With japonica rice 98-08, indica hybrids Shanyou 63, Gangyou 881, and X07S/Zihui 100, and sub-species hybrid Peiai 64S/9311 as materials, chlorophyll (Chl) content, Chl a fluorescence parameters, and membrane lipid peroxidation in flag leaf were measured at late developmental stages under natural conditions. Fv/Fm, qP, ΦPS2, and electron transport rate gradually decreased while qN increased conversely. Excessive photon energy led to the accumulation of active oxygen (O2-), H2O, malonyldialdehyde, and products of membrane lipid peroxidation, and resulted in reduced Chl content and early ageing subsequent to the photooxidation during flag leaf senescence. There was obvious diversification of these parameters among rice cultivars. In comparison with japonica cv. 98-08 (tolerant to photooxidation), Fv/Fm decreased in indica cv. Shanyou 63 (susceptible to photooxidation) with greater accumulation of active oxygen and a sharp drop in Chl content, which resulted in "yellowish" early ageing, and affected the filling and setting of rice grains. The mechanism for premature ageing in indica rice was related to irradiance and temperature at filling stages. On a sunny day at above 25 °C, the reaction centre of photosystem 2 (PS2) exhibited a dynamic change on reversible inactivation. Under the intense irradiance at noon, PS2 function in indica rice exhibited obvious down-regulation and photoinhibition. Under intense irradiance with lowered temperatures, PS2 resulted in photo-damage and early ageing, related to the degradation of PS2-D1 protein and the inhibition of endogenous protection systems such as the xanthophyll cycle and enzymes scavenging active oxygen. Hence for high-yield breeding, based on a good plant-type and utilising heterosis and tolerance of photooxidation, the selection of japonica rice or a sterile line with the japonica genotype as female is a strategy worthy of consideration. and Demao Jiao, Benhua Ji, Xia Li.
Seasonal and daily variations in chlorophyll (Chl) fluorescence were studied in two representative species of Mediterranean grasslands, Tuberaria guttata (an annual) and Chamaemelum nobile (a perennial), in order to assess physiological responses to climatically induced stresses during the growing season. The photochemical efficiency of photosystem (PS) 2 in dark-adapted leaves was measured by the Chl fluorescence ratio Fv/Fp. This ratio decreased progressively from December to July, as the effects of increasing solar radiation and summer drought became more severe. The seasonal decline was observed particularly as a depression of morning and midday values, when photoinhibition was more evident. In both species, the extent of this diurnal depression increased with midday irradiance throughout winter and spring. After sunset, there was complete recovery to optimum values. Towards the end of the life cycle, increased irradiance did not affect the midday decline further but Fv/Fp measurements in the morning and evening never regained their optimum values, indicating the accumulation of photodamage in the reaction centres of PS2. The half-rise time of Fp (T1/2), used to estimate the size of the plastoquinone pool, showed little daily variation in C. nobile throughout the most important part of its seasonal cycle. However, towards the end of its life cycle (June and July) T1/2 values ranged from ca. 200 ms before sunrise to near zero at midday on the same day. The annual species, T. guttata, showed similar disregulation in energy transmission rate both at the seedling stage and at end of its life cycle. Thus seedlings and reproductive plants in particular are sensitive to environmental conditions (extremes of temperature and drought) and cannot maintain consistent electron flow throughout the day. and L. Six months old in vitro-grown Anoectochilus formosanus plantlets were transferred to ex-vitro acclimation under low irradiance, LI [60 µmol(photon) m-2 s-1], intermediate irradiance, II [180 µmol(photon) m-2 s-1], and high irradiance, HI [300 µmol(photon) m-2 s-1] for 30 d. Imposition of II led to a significant increase of chlorophyll (Chl) b content, rates of net photosynthesis (PN) and transpiration (E), stomatal conductance (gs), electron transfer rate (ETR), quantum yield of electron transport from water through photosystem 2 (ΦPS2), and activity of ribulose-1,5-bisphosphate carboxylase/ oxygenase (RuBPCO, EC 4.1.1.39). This indicates that Anoectochilus was better acclimated at II