The rose grain aphid, Metopolophium dirhodum (Walker) (Hemiptera: Aphididae) is one of the three most important cereal aphid species in Europe. High temperature is detrimental for the survival of this species. Detailed experiments were conducted on the effect of high temperature (27, 28, 29, 30, 31, 31.5, 32.5, 33 and 34°C), period of exposure (2, 3, 4, 6 and 8 h per day for 1, 2, 4, 6 days) and developmental stage (2nd, 3rd, 4th instar nymph and adult) on the survival of the aphid. The results show that all three factors significantly affect survival. Temperatures over 29°C for 8 h significantly reduced survival, which decreased generally as the temperature increased. The survival was inversely related to the period of exposure. Exposing aphids to 32.5°C for 4 h or longer significantly reduced survival. Mature aphids had a lower tolerance of high temperatures than nymphs. Periods of high temperature experienced by 4th instar and adult aphids can greatly affect their survival. The value of these results for forecasting and determining control thresholds, the effect of global warming on cereal aphid abundance and the dropping off behaviour of the aphids are discussed.
Cholestasis is characterized by the elevation of serum total bile acids (TBA), which leads to the production of both free radicals and oxidative stress. Although they do not share the same mechanisms, membrane glycosphingolipids (GSL) and the antioxidant enzyme heme oxygenase-1 (HMOX1) both act against the pro-oxidative effect of TBA. The aim of the study was to assess the role of HMOX on GSL redistribution and composition within hepatocytes in the rat model of estrogen-induced cholestasis. Compared to the controls, an increase of total gangliosides in the liver homogenates of the cholestatic group (P=0.001) was detected; further, it paralleled along with the activation of their biosynthetic b-branch pathway (P<0.01). These effects were partially prevented by HMOX activation. Cholestasis was accompanied by a redistribution of GM1 ganglioside from the cytoplasm to the sinusoids; while HMOX activation led to the retention of GM1 in the cytoplasm (P=0.014). Our study shows that estrogen-induced cholestasis is followed by changes in the synthesis and/or distribution of GSL. These changes are not only triggered by the detergent power of accumulated TBA, but also by their pro-oxidant action. Several authors have reported the association of postprandial hypertriglyceridemia with oxidative stress, systemic inflammation and endothelial dysfunction. Our aim was to investigate the effect of high-calorie meal on blood markers of oxidative stress and endothelial dysfunction and the association of APOA5-1131T/C and -250G/A hepatic lipase (HL) polymorphisms with postprandial triglyceride response. This study included 102 healthy male volunteers. All participants consumed a high-calorie meal (823 calories, 50 g fat, 28 g protein, 60 g carbohydrates). Total cholesterol, triglycerides, HDL-cholesterol, LDL-cholesterol, hsCRP, TAS and ICAM-1 were measured at fasting state and postprandially. APOA5-1131T/C and -250G/A HL polymorphisms were also determined. Postprandial triglycerides were significantly increased (1.4 (1.1-2.1) vs. 2.4 (1.9-3.3) mmol/l, P<0.001). Average triglyceride increase was 1.0±0.7 mmol/l (65 %). Concentration of triglycerides, HDL- cholesterol, LDL-cholesterol, TAS and ICAM-1 differed significantly between the fasting state and postprandial measurements (P<0.001). However, those differences were within the limits of analytical imprecision. Other parameters did not change 3 h after the meal. Triglycerides response did not differ respective to the APOA5 and HL polymorphisms. Family history of hypertension and acut e myocardial infarction were associated with higher postprandial triglyceride concentrations. Postprandial hypertriglyceridemia is not associated with increased concentrations of hsCRP, TAS and ICAM-1. Results of the effect of subsidence of the Little Hungarian Plain on vertical movement of the riverbed and the downstream riverbed slope changes of the regulated flowchannel of Danube are presented. Subsidence of the flowchannel is also affected by the tectonic movements. The subsidence rate is attenuated by the andesitic Börzsöny and Visegrád Mountains since they function as natural barriers for the river. In the eastern vicinity of the Börzsöny and the Mátra, where the river flows in loose, unconsolidated sediments, the deepening of the flowchannel is considerable. and Príspevok prezentuje výsledky účinku poklesu Malej Uhorskej nížiny (geogr.) na vertikálny pohyb riečneho koryta a na zmeny pozdĺžneho sklonu upraveného koryta Dunaja. Pokles koryta toku je ovplyvnený aj tektonickými pohybmi. Intenzita poklesu je tlmená účinkom andezitovej línie horských celkov Börzsöny a Visegrád, ktoré pôsobia na rieku ako prírodné bariéry. Na východ od týchto horských celkov, kde rieka preteká cez voľnejšie uložené, nespevnené usadeniny, vyskytuje sa značné prehĺbenie riečneho koryta.
