We investigated the effects of supplementary KNO3 and NaCl on one-year-old, potted Valencia orange (Citrus sinensis) scions grafted on Iranian mandarin Bakraii [Citrus reticulate × Citrus limetta] (Valencia/Bakraii) and Carrizo citrange [C. sinensis × Poncirus trifoliata] (Valencia/Carrizo) rootstocks. After watering plants for 60 days with 50 mM NaCl, the lowest reduction in dry mass, stomatal conductance, and chlorophyll (Chl) content was found in Valencia/Bakraii. Bakraii accumulated more Cl- and Na+ in roots and transferred less to Valencia leaves compared with Carrizo rootstock. Moreover, higher net photosynthetic rate was found in Valencia/Bakraii than those on Carrizo rootstock. NaCl caused a decrease in the maximal efficiency of PSII photochemistry (Fv/Fm) and effective quantum yield (ΦPSII) but elevated coefficient of nonphotochemical quenching. Salinity reduced Ca2+, Mg2+, and total N contents, and increased Na+/K+ ratio in leaves and roots of both grafting combinations. Salinity increased K+ and proline content in leaves and decreased K+ concentrations in roots of both grafting combinations. In salinized plants, nitrate supplementation (10 mM KNO3) reduced leaf abscission, Cl-, Na+, Na+/K+, and Ca2+ concentrations in leaves and roots of both combinations. K+ and N concentrations and proline increased in leaves of the nitrate-supplemented salinized plants. Supplementary nitrate increased leaf number and area, stem elongation, Chl content, Fv/Fm, and ΦPSII and stimulated photosynthetic activity. Low temperature has negative effects on apple photosynthesis by inhibiting the accumulations of photosynthates and nitrogen. The interactive effects of low temperature and nitrogen application on photosynthetic parameters and the absorption and distribution of carbon and nitrogen in different organs were assessed to investigate if nitrogen application can relieve the low-temperature stress on gas exchange and the accumulations of carbon and nitrogen inside the apple plants. No matter under normal or low-temperature conditions, nitrogen application both improved the photosynthetic parameters including net photosynthetic rate, intercellular CO2 concentration, and quantum yield of regulated energy dissipation of PSII as well as the absorption of carbon and nitrogen in roots, stems, and leaves. Thus, we conclude that nitrogen application can relieve the effects of low-temperature stress on photosynthesis and is of benefit for the accumulations of carbon and nitrogen in multiple organs of apple seedlings.
Two methods of induced in vivo chlorophyll (Chl) fluorescence were used to investigate the effects of varying thallus temperature and hydration on the performance of photosynthetic apparatus of a foliar lichen Cetraria islandica: slow Chl fluorescence induction kinetic with the analysis of quenching mechanisms, and rapid irradiance response curves of photosynthesis derived from quantum yield of photochemical reactions of photosystem 2 (Φ2) recorded at increasing irradiances. We compared responses of photosynthetic apparatus in populations of C. islandica growing in lower altitude (LAP: 1 350 m a.s.l.) and in higher altitude (HAP: 2 000 m a.s.l.). At each altitude, the samples were collected both in fully irradiated sites (HI) and in shade (LI). Temperature optimum of photosynthetic processes was the same for LAP and HAP thalli of LI populations (18 °C), while it was significantly lower for HI HAP (14 °C). Gradual dehydration of fully hydrated thalli led to initial increase (up to 20 % of water saturation deficit, WSD) in FV/FM and Φ2, no change at 20-50 % WSD, and a dramatic decrease of the parameters within 50-80 % of WSD. LI HAP of C. islandica was the best adapted population to low temperature having higher rates of photochemical processes of photosynthesis than HI HAP within temperature range of -5 to +5 °C. Two-month-old seedlings of Sophora davidii were subjected to a randomized complete block design with three water (80, 40, and 20 % of water field capacity, i.e. FC80, FC40, and FC20) and three N supply [N0: 0, Nl: 92 and Nh: 184 mg(N) kg-1(soil)] regimes. Water stress produced decreased leaf area (LA) and photosynthetic