compared to LI treatment. On the other hand, HI acclimation led to a significant reduction of Chl a and b, PN, E, gs, photochemical quenching, dark-adapted quantum efficiency of open PS2 centres (Fv/Fm), probability of an absorbed photon reaching an open PS2 reaction centre (Fv'/Fm'), ETR, ΦPS2, and energy efficiency of CO2 fixation (ΦCO2/ΦPS2). This indicates that HI treatment considerably exceeded the photo-protective capacity and Anoectochilus suffered HI induced damage to the photosynthetic apparatus. Imposition of HI significantly increased the contents of antheraxanthin and zeaxanthin (ZEA), non-photochemical quenching, and conversion of violaxanthin to ZEA. Photoinhibition of photosynthesis was studied in young and mature detached sun needles of cypress under high irradiance (HI) of about 1 900 μmol m-2 s-1. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. Compared with the mature needles, the young needles, containing about half the amount of Chl a+b per unit area, exhibited a higher proportion of total carotenoids (Car) as xanthophyll cycle pigments and had an increased ratio of Car/Chl a+b. The potential efficiency of photosystem (PS) 2, Fv/Fm, markedly declined in HI-treated young needles without significant increase of F0 level. In contrast, the Fv/Fm ratio declined with significant increase of F0 level in mature needles. In isolated thylakoids, the rate of whole chain and PS2 activity markedly decreased in young HI-needles in comparison with mature needles. A smaller inhibition of PS1 activity was observed in both needles. In the subsequent dark incubation, fast recovery was found in both needle Types that reached maximum PS2 efficiencies similar to those observed in non-photoinhibited needles. The artificial exogenous electron donors DPC, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in mature needles, while DPC and NH2OH significantly restored it in young needles. Hence, HI-inactivation was on the donor side of PS2 in young needles and on the acceptor side of PS2 in mature needles. Quantification of the PS2 reaction centre proteins D1 and 33 kDa protein of water splitting complex following HI-exposure of needles showed pronounced differences between young and mature needles. Watermelon (Citrillus lanatus) plants were grown for two consecutive years in open-top chambers with three different ozone concentrations (O3-free air, O3 ambient, and air with additional O3; CFA, NFA, and NFA+O3) and three nitrogen fertilizer concentrations [0, 14.0, and 29.6 g N per pot; N0, N1, and N2). There was an interaction between ozone and N fertilizer for the major parameters studied. O3 and N2 treatments led to a significant decrease in maximum efficiency of photosystem 2 (PS2) photochemistry (Fv/Fm), and induced a significant decrease in the actual quantum yield of PS2 (ΦPS2), due mainly to the increased closure of PS2 reaction centres (qP) and to an increase in the non-photochemical quenching (NPQ). On the other hand, these plants exhibited an increased susceptibility to photoinhibition, which could be associated with an increased fraction of reduced QA. An increase in lipid peroxidation indicated that damage was occurring at the membrane levels. High N concentration enhanced the detrimental effects of ozone on the fluorescence parameter induction and lipid peroxidation. All these negative alterations led to a decreased yield. and A. The low chlorophyll b mutant of high yield rice had a lower light-harvesting complex 2 content than the wild type. The stability of oxygen evolution side of photosystem 2 was only slightly lower. A lower photon absorption rate and a stronger xanthophyll cycle capacity of this mutant led to a higher endurance to strong irradiance and a lower photoinhibition as compared with the wild type rice. and Xinbin Dai ... The concentrations of photosynthetic pigments decreased in both chilling stressed species but the ratios of chlorophyll (Chl) a/b and total carotenoids (Car)/Chls were depressed only in faba bean. The contents of α+β carotene and lutein+lutein-5,6-epoxide remained unaffected in both species, but the de-epoxidation state involving the components of xanthophyll cycle increased in pea. Under chilling stress the photosynthetic electron transport associated with photosystem 2, PS2 (with and without the water oxidising complex) decreased in both plant species, the inhibition being higher in faba bean. The intrachloroplast quinone pool also decreased in both