PAM (pulse amplitude modulation) fluorometers can be used to estimate the electron transport rate (ETR) [μmol(e-) m-2 s-1] from photosynthetic yield determinations, provided the absorptance (Abtλ) of the photoorganism is known. The standard assumed value used for absorptance is 0.84 (leaf absorptance factor, AbtF). We described a reflectance-absorptancetransmittance (RAT) meter for routine experimental measurements of the actual absorptance of leaves. The RAT uses a red-green-blue (RGB) LED diode light source to measure absorptances at wavelengths suitable for use with PAM fluorometers and infrared gas analysers. Results using the RAT were compared to Abtλ spectra using a Taylor integrating sphere on bird’s nest fern (Asplenium nidus), banana, Doryanthes excelsa, Kalanchoe daigremontiana, and sugarcane. Parallel venation had no significant effect upon Abt465 in banana, Doryanthes, a Dendrobium orchid, pineapple, and sugarcane, but there was a slight difference in the case of the fern A. nidus. The average Abt465 (approximately 0.96) and Abt625 (approximately 0.89) were approximately 14% and 6% higher than the standard value (AbtF = 0.84). The PAR-range Abt400-700 was only approximately 5% higher than the standard value (approximately 0.88) based on averaged absorptance from the blue, green, and red light data and from where the RGB-diode was used as a ‘white’ light source. In some species, absorptances at blue and red wavelengths are quite different (e.g. water lily). Reflectance measurements of leaves using the RAT would also be useful for remote sensing studies., R. Bean plants Phaseolus vulgaris L. (cv. Carioca and Negro Huasteco) and Vigna unguiculata L. Walp (cv. Epace-10) were grown in a growth chamber with a photosynthetic photon flux density of 200 μmol m-2 s-1 at leaf level and air temperature of 25+1 °C. Fully expanded, first pair leaves of 12-d-old plants were submitted for 90 min to high temperature (25, 30, 35, 40, 45, and 48 °C). Chlorophyll a fluorescence parameters (ETR, qP, qN, and F0) were investigated using a modulated fluorimeter at 25 °C during recovery considered here as 48 h after stress induction period. An accentuated decrease in qP and an increase in qN at 48 °C in Carioca and Negro Huasteco was not observed in Epace-10. In response to excitation irradiance a great potential for ETR was found in Negro Huasteco at 25 °C, also demonstrated by net photosynthetic rate. At 48 °C ETR was high for Epace-10 while it was equal to zero for Carioca and Negro Huasteco. Tolerance to high temperature observed in Epace-10 provided important information about the adaptative characteristics of Vigna cultivars to warm climates. and E. S. Costa ... [et al.].
Differences between sun (E) and shaded (S) foliage were studied in a Norway spruce (Picea abies [L.] Karst.) stand. Response curves describing the dependence of the CO2 assimilation rate (PN) on the CO2 concentration at the catalytic site of ribulose-1,5-bisphosphate carboxylase/oxygenase, RuBPCO (PN-Cc) were estimated using the simultaneous measurements of chlorophyll fluorescence and leaf gas exchange. Higher PN, higher electron transport (Ja), higher carboxylation capacity (Vc), and higher RuBPCO activity (τ) for sun acclimated needles was found. The S-needles had higher portion of internal limitation and higher CO2 compensation concentration (Γ) than the E-needles. Because higher degree of limitation of photosynthesis by carboxylation was ascertained, it can be assumed that photosynthesis in shade foliage is limited mainly by lower carboxylation capacity and by low chloroplastic CO2 concentration. and I. Tomato (Lycopersicon esculentum Mill. cv. Pearson) plants were grown in growth chambers for 25 days with cadmium (Cd) and then exposed briefly to ozone (O3). Gas exchange, chlorophyll a fluorescence, and pigment composition were analysed in leaves at the end of the treatment to assess the effects of a single pollutant and their combination on photosynthesis. The CO2 assimilation rate was dramatically reduced in plants subjected to the combined treatment, while the single effect of Cd appeared less severe than that of O3. The decline of CO2 photoassimilation found in all
O3-exposed plants was attributed to both stomatal and nonstomatal limitations. Tomato plants seemed to detoxify Cd to a great extent, but this resulted in growth suppression. In response to O3 exposure, the plants protected their photosystems by heat dissipation of excess energy via the xanthophyll cycle. Responses of the photosynthetic electron transport system of chloroplasts to exogenous proline application were evaluated in young and mature leaves of Arabidopsis thaliana plants under optimal growth conditions. Exogenous proline application (10 mM) during the 4th week of growth increased proline accumulation in young leaves more than in mature leaves, and possibly due to its degradation producing NADPH, decreased significantly the ratio of NADP+/NADPH in both leaf types compared with controls (without proline). However, the ratio of NADP+/NADPH remained significantly higher in the young leaves, suggesting lower proline degradation which resulted in less reduced plastoquinone pool than that in the mature leaves, under both low light [130 μmol(photon) m-2 s-1] and high light [1,200 μmol(photon) m-2 s-1] treatments. The young leaves seemed to adjust nonphotochemical fluorescence quenching in order to maintain a better PSII quantum yield. We concluded that under optimal growth conditions exogenous proline results in overreduction of the plastoquinone pool and blockage of photosynthetic electron flow due to accumulation of NADPH. The plants of Prosopis juliflora growing in northern India are