The rare and endangered plant, Begonia fimbristipula, shows red and green phenotypes, differentiated by a coloration of the abaxial leaf surface. In this study, we compared morphological and physiological traits of both phenotypes. The results showed that the red phenotype contained a significantly higher chlorophyll content, closer arrangement of chloroplasts, and a more developed grana. In addition, the red phenotype transferred significantly more light energy into the electron transport during the photoreaction. Similarly, the maximum photosynthetic rate, instantaneous water-use and light-use efficiencies of the red B. fimbristipula were all significantly higher than those of the green individuals. The differentiation between these two phenotypes could be caused by their different survival strategies under the same conditions; epigenetic variations may be in some correlation with this kind of phenotype plasticity. Red B. fimbristipula has an advantage in resource acquisition and utilization and possesses a better self-protection mechanism against changes in environmental conditions, therefore, it might adapt better to global climate change compared to the green phenotype. Further studies on the possible epigenetic regulation of those phenotypic differentiations are needed., Y. Wang, L. Shao, J. Wang, H. Ren, H. Liu, Q. M. Zhang, Q. F. Guo, X. W. We studied the expression of myosin heavy chain isoforms at mRNA and protein levels as well as fiber type composition in the fast extensor digitorum longus (EDL) and slow soleus (SOL) twitch muscles of adult inbred Lewis strain rats. Comparison of the results from Real Time RT-PCR, SDS-PAGE and fiber type analysis showed corresponding proportions of MyHC transcripts (MyHC-1, -2a, -2x/d, -2b), protein isoforms (MyHC-1, -2a, -2x/d, -2b) and fiber types (type 1, 2A, 2X/D, 2B) in both muscles. Furthermore, we found that slow MyHC-1 mRNA expression in the SOL was up to three orders higher than that of fast MyHC transcripts. This finding can explain the predominance of MyHC-1 isoform and fiber type 1 and the absence of pure 2X/D and 2B fibers in the SOL muscle. Based on our data presenting quantitative evidence of corresponding proportions between mRNA level, protein content and fiber type composition, we suggest that the Real Time RT-PCR technique can be used as a routine method for analysis of muscle composition changes and could be advantageous for the analysis of scant biological samples such as muscle biopsies in humans., J. The first attempt to artificially hybridize species of Potamogeton resulted in the hybrid P. perfoliatus × P. gramineus. The morphological features, reproductive behaviour and molecular markers of the offspring of this experimental hybridization were compared with those of the parental species and natural hybrids of the same assumed parentage. A phenotype corresponding to that of the natural hybrid P. × nitens was acquired from an experimental cross between P. perfoliatus and P. gramineus. All plants, both natural and artificial, of this hybrid were consistently sterile. They showed the ITS variants of both parental taxa, which is consistent with biparental inheritance of nuclear DNA. The experimental hybrid was used to test the maternal inheritance of chloroplast DNA in Potamogeton. Sequences of a chloroplast intergenic spacer (rpl20- rps12) were identical with those of the female parent. Then, the directions of the crosses resulting in the natural hybrids were investigated. Of five natural populations of P. × nitens, P. gramineus was the maternal parent of two and P. perfoliatus of three populations. The frequency of hybridization events and rise of hybrids are discussed.
We developed a new method for the quantitative determination of myosin heavy chain (MyHC) isoforms taking advantage of immunochemical differences and based on the ELISA principle. In the present paper we compare anal ysis of MyHC isoforms using the SDS-PAGE and the ELISA methods in the same samples of adult female inbred Lewis strain euthyroid, hyperthyroid and hypothyroid rats. In all thyroid states, the same composition and corresponding changes of MyHC isoforms were determined using both methodological approaches in the slow soleus and the fast extensor digitorum longus muscles. Our results showed that ELISA can be used for a “semi-quantitative” or “comparative” measurement of MyHC isoforms in multiple muscle samples, but that it is neither more exact no r faster compared to SDS-PAGE., Jan Říčný., and Obsahuje bibliografii a bibliografické odkazy
The aim of the present study was to compare the oscillations of oxygenation in skeletal muscle between early and late phases in prolonged exercise. During prolonged exercise at 60 % of peak oxygen uptake ( o2) for 60 min and at rest, oxygenated hemoglobin/myoglobin (Hb/MbO2) and total Hb/Mb (THb/Mb) were determined by near-infrared spectroscopy in the vastus lateralis. Power spectra density (PSD) for the difference between Hb/MbO2 and THb/Mb (−HHb/MbO2: deoxygenation) was obtained by fast Fourier transform at rest, in the early phase (1-6 min) and in the late phase (55-60 min) in exercise. Peak PSD in the early phase was significantly higher than that at rest. There were at least three peaks of PSD in exercise. The highest peak was a band around 0.01 Hz, the next peak was a band around 0.04 Hz, and the lowest peak was a band around 0.06 Hz. PSD in the early phase was not significantly different from that in the late phase in exercise. Heart rate (HR) showed a continuous significant increase from 3 min in exercise until the end of exercise. Skin blood flow (SBF) around the early phase was significantly lower than that around the late phase. It was concluded that oscillation of oxygenation in the muscle oxygen system in the early phase is not different from that in the late phase in prolonged exercise despite cardiovascular drift., T. The purpose of the present study was to examine whether the level of oxygen uptake (V.o2) at the onset of decrement-load exercise (DLE) is lower than that at the onset of constant-load exercise (CLE), since power output, which is the target of V.o2 response, is decreased in DLE. CLE and DLE were performed under the conditions of moderate and heavy exercise