pigment contents, inhibited photosynthetic efficiency, and induced photodamage in photosystem 2 (PS2), but increased specific leaf area (SLA). The decreased net photosynthetic rate (PN) under medium water stress (FC40) compared to control (FC80) might result from stomatal limitations, but the decreased PN under severe water deficit (FC20) might be attributed to non-stomatal limitations. On the other hand, N supply could improve photosynthetic capacity by increasing LA and photosynthetic pigment contents, and enhancing photosynthetic efficiency under water deficit. Moreover, N supply did a little in alleviating photodamages to PS2 caused by water stress. Hence water stress was the primary limitation in photosynthetic processes of S. davidii seedlings, while the photosynthetic characters of seedlings exhibited positive responses to N supply. In Leymus chinensis, mild water stress (soil moisture 60-65 % of field capacity) had no significant effects on nitrogen metabolism, photosynthesis, and chlorophyll fluorescence. Severe water stress (35-40 %) significantly decreased the activities of nitrate reductase, glutamine synthetase, and glutamate dehydrogenase, net photosynthetic rate, stomatal conductance, transpiration rate, maximal efficiency of photosystem 2 photochemistry (Fv/Fm), actual quantum yield, and photochemical quenching, but increased the endopeptidase activity and malondialdehyde contents. Broomcorn millet (Panicum miliaceum L.) is one of the important C4 crops in the semiarid regions of northern China. It is a close relative of biofuel crop switchgrass. Yet, there is no information on how these crops might respond to a climate change in China. In order to gain insight into such a response, we studied the effect of elevated CO2 concentration (EC) on broomcorn millet. The changes in leaf photosynthesis, chlorophyll fluorescence, morphological parameters, biomass and yield in response to EC [i.e., + 200 µmol(CO2) mol-1] over two years were determined at the open-top chamber (OTC) experimental facility in north China. EC increased net photosynthetic rate, stomatal conductance, intercellular CO2 concentration, transpiration rate, instantaneous transpiration efficiency, effective quantum yield of PSII photochemistry, and photochemical quenching coefficient of fully expanded flag leaves. Maximal quantum yield of PSII photochemistry declined under EC in 2013, but was not affected in 2014. EC significantly decreased intrinsic efficiency of PSII in 2013, but increased in 2014. Leaf nonphotochemical quenching decreased under EC both in 2013 and 2014. EC significantly enhanced the aboveground biomass and yield by average of 31.4 and 25.5% in both years, respectively. The increased yield of broomcorn millet under EC occurred due to the enhanced number of grains per plant. We concluded that photosynthesis of broomcorn millets was improved through increased stomatal conductance in leaves under EC, which led to an increase in height, stem diameter, aboveground biomass, and yield. The effect of drought stress on energy dissipation and antioxidant enzyme system in two sweet sorghum inbred lines (M-81E and Roma) was investigated. Results showed that the germination indicator increased more in M-81E than that in Roma under rehydration. Under drought stress, both the maximal photochemical efficiency of PSII (Fv/Fm) and oxidoreductive activity (ΔI/I0) of Roma decreased more than those in M-81E. Relative to Fv/Fm, the ΔI/I0 decreased markedly, which indicated that PSI was more sensitive to drought stress than PSII. Increases in the reduction state of QA (1-qp), nonphotochemical quenching (NPQ) and minimal fluorescence yield of the dark-adapted state (F0) were greater in Roma than those in M-81E; meanwhile, the H2O2 content was lower in M-81E than that in Roma. Our results suggested that the photoinhibition might be related to the accumulation of reactive oxygen species (ROS). Two rice chlorophyll (Chl) b-less mutants (VG28-1, VG30-5) and the respective wild type (WT) plant (cv. Zhonghua No. 11) were analyzed for the changes in Chl fluorescence parameters, xanthophyll cycle pool, and its de-epoxidation state under exposure to strong