A simple chlorophyll fluorescence (CF) measuring system has been implemented to study temporal characteristics of chlorophyll fluorescence induction (CFI) in dark-adapted freshwater algal cultures of Chlorella emersonii. There were two different decay time constants describing the CF quenching: τ0 (the faster) and τ1 (the slower) with amplitudes A0 and A1, respectively. The relative amplitude of the faster quenching component decreased once the sample was subject to deprivation from dissolved oxygen (DO). The DO concentration of samples was monitored to validate the effects of deprivation from air contact for up to 7 d and to the effect of adding DCMU to the culture (herbicide for blocking electron transport of photosystem 2). CFI analysis and DO measurements showed that the relative amplitude of A0 to (A0 + A1) and the DO concentration can be used as an indication of relative photosynthetic activity, thus allowing for the possibility to classify the physiological state of algal blooms into active and inactive states. and N. To explore the cause of difference in photosynthetic performance between different cultivars of crops, leaf net photosynt rate (PN) and photosystem 2 (PS2) photochemical efficiency (Fv/Fm), apparent quantum yield of carbon assimilation (φc), electron transport rate, photophosphorylation activity, etc. were measured in two soybean cultivars, Heinong 42 and Heinong 37. At pod setting and filling, significant differences in PN between them were observed. The former with a higher PN (from 7 to 38 %) had a significantly higher leaf thickness, leaf dry mass/area (LMA), chlorophyll content, soluble protein content, apparent quantum yield of electron transport through PS2 (φe), carboxylation efficiency (CE), and ribulose-1,5-bisphosphate carboxylase (RuBPC) activity. The significantly higher PN of Heinong 42 is mainly due to its higher content and activity of RuBPC. and Hua Jiang, Da-Quan Xu.
Wheat (Triticum aestivum L. cv. Sonalika) seedlings were grown in Hoagland solution. Primary leaves were harvested at 8, 12, and 15 d and cut into five equal segments. Contents of photosynthetic pigments and proteins, and photosystem 2 (PS2) activity increased from base to apex of these leaves. Chlorophyll (Chl) content was maximum at 12 d in all the leaf segments, but PS2 activity showed a gradual decline from 8 to 15 d in all leaf segments. In sharp contrast, the CO2 fixation ability of chloroplasts increased from 8 to 15 d. CO2 fixation ability of chloroplasts started to decline from base to apex of 15-d-old seedlings, where the content of ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit (RuBPCO-LSU) increased acropetally. RuBPCO-LSU content was maximum in all the leaf segments in 12-d-old seedlings. This shows a distinctive pattern of PS2, Chl, CO2 fixation ability of chloroplasts, and RuBPCO-LSU content along the axis of leaf lamina during development and senescence. RuBPCO-LSU (54 kDa) degraded to fragments of 45, 42, 37, 19, and 16 kDa products which accumulated along the leaf axis during ageing of chloroplasts. Thus the CO2 fixation ability of chloroplasts declines earlier than PS2 activity and photosynthetic pigment contents along the leaf lamina. and F. Dilnawaz ... [et al.].
Pisum sativum (L.) plants were grown under "white" luminescent lamps, W [45 µ mol(quantum) m-2 s-1] or under the same irradiation supplemented with narrow spectrum red light-emitting diodes (LEDs), RE [λmax = 660 nm, Δλ = 20 nm, 40 µmol(quantum) m-2 s-1]. Significant differences in the chlorophyll (Chl) a fluorescence parameters, degree of State 1-State 2 transition, and the pigment-protein contents were found in plants grown under differing spectral composition. Addition of red LEDs to the "white light" resulted in higher effective quantum yield of photosystem 2 (PS2), i.e. F'v/F'm, linear electron transport (ϕPS2), photochemical quenching (qP), and lower non-photochemical quenching (qN as well as NPQ). The RE plants were characterised by higher degree State 1-State 2 transition, i.e. they were more effective in radiant energy utilisation. Judging from the data of "green" electrophoresis of Chl containing pigment-protein complexes of plants grown under various irradiation qualities, the percentage of Chl in photosystem 2 (PS2) reaction centre complexes in RE plants was higher and there was no difference in the total Chl bound with Chl-proteins of light-harvesting complexes (LHC2). Because the ratio between oligomeric and monomeric LHC2 forms was higher in RE plants, we suggest higher LHC2 stability in these ones. and N. M. Topchyi ... [et al.].