intensities. Before and after these main exercises, previous exercise and post exercise were performed at 20 watts. DEL was started at the same power output as that for CLE and power output was decreased at a rate of 15 watts per min. V.o2 in moderate CLE increased at a fast rate and showed a steady state, while V.o2 in moderate DLE increased and decreased linearly. V.o2 at the increasing phase in DLE was at the same level as that in moderate CLE. V.o2 immediately after moderate DLE was higher than that in the previous exercise by 98±77.5 ml/min. V.o2 in heavy CLE increased rapidly at first and then slowly increased, while V.o2 in heavy DLE increased rapidly, showing a temporal convexity change, and decreased linearly. V.o2 at the increasing phase of heavy DLE was the same level as that in heavy CLE. V.o2 immediately after heavy DLE was significantly higher than that in the previous exercise by 156±131.8 ml/min. Thus, despite the different modes of exercise, V.o2 at the increasing phase in DLE was at the same level as that in CLE due to the effect of the oxygen debt expressed by the higher level of V.o2 at the end of DLE than that in the previous exercise., T. Pancreatic microcirculatory dysfunction emerged as a novel mechanism in the development of hypertension. However, the changes of pancreatic microcirculation profiles in hypertension remain unknown. Pancreatic microcirculatory blood distribution pattern and microvascular vasomotion of spontaneously hypertensive rats (SHRs) and Wistar Kyoto rats (WKYs) were determined by laser Doppler. Wavelet transform analysis was performed to convert micro-hemodynamic signals into timefrequency domains, based on which amplitude spectral scalograms were constructed. The amplitudes of characteristic oscillators were compared between SHRs and WKYs. The expression of eNOS was determined by immunohistochemistry, and plasma nitrite/nitrate levels were measured by Griess reaction. Additionally, endothelin-1, malondialdehyde, superoxide dismutase and interleukin-6 were determined by enzyme-linked immunosorbent assay. SHRs exhibited a lower scale blood distribution pattern with decreased average blood perfusion, frequency and amplitude. Wavelet transform spectral analysis revealed significantly reduced amplitudes of endothelial oscillators. Besides reduced expression of eNOS, the blood microcirculatory chemistry complements micro-hemodynamic profiles as demonstrated by an increase in plasma nitrite/nitrate, endothelin-1, malondialdehyde, interleukin-6 and a decrease of superoxide dismutase in SHRs. Here, we described abnormal pancreatic microcirculation profiles in SHRs, including disarranged blood distribution pattern, impaired microvascular vasomotion and reduced amplitudes of endothelial oscillators.
The parameters estimated from traditional A/Ci curve analysis are dependent upon some underlying assumptions that substomatal CO2 concentration (Ci) equals the chloroplast CO2 concentration (Cc) and the Ci value at which the A/Ci curve switches between Rubisco- and electron transport-limited portions of the curve (Ci-t) is set to a constant. However, the assumptions reduced the accuracy of parameter estimation significantly without taking the influence of Ci-t value and mesophyll conductance (gm) on parameters into account. Based on the analysis of Larix gmelinii's A/Ci curves, it showed the Ci-t value varied significantly, ranging from 24 Pa to 72 Pa and averaging 38 Pa. t-test demonstrated there were significant differences in parameters respectively estimated from A/Ci and A/Cc curve analysis (p<0.01). Compared with the maximum ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) carboxylation rate (Vcmax), the maximum electron transport rate (Jmax) and Jmax/Vcmax estimated from A/Cc curve analysis which considers the effects of gm limit and simultaneously fits parameters with the whole A/Cc curve, mean Vcmax estimated from A/Ci curve analysis (Vcmax-Ci) was underestimated by 37.49%; mean Jmax estimated from A/Ci curve analysis (Jmax-Ci) was overestimated by 17.8% and (Jmax-Ci)/(Vcmax-Ci) was overestimated by 24.2%. However, there was a significant linear relationship between Vcmax estimated from A/Ci curve analysis and Vcmax estimated from A/Cc curve analysis, so was it Jmax (p<0.05). and W. Zeng ... [et al.].
Some preliminary results of perturbation method applied to 5/2 asteroidal resonance with Jupiter are presented. The results are compared with those obtained by the author with the Wisdom method previously applied to this resonance by Šidlichovský and Melendo. Inferences for the origin of the corresponding Kirkwood gap are drawn.
The values of density and velocity in the samples of the predominant rocks from the Archaean section of the Kola superdeep borehole (SG-3) drilled in the northern frame of the Pechenga palaeorift structure have been studied. The main part of rocks of the SG-3 Archaean section is represented by gneisses, schists and amphibolites. Their main rock-forming minerals are plagioclase, hornblende, mica and quartz. The texture of the rocks is mainly medium-grained, the structure is nematogranoblastic and lepidogranoblastic. The parameters of elastic anisotropy were estimated by the acoustopolariscopy method, the density and velocity of compression and shear waves were determined in the lab and the mentioned characteristics were calculated by mineral composition. The measured sample velocities are unusually low, which can be explained by the decompaction effect of deep rocks. The values of the velocity characteristics that are close to those for the conditions of the rock deep occurrence were obtained by calculation with regard to their specific mineral composition. The average values of the velocity of compression and shear waves for gneisses were calculated by mineral composition are 6.38 ± 0.16 km/s and 3.52 ± 0.14 km/s, respectively. The average of the compression wave velocity for schists is 6.40 ± 0.13 km/s, of the shear wave velocity - 3.46 ± 0.09 km/s and for amphibolites 6.84±0.13 km/s and 3.82±0.08 km/s, respectively. The dynamic moduli in different rocks are lower in schists, medium in gneisses and higher in amphibolites., Olga Trishina, Feliks Gorbatsevich and Mikhail Kovalevskiy., and Obsahuje bibliografii
