Amoebae were found to cause severe gill tissue damage in turbot, Scophthalmus maximus L. from a grow-out facility in northwestern Galicia (Spain). The nature and extent of lesions along with negative results of bacteriological and virological examination made this agent responsible for mortalities in four turbot stocks supplied with water from a single source. We present our findings, although we failed to isolate amoebae, since there was a clear evidence of their primary role in the development of disease condition and occurrence of mortalities. In addition, this is a record both of a new host endangered by amoebae in intensive cultures and pathogenesis of the gill lesions.
Ninety four aquarium fishes were screened for the presence of amoebae in their internal organs. Five specimens of Ca-rassius auratus (L.) and one specimen of Xiphophorus hetleri Heckel were positive. Of the three strains which were isolated from C. auratus, successfully cloned and cultivated, one was identified as Vannella platypodia (Gläser, 1912) Page, 1976 and two strains as Rosculus ithacus Hawes, 1963. Both species are reported for the first time from organs of fish. None of them could be identified with the amoeba-like agent of goldfish granulomas described here.
From November 1997 to June 1998, 3,118 specimens of Echinogammarus stammeri (Karaman, 1931) (Amphipoda) were collected from the River Brenta (Northern Italy) and examined for larval helminths. Larvae of Polymorphus minuius (Goeze, 1782) singly infected the hemocoel of 23 (0.74%) crustaceans; all these larvae were cystacanth stages. This is the first record of Polymorphus minuius in E. stammeri. Some cystacanths had their forebody and hindbody fully inverted. Parasites were bright orange in colour and each was surrounded by a thin acellular envelope. This envelope likely protects the developing parasite larva from cellular responses of the amphipod. Hemocytes were seen adherent to the outer surface of the envelope. The sex ratio among the parasitised E. stammeri was almost 1:1. All Polymorphus minutus larvae were central in the amphipod body, made intimate contact with host internal organs, and frequently induced a marked displacement of them. None of the infected females of E. stammeri. carried eggs or juveniles in their brood pouch. In five hosts, Polymorphus minuius co-occurrcd with the cystacanth of another acanlhocephalan, Pomphorhynchus laevis (Millier, 1776), a parasite offish.
The object of the research was to investigate the spectral properties of Rayleigh-type surface waves, generated by shot-hole explosions during seismic refraction experiments which were carried out in the area of the Bohemian Massif and West Carpathians. The records of displacement amplitudes were spectrally analyzed and prevailing frequency fp, relative Δfr and absolute widths of the spectra Δfa were chosen as essential parameters. Whilst the prevailing frequencies were recorded within the interval f ÷ 0.80 - 3.70 Hz at the site of the observations, situated on the territory of the Bohemian Massif, the respecti ve frequency range f ÷ 0.80 - 2.6 Hz was found in the West Carpathians. Some functional dependences of the spectral amplitude parameters on epicentral distance were observed and regularities of their decrease were defined. Three adult nematode specimens, all ovigerous females belonging to the family Cystidicolidae Skryabin, 1946, were found for the first time in the subcutaneous tissue around the eye of the captive porcupinefish Diodon nichthemerus Cuvier at a public aquarium in Osaka, Japan. Because no male was available, these could not be identified to the genus and species. CD163 is a marker of macrophages with anti-inflammatory properties and its soluble form (sCD163) is considered a prognostic predictor of several diseases including type 2 diabetes mellitus (T2DM). We explored sCD163 levels at baseline and after very low-calorie diet (VLCD) or bariatric surgery in 32 patients with obesity (20 undergoing VLCD and 12 bariatric surgery), 32 obese patients with T2DM (22 undergoing VLCD and 10 bariatric surgery), and 19 control subjects. We also assessed the changes of CD163 positive cells of monocyte-macrophage lineage in peripheral blood and subcutaneous adipose tissue (SAT) in subset of patients. Plasma sCD163 levels were increased in obese and T2DM subjects relative to control subjects (467.2±40.2 and 513.8±37.0 vs. 334.4±24.8 ng/ml, p=0.001) and decreased after both interventions. Obesity decreased percentage of CD163+CD14+ monocytes in peripheral blood compared to controls (78.9±1.48 vs. 86.2±1.31 %, p=0.003) and bariatric surgery decreased CD163+CD14+HLA-DR+ macrophages in SAT (19.4±2.32 vs. 11.3±0.90 %, p=0.004). Our data suggest that increased basal sCD163 levels are related to obesity and its metabolic complications. Classical biological control is an important means of managing the increasing threat of invasive plants. It constitutes the introduction of natural enemies from the