- Nucleoside diphosphate kinases (NDPK) are key enzymes involved in the intracellular nucleotide maintenance in all living organisms, especially in trypanosomatids which are unable to synthesise purines de novo. Four putative NDPK isoforms were identified in the Trypanosoma cruzi Chagas, 1909 genome but only two of them were characterised so far. In this work, we studied a novel isoform from T. cruzi called TcNDPK3. This enzyme presents an atypical N-terminal extension similar to the DM10 domains. In T. cruzi, DM10 sequences targeted other NDPK isoform (TcNDPK2) to the cytoskeleton, but TcNDPK3 was localised in glycosomes despite lacking a typical peroxisomal targeting signal. In addition, TcNDPK3 was found only in the bloodstream trypomastigotes where glycolytic enzymes are very abundant. However, TcNDPK3 mRNA was also detected at lower levels in amastigotes suggesting regulation at protein and mRNA level. Finally, 33 TcNDPK3 gene orthologs were identified in the available kinetoplastid genomes. - Problems with parasitic infections and their interspecies transmissions are common in zoological gardens and could pose serious health damage to captive animals. This study presents results of eight-year monitoring of intestinal parasites in animals from Zoo Ljubljana, Slovenia. A total of 741 faecal samples from 40 animal species were collected two to four times per year and examined microscopically. Intestinal parasites were detected in 45% of samples, with detection of helminths (Cestoda, Nematoda - Ascaridida, Enoplida, Strongylida, Oxyurida, Rhabditida and Trichurida) and protists (Apicomplexa and Ciliophora) in 25% and 13% of samples, respectively; mixed infection was found in 7% of samples. The mostly infected were ungulates (61%), followed by reptiles (44%), ratites (29%), primates (22%) and carnivores (7%). During the observation period, the number of infected animal species increased from 8 to 25. This is the first long-term monitoring study of intestinal parasites in zoo animals from Slovenia. 4. Application of compound mixture of caprylic acid, iron and mannan oligosaccharide against Sparicotyle chrysophrii (Monogenea: Polyopisthocotylea) in gilthead sea bream, Sparus aurata

- We have evaluated the therapeutic effect of a compound mixture of caprylic acid (200 mg/kg fish), organic iron (0.2% of diet) and mannan oligosaccharide (0.4% of diet) in gilthead sea bream, Sparus aurata Linnaeus, infected with Sparicotyle chrysophrii Beneden et Hesse, 1863 in controlled conditions. One hundred and ten reared and S. chrysophrii-free fish (197 g) located in a cement tank were infected by the parasite two weeks following the addition of 150 S. chrysophrii-infected fish (70 g). Growth parameters and gill parasitic load were measured in treated against control fish after a ten-week-period. Differences in final weight, feed conversion ratio, specific growth rate and feed efficiency were not statistically significant between the experimental groups, suggesting no evident effect with respect to fish growth during the study period. Although the prevalence of S. chrysophrii was not affected by the mixture at the end of the experiment, the number of adults and larvae was significantly lower. The mean intensity encompassing the number of adults and larvae was 8.1 in treated vs 17.7 in control fish. Individual comparisons of gill arches showed that the preferred parasitism site for S. chrysophrii it the outermost or fourth gill arch, consistently apparent in fish fed the modified diet and in control fish. In conclusion, the combined application of caprylic acid, organic iron and mannan oligosaccharide can significantly affect the evolution of infection with S. chrysophrii in gilthead sea bream, being capable of reducing adult and larval stages of the monogenean. - The eye nematode Thelazia callipaeda Railliet et Henry, 1910 (Spirurida: Thelaziidae) is a vector-borne zoonotic nematode infecting a range of wild and domestic carnivores as well as humans. It is considered to be a causative agent of emerging and neglected disease and currently invades central part of Europe. Nematodes were collected from the eye of a dog living in Prague, which never travelled outside the Czech Republic. The nematodes were identified based on their morphology and partial sequence of the cox1 gene as T. callipaeda haplotype 1. This finding represents the northernmost record of autochthonous canine thelaziosis in Europe. The insufficient control of imported animals as well as free movement of dogs and wild carnivores within Europe probably facilitates spreading of T. callipaeda throughout the continent. 6. Castosyringophilus meropis sp. n. (Acariformes: Syringophilidae): a new quill mite species parasitising the world population of Merops apiaster Linnaeus (Coraciiformes: Meropidae)

- A new species, Castosyringophilus meropis sp. n., found in quills of feathers of the European bee-eater Merops apiaster Linnaeus (Coraciiformes: Meropidae) is described. This new species is close to C. claravis Skoracki et Glowska, 2008 and differs, in females, by the presence apunctate coxal fields (vs sparsely punctate in C. claravis) and by the lengths of setae d1 145-180 µm, f2 170-185 µm and ag3 190-215 µm (vs d1 200-220 µm, f2 230-250 µm and ag3 150-170 µm). We present a vast mite material collected from bee-eaters originated from different localities in Europe, Asia and Africa, both breeding and wintering grounds of this bird. 7. Ceratomyxa bohari sp. n. (Myxozoa: Ceratomyxidae) from the gall bladder of Lutjanus bohar Forsskål from the Red Sea coast off Saudi Arabia: morphology, seasonality and SSU rDNA sequence

