Arthrospira maxima is unique among cyanobacteria, growing at alkaline pH (<11) in concentrated (bi)carbonate (1.2 M saturated) and lacking carbonic anhydrases. We investigated dissolved inorganic carbon (DIC) roles within PSII of A. maxima cells oximetrically and fluorometrically, monitoring the light reactions on the donor and acceptor sides of PSII. We developed new methods for removing DIC based on a (bi)carbonate chelator and magnesium for (bi)carbonate ionpairing. We established relative affinities of three sites: the water-oxidizing complex (WOC), non-heme iron/QA-, and solvent-accessible arginines throughout PSII. Full reversibility is achieved but (bi)carbonate uptake requires light. DIC depletion at the non-heme iron site and solvent-accessible arginines greatly reduces the yield of O2 due to O2 uptake, but accelerates the PSII-WOC cycle, specifically the S2-S3 and S3-S0 transitions. DIC removal from the WOC site abolishes water oxidation and appears to influence free energy stabilization of the WOC from a site between CP43-R357 and Ca2+., G. Ananyev, C. Gates, G. C. In this work, the injuries caused by clethodim herbicide application as well as the use of exogenous salicylic acid (SA) as a protective agent against clethodim in Zea mays leaves were examined. Although the target for clethodim is the inhibition of acetyl coenzyme A carboxylase (ACCase) which is the key enzyme for fatty acid biosynthesis, it can indirectly affect the photosynthetic machinery, gaseous exchange and some biochemical parameters. Clethodim application caused chlorosis and yellowing of leaf-tip parts. Higher doses caused browning or reddening of leaves and sometimes dead parts of the leaf margins were observed. The rate of photosynthesis was significantly lowered and the pigments content was highly reduced as a response to clethodim spraying. Moreover, other gas-exchange properties were altered. Furthermore, accumulation of high amounts of carbohydrates, proteins and proline were detected. SA spraying three days prior clethodim application caused partially or totally disappearance of clethodim injuries and kept the leaves similar to those of control. Improved photosynthesis and enhanced pigments content were observed in leaves treated with SA. Other analyzed parameters showed values similar to those of the corresponding control. In spring and winter cultivars of oilseed rape (Brassica napus var. oleifera), acclimation of photosynthetic apparatus to cold was connected with the increase in activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and sucrose-phosphate synthase (SPS). Conversely, cold de-acclimation entailed the decline of RuBPCO and SPS activities. The rate of this photosynthetic de-acclimation might depend on day temperature. On the other hand, temperature rise during de-acclimation (identical during the day and night) resulted in the improvement of photosynthetic activity measured by means of chlorophyll fluorescence. An increase in SPS activity (and even transitory increase in RuBPCO activity) was observed when the elongation growth rate (EGR) accelerated during de-acclimation. Throughout re-acclimation, plants with high EGR were unable to maintain or recover higher photosynthetic capacity, despite the fact that SPS activity remained high or even increased during re-acclimation. and M. We studied the photosynthetic performance of sterile and fertile sporophytes in a natural population of the fern Dryopteris affinis growing within a riparian forest (Central Italy) using chlorophyll (Chl) a fluorescence transients, the OJIP phase, where O is for the minimum fluorescence, P is for the peak (the maximum), and J and I are inflections. The “vitality” of the samples was assessed by the maximum quantum yield of primary photochemistry obtained indirectly from the fluorescence data (Fv/Fm); in the same way, the so-called performance index (PIABS) was obtained from fluorescence data. The photosynthetic performance (inferred from PIABS) of D. affinis changed significantly with the seasonal development of the fronds. The highest photosynthetic performance was recorded in the summer, corresponding to the period of spore release. The photosynthetic performance decreased in the winter, down to the minimal values of senescent fronds reached at the end of the seasonal cycle (May-June). On the whole, during the seasonal development, sterile and fertile fronds had a similar photosynthetic behaviour, as inferred from fluorescence data. At the end of spore maturation and dispersal (September-October), the fertile fronds showed somewhat lower photosynthetic performance than the sterile fronds, as revealed by PIABS. Being a long-lived fern, confined to humid and undisturbed sites in the Mediterranean, D. affinis deserves to be further investigated as a potential indicator of ecological continuity in Mediterranean riparian forests., L. Tropical savanna ecosystems are extremely diverse and important for global carbon storage. In the state of Mato Grosso, tropical savanna (locally known as the Cerrado), turns from well-drained, upland areas into seasonally flooded areas within the Pantanal; however, the Cerrado and the Pantanal share many common tree species, such as Vochysia divergens, a flood-adapted tree native to the Amazon Basin, and Curatella americana, a tree, adapted native to the welldrained the Cerrado. We measured the photosynthetic light response of these species in the the Cerrado and the Pantanal over a 1-year period to determine how these species physiologically adjust to these hydrologically distinct habitats. We hypothesized that neither species would experience a significant decline in maximum, light-saturated photosynthetic