a1_Understanding of the diversity of species of Cryptosporidium Tyzzer, 1910 in tortoises remains incomplete due to the limited number of studies on these hosts. The aim of the present study was to characterise the genetic diversity and biology of cryptosporidia in tortoises of the family Testudinidae Batsch. Faecal samples were individually collected immediately after defecation and were screened for presence of cryptosporidia by microscopy using aniline-carbol-methyl violet staining, and by PCR amplification and sequence analysis targeting the small subunit rRNA (SSU), Cryptosporidium oocyst wall protein (COWP) and actin genes. Out of 387 faecal samples from 16 tortoise species belonging to 11 genera, 10 and 46 were positive for cryptosporidia by microscopy and PCR, respectively. All samples positive by microscopy were also PCR positive. Sequence analysis of amplified genes revealed the presence of the Cryptosporidium tortoise genotype I (n = 22), C. ducismarci Traversa, 2010 (n = 23) and tortoise genotype III (n = 1). Phylogenetic analyses of SSU, COWP and actin gene sequences revealed that Cryptosporidium tortoise genotype I and C. ducismarci are genetically distinct from previously described species of Cryptosporidium. Oocysts of Cryptosporidium tortoise genotype I, measuring 5.8-6.9 µm × 5.3-6.5 µm, are morphologically distinguishable from C. ducismarci, measuring 4.4-5.4 µm × 4.3-5.3 µm. Third-stage larvae of the nematode Serpinema trispinosum (Leidy, 1852) were collected from the midgut of four of five species of adult damselflies (Zygoptera) from a non-irrigated restored semipermanent wetland located in Stillwater, Oklahoma, USA. Of the four infected damselfly species, prevalence and mean abundance was highest for the southern spreadwing, Lestes disjunctus australis Walker (10%, 0.2 ± 0.8) and lowest for the familiar bluet, Enallagma civile (Hagen) (2.5%, 0.04 ± 0.3); whereas mean intensities were lowest for the citrine forktail, Ischnura hastata (Say) (1.5 ± 0.5) and the eastern forktail, Ischnura verticalis (Say) (1.0 ± 0). This is the first record of larvae of S. trispinosum from damselflies. Serpinema trispinosum adults have been reported from 18 species of North and Central American freshwater turtles, whereas microcrustaceans such as copepods serve as intermediate hosts and snails, fish and amphibians serve as paratenic hosts in this nematode's life cycle. However, dietary studies of the 18 species of freshwater turtles reported as definitive hosts for S. trispinosum indicate that aquatic insects including damselflies are more commonly reported in turtle diets than are fish or amphibians. Additionally, unlike snails and amphibians, larval damselflies predominantly feed on microcrustaceans, and our observation of S. trispinosum infecting damselflies may reflect the importance of these insects as paratenic hosts. The lung-dwelling nematode Rhabdias engelbrechti sp. n. was found in five of eight examined banded rubber frogs in Limpopo Province, South Africa. The species is differentiated from species of Rhabdias Stiles et Hassall, 1905 occurring in the Afrotropical Realm based on the presence of a globular cuticular inflation at the anterior end, the buccal capsule walls being distinctly divided into anterior and posterior parts, the buccal capsule size (6-9 μm × 16-18 μm), and the body length (3.8-6.1 mm). Rhabdias engelbrechti is the tenth species of the genus found in Afrotropical anurans. Our molecular phylogenetic analysis based on the complete sequences of the ITS region and partial sequences of large subunit (28S) gene of the nuclear ribosomal RNA demonstrates that the new species is more closely related to the Eurasian species Rhabdias bufonis (Schrank, 1788) than to two other species from sub-Saharan Africa represented in the tree. Two exogenous NO donors were used to act as substitutes for impaired endogenous nitric oxide (NO) production due to inhibition of NO synthase in rats. Six weeks' lasting inhibition of NO synthase by NG-nitro-L-arginine methyl ester (L-NAME) induced stabilized hypertension. Simultaneously administered isosorbide-5-mononitrate did not prevent the development of hypertension. Molsidomine, administered concomitantly with L-NAME, significantly attenuated the BP increase. However, BP was still found to be moderately increased compared to the initial values. Remarkable alterations in the geometry of the aorta, carotid and coronary artery found in NO-deficient hypertension were prevented in rats administered L-NAME plus molsidomine at the same time. In spite of 6 weeks' lasting inhibition of NOS, the NOS activators acetylcholine and bradykinin induced BP decrease; the maximum hypotensive value did not differ from the values recorded in the controls or in animals treated with L-NAME plus molsidomine. Notably enough, the hypotension was similar to that found in rats administered L-NAME alone for six weeks. Faecal samples from the rock hyrax (Procavia capensis jayakari Thomas) were collected from the Ibex Reserve in central Saudi Arabia. Eimerian oocysts, which are believed to represent a new species described here as Eimeria tamimi sp. n., were detected in 40 out of 93 samples. Oocysts were fully sporulated in 24-48 hours at 25 ± 2 °C. Sporulated oocysts of E. tamimi sp. n. were ovoid, measuring 35-42 × 19-25 μm (39 × 23 μm), a length/width ratio 1.5-2 (1.7). Oocyst wall was bilayered and measured 1.5 μm in thickness. Micropyle, oocyst residuum and polar granules were not present. Sporocysts are elongate, measuring 12-18 × 9-12 μm (15 × 10 μm), with a length/width ratio 1.1-1.8 (1.5) prominent Stieda bodies and sporocyst residuum. Experimental infection of two clinically healthy rock hyraxes with sporulated oocysts of E. tamimi sp. n. resulted in shedding unsporulated oocysts 5-10 days post infection. Partial sequences of 18S ribosomal RNA (18S rDNA) and cytochrome C oxidase subunit 1 (COI) regions were amplified using the polymerase chain reaction (PCR) and sequenced. Phylogenetic analysis based on 18S rDNA using maximum likelihood (ML) and Bayesian inference (BI) methods revealed that E. tamimi sp. n. grouped with Eimeria quokka Barker, O'Callaghan et Beveridge, 1988, E. mundayi Barker, O´Callaghan et Beveridge, 