The ladybird beetle, Cheilomenes sexmaculata (Fabricius), has been extending its distribution in and around urban areas at higher latitudes in Japan over the past 100 years. Between 2003 and 2011, we investigated the seasonal occurrence, aphid prey and population dynamics of this species in an urban park in Osaka City, central Japan. We found that C. sexmaculata completes three generations a year in Osaka. Overwintered adults emerge in March or April and produce two (or rarely one) generations by summer. Second-generation adults aestivate and subsequently produce another generation in autumn. This species feeds on five species of aphids that infest planted shrubs and alien weeds throughout the vegetative season. We analyzed the association between accumulated temperature and when overwintered adults first emerged. In addition, we determined the association between their time of emergence, peak abundance and last occurrence, and meteorological conditions. There was no association between the sum of effective temperatures and termination of adult overwintering. There was, however, an association between first occurrence and peak abundance, and climatic conditions, such as warm temperatures or low humidity. This species maintained a stable population in an urban park by becoming quiescent when climatic conditions were unfavourable or prey was scarce. Macrolophus pygmaeus (Hemiptera: Miridae) is an important predator of pests of horticultural crops and here its ability as a predator of Acyrthosiphon pisum (Hemiptera: Aphididae) is addressed for the first time. The percentage predation of the different aphid instars and the number partially consumed were studied. Our results, obtained using choice and no-choice tests, revealed that M. pygmaeus caught and consumed more young than later instars of A. pisum, which confirms results of previous studies using other species of aphids. We also studied the interactions between predators (male/female) foraging in the same patch. When the prey/predator ratio is kept constant at 10 : 1 the average percentage of aphids completely consumed by individual females or males does not change with increase in the number of foraging predators. However, the number of partially consumed aphids decreased when females shared the same patch. In contrast, there was an increase in the number of aphids partially consumed when two males shared the same patch. Intracellular bacteria of the genus Wolbachia (α-Proteobacteria) are the most widespread endosymbionts of insects. Host infection is usually associated with alterations in reproduction, such as cytoplasmic incompatibility, the induction of parthenogenesis and offspring sex ratio bias: all phenomena that may influence host speciation. In the present study, by using well-established molecular tools, we investigated the presence of Wolbachia in leaf beetles of the genus Crioceris and their host plants, which are various species of Asparagus. Multilocus sequence typing of bacterial genes showed that despite their occurrence in the same habitat and feeding on the same plant, two species of Crioceris, C. quinquepunctata and C. quatuordecimpunctata, are infected by two different strains of Wolbachia. C. asparagi, C. paracenthesis and C. duodecimpunctata, which are sympatric with the infected species, do not harbour the bacterium. Interestingly, DNA of Wolbachia was detected in host plant tissues that are exploited by the beetles, providing evidence for the horizontal transmission of the bacterium between beetles and their host plants. Babesia caballi (Nuttal, 1910) is one of the causative agents of equine piroplasmosis which causes economic losses to horse industry in China. There is an urgent need for rapid detection method for B. caballi infection in Xinjiang Province, China. To prepare monoclonal antibodies (mAbs) against Bc48 gene of B. caballi (Xinjiang local strains) and establish colloidal gold-immunochromatographic (ICT) assay for diagnosis of the disease, recombinant Bc48 was expressed and purified from Escherichia coli. With purified Bc48 as immunogen in mice, three hybridoma cells named 11F4, 1H2 and 7F4 secreting mAbs against Bc48 of B. caballi were obtained, which showed strong reaction with recombinant Bc48 and Bc48 gene transfected cells. Furthermore, colloidal gold labelled ICT assay based on purified Bc48 recombinant antigen and its mAb was developed. The ICT assay showed high sensitivity and specificity and no cross-reaction with Theileria equi (Laveran, 1901). Total of 56 horse serum samples collected from Xinjiang were tested by ICT and compared with the detection by commercial ELISA kit. The results showed that 32 out of 56 serum samples were positive by ICT and 33 were positive by ELISA. Some free-living amoebae are a potential threat to human health. The best known species are those of the genus Acanthamoeba Volkonsky, 1931, which cause Acanthamoeba keratitis, granulomatous amoebic encephalitis and other forms of tissue inflammation. The aim of the present study was to search for potential pathogenic genotypes of free-living amoeba in the sand in children's playgrounds. Our results confirmed that free-living amoebae were present in all examined playgrounds. Sequences of the 18S rDNA have shown that all isolated potentially pathogenic strains of amoebae belong to genotype T4 of Acanthamoeba. The potential pathogenicity of isolates was confirmed on mice. Plastination is a preservation method for biological specimens, with important advantages over classic conservation techniques with formaldehyde or alcohol. Plastinated specimens are dry, odourless, and