The effect of hormonal stimulation with FSH injection in the doses of 18 mg (360 IU) and 24 mg (480 IU FSH) on the levels of plasma catecholamines (dopamine, norepinephrine and epinephrine) was studied by radioenzymatic methods during synchronized oestrous cycles of the sheep. Catecholamines were determined in the blood plasma before and 24, 48, 96 and 120 hours after application of FSH. It follows from the results that the levels of plasma dopamine increased significantly (p<0.001) 24 and 48 hours after FSH application. Furthermore, the levels of dopamine (DA) during the other time intervals observed, compared with those of controls before hormonal stimulation, remained at a higher level. A lower dose of the hormone (18 mg) had a more pronounced effect on changes in the levels of plasma dopamine. Norepinephrine (NE) did not exhibit any significant changes in comparison with the plasma levels of dopamine in the sheep after hormonal stimulation with FSH. A statistically significant increase in plasma norepinephrine was recorded 24 hours after administration of 18 or 24 mg FSH. During the other time intervals observed, its levels did not differ from the control values. Plasma epinephrine (E) showed a significant increase 24 and 48 hours after FSH application but not later. The effect of FSH on plasma catecholamine levels was not dose-dependent and their increase was pronounced especially in the period of ovulation.
Genetic differentiation is a consequence of the combination of drift and restriction in gene flow between populations due to barriers to dispersal, or selection against individuals resulting from inter-population matings. In phytophagous insects, local adaptation to different kinds of host plants can sometimes lead to reproductive isolation and thus to genetic structuring, or even to speciation. Acanthoscelides obtectus Say is a bean bruchid specialized on beans of the Phaseolus vulgaris group, attacking both wild and domesticated forms of P. vulgaris and P. coccineus. This study reveals that the genetic structure of populations of this bruchid is explained mainly by their geographical location and is not related to a particular kind (wild or domesticated) of bean. In contrast, the species of bean might have led, to some extent, to genetic structuring in these bruchids, although our sampling is too limited to address such process unambiguously. If confirmed, it would corroborate preliminary results found for the parasitoid species that attack Acanthoscelides species, which might show a genetic structure depending on the species of host plant.
