The effects of chilling treatment (4 °C) under low irradiance, LI (100 μmol m-2 s-1) and in the dark on subsequent recovery of photosynthesis in chilling-sensitive sweet pepper leaves were investigated by comparing the ratio of quantum yields of photosystem (PS) 2 and CO2 assimilation, ΦPS2/ΦCO2, measured in normal air (21 % O2, NA) and low O2-air (2% O2, LOA), and by analyzing chlorophyll (Chl) a fluorescence parameters. Chilling treatment in the dark had little effect on Fv/Fm and ΦPS2/ΦCO2, but it caused the decrease of net photosynthetic rate (PN) under saturating irradiance after 6-h chilling treatment, indicating that short-term chilling alone did not induce PS2 photoinhibition. Furthermore, photorespiration and Mehler reaction also did not obviously change during subsequent recovery after chilling stress in the dark. During chilling treatment under LI, there were obvious changes in Fv/Fm and ΦPS2/ΦCO2, determined in NA or LOA. Fv/Fm could recover fully in 4 h at 25 °C, and ΦPS2/ΦCO2 increased at the end of the treatment, as determined in both NA and LOA. During subsequent recovery, ΦPS2/ΦCO2 in LOA decreased faster than in NA. Thus the Mehler reaction might play an important role during chilling treatment under LI, and photorespiration was an important process during the subsequent recovery. The recovery of PN under saturating irradiance determined in NA and LOA took about 50 h, implying that there were some factors besides CO2 assimilation limiting the recovery of photosynthesis. From the progress of reduced P700 and the increase of the Mehler reaction during chilling under LI we propose that active oxygen species were the factors inducing PS1 photoinhibition, which prevented the recovery of photosynthesis in optimal conditions because of the slow recovery of the oxidizable P700. and X.-G. The function of chloroplast ferredoxin quinone reductase (FQR)-dependent flow was examined by comparing a wild type tobacco and a tobacco transformant (ΔndhB) in which the ndhB gene had been disrupted with their antimycin A (AA)-fed leaves upon exposure to chilling temperature (4 °C) under low irradiance (100 µmol m-2 s-1 photon flux density). During the chilling stress, the maximum photochemical efficiency of photosystem (PS) 2 (Fv/Fm) decreased markedly in both the controls and AA-fed leaves, and P700+ was also lower in AA-fed leaves than in the controls, implying that FQR-dependent cyclic electron flow around PS1 functioned to protect the photosynthetic apparatus from chilling stress under low irradiance. Under such stress, non-photochemical quenching (NPQ), particularly the fast relaxing NPQ component (qf) and the de-epoxidized ratio of the xanthophyll cycle pigments, (A+Z)/(V+A+Z), formed the difference between AA-fed leaves and controls. In the pursuit of knowledge on the biological behavior of Brazilian Atlantic Forest tree species, this study evaluated the susceptibility of the light-demanding species, Schinus terebinthifolia Raddi., Pseudobombax grandiflorum (Cav.) A. Robyns and Joannesia princeps Vell., and of the shade-tolerant species, Hymenaea courbaril L. var. stilbocarpa and Lecythis pisonis Camb, to photoinhibition and acclimation capacity. These species were first cultivated under two irradiance conditions, I20 (20% direct sunlight radiation) and I100 (all-sky or direct sunlight) and then transferred from I20 to I100. The effects of the sudden increase in light radiation intensity on photosynthetic activity were then evaluated through chlorophyll (Chl) fluorescence imaging, HPLC xanthophylls analysis, and cell membrane lipid peroxidation measurements. Light-demanding species were found to present a higher photochemical efficiency and higher acclimation capacity under high light irradiance than shade-tolerant species. The higher photoinhibition tolerance observed in light-demanding species was associated to their higher capacity for photochemical dissipation and dissipation of excess excitation energy via the xanthophyll cycle, leading to a lower ROS generation. Photosynthetic organs are often characterized by anthocyanins being accumulated either in the epidermal or in the mesophyll cells making these tissues to turn reddish-brown in colour. It has been hypothesized that these pigments protect underlying chloroplasts from light-stress because they absorb photons of the photosynthetically active waveband. However, the photoprotective role of anthocyanins has not been undoubtedly shown on a broad range of species. In this study, green and anthocyanic areas of leaves of Pelargonium × hortorum, the latter possessing variable levels of anthocyanins, were compared using pigment analysis and pulse amplitude modulated in vivo chlorophyll (Chl) fluorescence. Quenching analysis of the induction and dark relaxation curves of slow Chl fluorescence kinetics showed that at photoinhibitory conditions [by applying above-saturation light intensity of 1,600 μmol(quantum) m-2 s-1 white light at low (4°C) temperature], anthocyanic areas were at least equally sensitive to photoinhibition as green leaf areas. In fact, the level of photoinhibition tended to be proportional to the level of anthocyanin accumulation suggesting that this characteristic was indicative of the photoinhibitory risk. Brassinosteroids (BRs), an important class of plant steroidal hormones, play a significant role in the amelioration of various biotic and abiotic stresses. 