In transgenic (TG) tomato (Lycopersicon esculentum Mill.) overexpressed ω-3 fatty acid desaturase gene (LeFAD7) was identified, which was controlled by the cauliflower mosaic virus 35S promoter and induced increased contents of unsaturated fatty acids in thylakoid membrane. Under chilling stress at low irradiance (4 °C, 100 µmol m-2 s-1) TG plants with higher linolenic acids (18: 3) content maintained a higher O2 evolution rate, oxidizable P700 content, and maximal photochemical efficiency (Fv/Fm) than wild type (WT) plants. Low temperature treatment for 6 h resulted in extensive changes of chloroplast ultrastructure: in WT plants most chloroplasts became circular, the number of amyloids increased, appressed granum stacks were dissolved, grana disappeared, and the number of grana decreased, while only a few grana were found in leaves of TG plants. Ozone (O3) is important air pollutant inducing severe losses of horticultural production. Cultivars of the same species, but with different leaf colors, may differ in their ozone sensitivity. However, it has not been clarified yet if different leaf coloration influences such a sensitivity. In this study, two purple-leafed and two green-leafed cultivars of Pakchoi were selected for ozone fumigation (240 ± 20 nmol mol-1, 09:00-16:00 h). Elevated O₃ decreased chlorophyll content, increased anthocyanin (Ant) content, damaged cell membrane integrity, enhanced antioxidative enzyme activities, depressed photosynthetic rate (PN) and stomatal conductance (gs), inhibited maximal quantum yield (Fv/Fm) and effective quantum yield [YII] of PSII photochemistry, and caused visible injury. Purple-leafed cultivars with higher Ant contents were more tolerant than green-leafed cultivars as indicated by lower relative enhancement in malondialdehyde content and lower relative losses in PN, gs, Fv/Fm, and YII. Leaves and other chlorophyllous tissues of plants often show transient or permanent anthocyanin coloration. The question of whether anthocyanins can function as effective light screens to modulate photosynthesis in plants was addressed by comparing photosynthetic responses in reddish-purple pods with those in green pods of the ornamental leguminous tree Bauhinia variegata. For these comparisons the actinic radiation employed was either red radiation (RR) which was poorly absorbed by anthocyanin or blue-green radiation (BGR) which was strongly absorbed by anthocyanin. Photon yields of photosystem 2 (PS2) photochemistry and photochemical chlorophyll fluorescence quenching coefficients (qp), measured over a range of photon flux densities (PFD) up to 1200 µmol m-2 s-1 at 23 °C and at five temperatures from 8 to 28 °C at a PFD of 260 µmol m-2 s-1, were almost identical in green pods irradiated with either RR or BGR and in purple pods irradiated with RR. However, qp values remained much higher in purple pods irradiated with BGR, e.g., 0.80 in BGR versus 0.29 in RR at a PFD of 1200 µmol m-2 s-1 at 23 °C, and 0.67 in BGR versus 0.28 in RR at a PFD of 260 µmol m-2 s-1 at 8 °C. The higher values of qp in BGR compared to RR indicated that photoabatement by anthocyanin allowed the first stable acceptor of PS2, QA, to be kept in a more oxidized state, thus decreasing the likelihood of photoinhibition. This was confirmed by demonstrating a lower susceptibility to photoinhibition in purple pods than in green pods in the sunlight, either naturally in pods on trees or in detached pods exposed to photoinhibitory conditions. The aim of this study was to evaluate the photochemistry of Luffa cylindrica (L.) Roem in fungal biocontrol interacting treatments. Healthy plants were infected with Pythium aphanidermatum before the biocontrol application. Biocontrol agents were selected in preliminary Petri-plate experiment evaluation against causative agent P. aphanidermatum. Photosynthetic performance traits were studied. We found that P. aphanidermatum infection caused significant reduction in photosynthetic performance, pigments, and in maximum quantum yield of primary photochemistry, photochemical quenching, and electron transport rate with increase in nonphotochemical quenching as compared with non-infected control. However, application of biocontrol agents substantially improved maximum quantum yield of PSII, performance index, and total content of photosynthetic pigments in infected plants. The low chlorophyll b mutant of high yield rice had a lower light-harvesting complex 2 content than the wild type. The stability of oxygen evolution side of photosystem 2 was only slightly lower. A lower photon absorption rate and a stronger xanthophyll cycle capacity of this mutant led to a higher endurance to strong irradiance and a lower photoinhibition as compared with the wild type rice. and Xinbin Dai ... The concentrations of photosynthetic pigments decreased in both chilling stressed species but the ratios of chlorophyll (Chl) a/b and total carotenoids (Car)/Chls were depressed only in faba bean. The contents of α+β carotene and lutein+lutein-5,6-epoxide remained unaffected in both species, but the de-epoxidation state involving the components of xanthophyll cycle increased in pea. Under chilling stress the photosynthetic electron transport associated with photosystem 2, PS2 (with and without the water oxidising complex) decreased in both plant species, the inhibition being higher in faba bean. The intrachloroplast quinone pool also decreased in both stressed species, yet an opposite trend was found for cytochrome b559LP. Under stress an increasing peroxidation of thylakoid acyl lipids was detected in pea, but higher protein/Chl ratio was detected in faba bean. Photoinhibition of photosynthesis was investigated in control (C) and chilling night (CN) leaves of grapevine under natural photoperiod at different sampling time in a day. