- This paper summarises the results of parasitological examinations of the European eel Anguilla anguilla (Linnaeus) in the Czech Republic, carried out at the Institute of Parasitology, Czech Academy of Sciences (previously the Czechoslovak Academy of Sciences) within the period of 50 years (1958-2008). Even though this survey is limited to the Czech Republic, it provides extensive data probably incomparable with any other study anywhere regarding the number of eels examined and parasites found. A total of 723 eels was examined from 42 localities that belong to all of the three main river drainage systems in the country, i.e. the Elbe, Danube and Oder river basins. Of the 31 species of adult and larval macroparasites including Monogenea (4 species), Trematoda (3), Cestoda (3), Nematoda (11), Acanthocephala (5), Hirudinea (1), Bivalvia (1), Copepoda (1), Branchiura (1) and Acariformes (1), most of them (30) were recorded from the Elbe River basin. These parasites can be divided into three main groups regarding their host specificity: parasites specific for eels (26%), non-specific adult parasites occurring also in other fishes (61%) and non-specific larvae (13%). The highest number (19) of parasite species was recorded in the Mácha Lake fishpond system in northern Bohemia. The parasite communities in eels from the individual localities exhibited large differences in their species composition and diversity depending on local ecological conditions. The parasite fauna of A. anguilla in the Czech Republic is compared with that in other European countries. - Alien phytophagous insects are often introduced along with their host plants, creating opportunities for troublesome invasions. Yet, not all of them are able to successfully colonize novel host plants. In this study, we investigated host selection by the alien leaf miner Phyllonorycter leucographella (Zeller, 1850) on both its original host and novel host plants in the insect's alien range. We predicted that this insect's percentage infestation of the original host would be positively related to its specific leaf area (SLA), because high-SLA leaves are nutritious and have thin cuticles, traits related to high offspring developmental success. We further hypothesized that this host selection process would apply in the selection of novel host plants. Our results show that this leaf miner selects leaves of its original host plant, Pyracantha coccinea, according to their SLA values. The SLA value was also positively related to the probability of P. leucographella infesting and successfully developing on novel host plants. The selection of high-SLA plants by the moth leads to a high developmental success on novel host plants in the first (summer) generation, but it is likely to be maladaptive in the second (overwintering) generation, because in temperate Europe, high SLA values are associated with deciduous plants that shed their leaves in autumn. - The ant genus Cardiocondyla is characterized by wingless, ergatoid males, which in some species replace the typical winged male. Depending on species, ergatoid males engage in lethal fighting for access to sexual females, establish territories within their nests, or are mutually tolerant. Here we investigate, whether the morphology of ergatoid males and in particular the shape of their mandibles reflect phylogeny or male behaviour. In contrast to the worker mandible, which is relatively similar in shape in the ten species examined, mandibles of ergatoid males show a great variation in size and shape not only between the two clades of Cardiocondyla but also among species belonging to Cardiocondyla clade A. This diversity appears to reflect the diverse reproductive tactics of ergatoid males of different species, with extremely long, anteriorly oriented mandibles associated with indirect killing of adult competitors, while particularly short mandibles possibly constitute an adaptation to a tactic in which ergatoid males crush the cuticle of young rivals., Christine V. - The trade-off between flight capability and reproduction is well known in adult males of insects with wing dimorphism but the reproductive advantage of short-winged (SW) males over long-winged (LW) males appears to vary across insect taxa. In the present study, we determined the difference in the mating ability of SW and LW males of Velarifictorus ornatus (Orthoptera: Gryllidae) in order to evaluate whether the SW male morph has a reproductive advantage. We found that the choice of a mate depended on the female. Compared with LW males, SW males had an obvious mating advantage when both SW and LW males courted females simultaneously, and that dealation significantly enhanced the mating ability of LW males. Losing the ability to produce songs reduced the mating advantage of SW males, thereby indicating that the greater mating advantage of SW males was related to the attractiveness of the song. In addition, the difference in the mating ability of LW and SW males was