24-epibrassinolide (EBR), an active brassinosteroid, was applied exogenously in different concentrations to characterize a role of BRs in tolerance of melon (Cucumis melo L.) to high temperature (HT) stress and to investigate photosynthetic performance of HT-stressed, Honglvzaocui (HT-tolerant) and Baiyuxiang (HTsensitive), melon variety. Under HT, Honglvzaocui showed higher biomass accumulation and a lower index of heat injury compared with the Baiyuxiang. The exogenous application of 1.0 mg L-1 EBR, the most effective concentration, alleviated dramatically the growth suppression caused by HT in both ecotypes. Similarly, EBR pretreatment of HTstressed plants attenuated the decrease in relative chlorophyll content, net photosynthetic rate, stomatal conductance, stomatal limitation, and water-use efficiency (WUE), as well as the maximal quantum yield of PSII photochemistry (Fv/Fm), the efficiency of excitation capture of open PSII center, the effective quantum yield of PSII photochemistry (ΦPSII), photochemical quenching coefficient, and the photon activity distribution coefficients of PSI (α). EBR pretreatment further inhibited the increase in intracellular CO2 concentration, leaf transpiration rate, minimal fluorescence of dark-adapted state, nonphotochemical quenching, thermal dissipation, and photon activity distribution coefficients of PSII. In the past decade, utilization of nanostructured materials has increased intensively in a wide range of applications. Titanium dioxide nanoparticles (TiO2 NPs), for instance, can be applied for the inactivation of various pathogens through photo-induced generation of reactive oxygen species. Although TiO2 NPs with high antimicrobial activity are of great importance, in practice, their phytotoxic effects have not yet been fully clarified. In this study, we investigated the potential phytotoxicity of TiO2 NPs on grapevine (Vitis vinifera L.) under field conditions. After foliar exposure, two particularly stress-sensitive parameters, photosynthetic function and the flavonol profile, were examined. Micro- and macroelement composition of the leaves was also studied by ICP-AES measurements. We found that TiO2 NPs significantly decreased the net CO2 assimilation and increased stomatal conductance, indicating metabolic (nonstomatal) inhibition of the photosynthesis. The effects of foliar spraying of the dithiocarbamate zineb on two cultivars of tomato grown in the field in a site with high ozone concentrations were studied by means of biomass assessment, antioxidant enzyme assays, lipid peroxidation, and chlorophyll fluorescence measurements. Zineb prevented the peroxidation of membrane lipids and decreased the activity of scavenging enzymes, which suggests that plants sprayed with zineb are subjected to lower oxidative stress than controls. The beneficial effects of zineb protection is the utilization of a larger fraction of absorbed radiant energy in photosynthesis and a larger fruit yield in plants of both cultivars. and Á. Gas exchange, photochemical efficiency, and leaf water potential (Ψl) of Salix matsudana (non-indigenous species), S. microstachya and S. gordejevii (indigenous species) were studied in Hunshandak Sandland, China. Ψl of all the three species decreased from 06:00 to 12:00, and increased afterwards. S. matsudana showed higher values of Ψl than others. Net photosynthetic rate (PN) and stomatal conductance (gs) of S. matsudana were the lowest among all, with the maximum PN at 10:00 being 75% of that of S. gordejevii. Compared with the indigenous species, the non-indigenous S. matsudana had also lower transpiration rate (E) and water use efficiency (WUE). The values of Fv/Fm in all the species were lower from 06:00 to 14:00 than those after 14:00, indicating an obvious depression in photochemical efficiency of photosystem 2 in both non-indigenous and native species. However, it was much more depressed in S. matsudana, the non-indigenous tree. PN was positively correlated to gs and negatively related to Ψl. Metabolite changes and senescence behaviour after mechanical phloem girdling were studied in leaf tissue of Quercus pubescens. Sugar accumulation is not only considered to be an important part of several developmental signalling pathways, but is also seen as one of the basic triggers for senescence induction, or at least an obligatory accessory phenomenon. Our survey showed that an accumulation of the soluble sugars, glucose and fructose, was not on its own obligatorily connected with the induction of leaf senescence, since no indication or even an onset of senescence could be observed during the course of the experiment. Instead, we observed an inhibition of leaf development with a decrease of photosynthesis and a slow-down of development in nearly all chlorophyll a fluorescence analysis parameters using the JIP-test. a1_To determine how the use of a given rootstock can influence the functioning of the photosynthetic apparatus of the scion under salt stress, the growth, gas exchange, photosystem II (PSII) efficiency, xanthophyll cycle, and chloroplast ultrastructure of nongrafted,
self-grafted, and pumpkin-grafted (hereafter referred to as rootstock-grafted) cucumber (Cucumis sativus L.) plants were investigated at day 15 after being treated with 90 mM NaCl. The reductions in plant growth of the rootstock-grafted plants were lower than those of the nongrafted and self-grafted plants under 90 mM NaCl. The net photosynthetic rate, stomatal conductance, maximal and effective quantum yield of PSII photochemistry, photochemical quenching coefficient, and effective quantum-use efficiency of PSII in the light-adapted state of the nongrafted and self-grafted plants were significantly decreased under 90 mM NaCl. However, these reductions were alleviated when the cucumber plants were grafted onto the pumpkin (Cucurbita moschata Duch.) rootstock. The intercellular CO2 concentrations were significantly increased in the nongrafted and self-grafted plants under 90 mM NaCl, whereas it was decreased in the rootstock-grafted plants. Nonphotochemical quenching (NPQ) and the deepoxidation state of the xanthophyll cycle were significantly increased under 90 mM NaCl, particularly in the rootstockgrafted plants, suggesting the rootstock-grafted plants had higher potential to dissipate excess excitation energy and reduce the probability of photodamage to PSII. Under 90 mM NaCl, the number of grana was reduced, the thylakoids were swollen, and starch granules accumulated in all plants.
