The development of smartphones, specifically their cameras, and imaging technologies has enabled their use as sensors/measurement tools. Here we aimed to evaluate the applicability of a fast and noninvasive method for the estimation of total chlorophyll (Chl), Chl a, Chl b, and carotenoids (Car) content of soybean plants using a smartphone camera. Single leaf disc images were obtained using a smartphone camera. Subsequently, for the same leaf discs, a Chl meter was used to obtain the relative index of Chl and the photosynthetic pigments were then determined using a classic method. The RGB, HSB and CIELab color models were extracted from the smartphone images and correlated to Chl values obtained using a Chl meter and by a standard laboratory protocol. The smartphone camera was sensitive enough to capture successfully a broad range of Chl and Car contents seen in soybean leaves. Although there was a variation between color models, some of the proposed regressions (e.g., the S and b index from HSB and Lab color models and NRI [RGB model]) were very close to the Chl meter values. The effect of ultraviolet B radiation (UV-B) on cellular ultrastructure, chlorophyll (Chl), carotenoids, and total phenolics of Acrostichum danaeifolium gametophytes was analyzed. The control group of spores was germinated under standard conditions, while the test group of spores was germinated with additional UV-B for 30 min every day for 34 d. The cell characteristics were preserved in gametophytes irradiated with UV-B, but the number of starch grains increased in the chloroplasts and the more developed grana organization in contrast to the chloroplasts of the control group. Chl a content decreased, while Chl b content increased in the gametophytes cultivated with UV-B for 34 d. Contents of lutein and zeaxanthin decreased and trans-β-carotene concentration was enhanced in the gametophytes irradiated with UV-B. The content of total phenolic compounds increased in the gametophytes cultivated with UV-B. Therefore our data suggest that the gametophytes of A. danaeifolium, a fern endemic to the mangrove biome, were sensitive to enhancement of UV-B radiation at the beginning of their development and they exhibited alterations in their ultrastructure, pigment contents, and protective mechanisms of the photosynthetic apparatus, when exposed to this radiation., A. The short-term acclimation (10-d) of Norway spruce [Picea abies (L.) Karst] to elevated CO2 concentration (EC) in combination with low irradiance (100 μmol m-2 s-1) resulted in stimulation of CO2 assimilation (by 61 %), increased total chlorophyll (Chl) content (by 17 %), significantly higher photosystem 2 (PS2) photochemical efficiency (Fv/Fm; by 4 %), and reduced demand on non-radiative dissipation of absorbed excitation energy corresponding with enhanced capacity of photon utilisation within PS2. On the other hand, at high cultivation irradiance (1 200 μmol m-2 s-1) both Norway spruce and spring barley (Hordeum vulgare L. cv. Akcent) responded to EC by reduced photosynthetic capacity and prolonged inhibition of Fv/Fm accompanied with enhanced non-radiative dissipation of absorbed photon energy. Norway spruce needles revealed the expressive retention of zeaxanthin and antheraxanthin (Z+A) in darkness and higher violaxanthin (V) convertibility (yielding even 95 %) under all cultivation regimes in comparison with barley plants. In addition, the non-photochemical quenching of minimum Chl a fluorescence (SV0), expressing the extent of non-radiative dissipation of absorbed photon energy within light-harvesting complexes (LHCs), linearly correlated with V conversion to Z+A very well in spruce, but not in barley plants. Nitrogen (N) starvation resulted in degreening, inhibition of photosynthetic oxygen evolution and dark respiration, reduced survival, and increased age-specific mortality in both Chlorella fusca and Chlorella vulgaris. Analysis of in vivo chlorophyll (Chl) fluorescence induction kinetics revealed the presence of N-starvation-induced changes at the level of degreened thylakoids in both species. These changes included decreased yield of the photochemistry of photosystem 2 (PS2), and a declined photosynthetic efficiency. Synthesis of secondary carotenoids represented a biochemical change in carotenogenesis that had a photoprotective effect in degreened C. fusca. This inferred photoprotection was reflected in the delayed inhibition of oxygen evolution and improved survival of C. fusca under N-starvation. The effect was further elucidated by comparison with C. vulgaris which was not able to synthesize secondary carotenoids under the same conditions.
7-day soil drought followed by 7-day rehydration was applied to potted German chamomile (Chamomilla recutita) plants at the beginning of their generative stage. Plants of a wild type (WT), plus two diploid (2n) and two tetraploid (4n) genotypes were studied, in order to examine the alterations in chlorophyll (Chl) and carotenoids (Car) contents, and chlorophyll fluorescence (CF) parameters during water shortage and rehydration. The fresh mass of the anthodia after the recovery was also studied.