not related to body size or age. - The influence of female mating status on ovarian development of the ladybird, Propylea dissecta (Mulsant) (Coleoptera: Coccinellidae), was investigated under laboratory conditions. We assessed the extent to which ovariole development was affected by mating and for that we initially created a base line by observing age specific ovariole development. Results show that the number of follicles in each ovariole increased with the age of both virgin and mated females up to the age of 3 days, thereafter, no increase in number of follicles was recorded. Ovariole width also increased with age in both virgin and mated females up to 4 days, thereafter, no increase in ovariole width was recorded. The ovariole width of mated females was significantly greater than that of virgin females. Egg maturation and the egg load started to increase at the age of 8 days in virgin females. Thereafter, it increased with increase in female age. While in mated females, immature eggs were recorded in their ovarioles from the age of 1 to 2 days. In mated females, however, the increase in the number of mature eggs per ovariole and egg load started when they were 3 days old. - The Asian ladybird Harmonia axyridis is an invasive insect in Europe and the Americas and is a great threat to the environment in invaded areas. The situation is exacerbated by the fact that non native species are resistant to many groups of parasites that attack native insects. However, very little is known about the complex microbial community associated with this insect. This study based on sequencing 16S rRNA genes in extracted metagenomic DNA is the first research on the bacterial flora associated with H. axyridis. Lady beetles were collected during hibernation from wind turbines in Poland. A mean ± SD of 114 ± 35 species of bacteria were identified. The dominant phyla of bacteria recorded associated with H. axyridis were Actinobacteria, Proteobacteria, Firmicutes and Bacteroidetes. Representatives of these phyla are common in the environment, e.g. in the soil, and are often identified as the dominant bacteria associated with arthropods. We also identified animal pathogenic bacteria, such as Burkholderia, Rhodococcus, Chlamydiae and Anaplasmataceae spp. (Neorickettsia helminthoeca and Ehrlichia ovina). We also identified Wolbachia pipientis in a single beetle. This bacterium is a causative agent of reproductive alterations in arthropods. These results support the enemy release hypothesis in the case of this ladybird invasion. Pathogenic bacteria were recorded in only a few samples. 219. Molecular characterisation of Cryptosporidium (Apicomplexa) in children and cattle in Romania

- To investigate the transmission of species of Cryptosporidium Tyzzer, 1907 in Timis County, Romania, 48 isolates of Cryptosporidium coccidia from 11 children, 29 calves and eight pigs were characterised by molecular analysis of two loci (SSU rRNA and 60-kDa glycoprotein gene). Overall, 22 isolates were amplified and sequence analyses revealed that all isolates were Cryptosporidium parvum Tyzzer, 1912. Two subtype families were identified, IIa and IId. Subtype IIdA22G1 (n = 4) was the single C. parvum subtype found in children. Subtypes found in calves included IIdA27G1 (n = 8), a novel subtype, IIdA25G1 (n = 5), IIdA22G1 (n = 2), IIdA21G1a (n = 1), and IIaA16G1R1 (n = 1). Subtype IIdA26G1 was found in a pig. These results were significantly different from previous Romanian reports, as the five subtypes of family IId identified in this study were never identified previously in this country. - Insect cellular immune reaction, which generally includes phagocytosis, encapsulation and nodule formation, is achieved by hemocytes circulating in insect haemolymph. The shift of hemocytes from the normal phase to the adhering phase is an important process in the cellular immune reaction, which includes the attachment of hemocytes to foreign surfaces or other hemocytes via adhesion factors. Neuroglian is one of the adhering factors associated with encapsulation in Manduca sexta and Drosophila melanogaster. Here we studied the localization of neuroglian (MsNrg) in Mythimna separata and its functional role in the cellular immune reaction. The distribution of MsNrg mRNA between hemocyte populations was determined using real-time quantitative reverse transcription PCR and in situ hybridization, which revealed that MsNrg was highly expressed in adhering hemocytes, especially in plasmatocytes. Unexpectedly, the transcript was observed as well in non-adhering hemocytes, implying neuroglian has a function in non-adhering hemocytes. Moreover, we showed that the amount of MsNrg mRNA was not changed by injections of either biotic or abiotic non-selves. Fewer latex beads were fully encapsulated by hemocytes in larvae treated with MsNrg double-stranded RNA than in control larvae, but their ability to achieve phagocytosis and nodule formation remained unchanged by the MsNrg knockdown.