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) and photosynthetic electron transport measurements. When the potential efficiency of photosystem (PS) 2, Fv/Fm was measured at midday, it markedly declined with significant increase of F0 in CN leaves. In isolated thylakoids, the rate of whole chain and PS2 activity were markedly decreased in CN leaves than control leaves at midday. A smaller inhibition of PS1 activity was also observed in both leaf types. Later, the leaves reached maximum PS2 efficiencies similar to those observed in the morning during sampling at evening. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the PS2 activity in both leaf types at midday. Thus CN enhanced inactivation on the acceptor side of PS2 in grapevine leaves. Quantification of the PS2 reaction centre protein D1 following midday exposure of leaves showed pronounced differences between C and CN leaves. The marked loss of PS2 activity in CN leaves noticed in midday samples was mainly due to the marked loss of D1 protein of the PS2 reaction centre. and M. Bertamini ... [et al.].
The degree of photoinhibition of sun and shade grown leaves of grapevine was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem 2 (PS2), Fv/Fm, markedly declined under high irradiance (HI) in shade leaves with less than 10 % of F0 level. In contrast, Fv/Fm ratio declined with about 20 % increase of F0 level in sun leaves. In isolated thylakoids, the rate of whole chain and PS2 activity in HI shade and sun leaves was decreased by about 60 and 40 %, respectively. A smaller inhibition of photosystem 1 (PS1) activity was also observed in both leaf types. In the subsequent dark incubation, fast recovery was observed in both leaf types that reached maximum PS2 efficiencies similar to non-photoinhibited control leaves. The artificial exogenous electron donors DPC, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in sun leaves, while DPC and NH2OH were significantly restored in shade leaves. Hence HI in shade leaves inactivates on the donor side of PS2 whereas it does at the acceptor side in sun leaves, respectively. Quantification of the PS2 reaction centre protein D1 and the 33 kDa protein of water splitting complex following HI-treatment of leaves showed pronounced differences between shade and sun leaves. The photoprotective function of leaf betacyanin in water-stressed Amaranthus cruentus plants was examined by comparing leaves of two strains which differ significantly in the amount of betacyanin. At 0, 1, and 2 days after the imposed water stress, leaves were subjected to high-light (HL) treatment to assess their photosynthetic capacity and photoinhibition susceptibility. The water stress equally reduced leaf relative water content (RWC),
gas-exchange rate and chlorophyll (Chl) contents in both leaves, indicating that the severity of water stress was comparable between the strains. Consequently, the extent of photoinhibition after the HL treatment increased in both strains as water stress developed; however, it was significantly greater in acyanic leaves than in betacyanic leaves, suggesting lower photoinhibition susceptibility in the betacyanic strain. The betacyanic leaves also exhibited approximately 30% higher values for photochemical quenching coefficient (qP) during the period of water stress despite the nonphotochemical quenching coefficient (qN) did not differ significantly between the strains. These results may be partially explained by the increased amount of leaf betacyanin under water stress. Moreover, a decrease in Chl content in betacyanic leaves might have enhanced light screening effect of betacyanin by increasing relative abundance of betacyanin to Chl molecule. In addition, reduced Chl content increased light penetrability of leaves. As a result, the extent of photoinhibition at the deeper tissue was exacerbated and the Chl fluorescence emitted from these tissues was more readily detected, facilitating assessment of photoinhibition at deeper tissues where the effect of betacyanic light screening is considered to be most apparent. Diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem 2 (PS2) as well as H2O2 content were analyzed in Reaumuria soongorica (Pall.) Maxim., a perennial semi-shrub. The rate of photorespiration was estimated by combined measurement of gas exchange and Chl fluorescence. The rate of photorespiration increased with the increasing drought stress (DS). The ratio of carboxylation electron flow to oxygenation electron flow (Jc/Jo) and the maximal photochemical efficiency of PS2 (variable to maximum fluorescence ratio, Fv/Fm) decreased with the increasing DS. Fv/Fm in isonicotinic acid hydrazide (INH)-sprayed plants was lower than that in normal plants under moderate DS, but no significant difference was observed under severe DS. H2O2 content in INH-sprayed plants was significantly lower than that in normal plants under severe DS.
