Changes in the chlorophyll (Chl) and carotenoid (Car) contents and photosynthetic activity of leaves of three-year-old maple trees were studied under an extremely high irradiance (HI) (5 000 and 7 000 pmol m-2 s‘*) and subsequent low irradiance (LI) (10 pmol m-2 s‘*). Speciál attention was paid to a possible linear correlation between zeaxanthin (z) accumulation and the decrease in variable Chl fluorescence (ratio Fy/FnO during photoinhibitory treatment. Hl-induced violaxanthin (v) transformation into z was a fast one-step response proceeding within 2-5 min. Changes in the Chl a and b contents or in non-xanthophyll cycle Car were either not observed (30 min at 5 000 pmol m*2 s‘i) or a slight decline of the Chl a, lutein, 3-carotene and z contents only occurred at prolonged exposure (60 min at 7 000 pmol m'^ s'i)- Photosynthetic activity measured via the ratios (Fy/F^, Fy/F^, Rfj 690, Rfj 735) sank during this treatment to a much higher extent at 7 000 than at 5 000 pmol m'^ s'*. Ratios Fy/F<„ Rfd 690 and Rfj 735 proved to be better indicators of photoinhibition with a larger amplitudě ťhan the ratio Fy/F,„. Unlike literatuře reports, no linear correlation was found between Hl-induced decrease of fluorescence ratios and the z accumulation. Decrease of the fluorescence ratios appears to be biphasic in nátuře: fast initial fall which parallels the z formation is followed by a phase when ratios Fy/F^,, Fy/F^,, Rj^ 690 and Rf^ 735 sink further without any changes in the z pool. Regeneration of photosynthetic activity under LI also proceeded in two steps; a relatively fast increase in ratios with little or no changes in the z pool followed by a slow many hours' restoring of the former fluorescence ratios, paralleled by the z transformation into v. The back-reaction of z to v under LI was increasingly retarded with higher irradiance. Z formation and decrease in the fluorescence ratios are thus probably two Hl-induced processes which may be mutually independent.
The role of the physical state of ovarian membranes was studied in immature rats after PMSG- and hCG-induced pseudopregnancy. Parallel changes in LH/hCG receptors, progesterone secretion and rigidity of membrane lipids were observed during pseudopregnancy. Possible structure-functional properties of the LH/hCG receptor were analyzed by the thermal perturbation technique. Thermal stability of the receptor was higher 5 days after hCG ovulatory injection to rats compared to days 11 or 18 and to control rats. Pseudopregnancy modified the quenching of protein fluorescence. The Stern-Volmer constants for controls and for rats on days 5, 11 and 18 of pseudopregnancy were found to be 2.4 and 4.6, 5.1 and 4.4, respectively, indicating that accessibility of fluorophores for the quencher was increased.
Changes in the content of pigments and rate of photosynthesis in Azolla microphylla Kaulf. fronds were measured during growth under solar and ultraviolet-C (UV-C) supplemented solar radiation. Maximum content of total chlorophyll (Chl) was observed on the 13th day (termination of the experiment) of treatment in both control and treated plants. The treated plants had significantly lower total Chl and carotenoid contents than the control plants during the 1st day of growth. After the 4th day of exposure to UV-C supplemented solar radiation, the Chl and carotenoids accumulation increased in treated plants, so that the pigment concentration in the treated fronds was nearer to the control values after the 13th day of treatment. Significant increase in UV absorbing pigments, anthocyanins, and flavonoids was observed at the 13th day of treatment. In spite of the roughly similar photosynthetic pigment concentration, the photosynthetic activity measured as the rate of electron transport at photosystem 2 was only 65 % of the control values after 13 d of UV-C exposure. and M. Leptin and adiponectin, two adipocytokines, may work together in regulating energy homeostasis and insulin action. Leptin gene expression has been investigated in term placental tissue complicated by gestational diabetes mellitus (GDM), but never in conjunction with all isoforms of the leptin receptor (LEPR A-D), or with adiponectin receptors (ADIPOR1 and 2). In this study we examined the association between changes in expression of these genes in placental tissue and GDM risk. We assessed placental gene expression of leptin, LEPR A-D and ADIPOR1 and 2 by real time PCR using mRNA from maternal and fetal biopsies. Tissues were collected from uncomplicated pregnancies (n=28) and those complicated by GDM (n=19). Gene expression was normalized to three endogenous housekeeping genes. Relative gene expression values were reported as fold change between groups. Adiponectin gene expression was out of the sensitive range of our assay. There were increases in leptin mRNA expression in GDM cases compared with controls for maternal-side (p=0.06), and fetal-side (p=0.09) placental biopsies. No significant changes were seen in GDM cases compared with controls in LEPR A-D or ADIPOR1 and 2. mRNA derived from maternal-side tissue was positively correlated with tissue from the fetal side for all genes studied (all p<0.01). Finally, we noted that absence or presence of GDM was a major factor in leptin mRNA expression after adjusting for maternal age, mode of delivery, parity and smoking status. In conclusion, increases in leptin mRNA expression in term placenta, but not that of its receptors, are associated with the diagnosis of GDM. Changes seen in the ligand, but not the receptor, of the leptin pathway in GDM-complicated pregnancies may also apply to the adiponectin pathway, as the ADIPOR1 and 2 mRNAs do not change with GDM diagnosis.
