The moths belong to the best-studied groups of insects in the Podyjí NP. In total, 2250 species of butterflies and moths (Lepidoptera) have hitherto been recorded here. Many species are closely associated with rocky steppe and heeathland, the most valuable natural habitats in this terriotory. Concurrently, the Podyjí NP is located on the western distribution border of many eastern and southern species. The Podyjí NP has very diverse vegetation which results in the occurrence of rich butterfly communities. To date, 142 species of butterflies have been recordeed in this area, of which 19 species (13 %) are considered regionally extinct and three species (2 %) are only migrants, not native to the Czech fauna. The main cause of the decline in butterflies is the abandonment of traditional techniques of habitat management. The Northern pine processionary moth, Thaumetopoea pinivora (Treitschke, 1834) shows a highly scattered distribution with fragmented populations across Europe. A previous study exploring the postglacial history of T. pinivora defined it as a cold-tolerant relict species and concluded that a progressive reduction of suitable habitats after the postglacial expansion from refugia in the southern Iberian peninsula best explained the distribution and genetic structure of populations of this species. However, recent records, both by us and others, challenge this view. Surprisingly, some of the newly found populations from southern Spain use black pine, Pinus nigra J.F. Arnold as a host plant despite the fact that the typical host of the species, Scots pine, Pinus sylvestris L. occurs in the area. We provide genetic data for one of these recently found southern populations where the larvae feed on P. nigra, and compare this with previously published data on individuals collected on P. sylvestris. This data reveals that populations from different host trees are no more genetically differentiated than populations sharing the same host plant. The findings of a wider diet breadth open the way to widen the search for the still unidentified glacial refugium of T. pinivora, and as such may contribute to a better understanding about how the species has spread across Europe., José A. Diapause is a common dormancy strategy exhibited by many species of invertebrates and insects to temporarily avoid seasonally recurring unfavourable conditions for their development, most usually in winter. Less frequently, a prolonged diapause lasting two or more years is described in species living in unpredictable environments where it is adaptive, but with significant costs. In this paper we examine the occurrence of prolonged diapause in the lycaenid butterfly Tomares ballus. Pupae of this species undergo an obligate diapause from mid-May to late January the following year. However, during our rearing experiments (from 2009 to 2016) the emergence of adults occurred sequentially and a fraction of the pupae remained in diapause for up to seven years. The annual percentage emergence after the first year of diapause was 45.6%, and only barely exceeded 50.0% in 2015. Remarkably, 12 pupae (11.4% of the initial brood) remained in diapause in their eighth year. The negative exponential equation fitted to the emergence data suggests that further emergences may occur within the next five years. Therefore, the potential for successful prolonged diapause of T. ballus pupae may be more than 10 years. The adaptive value of this strategy is discussed in relation to the effects of adverse and unpredictable weather during the flight period of the butterfly, intra-guild competition, parasitoids and changes in habitat quality. One of the best documented effects of climate change on biodiversity are shifts in phenology. However, long-term data quantifying and projecting the expected changes in phenology associated with climate warming are limited to a few well-recorded areas in the world. In the absence of temporal recording, an alternative approach is to determine the phenological response of species along marked gradients in climate or along latitudinal or altitudinal transects (space-for-time substitution). We studied the phenology (timing and duration of the flight period) of butterflies in 2006 along an altitudinal gradient (900-1680 m; estimated temperature lapse rate = -6.6°C/km) in the Serranía de Cuenca (central Spain) at the assemblage and individual species levels. Timing of the flight period was later for assemblages at high than at low altitudes. A similar trend of an increasing delay in the flight period with altitude was recorded for some individual species. However, there were also some exceptions to this pattern regardless of the number of sites and the altitudinal ranges of the species, suggesting possible local adaptation to regional climate. The duration of the flight period was shorter at high altitudes for assemblages, but this trend was not mirrored in the response of individual species. The results partly support substituting space-for-time when assessing the potential effect of climate change on phenophases such as the timing of the flight period, but we recommend extreme caution in extrapolating the results in the absence of information on how the responses of populations differ. and Juan Ignacio De Arce Crespo, David Gutiérrez.
