Growth, photosynthetic gas exchange, and chlorophyll fluorescence characteristics were investigated in wild type (WT) and Cd-sensitive mutant rice (Oryza sativa L.) plants using 50 µM Cd treatment for 12 d followed by a 3-d recovery. Under Cd stress, net dry mass and pigment contents were significantly lower in the mutant plants than in the WT. The mutant had lower net photosynthetic rate (P N), transpiration rate (E), and stomatal conductance (g s) than WT rice, however, it had higher intercellular CO2 concentration (C i), indicating that non-stomatal factors accounted for the inhibition of P N. Maximal photochemical efficiency of photosystem 2 (Fv/Fm), effective quantum yield of PS2 (ΦPS2), and photochemical quenching (qP) decreased much in the mutant under Cd stress. Cd content in roots and leaves of the mutant was significantly higher than those in the WT. Hence Cd toxicity was associated with the marked increases in Cd contents of plant tissue. Gas exchange, chlorophyll (Chl) fluorescence, and contents of photosynthetic pigments, soluble proteins (ribulose-1,5-bisphosphate carboxylase/oxygenase, RuBPCO), and antioxidant enzymes were characterized in the fully expanded 6th leaves in rice seedlings grown on either complete (CK) or on nitrogen-deficient nutrient (N-deficiency) solutions during a 20-chase period. Compared with the control plants, the lower photosynthetic capacity at saturation irradiance (Pmax) was accompanied by an increase in intercellular CO2 concentration (Ci), indicating that in N-deficient plants the decline in Pmax was not due to stomatal limitation but due to the reduced carboxylation efficiency. The fluorescence parameters ΦPS2, Fv'/Fm', electron transport rate (ETR), and qP showed the same tendency as Pmax in N-deficient plants. Correspondingly, a higher qN paralleled the rise of the ratio of carotenoid (Car) to Chl contents. However, Fv/Fm was still diminished, suggesting that photoinhibition did occur in the photosystem 2 (PS2) reaction centres. In addition, the activities of antioxidant enzymes on a fresh mass basis were gradually lowered, leading to the aggravation of membrane lipid peroxidation with the proceeding N-deficiency. The accumulation of malonyldialdehyde resulted in the lessening of Chl and soluble protein content. Analyses of regression showed PS2 excitation pressure (1 - qP) was linearly correlated with the content of Chl and inversely with soluble protein (particularly RuBPCO) content. There was a lag phase in the increase of PS2 excitation pressure compared to the decrease of RuBPCO content. Therefore, the increased excitation pressure under N-deficiency is probably the result of saturation of the electron transport chain due to the limitation of the use of reductants by the Calvin cycle. In Amaranthus tricolor the leaf structure included three layers of chlorenchyma on the vascular bundle periphery, namely, mesophyll cells (MSCs) with few chloroplasts, outer larger round bundle sheath cells (BSCs) with many chloroplasts in a centripetal position, and inner smaller BSCs with few chloroplasts around the vascular bundle cells. The ultra-thin sections showed that BSCs had abundant organelles, namely many large and round mitochondria with well-developed cristae in the cytoplasm. The chloroplasts in the BSCs were lens-like bodies, which seemed to be oval on cross sections. Granal and intergranal thylakoids were usually distinguished. Grana were stacked in parallel with prevailing plane of thylakoid lamellae. The chloroplasts in the MSCs appeared smaller than those in the BSCs and contained less stacked thylakoids but abundant peripheral reticulum. The ultra-thin sections of immunogold-labelled anti-ribulose-1,5-bisphosphate carboxylase/oxygenase (anti-RuBPCO) exhibited high density of RuBPCO labelling in the stroma region of chloroplasts of the BSCs. Some anti-RuBPCO immunogold particles were observed in the stromal region of MSCs chloroplasts. The anti-activase (A) immunogold-labelling indicated that RuBPCOA was mainly distributed in the stroma region of both BSCs and MSCs chloroplasts. From the chloroplast ultrastructure and localisation of RuBPCO and RuBPCOA we deduced that the photosynthetic carbon reduction cycle and the formation of assimilatory power function in both MSC and BSC chloroplasts of A. tricolor. and J. Tea tree (Melaleuca alternifolia) canopy was sprayed with low concentration of NaHSO3 or mixture of NaHSO3+ KH2PO4. The treatments significantly enhanced net photosynthetic rate (PN), carboxylation efficiency (CE), and the maximum response of PN to intercellular CO2 concentration. The enhancement of PN by foliar application of low concentrations of bisulfite was due to increasing CE relevant to ribulose-1,5-bisphosphate (RuBP) carboxylase/oxygenase activity and regeneration rate of RuBP depending on ATP formation. and W.-J. Miscanthus is one of the most promising bioenergy crops with high photosynthetic nitrogen-use efficiency (PNUE). It is unclear how nitrogen (N) influences the photosynthesis in Miscanthus. Among three Miscanthus genotypes, the net photosynthetic rate (PN) under the different light intensity and CO2 concentration was measured at three levels of N: 0, 100, and 200 kg ha-1. The concentrations of chlorophyll, soluble protein, phosphoenolpyruvate carboxylase (PEPC), ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) large subunit, leaf anatomy and carbon isotope discrimination (Δ) in the leaf were analyzed to probe the response of photosynthesis in Miscanthus genotypes to N levels. PN in all genotypes rose significantly as N application increased. The initial slope of response curves of PN to Ci was promoted by N application in all genotypes. Both stomatal conductance and Ci were increased with increased N supply, indicating that stomatal factors played an important role in increasing PN. At a given Ci, PN in all genotypes was enhanced by N, implying that nonstomatal factors might also play an important role in increasing PN. Miscanthus markedly regulated N investment into PEPC rather than the Rubisco large subunit under higher N conditions. Bundle sheath leakiness of CO2 was constant at about 0.35 for all N levels. To investigate how bisulfite promotes photosynthesis, a pot experiment was conducted with rice (Oryza sativa L.) plants to determine Rubisco activity and content, and Rubisco activase (RCA) gene expression after spraying NaHSO3 on rice leaves. The NaHSO3 treatment promoted significantly net photosynthetic rate (PN), carboxylation efficiency, maximum carboxylation rate, ribulose-1,5-bisphosphate regeneration rate, initial Rubisco activity, and RCA protein and mRNA concentrations. Therefore, the NaHSO3 enhancement of PN could be directly attributed to induction of RCA gene expression both at the transcription and translation levels.