The effects of phosphate concentration on plant growth and photosynthetic performance were examined in leaves of Zizania latifolia. Plants were grown for four weeks in a solution containing 0, 0.16, 0.64, and 2.56 mM orthophosphate. The results showed that the highest net photosynthetic rate (P N) was achieved at 0.64 mM orthophosphate, which corresponded to the maximum content of organic phosphorus in leaves. Low phosphorus (low-P) content in the culture solution inhibited plant growth, affecting plant height, leaf length, leaf number, tiller number, and fresh mass of leaf, sheath, culm, root, and total plant. In addition, we observed that low-P (0.16 mM) did not hinder the growth of roots but increased the root:shoot ratio, and significantly decreased the chlorophyll content, P N, stomatal conductance, and transpiration rate, but increased the intercellular CO2 concentration. Additionally, low-P significantly decreased the maximum carboxylation rate of Rubisco, the maximum rate of ribulose-1,5-bisphosphate regeneration, the effective quantum yield of PSII photochemistry, photochemical quenching coefficient, and electron transport rate, but increased the nonphotochemical quenching. However, the maximal quantum yield of PSII photochemistry was not significantly affected by low-P. High phosphorus (2.56 mM) caused only a slight decrease in gas-exchange parameters. In this study, we investigated the effects of Nw -nitro-L-arginine (L-NNA) on arterial blood pressure (BP), plasma noradrenaline (NA) and adrenaline (A) levels and angiotensin-converting enzyme (ACE) activity. L-NNA was applied with tap water (1 mg/ml) from the 3rd to the 8th week of age (group L-NNA1). In Experiment 1, long-term L-NNA application increased BP compared to the control group (group C1) (L-NNA1 = 131.4 ± 6.3, n=6; C1= 82.7 ± 4.7 mm Hg, n=7) but decreased plasma noradrenaline and adrenaline levels and ACE activity (NA levels: C1 = 15.5 ± 0.8, n=7; L-NNA1= 8,6 ± 0.5 ng/ml, n=7; A levels: C1 = 15.5 ± 0.8, n=7; L-NNA1 = 6.0 ± 0.5 ng/ml, n=7; ACE activities: C1= 87.3 ± 3.1, n=6; L-NNA1 = 46.2 ± 1.9 U/l, n=5). On the other hand, in Experiment 2 (carried out under the same conditions and in age-matched chickens), blood pressure, plasma noradrenaline levels and ACE activity were found to differ in the control group (C2) (BP=141.4 ± 15.5 mm Hg, n=7; NA =1.1 ± 0.4 ng/ml, n=7; ACE = 57.2 ± 5.3 U/l, n=7) as compared to C1, while plasma adrenaline levels were similar. In this series, long-term L-NNA application (group L-NNA2) did not change the BP, but surprisingly increased noradrenaline and ACE values (values of L-NNA2: BP = 165.7 ± 15.6 mm Hg, n=7; NA = 9.3 ± 1.3 ng/ml, n=8; ACE = 149.4 ± 16 U/l, n=8) while decreasing plasma adrenaline levels. L-arginine addition to L-NNA treatment completely reversed plasma noradrenaline and ACE activity values. Aiming at understanding the odd case of CAM expression by a C4 plant, some properties of phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31, orthophosphate: oxaloacetate carboxylyase, phosphorylating) were comparatively studied in leaves of CAM-expressing and non-expressing Portulaca oleracea L. plants. CAM expression was induced by growing plants under an 8-h photoperiod and under water-stress. CAM induction in leaves of these plants (designated as CAM) is indicated by the nocturnal acidification and by the clear diurnal oscillation pattern and amplitude of acidity, malic acid, and PEPC activity characteristic of CAM plants. Treatment of the other plant group (designated as C4) by growth under a 16-h photoperiod and well-watered conditions did not induce expression of the tested criteria of CAM in plants. In these C4 plants, the mentioned CAM criteria were undetectable. PEPC from CAM and C4Portulaca responded differently to any of the studied assay conditions or effectors. For example, extent and timing of sensitivity of PEPC to pH change, inhibition by malate, activation by glucose-6-phosphate or inorganic phosphate, and the enzyme affinity to the substrate PEP were reversed with induction of CAM from the C4-P. oleracea. These contrasting responses indicate distinct kinetic and regulatory properties of PEPC of the two modes. Thus by shifting to CAM in the C4Portulaca a new PEPC isoform may be synthesised to meet CAM requirements. Simultaneous occurrence of both C4 and CAM is suggested in P. oleracea when challenged with growth under stress.