T-2 toxin and its metabolite HT-2 toxin are one of the most toxic mycotoxins of type A-trichothecenes, which are produced mainly by Fusarium species. Therefore, study of Fusarium toxins T-2 toxin and HT-2 toxin is an essential issue because they could also play role in failures of reproductive functions as well as endocrine system of domestic animals. Assessment of the effect of A-trichothecene mycotoxin HT-2 toxin alone or combined with insulin-like growth factor (IGF-I), leptin and ghrelin on estradiol secretion by rabbit ovarian fragments in vitro was done. Rabbit ovarian fragments were incubated without (control group) or with HT-2 toxin, or its combinations with IGF-I, leptin and ghrelin at various concentrations for 24 h. Secretion of 17β-estradiol was determined by ELISA. Firstly, HT-2 toxin at the doses 10 and 100 ng.ml-1, but not at 1 ng.ml-1 decreased 17β-estradiol secretion by ovarian fragments. Secondly, 17β-estradiol secretion was not affected by HT-2 toxin exposure combined with growth factor IGF-I, metabolic hormones leptin and ghrelin. In conclusion, HT-2 toxin has potent direct dose-dependent effects on ovarian steroidogenesis in rabbits. Various protocols may be used for acute pancreatitis treatment. Recently, the benefit of hyperbaric oxygen (HBO) has been demonstrated. To clarify the mechanism of HBO on the process of the acute pancreatitis, we determined the levels of antioxidant enzymes in an acute pancreatitis model. Forty-five Sprague-Dawley rats were randomly divided into three groups: Group I: sham group (n=15), Group II: pancreatitis group (n=15), Group III: pancreatitis group undergoing HBO therapy (n=15). HBO was applied postoperatively for 5 days, two sessions per day at 2.5 fold absolute atmospheric pressure (ATA) for 90 min. Superoxide dismutase (Cu/Zn-SOD), malondialdehyde (MDA), and glutathione peroxidase (GSH Px) activity were measured in pancreatic tissue and erythrocyte lysate. MDA and GSH Px were also determined in plasma. In addition, amylase levels were measured in the serum. While serum amylase levels and MDA values in erythrocyte, plasma and pancreatic tissue were decreased, the levels of GSH Px and SOD were found to be significantly increased in the Group III as compared to those of the Group II. The dentate gyrus of the hippocampus is one of the few places in the brain where neurogenesis occurs in adulthood. Nowadays, an increasing number of children and young adults are affected by hypertension, one of the factors in the development of cerebrovascular diseases and age-related cognitive deficits. Since
these cognitive deficits are often hippocampus-dependent, it is possible that hypertension exerts this effect via decreasing adult neurogenesis which has been shown to be essential for a range
of cognitive tasks. We used spontaneously hypertensive rats, which develop hypertension in the first weeks of life. Half of them were treated with the antihypertensive drug captopril. We found that the drug-induced lowering of blood pressure in this period did not affect the rate of adult neurogenesis. In a second experiment, we used another animal model of hypertension – salt-sensitive and salt-resistant strains of Dahl rats. A high-salt diet induces hypertension in the salt-sensitive strain, but not in the salt-resistant strain. The high-salt diet led to salt-induced hypertension, but did not affect the level of adult neurogenesis in the dentate gyrus of the hippocampus. We
conclude that hypertension does not significantly affect the rate of hippocampal neurogenesis in young adult rats.
The aim of our study was to test the in fluence of short exposure (6 h) of preimplantation rabbit embryos to elevated temperatures (41.5 ºC or 42.5 ºC) in vitro on their developmental capacity. Fertilized eggs recovered from female oviducts at the pronuclear stage (19 hpc) were cultured at standard temperature (37.5 ºC) until the morula stage (72 hpc). Afterwards, the embryos were divided into two groups, cultured for 6 h either at hyperthermic (41.5 ºC or 42.5 ºC) or standard temperature (control 37.5 ºC), post-incubated overnight (16-20 h) at 37.5 ºC and then evaluated for developmental stages, apoptosis (TUNEL), proliferation (cell number), actin cytoskeleton and presence of heat-shock proteins Hsp70. It was observed that hyperthermia at 41.5 ºC did not alter progression of embryos to higher preimplantation stages (expanded and hatching/hatched blastocysts), rate of apoptosis, total cell number of blastocysts and structure of actin filament compared to 37.5 ºC. We stern-blotting revealed the presence of heat stress-induced 72 kDa fraction of Hsp70 proteins in granulosa cells (exposed to 41 ºC) and embryos (exposed to 41.5 ºC). Following the elevation of temperature to 42.5 ºC embryo development was dramati cally compromised. The embryos were arrested at the morula or early blastocyst stage, showed an increased rate of apoptosis and decreased total cell number compared to control. The structure of actin filaments in most of blastomeres was damaged and such blastomeres often contained apoptotic nuclei. In this group a presence of heat-stress-induced fraction of Hsp70 proteins had not been confirmed. This is the first report demonstrating a threshold of thermotolerance of rabbit preimplantation embryos to hyperthermic exposure in vitro. A detrimental effect of higher temperature on the embryo is probably associated with the loss of their ability to produce Hsp70 de novo, which leads to cytoskeleton alterations and enhanced apoptosis., A.