rate (Pmax) in their naturalized habitat. Physiological performance of each species was generally higher in the habitat that they were adapted to; however, our data indicated that both species have broad tolerance for seasonal variations in hydrology, allowing them to tolerate seasonal drought during the dry season in the Cerrado, and seasonal flooding during the wet season in the Pantanal. In V. divergens, flexible water-use efficiency, higher specific leaf area (SLA), and a greater ability to adjust mass-based Pmax (Pmax,m) to variations in leaf N and P concentration appeared to be key traits for withstanding prolonged drought in the Cerrado. In C. americana, increases in SLA and higher nutrient-use efficiency appeared to be important in maintaining high rates of Pmax,m in the seasonally flooded Pantanal. Physiological traits, which are positively associated with yield under salt-stress conditions, can be useful selection criteria in screening for salt tolerance. We examined whether chlorophyll (Chl) content can be used as screening criterion in wheat. Our study involved 5 wheat genotypes under both saline and nonsaline field conditions as well as in a sand-culture experiment. Salt stress reduced significantly biomass, grain yield, total Chl and both Chl a and b in all genotypes. In the sand-culture experiment, Chl accumulation was higher in PF70354/BOW, Ghods, and H499.71A/JUP genotypes at nonsaline control, moderate, and high salt concentrations, respectively. In the field experiment, genotype H499.71A/JUP belonged to those with the highest Chl density. The SPAD (Soil Plant Analysis Development) meter readings were linearly related to Chl content both in the sand-culture and in the field experiment. However, salt stress affected the calibration of SPAD meter. Therefore, separate Chl-SPAD equations were suggested for saline and nonsaline conditions. The correlation coefficients between the grain yield and SPAD were positive and significant both in the sand culture and in the field experiment. These findings suggested that SPAD readings could be used as a tool for rapid assessment of relative Chl content in wheat genotypes. The PsbH protein belongs to a group of small protein subunits of the photosystem 2 (PS2) complex and genes encoding PsbH homologues have been so far found in all studied oxygenic phototrophs. This single helix membrane protein is important for the proper function of the PS2 acceptor side and for stable assembly of PS2. Red alga contains four extrinsic proteins in photosystem II (PSII), which are PsbO, PsbV, PsbU, and PsbQ′. Except for the PsbQ′, the composition is the same in cyanobacterial PSII. Reconstitution analysis of cyanobacterial PSII has shown that oxygen-evolving activity does not depend on the presence of PsbQ′. Recently, the structure of red algal PSII was elucidated. However, the role of PsbQ′ remains unknown. In this study, the function of the acceptor side of PSII was analyzed in PsbQ′-reconstituted PSII by redox titration of QA and thermoluminescence. The redox potential of QA was positively shifted when PsbQ′ was attached to the PSII. The positive shift of QA is thought to cause a decrease in the amount of triplet chlorophyll in PSII. The photosynthetic parameters in leaves of three-year-old seedlings of Fraxinus rhynchophylla L. were studied under different soil water conditions and CO2 concentrations ([CO2]) with a
LI-COR 6400 portable photosynthesis system. The objective was to investigate the response of photosynthesis and stomatal conductance (gs) to various [CO2] and soil water conditions, and to understand the adaptability of F. rhynchophylla to such conditions. The results showed that the soil water content (RWC) required to maintain high photosynthetic productivity in F. rhynchophylla was 49.5-84.3%; in this range, net photosynthetic rate (PN) rose with [CO2] increasing from 500 to 1,400 μmol mol-1. Outside this RWC range, PN decreased significantly. The apparent maximum photosynthetic rate (Pmax,c) and carboxylation velocity (Vc) increased with increasing RWC and remained relatively high, when RWC was between 49.5 and 96.2%. CO2 compensation points and photorespiration rate exhibited a trend opposite to that of Pmax,c and Vc, indicating that moderate water stress was beneficial for increasing plant assimilation, decreasing photorespiration, and increasing production of photosynthates. gs declined significantly with increasing [CO2] under different water supplies, but the RWC range maintaining high gs increased. gs reached its maximum, when RWC was approximately 73% and then decreased with declining RWC. The maximal gs was found with increasing RWC. Thus, based on photosynthetic characteristics in artificial, vegetation construction in semiarid loess hill and gully area, F. rhynchophylla could be planted in habitats of low soil water content. and S. Changes in the temperature dependence of the maximum carboxylation capacity (VCmax) of Rubisco during thermal acclimation of PN remain controversial. I tested for acclimation of the temperature dependence of VCmax in quinoa, wheat, and alfalfa. Plants were grown with day/night temperatures of 12/6, 20/14, and 28/22°C. Responses of PN to substomatal CO2 (Ci) and CO2 at Rubisco (Cc) were measured at leaf temperatures of 10-30°C. VCmax was determined from the initial slope of the PNvs. Ci or Cc curve. Slopes of linear regressions of 1/VCmaxvs. 1/T [K] provided estimates the activation energy. In wheat and alfalfa the increases in activation energy with growth temperature calculated using Ci did not always occur when using Cc, indicating the importance of mesophyll conductance when estimating the activation energy.
