A simple method of a bicolor (multicolor), fast-Fourier, PAM chlorophyll fluorometry has been developed to obtain fluorescence induction curves. Quantum yields of PSII photochemistry were determined with blue and red simultaneously applied pulsed measuring lights for three subsequent 20-min periods of dark-, light-adaptation under actinic light and dark recovery. Measuring lights were cross-combined with blue and red actinic lights and saturation pulses. Coefficients of chromatic divergence were calculated as a ratio of the quantum yields obtained by red measuring light to that obtained by blue measuring light. Adaptation of Ficus benjamina and Hordeum vulgare leaves under blue (but not red) actinic light resulted in the sufficient increase of chromatic divergence. In addition, fraction of active, non(photo)inhibited, PSII centers was shown to be dependent on the color of measuring light. The development of smartphones, specifically their cameras, and imaging technologies has enabled their use as sensors/measurement tools. Here we aimed to evaluate the applicability of a fast and noninvasive method for the estimation of total chlorophyll (Chl), Chl a, Chl b, and carotenoids (Car) content of soybean plants using a smartphone camera. Single leaf disc images were obtained using a smartphone camera. Subsequently, for the same leaf discs, a Chl meter was used to obtain the relative index of Chl and the photosynthetic pigments were then determined using a classic method. The RGB, HSB and CIELab color models were extracted from the smartphone images and correlated to Chl values obtained using a Chl meter and by a standard laboratory protocol. The smartphone camera was sensitive enough to capture successfully a broad range of Chl and Car contents seen in soybean leaves. Although there was a variation between color models, some of the proposed regressions (e.g., the S and b index from HSB and Lab color models and NRI [RGB model]) were very close to the Chl meter values. The effect of ultraviolet B radiation (UV-B) on cellular ultrastructure, chlorophyll (Chl), carotenoids, and total phenolics of Acrostichum danaeifolium gametophytes was analyzed. The control group of spores was germinated under standard conditions, while the test group of spores was germinated with additional UV-B for 30 min every day for 34 d. The cell characteristics were preserved in gametophytes irradiated with UV-B, but the number of starch grains increased in the chloroplasts and the more developed grana organization in contrast to the chloroplasts of the control group. Chl a content decreased, while Chl b content increased in the gametophytes cultivated with UV-B for 34 d. Contents of lutein and zeaxanthin decreased and trans-β-carotene concentration was enhanced in the gametophytes irradiated with UV-B. The content of total phenolic compounds increased in the gametophytes cultivated with UV-B. Therefore our data suggest that the gametophytes of A. danaeifolium, a fern endemic to the mangrove biome, were sensitive to enhancement of UV-B radiation at the beginning of their development and they exhibited alterations in their ultrastructure, pigment contents, and protective mechanisms of the photosynthetic apparatus, when exposed to this radiation., A. Winter wheat is a grass species widely planted in northern and central China, where the increase of aerosols, air pollutants and population density are causing significant reduction in solar irradiance. In order to investigate the adaptation of winter wheat (Triticum aestivum L., cv. Yangmai 13) to low irradiance conditions occurring in the downstream plain of the Yangtze River (China), plants were subjected to four solar irradiance treatments (100%, 60%, 40%, and 20% of environmental incident solar irradiance). Significant increases in chlorophyll (Chl) and xanthophyll (Xan) pigments, and decreases in Chl a/b and Xan/Chl ratios were observed in plants under low light. Light-response curves showed higher net photosynthetic rates (PN) in fully irradiated plants, that also showed a higher light-compensation point. Shaded plants maintained high values of minimal fluorescence of dark-adapted state (F0) and maximum quantum efficiency of PSII photochemistry (Fv/Fm) that assess a lower degree of photoinhibition under low light. Reduced irradiance caused decreases in effective quantum yield of PSII photochemistry (ΦPSII), electron transport rate (ETR), and nonphotochemical quenching coefficient (qN), and the promotion of excitation pressure of PSII (1 - qP). The activities of the antioxidant enzymes superoxide dismutase and peroxidase were high under reduced light whereas no light-dependent