The influence of increased solar UV-B radiation on the photosynthetic characteristics in cowpea seedlings (Vigna unguiculata) grown at optimal (Mgs) and low (Mgd) Mg levels were studied. Both higher UV-B and Mgd treatments caused significant drops of photochemical activities and net CO2 uptake rates (PN). Yet the UV-B-induced decrease in the photosynthetic efficiency was lesser in Mgd seedlings. The leaf Chl a fluorescence measurements proved that after receiving an enhanced UV-B radiation these seedlings showed a significant enhancement in their variable parts. The PSM oscillation of slow fluorescence kinetics was remarkably altered by both treatments. The PN also followed a typical inhibitory pattern as seen in photochemical activities. Concentrations of several chloroplast proteins in trifoliate leaves were significantly reduced by Mgd treatment and unaffected by the other two treatments. Whereas the contents of 43-47 kDa polypeptides in primary leaves were markedly reduced with a maximal effect in Mgd seedlings, no major difference was noted for combined stress. and A. Premkumar, G. To further investigate the role of insulin during preimplantation embryo development, we compared the effects of insulin on the development of mouse and bovine preimplantation embryos and on cell proliferation during culture in vitro in simplex media. The influence of insulin on the development of mouse zygotes was determined during cultivation in mSOF medium, alone or supplemented with glucose. Similarly, the effects of insulin on the bovine preimplantation embryo development were studied in mSOF medium. The addition of insulin into mSOF medium enhanced significantly the number of cells per mouse blastocyst. Moreover, when mSOF medium was supplemented with insulin and 0.2 mmol.l-1 glucose, the percentage of hatched blastocysts and the mean cell number of mouse blastocysts were significantly higher. Insulin had no significant effect on the development of bovine embryos, produced by in vitro fertilization of in vitro matured oocytes. Neither the rates of developing embryos nor the mean number of cells in blastocysts were different in comparison with control embryos. Our results suggest that the in vitro development of mouse embryos could be enhanced by the addition of insulin to the culture medium and is further improved by the addition of glucose. In contrast to this our results indicate that insulin has no detectable beneficial effect on the preimplantation development of bovine embryos in mSOF medium., J. Mihalik, P. Rehák, J. Brain infections as well as peripheral challenges to the immune system lead to an increased production of interleukin-1beta (IL-1b), a cytokine involved in leukocyte-mediated breakdown of the blood-brain barrier. The effects of IL-1b have been reported to depend on whether the route of administration is systemic or intracerebral. Using 50-day-old male rats, we compared the effects of IL-1b on brain γ-glutamyl transpeptidase (GGT; an enzymatic marker of brain capillary endothelium) at 2, 24 and 96 h after either an intravenous (i.v.) injection of 5 μg IL-1β or an intracerebroventricular (i.c.v. - lateral ventricle) infusion of 50 ng IL-1β. When the i.v. route was used, the GGT activity underwent small but significant changes; decreasing in the hippocampus 2 h after the i.v. injection, increasing 24 h later and returning to control levels at 96 h. No significant changes in the hippocampal GGT activity were observed at 2 and 24 h following the i.c.v. infusion. The GGT activity in the hypothalamus remained unchanged regardless of the route of IL-1b administrations. Similar changes in GGT activity were revealed histochemically. The labeling was found mainly in the capillary bed, the changes being most evident in the hippocampal stratum radiatum and stratum lacunosum-moleculare. A transient increase in GGT activity at 24 h, together with a less sharp delineation of GGT-stained vessels, may reflect IL-1b induced increased turnover of glutathione and/or oxidative stress, that may in turn, be related to altered permeability of the blood-brain barrier in some neurological and mental disorders, including schizophrenia.
The abandonment of less productive agricultural land and the intensification of agricultural land use are the main features of the Common Agricultural Policy (CAP) that Croatia will enforce now as new member of the EU. Due to
demographic changes and the economic transition in Croatia resulting from
war in the 1990s, substantial tracts of agricultural land were abandoned. We investigated two habitat types in the protected floodplain landscape of Lonjsko polje in the continental part of the country: arable land and pastures.