irradiance, SI (1 700 µmol m-2 s-1). We also examined alterations in the chloroplast ultrastructure of the mutants induced by methyl viologen (MV) photooxidation. During HI (0-3.5 h), the photoinactivation of photosystem 2 (PS2) appeared earlier and more severely in Chl b-less mutants than in the WT. The decreases in maximal photochemical efficiency of PS2 in the dark (Fv/Fm), quantum efficiency of PS2 electron transport (ΦPS2), photochemical quenching (qP), as well as rate of photochemistry (Prate), and the increases in de-epoxidation state (DES) and rate of thermal dissipation of excitation energy (Drate) were significantly greater in Chl b-mutants compared with the WT plant. A relatively larger xanthophyll pool and 78-83 % conversion of violaxanthin into antheraxanthin and zeaxanthin in the mutants after 3.5 h of HI was accompanied with a high ratio of inactive/total PS2 (0.55-0.73) and high 1-qP (0.57-0.68) which showed that the activities of the xanthophyll cycle were probably insufficient to protect the photosynthetic apparatus against photoinhibition. No apparent difference of chloroplast ultrastructure was found between Chl b-less mutants and WT plants grown under low, LI (180 µmol m-2 s-1) and high, HI (700 µmol m-2 s-1) irradiance. However, swollen chloroplasts and slight dilation of thylakoids occurred in both mutants and the WT grown under LI followed by MV treatment. These typical symptoms of photooxidative damage were aggravated as plants were exposed to HI. Distorted and loose scattered thylakoids were observed in particular in the Chl b-less mutants. A greater extent of photoinhibition and photooxidation in these mutants indicated that the susceptibility to HI and oxidative stresses was enhanced in the photosynthetic apparatus without Chl b most likely as a consequence of a smaller antenna size. and Z.-F. In order to evaluate the photosynthetic activity of a C3 leaf from the electron transport rate (ETR) of photosystem 2 (PS2), a new method was devised and examined using leaves of sweet potato. In this method, both surfaces of a leaf were sealed with transparent films to stop the gas exchange between the leaf and the atmosphere; hence the functions of both photosynthetic assimilation (CO2 uptake) and photorespiration (CO2 release) are restricted to the inside of the leaf. After both functional rates became equally balanced, ETR of the sealed leaf (ETRseal) was determined from the chlorophyll fluorescence. The measurements were conducted at different irradiances and leaf temperatures and by using leaves of different age. Under each measurement condition, ETRseal showed a close positive relationship with the photosynthetic potential, or the gross photosynthetic rate measured in the air of 2 % O2 (PG2%) before sealing. The changes in growth and photosynthetic performance of two wheat (Triticum aestivum L.) cultivars (Bolal-2973 and Atay-85) differing in their sensitivity to boron (B) toxicity were investigated under toxic B conditions. Eight-day old seedlings were exposed to highly toxic B concentrations (5, 7.5, and 10 mM H3BO3) for 5 and 9 days. Fast chlorophyll a fluorescence kinetics was determined and analysed using JIP test. Growth parameters, tissue B contents, and membrane damage were measured at two stress durations. The photochemical performance of PSII was hindered more in the sensitive cultivar (Atay-85) than that of the tolerant one (Bolal-2973) under B toxicity. The increase in the B concentration and stress duration caused membrane leakage in both cultivars. However, higher membrane damage was observed in Atay-85 compared to Bolal-2973. Additionally, significant reduction of growth parameters was observed in both cultivars at toxic B concentrations. The accumulation of B was higher in shoots than in roots of both cultivars. Nevertheless, Atay-85 translocated more B from roots to leaves compared to Bolal-2973. The advantages of certain JIP test parameters were demonstrated for evaluation of PSII activity in plants exposed to B stress. Evaluation of photosynthetic performance by JIP test as well as assessment of growth and tissue B content might be used to determine the effects of B toxicity in wheat.
