Among the most extended ecosystems of the temperate zone, the seminatural, dry grasslands constitute a substantial proportion in the Carpathian Basin. The aim of our present study was to investigate the short-term effect of extensive fertilization on the species composition and CO2 exchange of loess grassland at community level. The in situ investigation of the latter parameter have not been yet carried out in Pannonian loess grasslands. Most of the parameters studied showed a considerable interannual variation both in the fertilized and in the control stands. As a result of the treatment, the average species number of the fertilized stand decreased by 22%, which was more significant in the autumn (26%) than in the spring. Diversity values, including Shannon index and species richness, increased by nearly 1.5 times in the year with adequate rainfall compared with the initial values. In general, species richness and the ratio of dicots decreased, while the ratio of therophytes, alien competitors, and C4 plants increased with the addition of fertilizers. Significant carbon sequestration potential was only detected during wet periods in the fertilized grass. The rate of CO2 uptake was found to be nearly five times higher in the fertilized stand and nearly three times higher in the control stand during the wet year compared with the previous, extremely dry year. The CO2 uptake potential of the fertilized grassland exceeded that of the control stand by 12% in the year with high rainfall, while the rate of CO2 exchange dropped by 50% in the dry year in the fertilized stand. Our study reinforced the idea that the decline in species richness was not necessarily followed by the reduction of stand level carbon uptake in a short period due to an insignificant change in ecophysiological functional groups. and S. Dust deposition on leaf surfaces can impact the growth and physiological traits of plants. We carried out a field experiment to investigate short-term effects of light surface dust on photosynthesis of cotton in the Tarim Basin using chlorophyll fluorescence and gas-exchange techniques. JIP-test analysis of OJIP curves showed that the total performance index for leaves without dust decreased by 32% at noon compared to the morning value. High irradiance at noon reduced actual quantum yield of PSII and increased nonphotochemical quenching for leaves without dust, showing photoinhibition. It suggested that light surface dust alleviated photoinhibition of cotton to high irradiance on a short-term basis. For the leaves without dust, high irradiance induced photoinhibition not only with respect to the photochemistry reactions but the biochemical pathways of CO2 fixation. The effect of three-day fasting on cardiac ischemic tolerance was investigated in adult male Wistar rats. Anesthetized open-chest animals (pentobarbitone 60 mg/kg, i.p.) were subjected to 20-min left anterior descending coronary artery occlusion and 3-h reperfusion for infarct size determination. Ventricular arrhythmias were monitored during ischemia and at the beginning (3 min) of reperfusion. Myocardial concentrations of beta-hydroxybutyrate and acetoacetate were measured to assess mitochondrial redox state. Short-term fasting limited the infarct size (48.5±3.3 % of the area at risk) compared to controls (74.3±2.2 %) and reduced the total number of premature ventricular complexes (12.5±5.8) compared to controls (194.9±21.9) as well as the duration of ventricular tachycardia (0.6±0.4 s vs. 18.8±2.5 s) occurring at early reperfusion. Additionally, fasting increased the concentration of beta-hydroxybutyrate and betahydroxybutyrate/acetoacetate ratio (87.8±27.0) compared to controls (7.9±1.7), reflecting altered mitochondrial redox state. It is concluded that three-day fasting effectively protected rat hearts against major endpoints of acute I/R injury. It was previously shown that 4 hours´ lasting inhibition of nitric oxide synthesis by administration of an L-arginine analogue, the NG-nitro-L-arginine methyl ester (L-NAME) changed the affinity of the Na-binding site of Na,K-ATPase thus resulting in elevation of enzyme activity especially at higher concentrations of sodium. Using the same experimental model, we focused our attention in the present study to the question of binding of ATP to the enzyme molecule in the left ventricle (LV), ventricular septum (S) and the right ventricle (RV) of the dog heart. Activation of the enzyme by increasing concentrations of ATP revealed a significant increase of the Vmax only in septum (by 38 %). The KM increased significantly in septum (by 40 %) and in left ventricle (by 56 %) indicating an altered sensitivity of the ATP-binding site of Na,K-ATPase in the hearts of NO-deficient animals. It is known that hypertension is accompanied by increased [Na+]i. The functional properties of Na,K-ATPase, which transports the Na+ out and K+ into myocardial cells during the relaxation phase, were investigated in the left ventricle (LV), septum (SV) and the right ventricle (RV) of anesthetized dogs with moderate acute blood pressure elevation elicited by short-term (4-hour) NO synthase inhibition. The NO-insufficiency was induced by administration of an L-arginine analogue, the NG-nitro-L-arginine methyl ester (L-NAME). Concerning the function of Na,K-ATPase under the conditions of lowered NO synthesis, we focused our attention to the binding of Na+ to the enzyme molecule. Activation of the enzyme by increasing Na+ concentrations revealed significant changes in both the maximal velocity (Vmax) and the affinity for Na+ (KNa) in all investigated heart sections. The Vmax increased by 27 % in LV, by 87 % in SV and by 58 % in RV. The KNa value increased by 86 % in LV, by 105 % in SV and by 93% in RV, indicating an apparent decrease in the sensitivity of the Na+-binding site in the Na,K-ATPase molecule. This apparently decreased pump affinity for Na+ together with the increase of Vmax suggest that, during the short-term inhibition of NO synthesis, the Na,K-ATPase is capable of extruding the excessive Na+ from the myocardial cells more effectively at higher [Na+]i as compared to the Na,K-ATPase of control animals., N. Adiponectin belongs to the group of biologically active substances secreted by adipocytes and referred to as adipokines. Disturbances in its secretion an d/or action are thought to be involved in the pathogenesis of some metabolic diseases. However, regulation of adiponectin secretion is poorly elucidated. In the present study, short-term regulation of adiponectin secretion in primary rat adipocytes was investigated. Isolated rat adipocytes were incubated in Krebs-Ringer buffer containing 5 mM glucose and insulin alone or in the combination with epinephrine, dibutyryl-cAMP, adenosine A 1 receptor antagonist (DPCPX), palmitate, 2-bromopalmitate or inhibitor of mitochondrial electron transport (rotenone). Adipocyte exposure for 2 h to insulin (1-100 nM) significantly increased secretion of adiponectin compared with secretion observed without insulin. Furthermore, secretion of adipon ectin from adipocytes incubated with glucose and insulin was reduced by 1 and 2 μ M epinephrine, but not by 0.25 and 0.5 μ M epinephrine. Under similar conditions, 1 and 2 mM dibutyryl-cAMP substantially diminished secretion of adiponectin, whereas 0.5 mM dibutyryl-cAMP was ineffective. Secretion of adiponectin was found to be effectively decreased by DPCPX. Moreover, ad ipocyte exposure to rotenone also resulted in a substantial diminution of secretory response of adipocytes incubated for 2 h with glucose and insulin. It was also demonstrated that palmitate an d 2-bromopalmitate (0.06-0.5 mM) failed to affect secretion of leptin. The obtained results indicated that in short-term regulation of adiponectin secretion, insulin and epinephrine exert the opposite effects. These effects appeared as early as after 2 h of exposure. Primary leaves of young plants of common bean (Phaseolus vulgaris cv. Carioca and Negro Huasteco) and cowpea (Vigna unguiculata Walp cv. Epace 10) were exposed to high irradiance (HI) of 2 000 µmol m-2 s-1 for 10, 20, and 30 min. The initial fluorescence (F0) was nearly constant in response to HI in each genotype except for Carioca. A distinct reduction of maximum fluorescence (Fm) was clearly observed in stressed genotypes of beans after 20 min followed by a slight recovery for the longer stress times. In common bean, the maximum quantum yield (Fv/Fm) was reduced slowly from 10 to 30 min of HI. In cowpea, only a slight reduction of Fv/Fm was observed at 20 min followed by recovery to normal values at 30 min. HI resulted in changes in the photochemical (qP) and non-photochemical (qN) quenching in