Growth, net photosynthetic rate (PN), chlorophyll fluorescence induction kinetics, and stromal fructose-1,6-bisphosphatase (sFBPase) in annual legumes native to the Mediterranean region, two clovers (Trifolium subterraneum L. ssp. oxaloides Nyman cv. Clare and T. michelianum Savi cv. Giorgia) and two Medicago species (M. polymorpha L. cv. Anglona and M. truncatula Gaertn. cv. Paraggio), shifted from 20 to 10 °C for 1 d or developed at 10 °C were compared with controls kept at 20 °C. Cold development produced a larger stimulation of growth in the clover cv. Giorgia and the Medicago cv. Paraggio. Transferring plants to low temperatures affected PN relatively less in clovers than in Medicago plants. Development at 10 °C relieved the inhibition of photosynthesis in Giorgia and Paraggio, but not in Clare and Anglona, which correlated with increases in the maximum rate of carboxylation by ribulose-1,5-bisphosphate carboxylase/oxygenase, RuBPCO (Vcmax), and the photon-saturated rate of electron transport (Jmax). In Medicago, transfer from high to low temperature inhibited photosynthesis in a lesser extent in Anglona than in Paraggio, which showed severe limitations at level of Vcmax and Jmax. Development at 10 °C in Paraggio produced an efficient photosynthetic cold acclimation, this being associated with a two-fold increase of quantum yield of photosystem 2 electron transport (ΔF/F'm) and with the activity of sFBPase. By contrast, Anglona showed an irreversible inhibition of PN coupled with the reduction of carbon metabolism by impairment of Calvin cycle enzyme activities such as RuBPCO and sFBPase, resulting in a poor cold acclimation of photosynthesis in this cultivar. and M. The epiphytic fern Platycerium bifurcatum grows in different habitats characterized by drought and high irradiance stress. The plant shows diurnal malate oscillations, indicative for CAM expression only in cover leaves, but not in sporotrophophyll. In P. bifurcatum cover leaves exposed to high irradiance and desiccation, the decrease in both CO2 assimilation (PN) and stomatal conductance (gs) was accompanied with occurrence of diurnal malate oscillations. Exogenously applied abscisic acid (ABA) induced the decrease in PN and gs, but no clear change in malate oscillations. The measurements of the maximum quantum efficiency of photosystem 2 (Fv/Fm) under high irradiance showed distinct photoinhibition, but no clear changes in Fv/Fm due to desiccation and ABA-treatment were found. and G. We investigated the strategies of four co-occurring evergreen woody species Quercus ilex, Quercus coccifera, Pinus halepensis, and Juniperus phoenicea to cope with Mediterranean field conditions. For that purpose, stem water potential, gas exchange, chlorophyll (Chl) fluorescence, and Chl and carotenoid (Car) contents were examined. We recognized two stress periods along the year, winter with low precipitation and low temperatures that led to chronic photoinhibition, and summer, when drought coincided with high radiation, leading to an increase of dynamic photoinhibition and a decrease of pigment content. Summer photoprotection was related to non-photochemical energy dissipation, electron flow to alternative sinks other than photosynthesis, decrease of Chl content, and proportional increase of Car content. Water potential of trees with deep vertical roots (Q. coccifera, Q. ilex, and P. halepensis) mainly depended on precipitation, whereas water potential of trees with shallow roots (J. phoenicea) depended not only on precipitation but also on ambient temperature. and F. Maize (Zea mays) seedlings were exposed for 6 h to strong irradiance (1 000 μmol m-1 s-1 of PPFD) at 5, 12, 17, or 25 °C, followed by an exposure to the darkness for 6 h at 22 °C. Leaf chlorophyll fluorescence, net photosynthetic rate (PN), and the amount of superoxide radicals (O2-⋅) in relation to chilling-induced photoinhibition were investigated. During