412. K13 propeller domain mutations and pfmdr1 amplification in isolates of Plasmodium falciparum collected from Thai-Myanmar border area in 2006-2010

- The K13 propeller domain mutation and pfmdr1 amplification have been proposed as useful molecular markers for detection and monitoring of artemisinin resistant Plasmodium falciparum Welch, 1897. Genomic DNA isolates of P. falciparum was extracted from 235 dried blood spot or whole blood samples collected from patients with uncomplicated falciparum malaria residing in areas along the Thai-Myanmar border during 2006-2010. Nested polymerase chain reaction (PCR) and sequencing were performed to detect mutations in K13 propeller domain of P. falciparum at codon 427-709. Pfmdr1 gene copy number was determined by SYBR Green I real-time PCR. High prevalence of pfmdr1 multiple copies was observed (42.5% of isolates). The presence of K13 mutations was low (40/235, 17.2%). Seventeen mutations had previously been reported and six mutations were newly detected. The C580Y was found in two isolates (0.9%). The F446I, N458Y and P574L mutations were commonly detected. Seven isolates had both K13 mutation and pfmdr1 multiple copies. - The aim of this study was to characterize karyotypes of central European spiders of the genera Arctosa, Tricca, and Xerolycosa (Lycosidae) with respect to the diploid chromosome number, chromosome morphology, and sex chromosomes. Karyotype data are reported for eleven species, six of them for the first time. For selected species the pattern in the distributions of the constitutive heterochromatin and the nucleolar organizer regions (NORs) was determined. The silver staining technique for detecting NORs of lycosid spiders was standardized. The male karyotype consisted of 2n = 28 (Arctosa and Tricca) or 2n = 22 (Xerolycosa) acrocentric chromosomes. The sex chromosome system was X1X20 in all species. The sex chromosomes of T. lutetiana and X. nemoralis showed unusual behaviour during late diplotene, namely temporary extension due to decondensation. C-banding technique revealed a small amount of constitutive heterochromatin at the centromeric region of the chromosomes. Two pairs of autosomes bore terminal NORs. Differences in karyotypes among Arctosa species indicate that the evolution of the karyotype in this genus involved autosome translocations and size changes in the sex chromosomes. 420. Key factors affecting the predation risk on insects on leaves in temperate floodplain forest

- The predation on insects on leaves was measured by direct observation using live larvae of Calliphora vicina (Diptera: Calliphoridae) as bait placed on 15 common species of woody plants in a floodplain forest in the temperate region. The predation rate was measured in terms of the proportion of the larvae that were missing or had been attacked after 30 min of exposure on leaves. Despite the fact that the important predators differed during the course of a season, the most frequently recorded predators were birds and ants and less frequently recorded wasps and spiders. Analysis of the pattern in the distribution of the attacks confirmed that it is best described by a negative binomial distribution, which corresponds to a clumped dispersal of predation. Based on the results of the best-fitted generalized additive model, we could not reject an equal probability of attacks on the different species of woody plants. Further, predation at the forest edge was notably higher than in the forest interior. The model also predicted marked variations in the incidence of attacks during the course of a day and a season, with the attacks occurring mainly in three periods during the year and two during the day.