WT plants adjust better to water stress than modern breeding genotypes, because drought resulted in the low fall in leaf water content of WT, the lowest decrease in the fresh mass of its anthodia (a 41% decrease from the control), and the most elastic response of the photosynthetic apparatus. 4n C11/2 strain plants suffered from the highest reduction in anthodia yield (87%), and had the lowest constitutive pigment contents. It was also the only genotype which revealed nontypical alterations in various CF parameters obtained on a dark- and light-adapted leaf. During drought, a big increase was noticed in minimal, maximal, and variable fluorescence of PSII reaction centres in the dark- adapted (F0, Fm and Fv, respectively), and in the light-adapted state (F0', Fm' and Fv')., It was accompanied by the biggest decline in linear electron transport rate (ETR), quantum efficiency of PSII electron transport (ΦPSII) and photochemical quenching coefficient (qP). These alterations were prolonged to the stage when the normal leaf water content was retained. On the contrary, C6/2 strain plants had the highest constitutive Chl and Car contents, which additionally increased after rehydration, similarly to the values of F0, Fm and Fv, which reflects the high photosynthetic potential of this genotype. It was accompanied by the relatively high yield of its anthodia after drought. Considering the drop in the yield triggered by drought, it seems to be the only parameter which may be linked with the ploidy level.
Although the yield formation of chamomile strains cannot simply be estimated by CF assay, this technique may serve as an additional tool in the selection of plants to drought. The following circumstances should be submitted; namely: measurement at the proper developmental stage of plants, in different water regimes, and an analysis of various CF parameters. In five genotypes of cowpea (Vigna unguiculata), the influence of salicylic acid (SA) on photosynthetic activity and biochemical constituents including peroxidase activity at the genotypic level was determined. After SA treatment the total free sugar content increased in IFC 8401 and IGFRI 450 genotypes, whereas the content of total leaf soluble proteins decreased significantly in IFC 902. The high chlorophyll (Chl) (a + b) content in IFC 902 showed a good correlation with the net photosynthetic rate (PN), as in this genotype a significant increase in PN was found after the SA treatment. and Amaresh Chandra,, R. During the development of the globular embryos via heart-shaped, torpedo-shaped, and cotyledonary embryos into plantlets, contents of chlorophyll (Chl) a and b and carotenoids, and activity of ribulose-1,5-bisphosphate carboxylase (RuBPC, EC 4.1.1.39) were investigated. In the solid cultivation (SC) the contents of Chl a, Chl b, Chl a/b, and total pigments increased up to plantlet stage. In the liquid cultivation (LC), contents of Chl a, Chl b, and total pigments increased till the torpedo-shaped stage, but decreased with the further development up to plantlets stage. During SC, RuBPC activity increased up to the torpedo-shaped embryo stage, but in the LC RuPBC activity increased continuously with the progress in the developmental stages. Over last decades, several studies have been focused on
short-term high light stress in lichens under laboratory conditions. Such studies reported a strong photoinhibition of photosynthesis accompanied by a partial photodestruction of PSII, involvement of photoprotective mechanisms, and resynthetic processes into gradual recovery. In our paper, we applied medium [800 μmol(photon) m-2 s-1] light stress to induce negative changes in PSII funcioning as well as pigment and glutathione (GSH) content in two Antarctic fruticose lichen species. Chlorophyll (Chl) fluorescence parameters, such as potential and effective quantum yield of photosynthetic processes and fast transients (OJIP) recorded during high light exposition and recovery, revealed that Usnea antarctica was less susceptible to photoinhibition than U. aurantiaco-atra. This might be supported by a more pronounced high light-induced reduction in Chl a and b contents in U. aurantiaco-atra compared with U. antarctica. In both experimental species, total GSH showed an initial increase during the first 30-40 min of high light treatment followed by a decrease (60 min) and an increase during dark recovery. Full GSH recovery, however, was not finished in U. aurantiaco-atra even after 5 h indicating lower capacity of photoprotective mechanisms in the species. OJIP curves showed high light-induced decrease in both species, however, the recovery of the OJIPs shape to pre-photoinhibitory values was faster and more apparent in U. antarctica than in U. aurantiaco-atra. Net photosynthetic rate, stomatal conductance, ratio of sub-stomatal to atmospheric CO2 concentration, transpiration rate, and water use efficiency changed significantly and assimilation capacity dropped continuously along the salinization and alkalinization process in the afternoon. Assimilation capacity of L. chinensis leaf correlated negatively with the degree of salinization and alkalinization. The photosynthetic characteristics of L. chinensis determined its community formation. By changing the ratio of chlorophyll a/b in leaves and accumulating soluble saccharides in rhizome, L. chinensis could adapt to the saline-alkali condition. and L. X. Shi, J. X. Guo.
Changes in the content of pigments and rate of photosynthesis in Azolla microphylla Kaulf. fronds were measured during growth under solar and ultraviolet-C (UV-C) supplemented solar radiation. Maximum content of total chlorophyll (Chl) was observed on the 13th day (termination of the experiment) of treatment in both control and treated plants. The treated plants had significantly lower total Chl and carotenoid contents than the control plants during the 1st day of growth. After the 4th day of exposure to UV-C supplemented solar radiation, the Chl and carotenoids accumulation increased in treated plants, so that the pigment concentration in the treated fronds was nearer to the control values after the 13th day of treatment. Significant increase in UV absorbing pigments, anthocyanins, and flavonoids was observed at the 13th day of treatment. In spite of the roughly similar photosynthetic pigment concentration, the photosynthetic activity measured as the rate of electron transport at photosystem 2 was only 65 % of the control values after 13 d of UV-C exposure. and M.