the photophase, a good correlation (r=-0.879) was observed between ΦPS2 (relative quantum efficiency of PS2 electron transport) and the amount of O2-⋅. Treatment with exogenous O2-⋅ reduced the PN and ΦPS2 as the chilling stress did, that was inhibited by specific scavenger of O2-⋅. Hence chilling-induced photoinhibition might be due to the production of O2-⋅. In contrast, in the dark period, PN and ΦPS2 of the seedlings treated with the exogenous O2-⋅ were enhanced, but they were inhibited by the specific scavenger of O2-⋅, showing the photoprotective role of O2-⋅ in the recovery phase. Furthermore, in terms of the effect of exogenous O2-⋅ on the xanthophyll cycle, the O2-⋅ production suggested a promotion effect for the de-epoxidation of violaxanthin during the photophase, the epoxidation of zeaxanthin at the dark stage, and the increase of the xanthophyll pool both in the photophase and dark phase, resulting in an enhancement of the ability of non-photochemical quenching to avoid or alleviate the damage to photosynthetic apparatus. and D. A greenhouse experiment examined whether clonal integration improves photosynthesis of ramets of alligator weed [Alternanthera philoxeroides (Mart.) Griseb.], a widespread invasive clonal plant in China, in heterogeneous (He) nutrient habitats. The connected pairs of ramets experienced different nutrient levels [high homogeneous (Ho) nutrient, low Ho nutrient, and two He nutrient treatments]. Clonal integration significantly improved the net photosynthetic rate, stomatal conductance, transpiration rate, and minimal and maximal chlorophyll fluorescence of ramets of alligator weed in low nutrient condition. These characteristics may contribute to the success of the ramets of alligator weed in invading contrasting habitats. The effects of simulated acid rain on gas exchange, chlorophyll fluorescence, and anti-oxidative enzyme activity in cucumber seedlings (Cucumis sativus L. cv. Jingchun No. 4) were investigated. Acid rain significantly reduced net photosynthetic rate and mainly non-stomatal factors contributed to the decrease of photosynthesis during the experimental period. The reduced photosynthesis was associated with a decreased maximal photochemical efficiency (Fv/Fm) and the average quantum yield of the photosystem 2 (PS2) reaction centres (ΦPS2). Meanwhile, acid rain significantly increased the activities of guaiacol peroxidase (GPX) and superoxide dismutase (SOD), but decreased the activity of catalase (CAT) together with an increased content of malonyldialdehyde (MDA), Hence the changes in photosynthesis in acid rain treatment might be a secondary effect of acidity damage probably due to lipid peroxidation of lipids and proteins in thylakoid membrane rather than direct effect on PS2 reaction centre. and Jing-Quan Yu, Su-Feng Ye, Li-Feng Huang.
In order to investigate the effect of large isoform of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) activase (RuBPCO-A) on photosynthesis, cDNA of the enzyme (rca) was transferred to rice cultivars (Oryza sativa f. japonica cv. Nipponbare) under the control of RuBPCO small subunit gene promoter (rbcS) via Agrobacterium tumefaciens-mediated transformation. Transgenic rice plants were identified by polymerase chain reaction (PCR) and Southern and Western blot analyses. Net photosynthetic rate (PN ) values of the T1 transgenic lines 34 (T34) and 40 (T40) were 45.26 and 46.32 % higher than that of the control plants, respectively. At the same time, their carboxylation efficiency and RuBPCO initial activity, quantum yield of electron transport in photosystem 2 (ΦPS2), and steady state photochemical fluorescence quenching (qP) increased. In addition, heading time of the transgenic rice was advanced. Thus increasing the amount of large isoform of RuBPCO-A in the transgenic rice might have a stimulatory effect on both photosynthesis and plant growth. and H. R. Wu ... [et al.].