Young plants of maize inbred lines CE777, CE704, and CE810 and their F1 hybrids displaying a positive heterotic effect in various photosynthetic characteristics were exposed to low temperature during their early growth developmental stage. The photochemical activity of isolated mesophyll chloroplasts and the contents of photosynthetic pigments in leaves of stressed and non-stressed plants were compared with the aim to find out the possible changes in the relationship between parents and hybrids, and to determine the genetic basis of heterosis in F1 generation. Strong decrease in the content of chlorophylls was observed for all genotypes examined when plants were subjected to low growth temperature. Similar change was recorded for Hill reaction activity (HRA) of inbred lines but not of their F1 hybrids, and no significant response at all was found for photosystem 1 (PS1) activity or the total carotenoids content. The intraspecific variation due to differences between genotypes was found for most of photosynthetic characteristics examined. This variation was caused by the additive and dominance genetic effects. Positive dominance was the main cause of positive heterosis in HRA and in the contents of photosynthetic pigments and was much more pronounced in the stressed plants compared to the non-stressed ones. The changes in thermoluminescence (TL) signals induced by short-term ozone exposure of leaves are characterized by a down-shift of the peak-temperature of the TLB-band and an increase of a TL band at 55°C. We investigated the relationship of these changes to photosystem 2 (PS2) photochemistry. The changes were not only detectable in the presence of ozone, but also after irradiation of dark-adapted leaves and after aging of irradiated detached leaf segments. The opposite effect on TL, an up-shift of the peak-temperature of the B-band and the decrease of the intensity of the band at 55°C were found after infiltration of leaves with nigericin, antimycin A, and diphenyleneiodonium chloride (DPI). Propyl gallate down-shifted the peak-temperature of the B-band. 2,5-dimethyl-1,4-benzoquinone up-shifted the peak-temperature of the B-band and decreased the intensity of the 55°C band. The intensity of the 55°C band did not change significantly in the presence of oxygen in comparison to that in nitrogen atmosphere. It decreased with time of dark adaptation (50% intensity was observed after 3 h of dark adaptation at room temperature), however, it was reactivated to its initial value (at 5 min of dark adaptation) after 1 single-turnover flash. The 55°C band was not significantly changed in the presence of DCMU. Thus the ozone-induced band at 55°C is assigned to charge recombination in PS2. Changes in the electron transport chain at the acceptor side of PS2, probably related to the cyclic electron transport around photosystem 1 and/or chlororespiration, could play an important role in the increase of the 55°C band and the down-shift of the B-band. The thermoluminescence signals from leaflets of the same pea plant varied gradually according to their developmental stage. The AG emission, due to a back flow of electrons towards photosystem 2 (PS2) along a cyclic/chlororespiratory pathway, was stronger in mature leaves than in the growing ones. These age-related variations could be explained by a higher capacity of cyclic electron flow in mature leaves.
Senescence-induced changes in the xanthophyll cycle activity and chlorophyll (Chl) fluorescence parameters were compared in detached barley (Hordeum vulgare L.) leaf segments kept for 6 d in darkness or under continuous " white light" (90 μmol m-2 s-1). Before detachment of the leaf segments, the plants were grown at periodic regime [12 h light (90 μmol m-2 s-1)/12 h dark]. The de-epoxidation state of the xanthophyll cycle pigments (DEPS) in the leaf samples was determined immediately (the actual DEPS), after 1 h of dark-adaptation (the residual DEPS), and during 14 min of a high-irradiance (HI) exposure (500 μmol m-2 s-1) (HI-induced DEPS). In the light-senescing segments, senescence was delayed pronouncedly compared to dark-senescing ones as the Chl content, the photosystem 2 photochemistry, and electron transport processes were highly maintained. Further, the actual DEPS increased, probably due to the increased mean photon dose. The HI-induced increase in the DEPS was stimulated in the light-senescing segments, whereas it was slowed down in the dark-senescing ones. However, after the 14 min HI-exposure of the dark-senescing segments the HI-induced DEPS was not markedly lower than in the mature leaves, which indicated the maintenance of the xanthophyll cycle operation. and M.