- A new myxozoan, Ceratomyxa bohari sp. n., infecting the gall bladder of two-spot red snapper, Lutjanus bohar Forsskål, in the Red Sea off Saudi Arabia, is described using light microscopy and characterised genetically. The infection was recorded as mature spores floating free in the bile. The overall prevalence of infection of the type host was 19% (67 fish infected of 360 examined), with the highest prevalence in autumn (31%; 28/90) and the lowest in winter at 12% (11/90). Mature spores are slender and slightly crescent-shaped in the frontal view, with anterior and posterior margins tapered gradually to rounded valvular tips. Spore valves are unequal with a prominent sutural line. The spore dimensions are 3-4 μm (mean 3.5 μm) in length and 16-19 μm (mean 17 μm) in thickness. Two polar capsules are spherical, equal in size, 1.5 μm in diameter. Coils of the polar filament are indiscernible. The sporoplasm is binucleated and fills nearly one third of the extracapsular space restricted to the area below the capsules. The molecular analysis based on the small subunit rDNA (SSU rDNA) sequence revealed a close relationship with majority of species of Ceratomyxa Thélohan, 1892 and phylogenetic clustering with species from different geographical location. Thus, the shorter spore of the present Ceratomyxa species and the divergence of the SSU rDNA sequences were the distinctive features that separate it from all previously described species and identified this parasite as a new species of Ceratomyxa., Lamjed Mansour, Abdel-Azeem S. 8. Genetic diversity of Echinococcus multilocularis in red foxes in Poland: the first report of a haplotype of probable Asian origin

- The aim of the present study was to estimate the genetic diversity of the cestode Echinococcus multilocularis Leuckart, 1863 in Poland based on sequence analysis of the mitochondrial genes of worms isolated from red foxes, Vulpes vulpes (Linnaeus). Overall, 83 adults of E. multilocularis from the same number of foxes in different parts of Poland were used for analysis. Sequences of the three mitochondrial genes, cytochrome b (cob), NADH dehydrogenase subunit 2 (nad2) and cytochrome c oxidase subunit 1 (cox1), were analysed. Seventy-four individual biological samples were successfully sequenced. Combined sequence analysis of these three genes exhibited fifteen Polish haplotypes (EmPL1-EmPL15). Most isolates (n = 29; 39%) were classified to the EmPL1 haplotype, which occurred mainly in the east, north and centre of Poland. Haplotype EmPL4 (n = 14; 19%) and other haplotypes appeared predominantly in the south and west area. Fourteen haplotypes were grouped in the European clade. One Polish haplotype (EmPL9) (n = 7, 10%) was assigned to the Asian clade with haplotypes from Japan and Kazakhstan. This haplotype was found only in northeast Poland and this is the westernmost report of haplotype of E. multilocularis belonging to the Asian clade in Europe. - Knowledge of blood parasites in Brazilian chelonians is limited, since they have been recorded in only six species. Mesoclemmys vanderhaegei (Bour) is a freshwater turtle with a wide geographic distribution in Brazil, but there is little information about its natural history. This paper reports on a study of the prevalence and infection intensity of a haemogregarine in two subpopulations of M. vanderhaegei. The study was conducted in two areas of Cerrado in the Upper Paraguay River basin in the state of Mato Grosso, Brazil, between November 2010 and August 2013. Ninety-five (53%) of the 179 turtles captured were positive for haemogregarine parasites. The parasitic forms observed were two morphotypes of intraerythrocytic gametocytes. The prevalence differed between size classes, increasing significantly according to the animals' body size. There was no significant difference between prevalence and sex, or between sampling periods. The mean parasite intensity was 9 parasites/2,000 erythrocytes (0.45%) and the parasite population presented an aggregated distribution, with an aggregation index of 19 and discrepancy of 0.772. 10. K13 propeller domain mutations and pfmdr1 amplification in isolates of Plasmodium falciparum collected from Thai-Myanmar border area in 2006-2010

- The K13 propeller domain mutation and pfmdr1 amplification have been proposed as useful molecular markers for detection and monitoring of artemisinin resistant Plasmodium falciparum Welch, 1897. Genomic DNA isolates of P. falciparum was extracted from 235 dried blood spot or whole blood samples collected from patients with uncomplicated falciparum malaria residing in areas along the Thai-Myanmar border during 2006-2010. Nested polymerase chain reaction (PCR) and sequencing were performed to detect mutations in K13 propeller domain of P. falciparum at codon 427-709. Pfmdr1 gene copy number was determined by SYBR Green I real-time PCR. High prevalence of pfmdr1 multiple copies was observed (42.5% of isolates). The presence of K13 mutations was low (40/235, 17.2%). Seventeen mutations had previously been reported and six mutations were newly detected. The C580Y was found in two isolates (0.9%). The F446I, N458Y and P574L mutations were commonly detected. Seven isolates had both K13 mutation and pfmdr1 multiple copies.