exposed to large variations of temperature, vapour pressure deficits (VPD), and photosynthetic photon flux density (PPFD) throughout the year. Under these conditions P. juliflora had two short periods of leaf production, one after the winter season and second after summer, which resulted in two distinct even aged cohorts of leaves. In winter with cold nights (2-8 °C) and moderate temperatures during the day, the plants showed high rates of photosynthesis. In summer the midday temperatures often reached <45 °C and plants showed severe inhibition of photosynthesis. The leaves of second cohort appeared in July and showed typical midday depression of photosynthesis. An analysis of diurnal partitioning of the absorbed excitation energy into photochemistry showed that a smaller fraction of the energy was utilised for photochemistry and a greater fraction was dissipated thermally, further the photon utilisation for photochemistry and thermal dissipation is largely affected by the interaction of irradiance and temperature. The plants showed high photochemical efficiency of photosystem 2 (PS2) at predawn and very little photoinhibition in all seasons except in summer. The photoinhibition in summer was pronounced with very poor recovery during night. Since P. juliflora exhibited distinct pattern of senescence and production of new leaves after winter and summer stress period, it appeared that the ontogenic characteristic together with its ability for safe dissipation of excess radiant energy in P. juliflora contributes to its growth and survival. and P. We conducted a hydroponic experiment in order to study effects of the ammonium/nitrate ratio (0:15, 5:10, 7.5:7.5, and 10:5) on photosynthetic characteristics and biomass accumulation in Brassica chinensis under low light intensity and water stress. Results showed that net photosynthetic rate, transpiration rate, intrinsic water-use efficiency, stomatal conductance, intercellular CO2 concentration, effective quantum yield of PSII photochemistry, electron transport rate, and nonphotochemical quenching were lower in the treatment (low light intensity and water deficit) than those in the control, whereas stomatal limitation increased. Minimum fluorescence, maximal quantum yield of PSII photochemistry, and photochemical quenching were largely unchanged. Pigment contents first increased and then decreased as the ammonium/nitrate ratios were altered, with significant differences between treatment and control observed at all ratios except for 10:5. Biomass first increased slightly and then decreased both in treated and control plants. The effects of metabolisable sugars sucrose and glucose along with non-metabolisable isomers of sucrose palatinose and turanose were tested. Rate of oxygen evolution (P), electron transport rate (ETR), and photochemical quenching (qp) showed substantial decrease after 24 and 48 h by glucose and sucrose treatments, whereas there was no effect on all these parameters by the treatment with palatinose and turanose. Also the Fv/Fm ratio remained constant through the time of studies revealing that the maximal photochemical capacity of the cells was unchanged. Non-photochemical quenching (qN) showed a decrease compared to the control values by all the treatments. The effects of calcium chloride solution (10 mmol L-1) on mesophyll cell ultrastructure, gas exchange, chlorophyll and carotenoid content, and PSII in tobacco leaf were studied by simulating water deficit conditions via treatment with 25% PEG-6000 for 24 h. The results showed that under drought stress, the mesophyll cell structure and morphology were destroyed, photosynthesis and gas-exchange processes changed, photosynthetic pigment content decreased, and the electron transfer efficiency in PSII reduced. However, compared with the control treatment, under drought conditions, the addition of exogenous calcium could stabilize the structure and function of the chloroplasts, mitochondria, and endomembrane system in the mesophyll cells, maintain normal leaf net photosynthetic rate and gas exchange, alleviate the degree of photosynthetic pigment degradation, and increase the electron transfer energy in the leaves in PSII. As a means of ensuring normal photosynthesis under drought stress, we discovered that the application of exogenous calcium was more important for stabilization of the structure of the organelles, regulation of the osmotic balance, and increase of the photosynthetic pigment content, and proved to be less important for regulation of stomatal opening and closing., W. The acclimation to high light, elevated temperature, and combination of both factors was evaluated in tomato (Solanum lycopersicum cv. M82) by determination of photochemical activities of PSI and PSII and by analyzing 77 K fluorescence of isolated thylakoid membranes. Developed plants were exposed for six days to different combinations of temperature and light intensity followed by five days of a recovery period. Photochemical activities of both photosystems showed different sensitivity towards the heat treatment in dependence on light intensity. Elevated temperature exhibited more negative impact on PSII activity, while PSI was slightly stimulated. Analysis of 77 K fluorescence emission and excitation spectra showed alterations in the energy distribution between both photosystems indicating alterations in light-harvesting complexes. Light intensity affected the antenna complexes of both photosystems stronger than temperature. Our results demonstrated that simultaneous action of high-light intensity and high temperature promoted the acclimation of tomato plants regarding the activity of both photosystems in thylakoid membranes., A.
