The mechanistic basis for protection of exogenous ascorbate against photoinhibition at low temperature was examined in leaves of rice (Oryza sativa L.). Exposure of intact leaves to chilling temperature resulted in a drastic decrease in the speed of development of non-photochemical fluorescence quenching (NPQ). This was related to the low temperature-imposed restriction on the formation of the fast relaxing component of NPQ (qf). Feeding with 20 mM ascorbate markedly increased the rate of qf development at chilling temperature due primarily to the enhanced rate of zeaxanthin (Z) formation. On the other hand, ascorbate feeding had no influence on photosystem 2 (PS2)-driven electron flow. The reduced state of the PS2 primary electron acceptor QA decreased in ascorbate-fed leaves exposed to high irradiance at chilling temperature owing to the increased Z-associated thermal energy dissipation in the light-harvesting antenna system of PS2. Furthermore, ascorbate feeding increased the photosynthetic apparatus of rice leaves to resist photoinhibition at low temperature. Our experiment was carried out in order to explore effects of plant growth regulators (PGR; thidiazuron, paclobutrazol, and ascorbic acid) on physiological traits of wheat genotypes under water surplus and deficit conditions. Study revealed that relative water content, membrane stability index, chlorophyll content, photosynthetic rate (PN), and maximal quantum yield of PSII improved with PGRs application across the genotypes both under irrigation and water stress. The response of HD 2733 genotype was more positive toward PGRs treatment as compared to other genotypes under water stress. Higher PN and chlorophyll contents were observed in HD 2987 followed by C 306 genotype under water-stress conditions. Moreover, Rubisco small subunit (SSU) expression was lower in wheat genotypes under water stress as compared to irrigated conditions. Application of PGRs led to upregulation of SSU under water stress, while no significant change was found in Rubisco level and activity under irrigated condition in dependence on PGRs treatments. Yield-related traits showed also significant reduction under water-stress conditions, while application of PGRs enhanced the yield and its components. In order to elucidate the drought resistance and high-yield mechanism of hybrid millet, we studied the influence of drought stress on the photosynthetic characteristics and dry matter accumulation. Our results revealed that drought stress caused lesser reduction in the net photosynthetic rate, maximal quantum yield of PSII photochemistry, excitation energy capture efficiency of PSII reaction centers and in the yield of hybrid millet compared to normal millet. When drought stress occurred in the jointing stage, the percentage decrease of PN, Fv/Fm, Fv′/Fm′, and the yield of Zhangzagu3 cultivar compared to control were 27.9%, 2.6%, 25. 5%, and 1.9%, respectively, the percentage decrease of Zhangzagu5 were 37.6%, 3.9%, 28.3%, and 16.7%, respectively, the decrease percentage of Datong29 were 60.1%, 6.4%, 4%, and 23.4%, respectively. Hybrid millet showed the similar reduction in the parameters referred above, when drought stress occurred at the heading stage, but the percentage decrease was much higher than that at the jointing stage. The growth response of Chlorella sorokiniana to certain irradiance, DO, and temperature demonstrated the possible causes of low productivity with this strain in outdoor cultures. The growth (biomass productivity) and chlorophyll fluorescence (Fv/Fm) were substantially reduced when the dissolved oxygen (above 200 % of air saturation) and temperature were elevated. and C. U. Influence of different phosphorus concentrations was studied in four rice varieties (Akhanphou, MTU1010, RP BIO 226, and Swarna) differing in their tolerance to low phosphorus. There was an increase in shoot and root dry mass with the increase in phosphorus concentration. At the low phosphorus concentration at both tillering and reproductive stages, Swarna, followed by Akhanphou, recorded maximum biomass for both roots and shoots, while the minimum was observed in RP BIO 226. Reduction in photosynthetic rate, stomatal conductance, transpiration rate, and internal CO2 concentration at low phosphorus concentrations were observed at both tillering and reproductive stages in all the genotypes. In low phosphorus, maximum photosynthetic rate was found in Swarna followed by Akhanphou. Phosphorus deficiency did not alter the maximum efficiency of PSII photochemistry, however, there was a reduction in effective PSII quantum yield, electron transport rate, and coefficient of photochemical quenching, while the coefficient of nonphotochemical quenching was higher in the low phosphorus-treated plants. Prolonged exposure to excessive energy and failure to utilize the energy in carbon-reduction cycle induced the generation of reactive oxygen species, which affected PSII as indicated by the fluorescence traits. The reduction was less severe in case of Swarna and Akhanphou. The activities of superoxide dismutase, peroxidase, and catalase increased in roots under low phosphorus concentration indicating that photoprotective mechanisms have been initiated in rice plants in response to phosphorus deficiency. Comparatively, Swarna and Akhanphou exhibited a higher biomass, higher photosynthetic rate, and better reactive oxygen species-scavenging ability which conferred tolerance under low phosphorus conditions., N. Low temperature significantly influences chloroplast development and chlorophyll (Chl) biosynthesis, so