stressed species, yet an opposite trend was found for cytochrome b559LP. Under stress an increasing peroxidation of thylakoid acyl lipids was detected in pea, but higher protein/Chl ratio was detected in faba bean. The influence of chilling (8 °C, 5 d) at two photon flux densities [PFD, L = 200 and H = 500 μmol(photon) m-2 s-1] on the gas exchange and chlorophyll fluorescence was investigated in chilling-tolerant and chilling-sensitive maize hybrids (Zea mays L., K383×K130, K185×K217) and one cultivar of field bean (Vicia faba L. minor, cv. Nadwiślański). The net photosynthetic rate (PN) for the both studied plant species was inhibited at 8 °C. PN of both maize hybrids additionally decreased during chilling. Changes in the quantum efficiency of PS2 electron transport (ΦPS2) as a response to chilling and PFD were similar to PN. Measurements of ΦPS2/ΦCO2 ratio showed that in field bean seedlings strong alternative photochemical sinks of energy did not appear during chilling. However, the high increment in ΦPS2/ΦCO2 for maize hybrids can indicate reactions associated with chill damage generation. At 8 °C the non-photochemical quenching (NPQ) increased in all plants with chilling duration and PFD. The appearance of protective (qI,p) and damage (qI,d) components of qI and a decrease in qE (energy dependent quenching) took place. NPQ components of field bean and maize hybrids differed from each other. The amount of protective NPQ (qE + qI,p) components as part of total NPQ was higher in field bean than in maize hybrids at both PFD. On 5th day of chilling, the sum of qE and qI,p was 26.7 % of NPQ in tolerant maize hybrids and 17.6 % of NPQ in the sensitive one (averages for both PFD). The increased PFD inhibited the ability of all plants to perform protective dissipation of absorbed energy. The understanding of the genotypic variation of NPQ components in maize may have implications for the future selection of plants with a high chilling tolerance. and J. Kościelniak, J. Citrus volkameriana (L.) plants were grown for 43 d in nutrient solutions containing 0, 2, 14, 98, or 686 µM Mn (Mn0, Mn2, Mn14, Mn98, and Mn686, respectively). To adequately investigate the combined effects of Mn nutrition and irradiance on photosystem 2 (PS2) activity, irradiance response curves for electron transport rate (ETR), nonphotochemical quenching (qN), photochemical quenching (qP), and real photochemical efficiency of PS2 (ΦPS2) were recorded under 10 different irradiances (66, 96, 136, 226, 336, 536, 811, 1 211, 1 911, and 3 111 µmol m-2 s-1, I66 to I3111, respectively) generated with the PAM-2000 fluorometer. Leaf chlorophyll content was significantly lower under Mn excess (Mn686) compared to Mn0; its highest values were recorded in the treatments Mn2-Mn98. However, ETR and ΦPS2 values were significantly lower under Mn0 compared to the other Mn treatments, when plants were exposed to irradiances ≥96 µmol m-2 s-1. Furthermore, Mn0 plants had significantly higher values of qN and lower values of qP at irradiances ≤226 and ≥336 µmol m-2 s-1, respectively, than those grown under Mn2-Mn686. Irrespective of Mn treatment, the values of ΦPS2 and qN decreased, while those of qP increased progressively by increasing irradiance from I136 to I3111. The effects of summer and winter stress on the chlorophyll and carotenoid contents and photosystem 2 efficiency were examined in six Mediterranean scrub species. These six species belong to two different plant functional types: drought semi-deciduous (Halimium halimifolium L., Rosmarinus officinalis L., Erica scoparia L.) and evergreen sclerophylls (Juniperus phoenicea L., Pistacia lentiscus L., Myrtus communis L.). Two sites with different water availability were chosen. In the xerophytic site, despite they belong to two different functional types, R. officinalis and J. phoenicea showed a similar response. These were the most affected species in summer. H. halimifolium showed optimal values of Fv/Fm and non-significant seasonal changes in xanthophyll content. In the mesic site, E. scoparia and M. communis were apparently the most affected species by winter climatic conditions. P. lentiscus presented a pattern similar to H. halimifolium, except for elevated F0 values. In all the studied species, lutein plus zeaxanthin content was negatively correlated with Fv/Fm in summer and with leaf water potential, thus indicating that the thermal dissipation of energy was a general pattern for all species.
