Greenhouse-grown susceptible 20-d-old seedlings of Theobroma cacao genotypes Catongo and tolerant genotype SCA6xCatongo were inoculated with a mixture of isolates of Crinipellis perniciosa, the causal agent of witches' broom. The characteristics of chlorophyll a fluorescence emission were monitored during leaf ontogeny using a portable system PAM-2000. In both inoculated and non-inoculated genotypes, significant differences were found for the effective quantum yield values of photosystem (PS) 2 (ΔF/Fm') at the B (7 to 14-d-old), D (21 to 30-d-old), and E (>30-d-old) stages of leaf development, and in quantum yield of the non-cyclic photosynthetic electron transport between PS2 and PS1 [qp(Fv/Fm)] and quencher efficiency [(Fm-Ft)/F0] at the B, C (15 to 20-d-old) and D stages. Intergenotypic differences were found only for the [qp(Fv/Fm)] and [(Fm-Ft)/F0] values at the E stage, and for fluorescence quenching (Fm-Ft) at the B and E stages. Highly significant inter- and intragenotype relationships were found between the rate of photosynthetic electron transport to PS2 (Amax) and maximum fluorescence during actinic irradiation (Fm'). Also, each of the highly significant relationships between (Fm-Ft) and Amax, [(Fm-Ft)/F0] and ΔF/Fm', and between [(Fm-Ft)/F0] and Amax were represented by a general model, independent of treatments. Therefore, alterations in energy distribution in the radiant energy collector complex interior of PS2 and reduction in absorption of photosynthetically active radiation were observed in the infected plants, mainly in the hybrid at the C stage. Species of the Theobroma genus are primarily known by their commercially valuable seeds, especially, T. cacao is one of the most important tropical perennial crops. Beside T. grandiflorum, T. bicolor, and T. angustifolium, T. cacao is the only species of the genus that has been better studied to obtain physiologically relevant information. The main objective of this work was to evaluate the leaf gas exchange in seedlings of seven species of the Theobroma genus, seeking to identify characteristics that could be used in T. cacao breeding programmes. The study was realized under greenhouse conditions using six-month-old seedlings, in which net photosynthetic rate (PN), stomatal conductance (gs), transpiration (E), as well as parameters derived from light curves (PN vs. photosynthetically active radiation) were evaluated. T. cacao, along with T. microcarpum, showed the lowest values of PN, gs, and E, while the highest values were presented by T. speciosum, which showed higher saturation irradiance and lower intrinsic and instantaneous water-use efficiencies, being considered the species less conservative in water use. Therefore, the parameters shown by the different evaluated species could serve to design T. cacao genotypes, through introgression of genes for specific environments such as the cabruca system widespread in southern Bahia, Brazil., A.-A. The recessive lethal character Luteus-Pa, expressed as a yellowing of leaves of young seedlings and followed by death approximately 60 d after emergence, presents a 3:1 segregation in crosses and/or selfpollinated plants. We evaluated quantitatively the fluorescence emission of chlorophyll (Chl), gas exchange, and chemical composition of normal and recessive homozygous cacao seedlings of the cross Pa 121×Pa 169. The characteristics of Chl fluorescence kinetics were studied in stages B2, B3, C, D, and E of leaf development, corresponding to plant ages of 9 to 12, 13 to 15, 16 to 20, 21 to 30, and >30 d, respectively. Gas exchanges were measured in mature leaves of both seedlings. In regular intervals of 3 d beginning at 33 d after emergence, the seedlings were separated into roots, stems, leaves, and cotyledons to determine the contents of saccharides (SAC) and free amino acids (FAA) and variation of the leaf Chl content. The Chl distribution in complexes of the photosynthetic apparatus was analysed by SDS-PAGE in mature leaves of both normal and recessive 32-d-old seedlings. There were variations in Chl fluorescence, gas exchanges and chemical composition of different parts of both types of seedlings. However, no significant differences were found in the Chl distribution through photosynthetic complexes of 32-d-old normal and recessive homozygous seedlings. After that period a decrease in the Chl concentration was observed in the recessive seedlings, and only minimum fluorescence (F0) was found. The F0 values were higher in the recessive seedlings than in the normal ones. The net photosynthetic rate of mature leaves was negative in agreement with low conductance, transpiration rate, and high internal CO2 concentration. These factors might have contributed to a depletion in SAC in different plant parts. Although F0 partially reflects the Chl concentration in leaf tissue, the increase in its value was probably due to a damage in reaction centres of photosystem 2. The Theobroma lethal character Luteus-Pa segregates in a 3:1 ratio, expresses in recessive homozygosis, initially inducing leaf chlorosis and finally provoking seedlings death. The objective of this work was to evaluate gas exchange, chlorophyll fluorescence emission, chemical composition and oxidative stress of wild and mutant seedlings resulting from the crosses Pa 30 × Pa 169 and its reciprocal, aiming to elucidate the seedlings death induced by Luteus-Pa. At 15 day after emergence (DAE) differences began to appear between the wild type and mutant. Mutant seedlings showed: (1) lack of photosynthesis and alterations in chloroplast morphology; (2) lower level of three abundant groups of proteins in leaves; (3) decrease in the content of chloroplastidic pigments (4) decrease in peroxidases activities and increase in leaf polyphenol oxidase activity; (5) decrease in carbohydrate and concentration of some nutrients and low dry mass in all plant parts. In leaves of mutant seedlings of both crosses damages occurred in the system responsible for the photochemical phase of photosynthesis.