Light trapping is the most widely used tool for determining the diversity of nocturnal Lepidoptera, but UV LEDs have yet to be used as light sources for the large-scale monitoring of Lepidoptera. We assessed the efficiency of this novel light source for sampling moths using a Heath type moth equipped with a strip of 150 high brightness UV LEDs (emission peak 398 nm, ~ 15 W) powered by a 12 V battery. We compared the number of individuals, the number of species and the Geometridae / Noctuidae ratio recorded for the samples collected using UV LED traps with those collected in two monitoring programs carried out in the same geographic region using two different light sources: a 200 W incandescent lamp (Rothamsted trap) and a 160 W mercury vapour lamp (manual catch). The total catch consisted of 61,120 individuals belonging to 699 species. The species richness rarefaction curves revealed that the Rothamsted trap collected fewer species and individuals than UV LED traps. Furthermore, the median numbers of species and individuals caught by UV LED traps fell within the range of those caught by mercury vapour lamp traps. In addition, the community composition recorded using incandescent lamps and UV LEDs was similar. The data obtained using UV LED traps, in absolute terms and in comparison with the other light sources and different sampling methods, clearly reveal that this light source is suitable for sampling macro-moth communities. Duponchelia fovealis Zeller is a polyphagous insect that has been recently reported attacking strawberry plants (Fragaria x ananassa Duchesne). Despite its economic importance there are few studies on this pest because it is difficult to rear it in the laboratory. With a constant supply of insects, studies on alternative methods of pest control can be conducted. This study aimed at developing an artificial diet for rearing D. fovealis with biological characteristics similar to those reared on their natural diet. This study was carried out in a climate-controlled room (25°C ± 2°C, RH 70% ± 10%, and 14L : 10D). The natural diet consisted of 'San Andreas' strawberry leaves (D1), while the artificial diet (D2) was developed in which beans, casein, soy protein, yeast and wheat germ are used as sources of protein. Five instars were identified. D. fovealis completed its life cycle in 38 and 40 days when fed D1 and D2, respectively. Survival was highest for the larvae fed the artificial diet. Females fed D1 lay a mean of 300.2 ± 62.3 eggs, while those fed D2, 220.3 ± 41.8 eggs. The artificial diet is suitable for the continuous rearing of D. fovealis in the laboratory., Maria A. C. Geographic isolation, altitude, climate, landscape and habitat are significant predictors of butterfly diversity in mountain ecosystems. Their diversity and its dependence on altitude, aspect (compass bearing) and biogeographic characteristics of the butterflies were surveyed on the karst mountain Biokovo in southern Croatia. The results affirm that there is a high diversity of butterflies in the study area and the species composition and biogeographic elements are more dependent on altitude than aspect of the mountain. The term cellular immune response refers to haemocyte-mediated responses, including phagocytosis, nodulation, and encapsulation. In the present study, we identified five types of circulating haemocytes in larvae of the haemolymph of the Asian corn borer, Ostrinia furnacalis (Guenée), including granulocytes, oenocytoids, plasmatocytes, prohaemocytes, and spherulocytes. The relative number of total free haemocytes per larva decreased significantly 0.5, 24, and 36 h after the injection of Beauveria bassiana conidia. Upon conidia challenge, both phagocytosis and nodulation were observed in the collected haemolymph from O. furnacalis larvae. In addition, plasma was found to be necessary for both phagocytosis and nodulation. Serpin is a broadly distributed superfamily of proteins that have a crucial role in regulating various immune reactions. Herein we identified a serpin-10 gene from Antheraea pernyi that encodes a 1557 amino acid residue protein with a predicted molecular weight of 58.76 kDa. Recombinant Apserpin-10 protein was expressed in a prokaryotic expression system (Escherichia coli) and the purified protein was used to prepare rabbit anti-Apserpin-10 polyclonal antibodies. Quantitative real-time polymerase chain reaction and western blot analysis indicate that Apserpin-10 was transcribed in all the tissues examined, including haemolymph, malpighian tubules, fat body, silk gland, integument and mid gut; the greatest expression level of Apserpin-10 was recorded in the fat body and haemocytes. The comparison of different developmental stages showed that Apserpin-10 transcript level was highest in 5th instar larvae, while the lowest expression was recorded at the egg stage. We also investigated the expression patterns of Apserpin-10 in fat body and haemocyte samples, following administration of heat-inactivated gram-positive bacteria (Micrococcus luteus), gram negative bacteria (Escherichia coli), a fungus (Beauveria bassiana) and virus (nuclear polyhedrosis virus, NPV). A substantial up-regulation of Apserpin-10 expression was recorded following pathogen challenge in both the tissues tested. Further the knock down of Apserpin-10 led to down regulation of antimicrobial peptide genes.
