A simple method of a bicolor (multicolor), fast-Fourier, PAM chlorophyll fluorometry has been developed to obtain fluorescence induction curves. Quantum yields of PSII photochemistry were determined with blue and red simultaneously applied pulsed measuring lights for three subsequent 20-min periods of dark-, light-adaptation under actinic light and dark recovery. Measuring lights were cross-combined with blue and red actinic lights and saturation pulses. Coefficients of chromatic divergence were calculated as a ratio of the quantum yields obtained by red measuring light to that obtained by blue measuring light. Adaptation of Ficus benjamina and Hordeum vulgare leaves under blue (but not red) actinic light resulted in the sufficient increase of chromatic divergence. In addition, fraction of active, non(photo)inhibited, PSII centers was shown to be dependent on the color of measuring light. The development of smartphones, specifically their cameras, and imaging technologies has enabled their use as sensors/measurement tools. Here we aimed to evaluate the applicability of a fast and noninvasive method for the estimation of total chlorophyll (Chl), Chl a, Chl b, and carotenoids (Car) content of soybean plants using a smartphone camera. Single leaf disc images were obtained using a smartphone camera. Subsequently, for the same leaf discs, a Chl meter was used to obtain the relative index of Chl and the photosynthetic pigments were then determined using a classic method. The RGB, HSB and CIELab color models were extracted from the smartphone images and correlated to Chl values obtained using a Chl meter and by a standard laboratory protocol. The smartphone camera was sensitive enough to capture successfully a broad range of Chl and Car contents seen in soybean leaves. Although there was a variation between color models, some of the proposed regressions (e.g., the S and b index from HSB and Lab color models and NRI [RGB model]) were very close to the Chl meter values. The effect of ultraviolet B radiation (UV-B) on cellular ultrastructure, chlorophyll (Chl), carotenoids, and total phenolics of Acrostichum danaeifolium gametophytes was analyzed. The control group of spores was germinated under standard conditions, while the test group of spores was germinated with additional UV-B for 30 min every day for 34 d. The cell characteristics were preserved in gametophytes irradiated with UV-B, but the number of starch grains increased in the chloroplasts and the more developed grana organization in contrast to the chloroplasts of the control group. Chl a content decreased, while Chl b content increased in the gametophytes cultivated with UV-B for 34 d. Contents of lutein and zeaxanthin decreased and trans-β-carotene concentration was enhanced in the gametophytes irradiated with UV-B. The content of total phenolic compounds increased in the gametophytes cultivated with UV-B. Therefore our data suggest that the gametophytes of A. danaeifolium, a fern endemic to the mangrove biome, were sensitive to enhancement of UV-B radiation at the beginning of their development and they exhibited alterations in their ultrastructure, pigment contents, and protective mechanisms of the photosynthetic apparatus, when exposed to this radiation., A. In this study, we hypothesized that colonization of olive trees (Olea europaea L.) with the arbuscular mycorrhizal fungus Rhizophagus irregularis could modify the profiles of rhizosphere microbial communities with subsequent effects on nutrient uptake that directly affects olive tree physiology and performance. In this context, a greenhouse experiment was carried out in order to study the effects of mycorrhizal colonization by R. irregularis on photosynthesis, pigment content, carbohydrate profile, and nutrient uptake in olive tree. After six months of growth, photosynthetic rate in mycorrhizal (M) plants was significantly higher than that of nonmycorrhizal plants. A sugar content analysis showed enhanced concentrations of mannitol, fructose, sucrose, raffinose, and trehalose in M roots. We also observed a significant increase in P, K, Ca, Mg, Zn, Fe, and Mn contents in leaves of the M plants. Anthropogenic activities and improper uses of phosphate fertilizers have led to an increase in cadmium concentrations in agricultural soils. Brassinosteroids are steroid hormones that are rapidly assimilated and metabolised with beneficial roles in physiological and biochemical processes in plants. Our aim was to ascertain whether exogenous treatment with 24-epibrassinolide (EBR) can mitigate the Cd toxicity, and whether this substance can reduce the Cd accumulation in plant tissues. Furthermore, the dose response to EBR was determined following exposure to Cd in Vigna unguiculata. The experiment was a completely randomised factorial design with two concentrations of Cd (0 and 500 μM) and three concentrations of EBR (0, 50, and 100 nM). Spraying plants exposed to Cd with EBR significantly reduced the concentrations of Cd and increased nutrient contents in all tissues. The EBR treatment caused significant enhancements in leaf, root, and total dry matter. Foliar application of EBR reduced the negative effects of Cd toxicity on chlorophyll fluorescence and gas exchange parameters. Pretreatment with EBR also increased contents of pigments in plants exposed to Cd, compared with the identical treatments without EBR. Cd elevated contents of oxidant compounds, inducing cell damages, while EBR significantly decreased the concentrations of these compounds. We confirmed that EBR mitigated the negative effects related to Cd toxicity, reduced the absorption and transport of Cd, and increased the contents of essential elements. Chloroplasts are commonly the site of the earliest abiotic injury visible in plant ultrastructure. In this study, six inbred lines of maize (Zea mays L.) were used to analyze changes in the ultrastructure of chloroplasts and related physiological parameters under conditions of drought stress simulated by 20% polyethylene glycol 6000 (-0.6 MPa) for two days. Chloroplasts of three maize lines proved to be more sensitive. They showed changes in the ultrastructure in response to drought, including damage of thylakoid membranes, an increase in the number and size of plastoglobuli, swelling of thylakoid membranes both stromal and granal, disorganization of the thylakoid membrane system, an obvious increase in the intrathylakoid space, and a decrease in the
