Tornádo zasáhlo území několika obcí na jižní Moravě ve večerních hodinách ve čtvrtek 24. června 2021. Ačkoliv se jednalo o relativně malé území, rozsahem škod šlo o mimořádnou událost nevídaných rozměrů. Velké množství lidí se ocitlo v přímém ohrožení života svého a svých blízkých. V psychologii nazýváme takovou událost jako potenciálně traumatizující a ze zkušenosti víme, že může mít závažné dopady na lidskou psychiku. Článek má za cíl představit, jak fungoval systém psychosociální krizové pomoci a spolupráce zajišťovaný ze strany Integrovaného záchranného systému ČR, který tvořil jádro psychosociální pomoci v akutní fázi katastrofy. Je uvedena návaznost na další organizace, které mají svou funkci především ve fázi stabilizace, ve střednědobé a dlouhodobé péči o zasažené osoby. Považujeme také za potřebné zopakovat principy psychosociální pomoci při mimořádných událostech. and The tornado hit the territory of several municipalities in South Moravia in the evening on Thursday, June 24, 2021. Although it was a relatively small area, the extent of the damage was extraordinary and of unprecedented proportions. Many people found themselves in direct danger of themselves and their loved ones. In psychology, we call such an event potentially traumatic, and we know from experience that it can have serious effects on the human psyche. The article aims to present how the system of psychosocial crisis intervention and psychological first aid provided by the Integrated Rescue System of the Czech Republic worked. This psychosocial assistance in the acute phase of a disaster builds the core of psychosocial help. There is a link to other organizations, which have their function mainly in the stabilization phase, in the medium and long-term care of the affected persons. We also consider it necessary to repeat the principles of psychosocial assistance in emergencies.
Due to emerging high spectral resolution, remote sensing techniques and ongoing developments to retrieve the spectrally resolved vegetation fluorescence spectrum from several scales, the light reactions of photosynthesis are receiving a boost of attention for the monitoring of the Earth's carbon balance. Sensor-retrieved vegetation fluorescence (from leaf, tower, airborne or satellite scale) originating from the excited antenna chlorophyll a molecule has become a new quantitative biophysical vegetation parameter retrievable from space using global imaging techniques. However, to retrieve the actual quantum efficiencies, and hence a true photosynthetic status of the observed vegetation, all signal distortions must be accounted for, and a high-precision true vegetation reflectance must be resolved. ESA's upcoming Fluorescence Explorer aims to deliver such novel products thanks to technological and instrumental advances, and by sophisticated approaches that will enable a deeper understanding of the mechanics of energy transfer underlying the photosynthetic process in plant canopies and ecosystems., S. Thioacetamide (TAA) is widely used in the production of drugs, pesticides and dyeing auxiliaries. Moreover, it is a chemical that can cause liver damage and cancer. TAA has recently been identified to cause bone damage in animal models. However, the type of bone damage that TAA causes and its potential pathogenic mechanisms remain unclear. The toxic effects of TAA on the femurs of New Zealand white rabbits and the underlying toxicity mechanism were investigated in this study. Serum samples, the heart, liver, kidney and femurs were collected from rabbits after intraperitoneal injection of TAA for 5 months (100 and 200 mg/kg). The New Zealand white rabbits treated with TAA showed significant weight loss and femoral shortening. The activities of total bilirubin, total bile acid and gamma-glutamyl transpeptidase in the serum were increased following treatment with TAA. In addition, the cortical bone became thinner, and the trabecular thickness decreased significantly in TAA-treated rabbits, which was accompanied by significantly decreased mineral density of the cortical and trabecular bone. Moreover, there was a significant decrease in modulus of elasticity and maximum load on bone stress in TAA-treated rabbits. The western blotting results showed that the expression of phosphorylated (p)-p38 and p-ERK in femur tissues of rabbits were increased after TAA administration. Collectively, these results suggested that TAA may lead to femoral damage in rabbits by activating the p38/ERK signaling pathway.
The aim of the study was to examine the potential impacts of bisphenol A (BPA) and its analogues BPB, BPF, and BPS on mice TM3 Leydig cells, with respect to basal cell viability parameters such as metabolic activity, cell membrane integrity, and lysosomal activity after 48-h exposure. In addition, monitoring of potential bisphenol´s actions included evaluation of ROS production and gap junctional intercellular communication (GJIC) complemented by determination of testosterone secretion. Obtained results revealed significant inhibition in mitochondrial activity started at 10 µg/ml of bisphenols after 48-h exposure. Cell membrane integrity was significantly decreased at 5 µg/ml of BPA and BPF and 10, 25, and 50 µg/ml of BPA and BPS. The lysosomal activity was significantly affected at 10, 25, and 50 µg/ml of applied bisphenols. A significant overproduction of ROS was recorded mainly at 5 and 10 µg/ml of tested compounds. In addition, significant inhibition of GJIC was observed at 5 µg/ml of BPB followed by a progressive decline at higher applied doses. In the case of testosterone production, a significant decline was confirmed at 10, 25 and 50 µg/ml.
