Transforming growth factor beta 1 (TGF-β1) is a pro-fibrotic cytokine with a key role in wound repair and regeneration, including induction of fibroblast-to-myofibroblast transition. Genistein is a naturally occurring selective estrogen receptor modulator with promising anti-fibrotic properties. In the present study we aimed to investigate whether genistein modulates TGF-β1 (canonical and non-canonical) signaling in normal dermal fibroblasts at the protein level (Western blot and immunofluorescence). We demonstrated that TGF-β1 induces the myofibroblast-like phenotype in the studied fibroblast signaling via canonical (SMAD) and non-canonical (AKT, ERK1/2, ROCK) pathways. Genistein induced only ERK1/2 expression, whereas the combination of TGF-β1 and genistein attenuated the ERK1/2 and ROCK signaling. Of note, the other studied pathways remained almost unaffected. Species of Acanthamoeba Volkonsky, 1931 are the commonest among free-living amoebae that are widespread in different water resources but with lacking phylogenetic data. This study aims at detecting molecular prevalence and genetic diversity of Acanthamoeba isolates in Kafrelsheikh Governorate, Egypt. Forty-eight water samples were collected from 12 swimming pools; four samples during each season over one year. Samples were filtered, cultivated on non-nutrient agar plates and examined microscopically. Polymerase chain reaction (PCR) and sequence analysis of positive samples targeting diagnostic fragment 3 (DF3) of the small subunit rRNA gene were done. Cultivation succeeded to detect 14 (29%) positive samples while PCR missed three positive samples. The obtained sequences were phylogenetically analysed. The phylogenetic tree was constructed for them with sequences of reference species from the NCBI database. The identified species were Acanthamoeba castellanii Douglas, 1930 (T4), A. astronyxis (Ray et Hayes, 1954) (T9) and A. hatchetti Sawyer, Visvesvara et Harke, 1977 (T11). The prevalence of species of Acanthamoeba was higher during summer and fall. Therefore, the control of the presence of Acanthmoeba spp. in swimming pools needs immediate, effective and practical measures to prevent and control infection with species of Acanthamoeba.
The Tongshan intrusion is located at the Guichi ore cluster field along the Lower Yangtze River metallogenic belt (LYRB) in eastern China. Elemental data and major elements of biotite are reported for Late Mesozoic plutons in the Guichi ore cluster in the Middle-Lower Yangtze River Region, attempting to explain their diagenetic age, petrogenesis, and tectonic implications. The Tongshan intrusion is dated at 146.3±3.2 Ma, with zircon εHf(t) average value of -5.23, illustrating that the Tongshan intrusion could be derived from crust mantle mixing magmatite provenance. The two-stage Hf model ages between 1184~1422 Ma is indicated magma source relationship with those of the Mesoproterozoic basement in south Anhui. The Tongshan intrusion is high-K calc-alkaline, and exhibit adakite-like geochemical feature, such as high Al2O3, Sr contents, Sr/Y value, low Y and Yb contents. The geochemical and reexplained published isotopic data indicate that these adakites are most likely from partial melting of subducted oceanic crust, and mixed with enriched lithospheric mantle components in the rising process.
The Mariánské Lázně Fault (western Bohemia, CZE) is a morphologically, geologically and tectonically prominent structure with that is 150 km long with an NNW-SSE orientation. Its tectonic activity, especially in the NW part and in the neighbouring Cheb basin, is well known and has been proven by present-day earthquake swarms, mantle-derived CO2 emanations, geophysical and paleoseismological research. It seems that other parts of the MLF (especially segments of NNW-SSE and N-S orientation) might also have been active during the Pleistocene, possible even in the Holocene. This study provides a robust set of morphometric analyses – mountain-front sinuosity, basin asymmetry, longitudinal stream profiles, SL-index, which assesses the possibility of recent tectonic activity. The results suggest that the activity of the central and southern part of the MLF could have been very young. A reconstruction of the evolution of the stream network of the Mže River, as a result of different timing of the activity of particular segments of the MLF, is also put forward. The first ideas about the evolution of terrain morphology and the stream network are proposed by this study, however subsequent field research (geophysics, paleoseismology) could prove and date the tectonic activity. Archaeological evidence has demonstrated that the Bujang Valley is Malaysia's richest archaeological site and served as the primary coastal centre. A study in the Bujang Valley found monuments related to trading activities and others that functioned as a temple related to the Hindu-Buddhist period. The main purpose of this study was to resolve issues and problems arising from previous studies related to the Bujang Valley civilisation, particularly in terms of iron studies. Geophysics plays a vital role in assisting archaeologists to obtain excellent preliminary results before they proceed with excavation and digging works. Therefore, the 2-D resistivity and ground-penetrating radar (GPR) methods were conducted to locate and map the potential iron smelting site at Site B2 (SB2). Three main characteristics that can be observed on the surface are a mound area, exposed clay bricks and surface finds. Two-D resistivity showed the resistivity values of a possible buried structure, with values > 800 Ωm. Radargram profiles showed the highest amplitude, indicating the reflections uncovered in the location in certain survey lines. This paper presents the first summary of research on the metallurgical sites in the Bujang Valley, the most important site in Malaysia. Geophysical methods, which rely on a physical contrast between buried archaeological features and the properties of the surrounding subsoil, can assist archaeological investigations.
We present here our adventures in research in photosynthesis with George C. Papageorgiou (1933-2020) focusing on George's initiative in the discovery of the protective effects of glycine betaine on the oxygen-evolving photosystem II complex. We end with a brief description of research on glycine betaine-synthesizing transgenic cyanobacteria. Two of us, Norio Murata (in Japan) and Kostas Stamatakis (in Greece), and all our collaborators, have the highest respect for George, and we miss him and our intense discussions with him on various topics of photosynthesis research.
Stone columns consist of granular material compacted in long cylindrical holes. They are used for improving the strength and consolidation characteristics of compressible soils. However, they are still less effective at supporting heavy loads, since they still cannot transfer applied stresses to deeper layers of soil. The main objective of this numerical study was to investigate the geotechnical performance of a combined foundation system composed of stone columns and piles grouped together under a rigid raft foundation in compressible soil. The failure mechanism of this hybrid foundation system was examined, and configurations optimizing the performance of the combined foundation system were explored. An analytical model was developed for predicting the ultimate carrying capacity of the combined system in compressible soils. It was deduced that combining stone columns and piles in one foundation system improved considerably the system’s carrying capacity. Moreover, the uppermost improvement was observed when the piles were installed on the periphery or edge of the raft foundation, while stones columns were placed at the center area of the raft. The failure of the combined foundation system started from the center of the raft and noticeably extended to its edges. Due to the presence of stone columns in the combined foundation system, the piles did not interact. The areas affected or influenced by the soil–pile interaction also did not overlap.