Both habitats were maintained by agricultural management and suffered from partial abandonment. Land abandonment increased the susceptibility to encroachment by the invasive plant species Amorpha fruticosa. Data on bird communities were obtained during the breeding season in 2010 while there were high water levels in the floodplain. Data were collected from 63 points, and a total of 1447 individuals from 70 species were recorded during
the study. We found that the bird community structure was primarily related to the presence/abandonment of agricultural land use and the habitat type. Further, we detected that the bird community structure in the same habitat type differed by management intensity. Open habitat specialists were most influenced by land abandonment. However, the conservation value (according to the Species of European Conservation Concern value, SPEC) of grazed pastures and abandoned pastures did not differ significantly, in part because the overgrown pastures with high water levels were found to be suitable for Acrocephalus species. The shift in bird community structure between abandoned and managed arable lands were smaller than those
detected in the pastoral communities. Because land abandonment is a widespread phenomenon in Croatia, we emphasize the urgent need
for a nationwide monitoring program for farmland birds to register the resulting changes in farmland bird communities and to develop appropriate agri-environment measures to mitigate the process.
The present study focuses on the possible influence of large-scale climatic patterns on precipitation, temperature, and discharge in two distinct river basins in the Czech Republic. The first one is a flat lowland basin of the Cidlina River and the second one represents a more mountainous type of climate (the Blanice River). The large-scale climatic patterns used comprise monthly averages of teleconnection patterns, sea level pressure values, and two geopotential heights (850 hPa and 500 hPa). The correlations for corresponding months and also up to a three months lead time were investigated. The relations concerning the corresponding months proved to be most reliable in the winter period of the year. The behaviour of both selected river basins with respect to these predictors is rather similar in character. However, higher values of correlation coefficients are reached in the case of the Cidlina River basin. Considering the prediction abilities with a certain lead time, the results seem to be more promising for the winter and spring periods in both basins. In the Cidlina River basin, they are likely to be beneficial also for the late summer and autumn period.
Several studies have investigated the association between infection with Toxoplasma gondii (Nicolle et Manceaux, 1908), pregnancy and fertility, but the results of studies focused on the fertility are rather ambiguous. Here we report results of four new cross-sectional studies. The studies were performed in the General University Hospital, Prague (study A with n = 1 165, and study C with n = 317), in private clinics of the Centre of Reproductive Medicine, Prague (study B with n = 1 016), and in a population of Czech and Slovak volunteers from the Facebook page 'Guinea Pigs' willing to participate in various basic science studies (study D with n = 524). In studies A and B, the clinical records were used to assess the fertility problems, whereas in studies C and D, the women were asked to rate their fertility problems using a six-point scale. Pregnant T. gondii-infected women were older than T. gondii-free women (study A: 33.1 vs 31.2, P < 0.001; study B: 30.6 vs 29.6, P = 0.012) and more often used assisted reproductive technology to conceive (study A: 17.2% vs 12.4%, P = 0.041; study B: 13.4% vs 9.2%, P = 0.317). Pregnant T. gondii-infected primiparous women were older than T. gondii-free primiparas (study A: 31.1 vs 29.5, P < 0.001; study B: 29.7 vs 28.9, P = 0.064) and more often used assisted reproductive technology to conceive (study A: 24.7% vs 14.4%, P = 0.010; study B: 15.9% vs 15.5%, P = 0.888). T. gondii-infected women reported to take a longer time to conceive than T. gondii-free women (P = 0.015). They also claimed to have more fertility problems than T. gondii-free women (P < 0.0001). Our results suggest that 'asymptomatic' latent toxoplasmosis could be a more serious source of fertility problems and health-associated burden than more severe but far rarer congenital toxoplasmosis., Šárka Kaňková, Jaroslav Flegr, Pavel Calda., and Obsahuje bibliografii
Net CO2 exchange rate (PN) of shoots and diel fluctuations in titratable acidity of leaves of Senecio rowleyanus were measured to determine whether penetration of radiant energy through leaf "windows" (narrow, translucent strips on the leaf epidermis) resulted in increased CAM. Nocturnal PN and nighttime increases in acidity were compared among plants with windows covered with reflective adhesive tape, transparent adhesive tape (to control for potential effects of the adhesive), and no tape. The windows did not significantly enhance the degree of CAM in S. rowleyanus. and K. J. Egbert, C. E. Martin.
