The effect of sulphur deprivation and irradiance (180 and 750 µmol m-2 s-1) on plant growth and enzyme activities of carbon, nitrogen, and sulphur metabolism were studied in maize (Zea mays L. Pioneer cv. Latina) plants over a 15-d-period of growth. Increase in irradiance resulted in an enhancement of several enzyme activities and generally accelerated the development of S deficiency. ATP sulphurylase (ATPs; EC 2.7.7.4) and o-acetylserine sulphydrylase (OASs; EC 4.2.99.8) showed a particular and different pattern as both enzymes exhibited maximum activity after 10 d from the beginning of deprivation period. Hence in maize leaves the enzymes of C, N, and S metabolism were differently regulated during the leaf development by irradiance and sulphur starvation. and S. Astolfi, M. G. de Biasi, C. Passera.
Differences in acclimation to elevated growth CO2 (700 µmol mol-1, EC) and elevated temperature (ambient +4 °C, ET) in successive leaves of wheat were investigated in field chambers. At a common measurement CO2, EC increased photosynthesis and the quantum yield of electron transport (Φ) early on in the growth of penultimate leaves, and later decreased them. In contrast, EC did not change photosynthesis, and increased Φ at later growth stages in the flag leaf. Contents of chlorophyll (Chl), ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and total soluble protein were initially higher and subsequently lower in penultimate than flag leaves. EC decreased RuBPCO protein content relative to soluble protein and Chl contents throughout the development of penultimate leaves. On the other hand, EC initially increased the RuBPCO:Chl and Chl a/b ratios, but later decreased them in flag leaves. In the flag leaves but not in the penultimate leaves, ET initially decreased initial and specific RuBPCO activities at ambient CO2 (AC) and increased them at EC. Late in leaf growth, ET decreased Chl contents under AC in both kinds of leaves, and had no effect or a positive one under EC. Thus the differences between the two kinds of leaves were due to resource availability, and to EC-increased allocation of resources to photon harvesting in the penultimate leaves, but to increased allocation to carboxylation early on in growth, and to light harvesting subsequently, in the flag leaves. and P. The plant growth, net photosynthetic rate (PN), intercellular CO2 concentration (ci), and dry matter production of Ceriops roxburghiana Arn. were significantly increased with increasing salinity from 0 to 400 mM NaCl. At 600 mM NaCl, shoot and root lengths, and dry mass were significantly depressed with respect to control. Absence of diurnal fluctuation of concentrations of organic acids, and the low activity of phosphoenolpyruvate carboxylase and high activity of ribulose-1,5-bisphosphate carboxylase confirmed the operation of C3 pathway in Ceriops even at increasing salinity. and A. Net photosynthetic rate (PN) measured at the same CO2 concentration, the maximum in vivo carboxylation rate, and contents of ribulose-1,5-bisphosphate (RuBP) carboxylase/oxygenase (RuBPCO) and RuBPCO activase were significantly decreased, but the maximum in vivo electron transport rate and RuBP content had no significant change in CO2-enriched [EC, about 200 µmol mol-1 above the ambient CO2 concentration (AC)] wheat leaves compared with those in AC grown wheat leaves. Biao 810S is a chlorina mutant of the thermosensitive genic male sterile (TGMS) rice. We compared photosynthetic characteristics of these two lines. The contents of chlorophylls and carotenoids in Biao 810S were approximately half of those in 810S. However, the net photosynthetic rate (PN) of Biao 810S was higher than that of 810S under high irradiance or low concentration of carbon dioxide, and the photon quantum efficiency was higher than that of 810S. The activity of ribulose-1,5-bisphosphate carboxylase/oxygenase in Biao 810S was only 69.80 % of that in 810S, but the activities of phosphoenolpyruvate carboxylase and NADP-malic enzyme were 79.50 and 69.06 % higher than those of 810S, respectively, suggesting that the efficiency of photon energy utilization in Biao 810S was enhanced by reduction of thermal dissipation and increase of electron transfer rate to generate sufficient assimilation power for the dark reactions. Consequently, the increased activities of C4 photosynthetic enzymes lead to more effective fixation of CO2 and the synergistic effect of light and dark reactions contributed to the higher PN of Biao 810S. and L.