Direct effects and after-effects of soil drought for 7 and 14 d were examined on seedling dry matter, leaf water potential (ψ), leaf injury index (LI), and chlorophyll (Chl) content of drought (D) resistant and sensitive triticale and maize genotypes. D caused higher decrease in number of developed leaves and dry matter of shoots and roots in the sensitive genotypes than in the resistant ones. Soil D caused lower decrease of ψ in the triticale than maize leaves. Influence of D on the Chl b content was considerably lower than on the Chl a content. In triticale the most harmful D impact was observed for physiologically younger leaves, in maize for the older ones. A period of 7-d-long recovery was too short for a complete removal of an adverse influence of D. and M. T. Grzesiak ... [et al.].
There is public concern that large-scale disturbances to forest cover caused by insects and storm winds in the Bohemian Forest could intensify high water flows and enhance the expected flooding risks predicted in current regional climate change scenarios. We analysed stream discharge in Upper Vydra and Große Ohe, neighbouring catchments in the Bohemian Forest, the largest contiguous forested area in Central Europe. Upper Vydra, in the Šumava National Park, and Große Ohe (including the Upper Große Ohe headwater catchment in the Bavarian Forest National Park) are similar in size, but differ in land use cover and the extent of disturbed Norway spruce stands. Publicly available runoff and meteorological data (1978-2011) were examined using non-parametric trend and breakpoint analysis. Together with mapped vegetation cover changes, the results were used to address the following questions: 1) are there significant changes in the hydrological cycle and, if so, do these changes relate to 2) the extent and expansion of disturbance in forests stands and/or 3) altered precipitation dynamics and thermal conditions? We found no marked overall change in annual runoff or in annual or seasonal precipitation, but a significant increase in high flows in March. This overall trend related to the marked warming in late winter and early spring (+~4 K in April, p < 0.01), irrespective of altitude and slope position. It significantly shifted the end of the snow cover period by more than three weeks to the beginning/middle of April depending on altitude, and intensified snow melt. In the Upper Große Ohe catchment, a significant decrease in catchment balance, the difference between the long term precipitation and runoff (-72 mm, 11%) was found when the loss of tree cover reached 30% of catchment area. Diminished evapotranspiration losses from severely disturbed stands increased groundwater recharge during summer and caused a significant rise in low flows in autumn. However, observed increases in late winter high flows were due to warming only. They could be further intensified by the increasing winter precipitation predicted under present climate change scenarios, and would therefore increase the risk of flooding at lower elevations.
Hypoxic pulmonary vasoconstriction (HPV) occurs in smooth muscle cells (SMC) from small pulmonary arteries (SPA) and is accompanied by increases in free cytoplasmic calcium ([Ca2+]i) and cytoplasmic pH (pHi). SMC from large pulmonary arteries (LPA) relax during hypoxia, and [Ca2+]i and pHi decrease. Increases in pHi and [Ca2+]i in cat SPA SMC during hypoxia and the augmentation of hypoxic pulmonary vasoconstriction by alkalosis seen in isolated arteries and lungs suggest that cellular mechanisms, which regulate inward and outward movement of Ca2+ and H+, may participate in the generation of HPV. SMC transport systems that regulate pHi include the Na+-H+ transporter which regulates intracellular Na+ and H+ and aids in recovery from acid loads, and the Na+-dependent and Na+-independent Cl-/HCO3- transporters which regulate intracellular chloride. The Na+-dependent Cl-/HCO3- transporter also aids in recovery from acidosis in the presence of CO2 and HCO3-. The Na+-independent Cl-/HCO3- transporter aids in recovery from cellular alkalosis. The Na+-H+ transporter was present in SMC from SPA and LPA of the cat, but it seemed to have little if any role in regulating pHi in the presence of CO2 and HCO3-. Inhibiting the Cl-/HCO3- transporters reversed the normal direction of pHi change during hypoxia, suggesting a role for these transporters in the hypoxic response. Future studies to determine the interaction between pHi, [Ca2+]i and HPV should ascertain whether pHi and [Ca2+]i changes are linked and how they may interact to promote or inhibit SMC contraction., J.