effect of coldness on Chl content and Chl fluorescence characteristics was investigated in C. bungeana (Chorispora bungeana Fisch. & C.A. Mey). The levels of transcript and protein of an enzymatic step during Chl biosynthesis in response to chilling (4°C) and freezing (-4°C) were also examined in this work. Significant reduction in total Chl content was observed, but the reduction was much less at 4°C than that at -4°C. Moreover, the maximal quantum efficiency of photosystem II (PSII) photochemistry, indicated by Fv/Fm, decreased in the first 12 h, but then started to increase and reached higher levels than the control at 24 h and 48 h at 4°C, but decreased continuously at -4°C. Whereas quantum yield of PSII (ΦPSII) showed no significant difference between the chilling-stressed and the control seedlings, at -4°C, ΦPSII was markedly reduced with the prolonged treatment. In general, there were no significant responses of photochemical quenching (qP) and non-photochemical quenching (NPQ) to cold treatment. Meanwhile, the full-length cDNA of NADPH:protochlorophyllide oxidoreductase (POR, EC 1.3.1.33) was isolated and termed CbPORB (GenBank Accession No. FJ390503). Its transcript and protein content only slightly declined at 4°C, but dramatically reduced at -4°C with the time. These results strongly suggest that CbPORB possesses certain resistant characteristics and is a major player in Chl biosynthesis process involved in plant growth and development of C. bungeana under cold environmental conditions. and Y. Photosynthetic activity of leaf disks from chlortoluron (2 µmol per plant) treated and non-treated non-transgenic and transgenic (PGF-6) tobacco plants was measured from 1 up to 21 d after treatment under greenhouse conditions. PGF-6 plants, expressing the fused rat cytochrome P4501A1/yeast reductase genes were used. PGF-6 plants were much more chlortoluron-resistant than control plants. In non-transgenic tobacco plants the electron transport flow to PQ pool was strongly inhibited 1 d after treatment with herbicide whereas it was still existing in PGF-6 plants although some reduction was observed. The quantum yield of photosystem 2 (ΦPS2) which is related to the quantum yield of whole-chain electron transfer was much more inhibited by chlortoluron than the primary PS2 photochemistry, measured by the ratio Fv/Fm. Lower PS2 activity was found for herbicide-treated non-transgenic plants up to the 9th day. Then it started to increase in both control and PGF-6 plants, but more rapidly in PGF-6 ones, and its values were near to the control level at the 21st d after chlortoluron treatment. and E. The combined effects of water stress (WS) and low irradiance (LI) on growth, photosynthesis, osmotic adjustment, and lipid peroxidation were studied in dragon spruce (Picea asperata Mast.) seedlings grown under two water treatments (well watered, 100 % of field capacity, and water stressed, 30 % of field capacity) and two irradiances (HI, 100 % of full sunlight and LI, 15 % of full sunlight). WS reduced growth, chlorophyll (Chl) a and b contents, net photosynthetic rate, transpiration rate, stomatal conductance, and effective quantum yield of photosystem 2 (Y) but increased free proline and malondialdehyde contents. LI increased Chl contents and decreased Y, photochemical quenching (qP), and non-photochemical quenching (qN) under both water treatments. A gradual reduction in leaf water potential (Ψleaf), net photosynthetic rate (PN), stomatal conductance, and transpiration rate was observed in two drought tolerant (C 306 and K 8027) and two susceptible (RW 893 and 899) genotypes subjected to water stress. The extent of reduction was lower in K 8027 and C 306 and higher in RW 893 and RW 899. Rewatering the plants after 5 d of stress restored PN and other gas exchange traits in all four cultivars. Water stress had no significant effect on variable to maximum fluorescence ratio (Fv/Fm) indicating that water stress had no effect on primary photochemistry of photosystem 2 (PS2). However, water stress reduced the efficiency of excitation energy transfer (F'v/F'm) and the quantum yield of electron transport (ΦPS2). The reduction was more pronounced in susceptible cultivars. Chlorophyll fluorescence kinetics was used to investigate the effect of 1,4-dithiothreitol (DTT) on the distribution of excitation energy between photosystem 1 (PS1) and photosystem 2 (PS2) in soybean leaves under high irradiance (HI). The maximum PS2 quantum yield (Fv/Fm) was hardly affected by the presence of DTT, however, photon-saturated photosynthesis was depressed distinctly. Photochemical efficiency of open PS2 reaction centres during irradiation (Fv'/Fm') was enhanced by about 30-40 % by DTT treatment, whereas photochemical quenching (qP) was depressed by about 40 % under HI. DTT treatment caused a 30 % decrease in allocation of excitation energy to PS1 under HI and a 20 % increase to PS2. An obvious shift in the balance of excitation energy distribution between photosystems was observed in DTT-treated leaves. Though high excitation pressure (1 - qP) resulted from DTT treatment, non-photochemical quenching (qN) was lower. DTT completely inhibited the formation of zeaxanthin and also distinctly depressed the state transition (qT). The shift in the balance of excitation distribution between the two photosystems induced by DTT was mainly due to the enhancement of excitation energy capture by PS2 antenna and the inhibition of state transition. It might be the shift in the balance between the two photosystems that mainly induced the depression of photosynthesis.