length-to-width ratio and area of chloroplasts. In addition, the contents of malondialdehyde increased markedly in the sensitive lines. Contrary to the sensitive lines, stable structures and shapes of chloroplasts were observed in the drought-resistant lines; it could be considered as an advantage contributing to drought tolerance in the plants. Effíciency of the energy transformation for CO2 fixation (E), and kinetics of the initial 02-mediated electron transport of Spimlina platemis (Gom.) Geitl. and Chlorella vulgaris Beijerinck cells were measured after adaptation to various growth irradiances (7) by means of the delayed fluorescence (DF) induction curves. Maxima of the membrane potential expenses during induction period were observed at I half saturating oxygen evolution; they were shifted according to growth 1 remaining higher in Spirulina than in Chlorella. The alterations of absorbance and fluorescence spectra at 25 oC after adaptation to / demonstrated changes in composition of pigments of algae, created to compensate for the imbalance in radiation absorption between the two photosystems. For Spirulina cells, the value of E was higher after growing under low /, or under blue radiation absorbed mainly by photosystem (PS) 1 (400-500 nm) with excitation by yellow (570 nm) radiation. For Chlorella cells, it was also higher after growing under low I. Under such conditions the half-rise time for DP-phase of DF induction curve decreased, which reflected an acceleration of kinetics of the initial electron transport between photosystems. An opposite situation was observed with Spirulina cells grown under high I or yellow radiation, and Chlorella cells from high I. Enhancement of effective PS2/PS1 ratio associated with decrease of reaction centre (RC) 2/RCl stoichiometry may be a cause of the increase of E and high membrane energization under saturating I in algae adapted to low 1.
We tested whether cheap and quick chlorophyll (Chl) fluorescence can be used in ecophysiological field studies as proxies for
gas-exchange measurements. We measured net photosynthetic rate at saturating irradiance and ambient atmospheric CO2 concentrations (PNsat), maximum carboxylation rate (Vcmax), maximum quantum yield of PSII (Fv/Fm), the performance index (PIabs), leaf nitrogen (Narea), and carbon isotope discrimination (Δ13C) within four herbaceous species along two elevational gradients. We analysed the relationship between Chl fluorescence and gas-exchange parameters and their link to indirect assessment of plant performance via ecophysiological traits. Fv/Fm showed no relationship to PNsat and only weak relationships to Vcmax. PIabs was positively related to PNsat and Vcmax. PIabs, PNsat, and Vcmax were positively associated with Narea and negatively to Δ13C, whereas Fv/Fm showed no relationship to Narea and a positive to Δ13C. A field study was conducted with the aim to elucidate photosynthetic responses of five emmer hulled wheat (Triticum turgidum ssp. dicoccum) accessions to 30 (N-limited) and 100 kg(N) ha-1
(N-sufficient) conditions at control and drought stress (irrigation after 30-40% and 60-70% depletion of available soil water, respectively). Chlorophyll (Chl) a and Chl b concentrations of the emmer wheats remained unchanged but net photosynthetic rate and dry mass increased and decreased, respectively, when received a sufficient amount of N. Smaller drought-induced decreases in Chl concentration, membrane stability index, and dry mass were concomitant to a greater decrease in intercellular CO2 concentration of emmer compared to the durum (Triticum turgidum) and bread wheats (Triticum aestivum). The lack of negative effect of insufficient N on Chl concentration and dry mass of emmer wheat suggests that this type of wheat possesses an obvious potential for organic farming., M. Vaghar, P. Cotton (Gossypium hirsutum L.) yields are impacted by overall photosynthetic production. Factors that influence crop photosynthesis are the plants genetic makeup and the environmental conditions. This study investigated cultivar variation in photosynthesis in the field conditions under both ambient and higher temperature. Six diverse cotton cultivars were grown in the field at Stoneville, MS under both an ambient and a high temperature regime during the 2006-2008 growing seasons. Mid-season leaf net photosynthetic rates (PN) and dark-adapted chlorophyll fluorescence variable to maximal ratios (Fv/Fm) were determined on two leaves per plot. Temperature regimes did not have a significant effect on either PN or Fv/Fm. In 2006, however, there was a significant cultivar × temperature interaction for PN caused by PeeDee 3 having a lower PN under the high temperature regime. Other cultivars' PN were not affected by temperature. FM 800BR cultivar consistently had a higher PN across the years of the study. Despite demonstrating a higher leaf Fv/Fm, ST 5599BR exhibited a lower PN than the other cultivars. Although genetic variability was detected in photosynthesis and heat tolerance, the differences found were probably too small and inconsistent to be useful for a breeding program., W. T.
