Drought stress has multiple effects on the photosynthetic apparatus. Herein, we aimed to study the effect of drought stress on fluorescence characteristics of PSII in leaves of Plectranthus scutellarioides and explore potentially underlying mechanisms. Plants of P. scutellarioides were grown in a greenhouse and subjected to drought (DS, drought-stressed) or daily irrigation (control group). Leaf chlorophyll (Chl) index and induction kinetics curves of Chl a fluorescence and the JIP-test were used to evaluate effects of drought lasting for 20 d. Our results showed that both the leaf and soil relative water content decreased with increasing treatment duration. The leaf Chl index was reduced to half in the DS plants compared with the control group after 20 d. The minimal fluorescence in the DS plants was higher than that in the control plants after 10 d of the treatment. Maximum photochemical efficiency and lateral reactivity decreased with increasing treatment duration in the DS plants. With the continuing treatment, values of absorption flux per reaction center (RC), trapped energy flux per RC, dissipated energy flux per RC, and electron transport flux per RC increased in the earlier stage in the DS plants, while obviously decreased at the later stage of the treatment. Water deficit is one of the major limiting factors in vegetation recovery and restoration in loess, hilly-gully regions of China. The light responses of photosynthesis in leaves of two-year old Prunus sibirica L., Hippophae rhamnoides L., and Pinus tabulaeformis Carr. under various soil water contents were studied using the CIRAS-2 portable photosynthesis system. Light-response curves and photosynthetic parameters were analyzed and fitted using the rectangular hyperbola model, the exponential model, the nonrectangular hyperbola model, and the modified rectangular hyperbola model. Under high light, photosynthetic rate (PN) and stomatal conductance (gs) were steady and photoinhibition was not significant, when the relative soil water content (RWC) varied from 56.3-80.9%, 47.9-82.9%, and 33.4-92.6% for P. sibirica, H. rhamnoides, and P. tabulaeformis, respectively. The light-response curves of PN, the light compensation point (LCP), and the dark respiration rate (RD) were well fitted using the above four models. The nonrectangular hyperbola was the best model in fitting the data; the modified rectangular hyperbola model was the second, and the rectangular hyperbola model was the poorest one. When RWC was higher or lower than the optimal range, the obvious photoinhibition and significant decrease in PN with increasing photosynthetic photon flux density (PPFD) were observed in all three species under high light. The light saturation point (LSP) and apparent quantum yield also decreased significantly, when the upper limit of PPFD was 200 μmol m-2 s-1. In this work, photosystem II (PSII) photochemistry, leaf water potential, and pigment contents of male and female Pistacia lentiscus L. were investigated during a seasonal cycle at three different, arid locations: superior semiarid, inferior semiarid, and arid. The results showed that the gender, season, and the site conditions interacted to influence the quantum yield and pigment contents in P. lentiscus. Predawn leaf water status was determined only by the site and season. The annual patterns of PSII maximum quantum efficiency (Fv/Fm) were characterized by a suboptimal activity during the winter, especially, populations with the more negative water potential exhibited a lower chlorophyll (Chl) a content and chronic photoinhibition irrespective of a gender. We also demonstrated that both photochemical or nonphotochemical mechanisms were involved to avoid the photoinhibition and both of them depended on the season. This plasticity of photosynthetic machinery was accompanied by changes in carotenoids and Chl balance. In the spring, the female Fv/Fm ratio was significantly higher than in male individuals, when the sexual dimorphism occurred during the fruiting stage, regardless of site conditions. P. lentiscus sex-ratio in Mediterranean areas, where precipitations exceeded 500 mm, was potentially female-biased. Among the fluorescence parameters investigated, nonphotochemical quenching coefficient appeared as the most useful one and a correlation was found between Chl a content and Fv/Fm. We aimed to find out relations among nonphotochemical quenching (NPQ), gross photosynthetic rate (PG), and photoinhibition during photosynthetic light induction in three woody species (one pioneer tree and two understory shrubs) and four ferns adapted to different light regimes. Pot-grown plants received 100% and/or 10% sunlight according to their light-adaptation capabilities. After at least four months of light acclimation, CO2 exchange and chlorophyll fluorescence were measured simultaneously in the laboratory. We found that during light induction the formation and relaxation of the transient NPQ was closely related to light intensity, light-adaption capability of species, and PG. NPQ with all treatments increased rapidly within the first 1-2 min of the light induction. Thereafter, only species with high PG and electron transport rate (ETR), i.e., one pioneer tree and one mild shade-adapted fern, showed NPQ relaxing rapidly to a low steady-state level within 6-8 min under PPFD of 100 μmol(photon) m-2 s-1 and ambient CO2 concentration. Leaves with low PG and ETR, regardless of species characteristics or inhibition by low CO2 concentration, showed slow or none NPQ relaxation up to 20 min after the start of low light induction. In contrast, NPQ increased slowly to a steady state (one pioneer tree) or it did not reach the steady state (the others) from 2 to 30 min under PPFD of 2,000 μmol m-2 s-1. Under high excess of light energy, species adapted to or plants acclimated to high light exhibited high NPQ at the initial 1 or 2 min, and showed low photoinhibition after 30 min of light induction. The aim of this study was to assess the impact of the mitochondrial alternative oxidase (AOX) pathway on energy metabolism in chloroplasts, and evaluate the importance of the AOX in alleviating drought-induced photoinhibition in pepper (Capsicum annuum L.). Inhibition of AOX pathway decreased photosynthesis and increased thermal energy