both species, but to a different extent. In cowpea plants, more efficiency in the use of the absorbed energy under photoinhibitory conditions was related to increase in qP and decrease in qN. In addition, lipid peroxidation changed significantly in common bean genotypes with an evident increase after 20 min of HI. Hence the photosynthetic apparatus of cowpea was more tolerant to HI than that of common bean and the integrity of cowpea cell membranes was apparently maintained under HI. and L. C. S. Ferreira ... In addition to a number of deleterious effects on cellular integrity and functions, diabetic metabolic milieu has been implicated in a rapidly growing number of alterations in signal transduction. In this review we focus on Akt kinase physiology, its alterations in diabetes mellitus (DM), and on the emerging role of this signaling system in the pathophysiology of diabetic microvascular complications. Studies focusing on Akt in diabetes suggest both decrease and increase of Akt activity in DM. Alterations of Akt activity have been found in various tissues and cells in diabetes depending on experimental and clinical contexts. There is convincing evidence suggesting defective Akt signaling in the development of insulin resistance. Similar defects, as in insulin-sensitive tissues, have been reported in endothelia of DM Type 2 models, possibly contributing to the development of endothelial dysfunction under these conditions. In contrast, Akt activity is increased in some tissues and va
scular beds affected by complications in DM Type 1. Identification of the role of this phenomenon in DM-induced growth and hemodynamic alterations in affected vascular beds remains one of the major challenges for future research in this area. Future studies should include the evaluation of
therapeutical benefits of pharmacological modulators of Akt activity.
Open area rainfall and throughfall measurements in the Western Tatra Mountains (altitude about 1500 m a.s.l.) made by tipping bucket gauges were used to estimate the usefulness of the short-time data in analysis of spruce interception. The 10-minute data from period 13 May-13 October 2009 did not reveal meaningful correlations between the open area rainfall and throughfall. Aggregated measurements representing individual rainfall events were more useful. They showed linear relationship between open area rainfall and throughfall for events with total rainfall depth in the open area exceeding 5 mm. Correlation between open area rainfall and throughfall for rainfall events with duration above 120 minutes was significantly better than for the shorter ones. Mean values of interception (percentage of open area rainfall which did not appear in throughfall) of individual rainfall events was high. When we excluded events for which throughfall was higher than the open area rainfall, mean interception for larger and longer rainfall events was 46% and 48%, respectively. For smaller (runoff depth below 5 mm) and shorter events (duration below 2 hours) the mean interception was 70% and 72%, respectively. However, the data revealed very high variability of interception. and Príspevok sa zaoberá hodnotením užitočnosti krátkodobých meraní zrážok na otvorenej ploche a v smrekovom lese pri určovaní intercepcie. Vychádza z merania zrážok preklápacími zrážkomermi v Západných Tatrách v nadmorskej výške okolo 1500 m n.m. Desaťminútové údaje merané v období 13.5.- 13.10.2009 neposkytli použiteľné korelácie medzi dažďom na otvorenej ploche a v lese. Lepšie výsledky boli získané pre sumárne úhrny zrážok pre jednotlivé dažde. Ak úhrn dažďa na voľnej ploche prekročil približne 5 mm, veľkosť podkorunových zrážok rástla lineárne s veľkosťou zrážok na voľnej ploche. Korelácia medzi zrážkami na voľnej ploche a v lese pre dažde s dĺžkou trvania nad 120 minút bola podstatne lepšia, ako pre kratšie dažde. Priemerná hodnota intercepcie (vyjadrenej ako percento zrážok na voľnej ploche, ktoré sa neobjavilo v lese) pre jednotlivé zrážkové udalosti bola vysoká. Po vylúčení udalostí, pre ktoré bol v lese nameraný vyšší úhrn zrážok ako na voľnej ploche, bola priemerná intercepcia pre väčšie dažďe 46 % a pre dlhšie dažde 48 %. Pre menšie (úhrn pod 5 mm) a kratšie (trvanie pod 2 hodiny) dažde bola priemerná intercepcia 70 % a 72%. Hodnoty intercepcie pre jednotlivé udalosti však mali veľkú variabilitu.