native range of the target plant into the invaded area. This method may be the only cost-effective solution to control the rapidly expanding common ragweed, Ambrosia artemisiifolia, in non-crop habitats in Europe. Therefore, candidate biocontrol agents urgently need to be assessed for their suitability for ragweed control in Europe. A previous literature review prioritized the host-specific leaf beetle Ophraella slobodkini as a candidate agent for ragweed control in Europe, whereas it rejected its oligophagous congener O. communa. Meanwhile, O. communa was accidentally introduced and became established south of the European Alps, and we show here that it is expanding its European range. We then present a short version of the traditional pre-release risk-benefit assessment for these two candidate agents to facilitate fast decision-making about further research efforts. We selected two complementary tests that can be conducted relatively rapidly and inform about essential risks and benefits. We conducted a comparative no-choice juvenile performance assay using leaves of ragweed and sunflower, the most important non-target plant, in Petri dishes in climatic conditions similar to that in the current European range of O. communa. This informs on the fundamental host range and potential for increasing abundance on these host plants. The results confirm that O. slobodkini does not survive on, and is hence unlikely to cause severe damage to sunflower, while O. communa can survive but develops more slowly on sunflower than on ragweed. In parallel, our species distribution models predict no suitable area for the establishment of O. slobodkini in Europe, while O. communa is likely to expand its current range to include a maximum of 18% of the European ragweed distribution. Based on this early assessment, the prioritization and further assessment of O. slobodkini seem unwarranted whereas the results urgently advocate further risk-benefit analysis of O. communa. Having revealed that most of the European area colonized by ragweed is unlikely to be suitable for these species of Ophraella we suggest the use of such relatively short and cheap preliminary assessment to prioritise other candidate agents or strains for these areas., Suzanne T. Problems with parasitic infections and their interspecies transmissions are common in zoological gardens and could pose serious health damage to captive animals. This study presents results of eight-year monitoring of intestinal parasites in animals from Zoo Ljubljana, Slovenia. A total of 741 faecal samples from 40 animal species were collected two to four times per year and examined microscopically. Intestinal parasites were detected in 45% of samples, with detection of helminths (Cestoda, Nematoda - Ascaridida, Enoplida, Strongylida, Oxyurida, Rhabditida and Trichurida) and protists (Apicomplexa and Ciliophora) in 25% and 13% of samples, respectively; mixed infection was found in 7% of samples. The mostly infected were ungulates (61%), followed by reptiles (44%), ratites (29%), primates (22%) and carnivores (7%). During the observation period, the number of infected animal species increased from 8 to 25. This is the first long-term monitoring study of intestinal parasites in zoo animals from Slovenia. 5-hydroxytryptamine (5-HT) is involved in the stress-induced alteration of colonic functions, specifically motility and secretion, but its precise mechanisms of regulation remain unclear. In the present study, we have investigated the effects of 5-HT on rat colonic mucosal secretion after acute water immersion restraint stress, as well as the underlying mechanism of this phenomenon, using short circuit current recording (ISC), real-time polymerase chain reaction, Western blot analysis, and enzyme-linked immunosorbance assays. After 2 h of water immersion restraint stress, the baseline ISC and 5-HT-induced ISC responses of the colonic mucosa were significantly increased. Pretreatment with selective 5-HT4 receptor antagonist, SB204070, inhibited the 5-HT-induced colonic ISC response by 96 % in normal rats and 91.2 % in acute-stress rats. However, pretreatment with the selective antagonist of 5-HT3 receptor, MDL72222 or Y-25130, had no obvious effect on 5-HT-induced ISC responses under either set of conditions. Total protein expression of both the mucosal 5-HT3 receptors and the 5-HT4 receptors underwent no significant changes following acute stress. Both colonic basal cAMP levels and foskolin-induced ISC responses were significantly enhanced in acute stress rats. 5-HT significantly enhanced the intracellular cAMP level via 5-HT4 receptors in the colonic mucosa from both control and stressed animals, and 5-HT-induced cAMP increase in stressed rats was not more than that in control rats. Taken together, the present results indicate that acute water immersion restraint stress enhances colonic secretory responses to 5-HT in rats, a process in which increased cellular cAMP accumulation is involved., Y.