Super-rice hybrids are two-line hybrid rice cultivars with 15 to 20 % higher yields than the raditional three-line hybrid rice cultivars. Response of photosynthetic functions to midday photoinhibition was compared between seedlings of the traditional hybrid rice (Oryza sativa L.) Shanyou63 and two super-rice hybrids, Hua-an3 and Liangyoupeijiu. Under strong midday sunlight, in comparison with Shanyou63, the two super-rice hybrids were less photoinhibited, as indicated by the lower loss of the net photosynthetic rate (PN), the quantum yield of photosystem 2 (ΦPS2), and the maximum and effective quantum yield of PS2 photochemistry (Fv/Fm and Fv'/Fm'). They also had a much higher transpiration rate. Hence the super-rice hybrids could protect themselves against midday photoinhibition at the cost of water. The photoprotective de-epoxidized xanthophyll cycle components, antheraxanthin (A) and zeaxanthin (Z), were accumulated more in Hua-an3 and Liangyoupeijiu than in Shanyou63, but the size of xanthophyll cycle pool of the seedlings was not affected by midday photoinhibition. Compared to Shanyou63, the super-rice hybrids were better photoprotected under natural high irradiance stress and the accumulation of Z and A, not the size of the xanthophyll pool protected the rice hybrids against photoinhibition. and Q. Employing the non-invasive techniques of infra-red gas analysis and pulse amplitude modulated chlorophyll fluorometry, we determined the partitioning of photosynthetic electrons between photosynthetic carbon reduction and other reductive processes resulting in the formation of active oxygen species (AOS) in intact green leaves. This we studied in plant species that are adapted to two different agro-climatic conditions, namely the warm plains (76°36'E, 9°32'N) and the cool mountains (1 600 m a.s.l.) in the south Indian state of Kerala. Ground frost and low temperature were more harmful to those species adapted to the warm plains than the ones adapted to the cool mountains. Exposure to low temperature decreased leaf photosynthetic carbon assimilation rates and quantum yield of photochemical activity in species naturally adapted to the warm plains. High irradiances further aggravated the harmful effects of low temperature stress possibly by overproducing AOS. This resulted in severe peroxidative damage as inferred by the accumulation of malondialdehyde (MDA) in the leaves. and B. Alam, J. Jacob.
The responses to irradiance of photosynthetic CO2 assimilation and photosystem 2 (PS2) electron transport were simultaneously studied by gas exchange and chlorophyll (Chl) fluorescence measurement in two-year-old apple tree leaves (Malus pumila Mill. cv. Tengmu No.1/Malus hupehensis Rehd). Net photosynthetic rate (PN) was saturated at photosynthetic photon flux density (PPFD) 600-1 100 (μmol m-2 s-1, while the PS2 non-cyclic electron transport (P-rate) showed a maximum at PPFD 800 μmol m-2 s-1. With PPFD increasing, either leaf potential photosynthetic CO2 assimilation activity (Fd/Fs) and PS2 maximal photochemical activity (Fv/Fm) decreased or the ratio of the inactive PS2 reaction centres (RC) [(Fi - Fo)/(Fm - Fo)] and the slow relaxing non-photochemical Chl fluorescence quenching (qs) increased from PPFD 1 200 μmol m-2 s-1, but cyclic electron transport around photosystem 1 (RFp), irradiance induced PS2 RC closure [(Fs - Fo')/Fm' - Fo')], and the fast and medium relaxing non-photochemical Chl fluorescence quenching (qf and qm) increased remarkably from PPFD 900 (μmol m-2 s-1. Hence leaf photosynthesis of young apple leaves saturated at PPFD 800 μmol m-2 s-1 and photoinhibition occurred above PPFD 900 μmol m-2 s-1. During the photoinhibition at different irradiances, young apple tree leaves could dissipate excess photons mainly by energy quenching and state transition mechanisms at PPFD 900-1 100 μmol m-2 s-1, but photosynthetic apparatus damage was unavoidable from PPFD 1 200 μmol m-2 s-1. We propose that Chl fluorescence parameter P-rate is superior to the gas exchange parameter PN and the Chl fluorescence parameter Fv/Fm as a definition of saturation irradiance and photoinhibition of plant leaves. and Husen Jia, Dequan Li.