1988, E. potoroi Barker, O'Callaghan et Beveridge, 1988 and E. gaimardi Barker, O'Callaghan et Beveridge, 1988 marsupials. The rat strain transgenic for the murine Ren-2 renin gene (TGR) is defined as a monogenic model of angiotensin II-dependent hypertension with endogenous activation of the renin-angiotensin system. Homozygous males TGR develop malignant hypertension with a strong salt-sensitive component. These animals show severe hypertension, proteinuria and high mortality. Morphological changes of renal parenchyma correspond to chronic ischemic glomerular changes. Heterozygous TGR develop only mild hypertension and thus provide a more suitable model of hypertension regarding to clinic al studies. Within the renal parenchyma, secondary focal segmental glomerulosclerosis (FSGS) predominates. High-salt diet in heterozygous animals induces transition from benign to malignant phase of hypertension. In this case, ischemic glomerular changes are superimposed on preexisting secondary FSGS. In the regression model of hypertension (late-onset treatment) the effect of salt intake is attenuated. In homozygous TGR, early selective ET A receptor blockade decreased blood pressure and ameliorated end-organ damage. Late selective ET A receptor blockade reduced podocyte injury despite final severe hypertension. Survival rate was markedly improved in both regimens with ETA selective blockade, while there was only partial improvement with early non-selective blockade. Both bosentan and atrasentan decreased ET-1 levels in both regimens. In heterozygous TGR, early and late ETA treatment substantially while ETA/ETB treatment partially improved survival rate. Significant effect on BP was found with early and late ETA blockade, while ETA/ETB blockade had no effect. Bosentan and at rasentan similarly decreased ET-1 levels on both regimens. In conclusion, selective ETA receptor blockade is superior to nonselective ETA/ETB receptor blockade in attenuating hypertension and end-organ damage. The response of some photosynthetic parameters (CO2 assimilation, transpiration rate, stomatal conductance, intercellular CO2 concentration, water-use efficiency, and chlorophyll content), shoot development, and the morphological features of the root system to differentiated conditions of nitrogen supply was tested in festulolium (Festulolium braunii K. Lack of nitrogen resulted in formation of longer and finer roots and lowered chlorophyll content, CO₂ assimilation, and water-use efficiency, resulting in lower dry matter accumulation. Application of both dosages of nitrogen resulted in improved aboveground features, while root features were enhanced without nitrogen fertilization. A parasitic nematode from the stomach of Japanese eel Anguilla japonica Temminck et Schlegel in western Japan, previously identified as Heliconema longissimum (Ortlepp, 1922), was morphologically re-examined and compared with the previous descriptions. In addition, the third-stage larva of this nematode is described, based on the specimens of encapsuled larvae found in musculature of two crabs, Hemigrapsus sp. and Perisesarma bidens (De Haan), caught from the upper-intertidal zone of the same locality. As a result of the morphological observation, seven pairs of postcloacal papillae in adult males are confirmed. This matches with the character of H. longissimum, but the shape of the fifth postcloacal papillae differs between the present material and H. longissimum; the former possesses pedunculate papillae in the fifth pair whereas the latter has sessile papillae. Since the pedunculate papillae can be found in the original description and the syntype specimens of H. anguillae Yamaguti, 1935 that has been synonymised with H. longissimum, we thus here resurrect H. anguillae as an accepted species. Parasitological examinations of 102 specimens of the lesser great cormorant Phalacrocorax carbo sinensis (Blumenbach) from two nesting regions in the Czech Republic (South Bohemia and South Moravia) were carried out at the Institute of Parasitology, Czech Academy of Sciences (previously the Czechoslovak Academy of Sciences) in the years 1987-1992. In them, a total of 19 species of helminth parasites was found, including Trematoda (11 species), Cestoda (2), Nematoda (4) and Acanthocephala (2), which can be divided into three main groups regarding their host specificity: parasites specific for cormorants (Phalacrocorax spp.) (37%), those parasitic mainly in cormorants (16%) and non-specific parasites (47%). Of the 19 species recorded, 100% were found in South Moravia, but only 47% of these 19 species in South Bohemia. The higher number of helminth species in cormorants from South Moravia and a higher proportion of non-specific species may be associated with the presence of the large Nové Mlýny water reservoir, in addition to better ecological and environmental conditions in this warmer region. Morphometric data from spores of ten myxosporean species were statistically analysed to explore myxosporean intraspecific variation in measurements when obtained from a sample from: (1) the same plasmodium, (2) different plasmodia from the same host and (3) different host individuals and localities. In some cases, significant differences in spore dimensions were found between samples from the same plasmodium, highlighting the difficulty of obtaining representative measurements of myxosporean spore. In addition, significant differences in spore dimensions were found when plasmodia from the same site of infection were compared, suggesting that measurements of spores should come from several different plasmodia of the sampling to increase the reliability of the morphology data. Moreover, significant differences in spore dimensions were observed for most spore dimensions when data were compared between localities. In all cases, there was clear overlap in ranges of dimensions even when means differed significantly. The present statistical analysis shows that intraspecific morphometric variation of myxosporean species commonly occurs, highlighting the importance of reporting ranges of measurements for a species, not just the mean dimensions, and taking into account all evidence when assigning or describing myxosporean species., Yanhua Zhai, Christopher M.
