2. Effect of specific translation inhibitors on polypeptide composition and spectral characteristics ofwheat thylakoid membranes

- Both the synthesis of apoprotein P700 with molecular mass of 72 kDa and asseinbly of chlorophyll (Chl) a-protein of photosystem (PS) 1 were blocked by lincoinycin (inhibitor of translation in chloroplasts). Synthesis of 48 and 43,5 kDa polypeptides belonging to the PS2 complex were also suppressed. Chl a forms of PS2 reaction centi e (RC) at 683 nin and of PS 1 RC at 690, 696 and 705 nm disappeared, Relative Chl h content and fluorescence bands ratio F736/F686 increased significantly Cycloheximide (inhibitor of translation in cytoplasin) ti eatinent resulted in reduction of polypeptides of the light-harvesting Chl a//i-protein complex (LFIC) in the 28.5- 25.5 kDa region. At the same time a relatively smaller decrease was observed of two Chl foirns of the LHC: Chl by 3.8 % and Chl by 10.2 % (12.4 and 34.3 % in the contiol, respectively). The fluorescence bands ratio F736/F686 decrease in low temperature spectra was possibly connected with the LIlC-1 and LIlC-2 synthesis blocking.

- The method of processing of laser ranging data collected using the passively mode locked YAG train laser is described. The algorithm for resolving of individual peaks in measured ranges histogram and system internal noise determination is explained together with the crosscorrelation methods for system calibration constant evaluation. 5. The effect of pre-treatment with proline on the responses of Pisum sativum to salt stress

- 10-d-old pea plants {Pisum sativum L. cv. Ran 1) were treated for 24 h with proline (10-6 M oř 10*5 M) before salinization with 50 mM NaCl for 2 d. Salt stress resulted in an increase of endogenous ffee proline content, CO2 compensation concentration, photorespiration and glycollate oxidase activity; net photosynthetic rate (P^) was inhibited, but dark respiration rate (Pp) was not affected. •‘♦CO2 fixation by protoplasts isolated from salt stressed plants was inhibited by 60 %, however, the *‘*C02 fixation by protoplasts, isolated firom plants treated with proline before salinization, was only slightly reduced by NaCl. Proline alleviated the inhibitory effect of NaCl in a concentration-depending manner. Pre-treatment with proline decreased Na+ and CP accumulation in the shoot; the root content of these ions was increased.