changes in catalase activity were observed. Thiobarbituric acid reactive species content and electrolyte leakage decreased under shaded plants that showed a lower photooxidative damage. The results suggest that winter wheat cv. Yangmai 13 is able to maintain a high photosynthetic efficiency under reduced solar irradiance and acclimates well to shading tolerance. The effect of arbuscular mycorrhizal fungi (AMF) inoculation and organic slow release fertilizer (OSRF) on photosynthesis, root phosphatase activity, nutrient acquisition, and growth of Ipomoea carnea N. von Jacquin ssp. fistulosa (K. Von Martinus ex J. Choisy) D. Austin (bush morning glory) was determined in a greenhouse study. The AMF treatments consisted of a commercial isolate of Glomus intraradices and a non-colonized (NonAMF) control. The OSRF was applied at 10, 30, and 100 % of the manufacturer's recommended rate. AMF plants had a higher net photosynthetic rate (PN), higher leaf elemental N, P, and K, and generally greater growth than NonAMF plants. Total colonization levels of AMF plants ranged from 27 % (100 % OSRF) to 79 % (30 % OSRF). Root acid phosphatase (ACP) and alkaline phosphatase (ALP) activities were generally higher in AMF than non-AMF plants. When compared to NonAMF at 100 % OSRF, AMF plants at 30 % OSRF had higher or comparable ACP and ALP activity, higher leaf elemental P, N, Fe, Cu, and Zn, and a greater PN (at the end of the experiment), leading to generally greater growth parameters with the lower fertility in AMF plants. We suggest that AMF increased nutrient acquisition from an organic fertilizer source by enhancing ACP and ALP activity thus facilitating P acquisition, increasing photosynthesis, and improving plant growth. and L. In this study, we hypothesized that colonization of olive trees (Olea europaea L.) with the arbuscular mycorrhizal fungus Rhizophagus irregularis could modify the profiles of rhizosphere microbial communities with subsequent effects on nutrient uptake that directly affects olive tree physiology and performance. In this context, a greenhouse experiment was carried out in order to study the effects of mycorrhizal colonization by R. irregularis on photosynthesis, pigment content, carbohydrate profile, and nutrient uptake in olive tree. After six months of growth, photosynthetic rate in mycorrhizal (M) plants was significantly higher than that of nonmycorrhizal plants. A sugar content analysis showed enhanced concentrations of mannitol, fructose, sucrose, raffinose, and trehalose in M roots. We also observed a significant increase in P, K, Ca, Mg, Zn, Fe, and Mn contents in leaves of the M plants. 7-day soil drought followed by 7-day rehydration was applied to potted German chamomile (Chamomilla recutita) plants at the beginning of their generative stage. Plants of a wild type (WT), plus two diploid (2n) and two tetraploid (4n) genotypes were studied, in order to examine the alterations in chlorophyll (Chl) and carotenoids (Car) contents, and chlorophyll fluorescence (CF) parameters during water shortage and rehydration. The fresh mass of the anthodia after the recovery was also studied.
WT plants adjust better to water stress than modern breeding genotypes, because drought resulted in the low fall in leaf water content of WT, the lowest decrease in the fresh mass of its anthodia (a 41% decrease from the control), and the most elastic response of the photosynthetic apparatus. 4n C11/2 strain plants suffered from the highest reduction in anthodia yield (87%), and had the lowest constitutive pigment contents. It was also the only genotype which revealed nontypical alterations in various CF parameters obtained on a dark- and light-adapted leaf. During drought, a big increase was noticed in minimal, maximal, and variable fluorescence of PSII reaction centres in the dark- adapted (F0, Fm and Fv, respectively), and in the light-adapted state (F0', Fm' and Fv')., It was accompanied by the biggest decline in linear electron transport rate (ETR), quantum efficiency of PSII electron transport (ΦPSII) and photochemical quenching coefficient (qP). These alterations were prolonged to the stage when the normal leaf water content was retained. On the contrary, C6/2 strain plants had the highest constitutive Chl and Car contents, which additionally increased after rehydration, similarly to the values of F0, Fm and Fv, which reflects the high photosynthetic potential of this genotype. It was accompanied by the relatively high yield of its anthodia after drought. Considering the drop in the yield triggered by drought, it seems to be the only parameter which may be linked with the ploidy level.
