Steroid hormone 20-hydroxyecdysone and the sesquiterpenoid juvenile hormone are the main regulators of insect development; however, it is unclear how they interact in the regulation of metamorphic events. Using the silkworm, Bombyx mori, we show that the juvenile hormone analogue fenoxycarb affects the cascade of ecdysone regulated genes that control the programmed cell death in the larval midgut. Morphological changes that occur during cell death were investigated by studying cross-sections of the midgut stained with hematoxylin and eosin. Apoptosis-specific DNA fragmentation was detected using TUNEL assay. Expression patterns of genes ATG8 and ATG12, which were used as indicators of autophagy, and genes of the ecdysone-regulated gene cascade were examined using real-time quantitative polymerase chain reaction. Fenoxycarb application on day 0 of the 5th larval instar extended the feeding period and postponed programmed cell death in mature larval midgut. This effect was probably due to a delay in ecdysone secretion and associated changes in gene expression were mostly not a direct response to the fenoxycarb. However, differences in the gene expression patterns in the control and fenoxycarb treated insects during the prepupal and early pupal stages indicated that fenoxycarb may also exert a more direct effect on some genes of the ecdysone regulated gene cascade., Ebru Goncu, Ramazan Uranli, Osman Parlak., and Obsahuje bibliografii
The K13 propeller domain mutation and pfmdr1 amplification have been proposed as useful molecular markers for detection and monitoring of artemisinin resistant Plasmodium falciparum Welch, 1897. Genomic DNA isolates of P. falciparum was extracted from 235 dried blood spot or whole blood samples collected from patients with uncomplicated falciparum malaria residing in areas along the Thai-Myanmar border during 2006-2010. Nested polymerase chain reaction (PCR) and sequencing were performed to detect mutations in K13 propeller domain of P. falciparum at codon 427-709. Pfmdr1 gene copy number was determined by SYBR Green I real-time PCR. High prevalence of pfmdr1 multiple copies was observed (42.5% of isolates). The presence of K13 mutations was low (40/235, 17.2%). Seventeen mutations had previously been reported and six mutations were newly detected. The C580Y was found in two isolates (0.9%). The F446I, N458Y and P574L mutations were commonly detected. Seven isolates had both K13 mutation and pfmdr1 multiple copies. It needs to be confirmed whether parasites harbouring both K13 mutation and pfmdr1 multiple copies and/or the observed new mutations of K13 propeller domain are associated with clinical artemisinin resistance., Papichaya Phompradit, Wanna Chaijaroenkul, Phunuch Muhamad, Kesara Na-Bangchang., and Obsahuje bibliografii
Celá problematika je ještě daleko od svého kompletního vyřešení, ale současný pohled nám může pomoci v lepší péči o vodní prostředí., This article deals with the taxonomy, reproductive biology, phylogenetics, phylogeography and introduction history of the fishes of the genus Carassius. It summarizes findings associated with their unique mode of reproduction, challenging species delimitation and insufficient understanding of the biology and systematics of these fishes in the past. Deeper insight into the biology of Prussian Carp (Carassius gibelio) will require more research, but the available results can improve our understanding and management of the aquatic environment., Lukáš Kalous., and Obsahuje seznam literatury
The aim of this study was to characterize karyotypes of central European spiders of the genera Arctosa, Tricca, and Xerolycosa (Lycosidae) with respect to the diploid chromosome number, chromosome morphology, and sex chromosomes. Karyotype data are reported for eleven species, six of them for the first time. For selected species the pattern in the distributions of the constitutive heterochromatin and the nucleolar organizer regions (NORs) was determined. The silver staining technique for detecting NORs of lycosid spiders was standardized. The male karyotype consisted of 2n = 28 (Arctosa and Tricca) or 2n = 22 (Xerolycosa) acrocentric chromosomes. The sex chromosome system was X1X20 in all species. The sex chromosomes of T. lutetiana and X. nemoralis showed unusual behaviour during late diplotene, namely temporary extension due to decondensation. C-banding technique revealed a small amount of constitutive heterochromatin at the centromeric region of the chromosomes. Two pairs of autosomes bore terminal NORs. Differences in karyotypes among Arctosa species indicate that the evolution of the karyotype in this genus involved autosome translocations and size changes in the sex chromosomes. Based on published results and those recorded in this study it is suggested that the ancestral male karyotype of the superfamily Lycosoidea consisted of 28 acrocentric chromosomes. and Petr DOLEJŠ, Tereza KOŘÍNKOVÁ, Jana MUSILOVÁ, Věra OPATOVÁ, Lenka KUBCOVÁ, Jan BUCHAR, Jiří KRÁL.