-J. Wheat (Triticum aestivum L. cv. HD 2285) was grown in control (C) and heated (H) open top chambers (OTCs) for entire period of growth and development till maturity. The mean maximum temperature of the entire period was 3 °C higher in H-compared to C-OTCs. Net photosynthetic rate (P N) measured at different temperature (20-40 °C) of C-and H-grown plants showed greater sensitivity to high temperature in H-plants. PN measured at respective growth temperature was lower in H-compared to C-plants. The CO2 and irradiance response curves of photosynthesis also showed lesser response in H-compared to C-plants. The initial slope of PN versus internal CO2 concentration (PN/Ci) curve was lower in H-than C-plants indicating ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) limitation. In irradiance response curve, the plateau was lower in H-compared to C-plants which is interpreted as RuBPCO limitation. RuBPCO content in the leaves of C-and H-plants, however, was not significantly different. Ribulose-1,5-bisphosphate carboxylase (RuBPC) initial activity was lower in H-plants, whereas activity of fully activated enzyme was not affected, indicating a decrease in activation state of the enzyme. This was further substantiated by the observed decrease in RuBPCO activase activity in H-compared to C-plants. RuBPCO activase was thus sensitive even to moderate heat stress. The contents of chlorophyll (Chl) and carotenoids (Car) per fresh mass were lower in shade needles than in sun needles. Ribulose-1,5-bisphosphate carboxylase (RuBPC) activity and contents of soluble proteins were also significantly lower in shade needles. In isolated thylakoids, a marked lower rate of whole chain and photosystem (PS) 2 activities were observed in shade needles. Smaller lower rate of PS1 activity was also observed in shade needles. The artificial exogenous electron donors, diphenyl carbazide (DPC) and NH2OH, significantly restored the loss of PS2 activity in shade needles. Similar results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements. The marked lower rate of PS2 activity in shade needles was due to the lower contents of 47, 33, 28-25, 23, and 17 kDa polypeptides. This conclusion was confirmed by immunological studies showing that the content of the 33 kDa protein of the watersplitting complex was diminished significantly in shade needles. and M. Bertamini, K. To explore the cause of difference in photosynthetic performance between different cultivars of crops, leaf net photosynt rate (PN) and photosystem 2 (PS2) photochemical efficiency (Fv/Fm), apparent quantum yield of carbon assimilation (φc), electron transport rate, photophosphorylation activity, etc. were measured in two soybean cultivars, Heinong 42 and Heinong 37. At pod setting and filling, significant differences in PN between them were observed. The former with a higher PN (from 7 to 38 %) had a significantly higher leaf thickness, leaf dry mass/area (LMA), chlorophyll content, soluble protein content, apparent quantum yield of electron transport through PS2 (φe), carboxylation efficiency (CE), and ribulose-1,5-bisphosphate carboxylase (RuBPC) activity. The significantly higher PN of Heinong 42 is mainly due to its higher content and activity of RuBPC. and Hua Jiang, Da-Quan Xu.
Among four mulberry (Morus alba L.) cultivars (K-2, MR-2, BC2-59, and S-13), highest net photosynthetic rate (PN) was observed in BC2-59 while the lowest rates were recorded with K-2. Significant differences among the four cultivars were found in leaf area, biomass production, activities of ribulose-1,5-bisphosphate carboxylase and sucrose phosphate synthase, and glucose and sucrose contents. The PN and the activities of photosynthetic enzymes in the four cultivars were significantly correlated with the growth and biomass production measured as leaf yield, total shoot mass, and aerial plant biomass. and K.