dissipation in plants under normal conditions. It indicated that AOX pathway could influence chloroplast energy metabolism. Drought reduced carbon assimilation. Photoinhibition was caused by excess of absorbed light energy in spite of the increase of thermal energy dissipation and cyclic electron flow around PSI (CEF-PSI). Upregulation of AOX pathway in leaves experiencing drought would play a critical role in protection against photoinhibition by optimization of carbon assimilation and PSII function, which would avoid over-reduction of photosynthetic electron transport chain. However, inhibition of AOX pathway could be compensated by increasing the thermal energy dissipation and CEF-PSI under drought stress, and the compensation of CEF-PSI was especially significant., W. The specific features of the structural and functional organisation of the photosynthetic apparatus (PSA) were studied in wild halophytes representing three strategies of salt tolerance: euhalophyte Salicornia perennans, crynohalophyte Limonium gmelinii, and glycohalophyte Artemisia santonica. The sodium content in aboveground parts of the plants corresponded to the strategy of salt tolerance. The photosynthetic cells of the euhalophyte were large and contained a higher number of chloroplasts than those in other species. In contrast, the number of cells per a leaf area unit was lower in S. perennans as compared to cryno- and glycohalophytes. Thereupon, the cell and chloroplast surface area per leaf area unit declined in the following sequence: A. santonica > L. gmelinii > S. perennans. However, the large cells of euhalophyte contained chloroplasts of larger sizes with 4- to 5-fold higher chlorophyll (Chl) content per chloroplast and Chl concentration in chloroplast volume unit. Also, chloroplasts of S. perennans were characterised by the higher content of glyco- and phospholipids. Qualitative composition of fatty acids (FA) in lipids isolated from the chloroplast-enriched fraction was similar in all three species; however, the index of unsaturation of FA was higher in glycohalophyte A. santonica than those in two other species. Under natural condition, PSA of all three halophytes showed high resistance to soil salinity. The results indicated tolerance of PSII to the photodamage in halophytes. The high rate of electron transport through PSII can be important to prevent oxidative damage of PSA in halophytes under strong light and hight temperature in vivo. Thus, the strategy of salt tolerance is provided by both the leaf anatomical structure and the ultrastructure of photosynthetic membranes, which is determined in particular by the specific composition of lipids., O. High irradiance promotes decreases in the quantum yield in plants, which reduce the photosynthetic rate. The excess of light in combination with water deficit can intensify the response of plants to stress, especially in species susceptible to those factors. The aim of the present study was to characterize the photosynthetic activity of young jatobá-do-cerrado (Hymenaea stigonocarpa Mart. ex Hayne) trees under different irradiance conditions, both alone and/or in combination with water deficit. Four irradiances [45, 230, 510, and 1,700 μmol(photon) m-2 s-1] and two levels of water in soil (90% and 50% of field capacity) were used. Gas exchange, water potential, and chlorophyll a fluorescence were measured. The highest rates of photosynthesis were observed under irradiances of 230 and 510 μmol(photon) m-2 s-1. Irradiance of 1,700 μmol(photon) m-2 s-1 led to the photoinhibition of photosynthesis, as indicated by a reduced maximum quantum yield of PSII, effective quantum yield ratio, and electron transport rate, as well as higher nonphotochemical quenching. Red alga contains four extrinsic proteins in photosystem II (PSII), which are PsbO, PsbV, PsbU, and PsbQ′. Except for the PsbQ′, the composition is the same in cyanobacterial PSII. Reconstitution analysis of cyanobacterial PSII has shown that oxygen-evolving activity does not depend on the presence of PsbQ′. Recently, the structure of red algal PSII was elucidated. However, the role of PsbQ′ remains unknown. In this study, the function of the acceptor side of PSII was analyzed in PsbQ′-reconstituted PSII by redox titration of QA and thermoluminescence. The redox potential of QA was positively shifted when PsbQ′ was attached to the PSII. The positive shift of QA is thought to cause a decrease in the amount of triplet chlorophyll in PSII. Environmental pollution by antibiotics poses a potential ecological risk to aquatic photosynthetic organisms. In the present study, toxic effects of erythromycin on PSI and PSII were investigated in cyanobacteria culture medium of Microcystis aeruginosa. The activity and electron transport of both photosystems were affected by erythromycin in a concentrationdependent manner. The quantum yield of PSII (YII) was reduced at 0.1 mg L-1 of erythromycin, while the quantum yield of PSI (YI) significantly decreased at concentration of 5-25 mg L-1. The decline of YII was accompanied by an increase of nonregulated energy dissipation (YNO). At 10 mg L-1 of erythromycin, YII decreased by 55%, while YNO increased by 18%. The decrease of YI induced by erythromycin was caused by donor-side limitation of PSI (YND). YND was markedly enhanced with elevated erythromycin concentration. At 10 mg L-1 of erythromycin, YI and YNA (PSI acceptor-side limitation) decreased by 8 and 82%, respectively, while YND rose by 314%. The quantum yield of cyclic electron flow increased significantly at 0.1-1 mg L-1 of erythromycin; it decreased but remained higher than that of the control at 5-25 mg L-1 of erythromycin. The contribution of cyclic electron flow to YI, and to linear electron flow rose significantly with the increasing erythromycin concentration. The maximum values of electron transport rates in PSII and PSI decreased by 71 and 24.3%, respectively, at 25 mg L-1 of erythromycin. Compared with the untreated control, the light saturation of PSII and PSI decreased significantly with increasing erythromycin concentration. We showed that concentrations of erythromycin >- 5 mg L-1 could exert acute toxicity to cyanobacteria, whereas the chronic toxicity caused by concentrations of ng or μg L-1 needs further research., C.-N.
