In five genotypes of cowpea (Vigna unguiculata), the influence of salicylic acid (SA) on photosynthetic activity and biochemical constituents including peroxidase activity at the genotypic level was determined. After SA treatment the total free sugar content increased in IFC 8401 and IGFRI 450 genotypes, whereas the content of total leaf soluble proteins decreased significantly in IFC 902. The high chlorophyll (Chl) (a + b) content in IFC 902 showed a good correlation with the net photosynthetic rate (PN), as in this genotype a significant increase in PN was found after the SA treatment. and Amaresh Chandra,, R. Anthropogenic activities and improper uses of phosphate fertilizers have led to an increase in cadmium concentrations in agricultural soils. Brassinosteroids are steroid hormones that are rapidly assimilated and metabolised with beneficial roles in physiological and biochemical processes in plants. Our aim was to ascertain whether exogenous treatment with 24-epibrassinolide (EBR) can mitigate the Cd toxicity, and whether this substance can reduce the Cd accumulation in plant tissues. Furthermore, the dose response to EBR was determined following exposure to Cd in Vigna unguiculata. The experiment was a completely randomised factorial design with two concentrations of Cd (0 and 500 μM) and three concentrations of EBR (0, 50, and 100 nM). Spraying plants exposed to Cd with EBR significantly reduced the concentrations of Cd and increased nutrient contents in all tissues. The EBR treatment caused significant enhancements in leaf, root, and total dry matter. Foliar application of EBR reduced the negative effects of Cd toxicity on chlorophyll fluorescence and gas exchange parameters. Pretreatment with EBR also increased contents of pigments in plants exposed to Cd, compared with the identical treatments without EBR. Cd elevated contents of oxidant compounds, inducing cell damages, while EBR significantly decreased the concentrations of these compounds. We confirmed that EBR mitigated the negative effects related to Cd toxicity, reduced the absorption and transport of Cd, and increased the contents of essential elements. In plants exposed to Cd, the most apparent dose response was found for 100 nM EBR, with beneficial repercussions on growth, gas exchange, primary photosynthetic processes, and photosynthetic pigments, which were intrinsically connected to lower production of oxidant compounds and cell damage., L. The influence of increased solar UV-B radiation on the photosynthetic characteristics in cowpea seedlings (Vigna unguiculata) grown at optimal (Mgs) and low (Mgd) Mg levels were studied. Both higher UV-B and Mgd treatments caused significant drops of photochemical activities and net CO2 uptake rates (PN). Yet the UV-B-induced decrease in the photosynthetic efficiency was lesser in Mgd seedlings. The leaf Chl a fluorescence measurements proved that after receiving an enhanced UV-B radiation these seedlings showed a significant enhancement in their variable parts. The PSM oscillation of slow fluorescence kinetics was remarkably altered by both treatments. The PN also followed a typical inhibitory pattern as seen in photochemical activities. Concentrations of several chloroplast proteins in trifoliate leaves were significantly reduced by Mgd treatment and unaffected by the other two treatments. Whereas the contents of 43-47 kDa polypeptides in primary leaves were markedly reduced with a maximal effect in Mgd seedlings, no major difference was noted for combined stress. and A. Premkumar, G. The kinetics and other characteristics of nitrate reductase (NR, EC 1.6.6.1) in cowpea [Vigna unguiculata (L.) Walp.] seedlings irradiated with biologically effective UV-B radiation (280-320 nm, 3.2 W m-2 s-1) were recorded. The in vivo and in vitro NR activities were inhibited by 34 and 41 % under UV-B treatment, respectively. Both Vmax and Km for the substrate were enhanced by UV-B radiation. The Km for nitrate increased from 1.2 to 1.7 mM after the UV-B irradiation. The change in Km for NADH was from 0.12 to 0.17 mM. The increases in Km indicate that UV-B radiation seriously changes the topology of NR, particularly with respect to the nitrate and NADH binding sites. The rate of NR turnover indicates the extent of damage inflicted by UV-B radiation on the nitrate metabolism. The half-life (t1/2) of NR was reduced from 7 to 4 h in the UV-B treated seedlings. Cowpea [Vigna unguiculata (L.) Walp. cv. Co 4] seedlings were subjected to a weighted irradiance of 3.2 W m-2 s-1 of biologically effective ultraviolet-B radiation (UV-B, 280-320 nm) and the changes in the kinetic and other characteristics of nitrite reductase (NiR) were recorded. The activity of NiR was hampered by 19 % under UV-B irradiation compared to the control. The UV-B treated plants required higher concentrations of nitrate for the induction of NiR synthesis than the controls. The NiR activity decay kinetics showed that the UV-B treatment significantly lowers the t1/2 of the enzyme, thereby indicating a reduced rate of enzyme turnover. The comparison of kinetic characteristics of nitrate reductase (NR) and NiR under UV-B treatment showed that NiR was not so sensitive to UV-B radiation as NR. As shown by enzyme turnover rates, NiR extracted from plants irradiated by UV-B in situ was less sensitive to UV-B radiation than the enzyme extract subjected to in vitro UV-B irradiation. Characterisation of proteases degrading ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO, EC: 4.1.1.39) was studied in the cowpea leaf during monocarpic senescence 3 and 9 d after flowering (DAF), representing early and mid pod fill. The stage at 3 DAF coincided with decrease in the metabolic parameters characterising senescence, i.e., contents of total soluble proteins, RuBPCO, and leaf nitrogen. At 9 DAF, there was a decline in total soluble proteins and an appearance of a 48 kDa cysteine protease. Characterisation of the proteases was done using specific inhibitors. Subcellular localisation at 3 DAF was studied by following the degradation of RuBPCO large subunit (LSU) in the vacuole lysates using immunoblot analyses. Cysteine proteases played a predominant role in the degradation of RuBPCO LSU at the crude extract level. At 9 DAF, expression of cysteine protease isoforms was monitored using polyclonal antibodies against papain and two polypeptides of molecular masses 48 and 35 kDa were observed in the vacuole lysates.