free of carcinogenic and toxic solutions. There are only few references about the plastination of parasites. Moreover, there is no information on the effect of plastination on the morphology and morphometry of these animals. The aim of this study was to define a plastination protocol to preserve various species of parasites, namely the nematodes Parascaris equorum (Goeze, 1782); Ascaris suum Goeze, 1782 and Dirofilaria immitis (Leidy, 1856); the acanthecephalan Macracanthorhynchus hirudinaceus (Pallas, 1781); the trematodes Fasciola hepatica Linnaeus, 1758 and Dicrocoelium dendriticum (Rudolphi, 1819) and the tapeworm Taenia sp. in the best morphological and morphometric conditions. Results showed that some individuals suffered collapse (P. equorum, A. suum, and D. dendriticum). However, other parasites presented good results with almost no change after plastination (D. immitis, M. hirudinaceus and F. hepatica). Bats of the family Phyllostomidae are common hosts to streblids known as bat flies. Here, we discuss the component community, prevalence and intensity of infection with species of Streblidae on an assemblage of phyllostomid bats in the Cafuringa Environmental Protection Area (APA Cafuringa) in the core area of the Cerrado in Central Brazil. A total of 1 841 streblid individuals of 24 species occurred on 752 bats of 14 species. Ten species of streblids infected Glossophaga soricina (Pallas), whereas seven or fewer streblid species infected the other bat species. Nine bat fly species presented a prevalence of more than 50%, whereas some differences in the abundance of bat flies among hosts were observed. Strebla wiedemanni Kolenati, 1856 and Trichobius furmani Wenzel, 1966 were more host-specific compared to the other streblids, and they occurred in greater abundance on their preferred hosts. Faecal samples from 162 wild animals were collected from 32 distinct sites of Łęczyńsko-Włodawskie Lakeland (eastern Poland). The presence of Giardia duodenalis (Stiles, 1902) was assessed by a Direct Fluorescence Assay (DFA) and by Polymerase Chain Reaction (PCR) and sequencing of a fragment of the beta-giardin gene. DFA showed the presence of cysts of G. duodenalis in 12 of 162 faecal samples (7%), namely in four wild boars (15%), four foxes (19%), two roe deer (4%), and two wolves (29%). PCR identified 34 of the 162 (21%) samples as positive, including 11 wild boars (41%), five red deer (18%), 11 roe deer (23%), four moose (17%), two wolves (29%) and a single sample from the European badger. Thus, PCR detected a significantly higher number of infection than DFA (P = 0.0005). However, 14 of 34 PCR products could not be sequenced because of their insufficient amount; the low number of cysts, poor conservation of the faeces or presence of PCR inhibitors may have contributed to weak DNA amplification. Sequence analysis of the remaining 20 products showed the presence of assemblage B in wild boars, red deer and roe deer, whereas samples from wolves were identified as assemblage D. This is the first detection of assemblage B in wild boars and deer. The present study was undertaken to identify potentially immunoreactive proteins of the muscle larvae (ML) and adult stage (Ad) of the nematode Trichinella spiralis Owen, 1835. To identify immunoreactive proteins that are specifically recognised by anti-Trichinella antibodies, ML and Ad crude extracts and their excretory-secretory (E-S) products were subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblot with serum samples from pigs experimentally infected with T. spiralis. A total of 18 bands were selected for final identification by liquid chromatography-tandem mass spectrometry. To further understand the functions of the proteins identified in this study, gene ontology terms were applied. Results showed that the specific antibodies against T. spiralis reacted with protein bands matching heat shock proteins, aminopeptidase, enolase, isocitrate dehydrogenase NADP-dependent, tropomyosin, P49 antigen, serine proteinase, secreted 5'-nucleotidase, antigen targeted by protective antibodies, 53 kDa E-S antigen, putative trypsin and paramyosin. Three proteins common for both adult stage and muscle larvae, including heat shock proteins, enolase and 5'-nucleotidase, might play important role during T. spiralis infection. These proteins are presumably presented to the host immune system and may induce humoral immune response. The effectiveness of natural enemies in controlling pests may be determined by many traits linked to their ability to regulate the density of their prey. In this respect, the phenomenon of pseudoparasitism, in which female parasitoids reject a host after inserting their ovipositor into it, is fairly common among hymenopteran parasitoids. However, in spite of this its effect on hosts is rarely reported in entomological and biological control literature. For this reason, in the present study, the pseudoparasitism by the parasitoid Spalangia cameroni Perkins of the Mediterranean pest Ceratitis capitata (Wiedemann) and its effect on several biological parameters of the host were studied under laboratory conditions. The results indicate that the percentage pseudoparasitism by S. cameroni of medfly in the laboratory is high, even slightly higher than host-feeding, which is commonly used to evaluate the potential of parasitoids as biological control agents. In addition, the adults that emerge from pseudoparasitized medfly pupae have a male-biased sex ratio, low levels of survival and are frequently damaged, which results in small adults and an inability to mate successfully.