Although the yield formation of chamomile strains cannot simply be estimated by CF assay, this technique may serve as an additional tool in the selection of plants to drought. The following circumstances should be submitted; namely: measurement at the proper developmental stage of plants, in different water regimes, and an analysis of various CF parameters. The increase in F0 and F0', and the reduction in ETR, Fv'/Fm', ΦPSII and qP values in response to water deficit should be an indicator of the impairment of the photosynthetic apparatus through drought., and R. In five genotypes of cowpea (Vigna unguiculata), the influence of salicylic acid (SA) on photosynthetic activity and biochemical constituents including peroxidase activity at the genotypic level was determined. After SA treatment the total free sugar content increased in IFC 8401 and IGFRI 450 genotypes, whereas the content of total leaf soluble proteins decreased significantly in IFC 902. The high chlorophyll (Chl) (a + b) content in IFC 902 showed a good correlation with the net photosynthetic rate (PN), as in this genotype a significant increase in PN was found after the SA treatment. and Amaresh Chandra,, R. Anthropogenic activities and improper uses of phosphate fertilizers have led to an increase in cadmium concentrations in agricultural soils. Brassinosteroids are steroid hormones that are rapidly assimilated and metabolised with beneficial roles in physiological and biochemical processes in plants. Our aim was to ascertain whether exogenous treatment with 24-epibrassinolide (EBR) can mitigate the Cd toxicity, and whether this substance can reduce the Cd accumulation in plant tissues. Furthermore, the dose response to EBR was determined following exposure to Cd in Vigna unguiculata. The experiment was a completely randomised factorial design with two concentrations of Cd (0 and 500 μM) and three concentrations of EBR (0, 50, and 100 nM). Spraying plants exposed to Cd with EBR significantly reduced the concentrations of Cd and increased nutrient contents in all tissues. The EBR treatment caused significant enhancements in leaf, root, and total dry matter. Foliar application of EBR reduced the negative effects of Cd toxicity on chlorophyll fluorescence and gas exchange parameters. Pretreatment with EBR also increased contents of pigments in plants exposed to Cd, compared with the identical treatments without EBR. Cd elevated contents of oxidant compounds, inducing cell damages, while EBR significantly decreased the concentrations of these compounds. We confirmed that EBR mitigated the negative effects related to Cd toxicity, reduced the absorption and transport of Cd, and increased the contents of essential elements. The metabolic pathway of primary carbon fixation was studied in a peculiar pennate marine diatom, Haslea ostrearia (Bory) Simonsen, which synthesizes and accumulates a blue pigment known as "marennine". Cells were cultured in a semi-continuous mode under saturating [350 µmol(photon) m-2 s-1] or non-saturating [25 µmol(photon) m-2 s-1] irradiance producing "blue" (BC) and "green" (GC) cells, characterized by high and low marennine accumulation, respectively. Growth, pigment contents (chlorophyll a and marennine), 14C accumulation in the metabolites, and the carbonic anhydrase (CA) activity of the cells were determined during the exponential growth phase. Growth rate and marennine content were closely linked to irradiance during growth: higher irradiance increased both growth rate and marennine content. On the other hand, the Chl a concentration was lower under saturating irradiance. The distribution between the Calvin-Benson (C3) and β-carboxylation (C4) pathways was very different depending on the irradiance during growth. Metabolites of the C3 cycle contained about 70 % of the total fixed radioactivity after 60 s of incorporation into cells cultured under the non-saturating irradiance (GC), but only 47 % under saturating irradiance (BC). At the same time, carbon fixation by β-carboxylation was 24 % in GC versus about 41 % in BC, becoming equal to that in the C3 fixation pathway in the latter.