Four fluorescence parameters [Fv/Fm = the intrinsic efficiency of energy conversion via photosystem 2 (PS2); Fv'/Fm'= the efficiency of energy conversion via PS2 in the light; P = fraction of absorbed radiant energy utilized for photosynthesis; and D = fraction of absorbed radiant energy dissipated as heat] were measured on leaves of seven species of succulents having epidermal windows. While the function of leaf windows has reportedly been to increase absorption of radiant energy and, hence, the rate of photosynthesis in these species, recent evidence indicates that this translucent portion of epidermal tissue, lacking chlorophyll, may also result in photoinhibition in these species, especially for those with growth habits aboveground. Species with aboveground and belowground growth habits were compared with their leaf windows covered with reflective tape and with windows unobstructed. Results showed no increase in photoinhibition for these species resulting from the radiant energy penetrating the window tissue. Although the efficiency of the photosynthetic mechanism was not significantly influenced by the additional radiant energy provided by the window for individual species, there were significant differences in the efficiencies of radiant energy capture (Fv'/Fm') and utilization (P) between the two growth habits. Species with an aboveground growth habit were less efficient in radiant energy utilization compared with the species having a belowground growth habit. and K. J. Egbert, C. E. Martin.
Low temperature (LT) is one of the major factors that limit crop production and reduce yield. To better understand the cold-tolerance mechanism in the plantains, a sensitive cultivar Williams (Musa acuminata AAA cv. Williams) and a tolerant cultivar Cachaco (Musa paradisiaca ABB cv. Dajiao) were used. LT resulted in increased malondialdehyde (MDA) content, elevated contents of hydrogen peroxide (H2O2) and superoxide radical (O2.-), and decreased photochemical efficiency (Fv/Fm) and net photosynthetic rate (PN), but cv. Cachaco showed better LT tolerance than cv. Williams. After LT treatment for 120 h, total scavenging capability (DPPH. scavenging capability) in Williams showed a significant decrease but no significant alternations was found in Cachaco. Ascorbate peroxidase (APX) and peroxidase (POD) displayed a significant increase but superoxide dismutase (SOD) showed no significant alternations and catalase (CAT) showed a significant decrease in Cachaco after 120 h of LT treatment. All the four antioxidant enzymes above showed a significant decrease in Williams after 120 h of LT treatment. Our results suggest that higher activities of APX, POD, SOD, and DPPH. scavenging capability to a certain extent can be used to explain the higher cold tolerance in the plantain, which would provide a theoretical guidance for bananas production and screening cold-resistant variety. and Q. Brain imaging studies suggest localization of verbal working memory in the left dorsolateral prefrontal cortex (DLPFC) while face processing and memory is localized in the inferior temporal cortex and other brain areas. The goal of this study was to assess the effect of left DLPFC low-frequency repetitive transcranial magnetic stimulation (rTMS) on
verbal recall and face recognition. The study revealed a significant decrease of free recall in word encoding under rTMS (110 % of motor threshold, 0.9 Hz) in comparison with sham stimulation (p=0.03), while no significant difference was found with facial memory tests. Our findings support the essential role of the left DLPFC in word but not facial memory and confirm the content specific arrangement of cortical areas involved in semantic memory. As a non-invasive tool, rTMS is useful for cognitive brain mapping and the functional localization of the category specific memory system.
