Because nutrients accrued during larval stages represent the major limiting factor for egg production, the use of adult feeding to enhance the reproductive output in moths is considered to be largely weight-dependent. It is hypothesized, however, that feeding by adults could be adaptive and an effective means of increasing their reproductive success. In order to test this, the calling behaviour of Spodoptera littoralis females that differed in body weight and whether they had fed or not were recorded. Two experiments were carried out. In the first, the calling behaviour of food-deprived females of different body weights was recorded. A strong positive correlation was found between body weight at emergence and the total duration of calling of females on the second to the fifth night after emergence. In the second experiment, groups of female moths that varied in body weight were given access to water or sucrose. Feeding on sucrose significantly reduced the pre-calling period and increased the total time spent calling on the six nights after emergence. The increase in time spent calling associated with ingesting sucrose were proportionately similar for both small and large females, implying that feeding by adults can result in an increase in the time spent calling by moths irrespective of larval nutritional status. Female longevity was also correlated with moth weight at emergence and/or sucrose availability. It is concluded that it is advantageous for female S. littoralis to be large and/or have access to sucrose-rich food in the adult stage as they can spend more time attracting a mate, which increases their chances of mating in early adult life, and their longer adult life may indirectly result in an increase in fecundity., Medhat M. This study determined the effect of larval density-dependent competition for food on development and adult fitness in Sesamia nonagriodes Lef. (Lepidoptera: Noctuidae). Different numbers (5, 10, 15, 20 and 30 individuals) of larvae of the pink stalk borer were reared on a constant amount of food. Although crowding during the immature stages did not significantly increase mortality, it prolonged the larval developmental period and resulted in reduced pupal weight. Females were more adversely affected by high density than males, resulting in lighter females, indicating that female growth is more sensitive to density. The fecundity of the adults reared in the various larval crowding treatments was analysed. Total female fecundity was correlated negatively with increasing larval density. The effects of crowding on fecundity were not caused by the reduced pupal weight, indicating that food shortage during larval development may affect adult traits. Female longevity was negatively affected by density and positively related to pupal weight. Thus, larval density may affect the allocation of food resources and adult fitness. We conclude that crowding related changes during larval development directly affect larval life and reduce female fitness.
In order to determine when during the development of H. armigera CYP6B6 is expressed, we measured the width of the head capsule, the length of the body and weight of the different life stages of the cotton bollworm reared under laboratory condition. The larvae took about 20 d to complete their development under these conditions, the pre-pupae about 4 d, pupae about 9 d, and the adults lived for about 9 d and the eggs took about 4 d to hatch. Knowing this it was possible to determine the pattern of cytochrome P450 CYP6B6 expression at different stages in the life cycle of H. armigera using real-time quantitative polymerase chain reaction (RT-qPCR). The CYP6B6 gene was expressed in all the developmental stages of the cotton bollworm. The results indicate that the level of expression was lower in the egg, pre-pupal, pupal and adult stages than in larvae. The level of expression of CYP6B6 decreased from the 1st to the 3rd instar larva, with the lowest level recorded in the 3rd instar larvae and then increased reaching its highest value in 6th instar larvae. A similar trend in the expression of CYP6B6 was detected using immunohistochemistry. As the results show, more efficient control of the cotton bollworm can be achieved if insecticides are applied when the larvae of H. armiger are in the 3rd instar, because at that stage the eat little food and there is a low level of detoxifying enzymes in the larvae., Xuetao Zhang ... The host recognition and acceptance behaviour of two braconid larval parasitoids (Cotesia sesamiae and C. flavipes) were studied using natural stemborer hosts (i.e., the noctuid Busseola fusca for C. sesamiae, and the crambid Chilo partellus for C. flavipes) and a non-host (the pyralid Eldana saccharina). A single larva was introduced into an arena together with a female parasitoid and the behaviour of the wasp recorded until it either stung the larva or for a maximum of 5 min if it did not sting the larva. There was a clear hierarchy of behavioural steps, which was similar for both parasitoid species. In the presence of suitable host larvae, after a latency period of 16-17 s, the wasp walked rapidly drumming the surface with its antennae until it located the larva. After location and antennal examination of the host, which lasted 60-70 s and 30 s, respectively, the parasitoid inserted its ovipositor. Stinging that resulted in successful oviposition usually lasted 5-6 s. In the presence of non-host larvae, the latency period was between 25-70 s, and parasitoids spent significantly more time walking and antennal drumming on larvae without ovipositing. It is likely that these two parasitoid species use their antennae for host recognition, and both their antennae and tarsi for final acceptance of a host for oviposition. In both C. sesamiae and C. flavipes tactile and contact-chemoreception stimuli from the hosts seemed to play a major role in the decision to oviposit.