The apicomplexan Toxoplasma gondii (Nicolle et Manceaux, 1908) secretes a group of serine/threonine kinases from rhoptries, which play vital roles in boosting intracellular infection. Toxoplasma gondii rhoptry organelle protein 17 (ROP17) is one of these important kinase proteins. Nevertheless, its function remains unclear. Here, we showed that ROP17 induced autophagy in vitro and in vivo. The autophagy of small intestine tissues of T. gondii tachyzoite (RH strain)-infected mice was detected by the immunohistochemistry staining of LC3B, Beclin 1 and P62. ROP17 overexpression augmented starvation-induced autophagy in HEK 293T cells as measured by MDC staining, transmission electron microscopy (TEM), fluorescence microscopy and Western blot analysis. Moreover, the interaction of ROP17 and Bcl-2 was confirmed using co-immunoprecipitation analysis, and the data demonstrated that ROP17 had an autophagic role dependent on the Beclin 1-Bcl-2 pathway, which was also revealed in an in vivo model through immunohistochemical staining. Pearson coefficient analysis showed that there existed strong positive correlations between the expression of ROP17 and LC3B, Beclin 1 and phosphorylation of Bcl-2, while strong negative correlations between the expression of ROP17 and p62 and Bcl-2. Collectively, our findings indicate that ROP17 plays a pivotal role in maintaining T. gondii proliferation in host cells via the promotion of autophagy-dependent survival.
Rice being the major food crop for more than half of the world population is severely affected by drought stress starting from the establishment of the seedling. We focused on the UV-B priming mediated transgenerational drought tolerance of a drought-tolerant rice variety (Vaisakh) towards polyethylene glycol (PEG) 6,000 (20%)-induced drought. Results showed that priming in F0 generation and re-priming in F1 generation with UV-B enhanced the PEG stress tolerance potential of rice seedlings with increased expression of genes encoding antioxidant enzymes and stress-related proteins offering better protection to primed plants. UV-B priming maintained oxidative homeostasis of the plant cell thus ensuring uninterrupted mitochondrial and photosynthetic activities. Cumulatively, our results suggest that the transgenerational priming memory retained in the seeds is transferred to offspring without any loss. Moreover, re-priming in F1 generation further boosted the innate tolerance potential of a tolerant variety resulting in stable cellular redox homeostasis.
High incidence of infertility along with low vitamin D levels was detected in otherwise healthy young men. The aim is to observe the effect of vitamin D supplementation on semen parameters as assessed by semen analysis in infertile men. In total, 45 men (mean age 36.6 years) in consecutive order were included, of whom 34 finished the study. Subjects were supplemented by vitamin D (cholecalciferol) 2500 IU/day. Vitamin D levels were assessed by HPLC. Semen analysis was performed strictly following 2010 WHO guidelines. Study periods were baseline and month 6. During follow-up, 20 %, 7.4 %, 22 % and 0.7 % increase in serum vitamin D levels, progressive sperm motility, sperm concentration and sperm morphology, respectively, were observed (all p<0.05). At follow-up end, 9 patients (26 %) reached normal sperm parameters of whom 2 fertilized their partner. There was no correlation between vitamin D and semen parameters observed. This study proves that vitamin D supplementation is possibly a modulator of sperm parameters in vitamin D deficient, otherwise healthy men. Rheumatoid arthritis (RA) and its animal model adjuvant arthritis (AA) are inflammatory diseases characterized by chronic inflammation, systemic oxidative stress and disturbed mitochondrial bioenergetics of skeletal muscle. The present study aimed to evaluate the effects of coenzyme Q10 - CoQ10 (100 mg/kg b.w.), omega-3-polyunsaturated fatty acids - ω-3-PUFA (400 mg/kg b.w.) and their combined treatment in AA on impaired skeletal muscle mitochondrial bioenergetics, inflammation and changes in levels CoQ9 and CoQ10 in plasma. Markers of inflammation (C-reactive protein, monocytechemotactic protein-1), antioxidant capacity of plasma, respiratory chain parameters of skeletal muscle mitochondria and concentrations of CoQ9 and CoQ10 in plasma and in muscle tissue were estimated. Treatment of the arthritic rats with CoQ10, ω-3-PUFA alone and in combination partially reduced markers of inflammation and increased antioxidant capacity of plasma, significantly increased concentrations of coenzyme Q in mitochondria and improved mitochondrial function in the skeletal muscle. Combined treatment has similar effect on the mitochondrial function as monotherapies; however, it has affected inflammation and antioxidant status more intensively than monotherapies. Interactions involving host plants (cowpea, groundnut, cotton, sunflower, greengram, blackgram) an insect herbivore, black legume aphid Aphis craccivora Koch (Hemiptera: Aphididae) and a predator, the zigzag beetle Cheilomenes sexmaculata (Fabricius) (Coleoptera: Coccinellidae) were investigated during 2014-2015. The preference of the aphid A. craccivora for host plants measured in terms of growth and multiplication was cowpea > groundnut > greengram > blackgram > sunflower > cotton (most preferred to least preferred). Cowpea was the most preferred host plant for growth and multiplication of A. craccivora. Aphid's oviposition period was longest, fecundity greatest; adult longevity longest (188.40 ± 28.87 h; 52.00 ± 10.92; 231.60 ± 40.41 h), and nymphal mortality was lowest (0%) when reared on cowpea followed by groundnut, greengram and blackgram. Aphids had highest nymphal mortality (100%), with very few or no live adults produced, when reared on sunflower and cotton, the least preferred host of A. craccivora in this study. At the third trophic level, both the larvae and the adults of the coccinellid, Cheilomenes sexmaculata, consumed more of the aphids reared on groundnut than of those reared on blackgram, greengram, cowpea, sunflower and cotton. The biochemical constituents (phenols) present in cotton and sunflower, which contributed to the aphid's nymphal mortality, also affected the feeding behaviour of the coccinellid., Snehasish Routray, Karnam V.
