exhibited greater specific leaf mass referred to dry mass, leaf area and soluble protein at the beginning of the leaf development. This might have resulted from the increased CO2 fixation rate observed in HI plants, during early development of primary leaves. Chlorophyll a and b contents in HI plants were lower than in LI plants in young leaves. By contrast, the carotenoid content was significantly higher in HI plants. Glucose concentration increased with the leaf age in both treatments (HI and LI), while the starch content decreased sharply in HI plants, but only slightly in LI plants. Glucose contents were higher in HI plants than in LI plants; the differences were statistically significant (p<0.05) mainly at the beginning of the leaf senescence. On the other hand, starch contents were higher in HI plants than in LI plants, throughout the whole leaf development period. Nitrate reductase (NR) activity decreased with leaf ageing in both treatments. However, the NR activation state was higher during early leaf development and decreased more markedly in senescent leaves in plants grown under HI. GS activity also decreased during sunflower leaf ageing under both PFD conditions, but HI plants showed higher GS activities than LI plants. Aminating and deaminating activities of glutamate dehydrogenase (GDH) peaked at 50 days (senescent leaves). GDH deaminating activity increased 5-fold during the leaf development in HI plants, but only 2-fold in LI plants., a2_ The plants grown under HI exhibited considerable oxidative stress in vivo during the leaf senescence, as revealed by the substantial H2O2 accumulation and the sharply decrease in the antioxidant enzymes, catalase and ascorbate peroxidase, in comparison with LI plants.
