Hladiny serotoninu u pacientů i v kontrolní skupině byly stanoveny pomocí enzymové imunoanalýzy (EIA, Immunotech). Výsledky ukazují pokles hladin arteriálního serotoninu během 24 hodin po operaci., Objective: Percutaneous closure of patent foramen ovale (PFO) has been associated with improvement of migraine headache. Tissues-borne serotonin bypassing the metabolic active pulmonary circulation through the PFO is the proposed trigger of migraine attacks in susceptible patients. Therefore a hypothetical decrease of arterial serotonin levels after PFO closure might explain the migraine improvement. Aim of the study: Aim of the study was the investigation of suitability of direct serotonin assay in venous and arterial blood in patients undergoing percutaneous PFO closure for further studying the mechanism of migraine improvement after PFO closure. Materials and methods: Arterial and venous serum serotonin levels were investigated in eight patients undergoing correctional surgery operation. The samples were collected as follows: Venous and arterial blood before operation, arterial blood during operation, venous sample from right atrial central catheter and then peripheral venous and arterial sample 24 hours after the operation. Serotonin levels were investigated in control group of 6 healthy patients. Serotonin was measured by commercially available enzyme immunoassay (EIA, Immunotech). Results: The results (mean ± SD) were as follows: Control group: 906.0 ± 229.4 nmol/l, Venous blood before the operation: 126.0 ± 69.6 nmol/l, venous blood after the operation: 140 ± 40 nmol/l, arterial blood before the operation: 165± 25 nmol/l, arterial blood during operation: 182 ± 44 nmol/l, arterial blood after the operation: 130 ± 36 nmol/l. Conclusion: Direct assay of serotonin with commercially available EIA is suitable for studying changes of serotonin levels after PFO closure. Our preliminary results support the theory of the decrease of arterial blood levels of serotonin after PFO closure., Šramko M., Kotaška K., Veselka J., Linhartová K., Průša R., and Lit.: 14
Soubor tvořilo 301 pacientů (161 diabetiků a 140 nediabetiků, 182 mužů a 119 žen, věk 44,1 ± 25,3 let). Uvedená HPLC metoda je dostatečně specifická pro stanovení albuminu v moči, nedochází k významným interferencím s dalšími bílkovinami., Objective: The aim of our study was the implementation of HPLC method for assessing albuminuria, confirming or refusing the hypothesis about the existence of co-eluting proteins, and comparison of urine albumin concentrations assessed using HPLC and immunoturbidimetric methods in patient samples. Material and Methods: We developed the HPLC method under these chromatographic conditions: multilinear gradient of mobile phase consisting of water (solvent A), acetonitrile (solvent B), 100 mM NaH2PO4 (solvent C), 1 % trifluoroacetic acid (solvent D), flow rate 2 mL/min, temperature 22 ˚C, UV detection in the wavelength 280 nm, chromatographic column Zorbax 300 SB-C3, liquid chromatograph Agilent 1200 (Agilent Technologies, USA). We analyzed two mixtures of urine. The first one was prepared from 30 patient urine samples with immunoturbidimetrically physiological albuminuria, to which we added albumin standard. The second mixture was prepared from 30 patient urine samples with mild albuminuria. We analyzed and compared albuminuria in a group of 301 patients (161 diabetics and 140 nondiabetics, 182 males and 119 females, age 44.1 ± 25.3 years). Results: Transferrin, α-1-acid glycoprotein, α-1-antitrypsin, α-1-antichymotrypsin and hemopexin do not interfere with albumin in the presented HPLC method, whereas the elution curve of prealbumin splits into several peaks, of which a few interfere with albumin. With regard to very low prealbumin concentrations in serum and in urine, we can assume that this interference is not of clinical importance. In mixture 1 we did not find a significant difference between the albuminuria assessed using both methods (79.1 mg/L immunoturbidimetrically vs. 82.5 mg/L HPLC), while in mixture 2 we measured over 26 % greater albuminuria using HPLC (79.5 mg/L immunoturbidimetrically vs. 99.9 mg/L HPLC). These results suggest that immunounreactive albumin really exists. We found a statistically significant difference between the methods in patient urine samples (entire patients´ group: [median ± SEM]: 21.1 ± 38.2 mg/L immunoturbidimetrically vs. 30.4 ± 42.2 mg/L HPLC, p < 0.0001, Mann-Whitney test), which was most remarkable in the group of diabetic patients with immunoturbidimetrically physiological albuminuria. Conclusion: Our results prove that the HPLC method for albumin detection is more sensitive than immunoturbidimetry and sufficiently specific and there are no significant interferences with other proteins., and Fořtová M., Klapková E., Průša R.
Lipokaliny tvoří velkou skupinu malých extracelulárních proteinů. Vykazují velkou rozmanitost na úrovni sekvenčního uspořádání, terciární struktura je oproti tomu vysoce konzervovaná a představuje ji β-válec tvořený osmi antiparalelními strukturami β-skládaného listu. Vazebné místo ligandu se nachází uvnitř β-válce. Zastávají rozličné funkce, např. transportují retinoidy, steroidy, biliny, feromony, účastní se syntézy prostaglandinů, tvorby ochranného zbarvení bezobratlých, vytváření čichových vjemů, jsou zapojené do regulace buněčné homeostázy a modulace imunitní odpovědi. Lipokalin-2 (neutrofilní s gelatinázou asociovaný lipokalin), časný marker různých typů renálního poškození, vykazuje pleiotropní biologické aktivity v různých typech buněk a tkání. Hraje významnou roli v angiogenezi, apoptóze, organogenezi, zánětu, hematopoeze, v procesu hojení ran, v renální fyziologii, v nádorové a reprodukční biologii., The lipocalin protein family is a large group of small extracellular proteins. The family demonstrates great diversity at sequence level; however, lipocalin crystal structures are highly conserved and comprise a single eight-stranded antiparallel β-barrel, which encloses an internal ligand-binding site. They have been associated with a variety of different functions, among them transport of retinoids, steroids, bilins, pheromones, enzymic synthesis of prostaglandins, cryptic coloration, olfaction, immune response, cell regulation.