In Lepidoptera, larval dietary experience of volatile cues can induce adult preference for these cues. However, such induction may require several generations, depending in part on the degree of specialization of the insects. In a previous study, using species of noctuid stem borers with different diet breadths, namely the polyphagous Sesamia nonagrioides, the oligophagous Busseola fusca and monophagous Busseola nairobica, it was shown that in S. nonagrioides, one generation was enough to induce a preference for vanillin in the resulting gravid females, whereas even two generations failed to induce a response in adults of Busseola spp. In this study, we checked whether a higher number of generations of exposure to a vanillin-enriched medium could induce a significant olfactory preference for this medium in the species B. fusca and B. nairobica, which have narrower host-plant utilization ranges than S. nonagrioides. Larvae were reared to the adult stage on an artificial diet enriched with vanillin for periods of from 2 to 7 or 8 generations, followed by two-choice tests on gravid females using a Y-tube olfactometer. The results showed different responses according to the species: for the oligophagous B. fusca, there was no significant effect of the conditioning, while a significant preference for the odours emitted by this new vanillin-enriched diet was recorded for the polyphagous S. nonagrioides and monophagous B. nairobica, but after different numbers of generations. In conclusion, different diet breadths seem to be associated with different levels of plasticity in the odour preference of adults. Insect cellular immune reaction, which generally includes phagocytosis, encapsulation and nodule formation, is achieved by hemocytes circulating in insect haemolymph. The shift of hemocytes from the normal phase to the adhering phase is an important process in the cellular immune reaction, which includes the attachment of hemocytes to foreign surfaces or other hemocytes via adhesion factors. Neuroglian is one of the adhering factors associated with encapsulation in Manduca sexta and Drosophila melanogaster. Here we studied the localization of neuroglian (MsNrg) in Mythimna separata and its functional role in the cellular immune reaction. The distribution of MsNrg mRNA between hemocyte populations was determined using real-time quantitative reverse transcription PCR and in situ hybridization, which revealed that MsNrg was highly expressed in adhering hemocytes, especially in plasmatocytes. Unexpectedly, the transcript was observed as well in non-adhering hemocytes, implying neuroglian has a function in non-adhering hemocytes. Moreover, we showed that the amount of MsNrg mRNA was not changed by injections of either biotic or abiotic non-selves. Fewer latex beads were fully encapsulated by hemocytes in larvae treated with MsNrg double-stranded RNA than in control larvae, but their ability to achieve phagocytosis and nodule formation remained unchanged by the MsNrg knockdown. Laboratory experiments were conducted to examine the effects of photoperiod and temperature on the pupation behaviour of the parasitoid, Microplitis mediator (Haliday) (Hymenoptera: Braconidae), parasitizing larvae of Mythimna separata Walker (Lepidoptera: Noctuidae). A combination of long photoperiod (14 + h L) and warm temperatures (20–24°C) caused parasitized caterpillars to climb to upper plant parts where the parasitoid produced a green, non-diapausing cocoon on a green leaf, initially retaining the dying caterpillar host as a protective covering. In contrast, short photoperiod (8–10 h L) and low temperature (16–18°C) induced host caterpillars to descend the plant where the parasitoid produced a brown, diapausing cocoon either hanging by silk from a senescing leaf or simply lying on the soil, but without any continued association with the host. Nocturnal pollinators such as moths have received less attention than diurnal insects. To elucidate whether nocturnal moths are important pollinators, we observed both the diurnal and nocturnal visitors to the flowers of Uncaria rhynchophylla (Rubiaceae) in a warm-temperate forest in central Japan. The diurnal visitors included various taxonomic groups (e.g., bees, hoverflies and butterflies). The nocturnal visitors were exclusively moths (Geometridae, Erebidae, Noctuidae and Crambidae). Pollen grains of U. rhynchophylla were attached to both diurnal and nocturnal visitors. Although diurnal flower visitors carried pollen grains of other plant species, nocturnal moths did not carry heterospecific pollen grains. When insect herbivores develop over many generations on the same plant species, their descendants may evolve physiological adaptations that enable them to develop more successfully on that plant species than naïve conspecifics. Here, we compared development of wild and lab-reared caterpillars of the cabbage moth, Mamestra brassicae, on a cultivar of cabbage Brassica oleracea (cv. Cyrus) and on a wild plant species, sorrel, Rumex acetosa, on which the wild strain had been collected and reared for two earlier consecutive generations. The lab strain had been reared on the same cabbage cultivar for more than 20 years representing > 200 generations. Survival to adult did not vary with strain or plant species. Both strains, however, developed significantly faster when reared on R. acetosa than B. oleracea. Pupae from the field strain were larger when reared on B. oleracea than on R. acetosa, whereas the identity of the plant species did not matter for the lab strain. Our results show that long-term rearing history on cabbage had little or no effect on M. brassicae performance, suggesting that some generalist herbivores can readily exploit novel plants that may be chemically very different from those on which they have long been intimately associated., Jeffrey A. Membrane-bound proteases from preparations of the midgut of 5th instar velvetbean caterpillars, Anticarsia gemmatalis (Hübner) were obtained by resuspension of the pellet obtained after 100,000 g centrifugation. As expected of trypsin-like proteases, they hydrolyzed casein and the synthetic substrates N-α-benzoyl-L-Arg-p-nitroanilidine (L-BApNA) and N-α-p-tosyl-L-Arg methyl ester (L-TAME). Higher activities were observed at 50°C, and at pH 8.5 and 8.0 for both synthetic substrates L-BApNA and L-TAME. The membrane-bound proteases were inhibited by EDTA, phenylmethan sulphonyl fluoride (PMSF), tosyl-L-lysine chloromethyl ketone (TLCK), benzamidine and aprotinin. TLCK and benzamidine were particularly active inhibitors. The KM-values obtained were 0.23 mM for L-BApNA and 92.5 µM for L-TAME. These results provide evidence for the presence of membrane-bound trypsin-like proteases in the midgut of the velvetbean caterpillar, a key soybean pest in warm climates. The interaction between A. gemmatalis digestive proteases and soybean protease inhibitors has potentially important consequences for soybean breeding programs.
