Multidrug resistance of cancer cells is often accompanied by the (over)expression of integral plasma membrane P-glycoprotein, an ATP-dependent transport pump for diverse unrelated compounds. The glutathione detoxification system represents another mechanism that may be involved in multidrug resistance. In the multidrug-resistant L1210/VCR cell line obtained by long-term adaptation of parental L1210 cells to vincristine, an increased expression of P-glycoprotein has previously been established. In this paper, we investigated if the glutathione detoxification system is also involved in the multidrug resistance of these cells. L1210/VCR cells with resistance induced by adaptation to vincristine were also found to be cross-resistant to vinblastine, actinomycin D, mitomycin C, doxorubicin and cyclophosphamide. The resistance of the above cells to vincristine and doxorubicin was accompanied by a depression of drug accumulation (which has not yet been established for other drug). L1210/VCR cells are able to survive better than sensitive cells under conditions when glutathione was depleted by L-buthionine sulfoximine. Nevertheless, L-buthionine sulfoximine did not influence the resistance of L1210/VCR cells to vincristine. Moreover, the presence of sublethal concentrations of cytostatics neither changed the IC50 value of resistant cells to L-buthionine sulfoximine nor the cytoplasmic activity of glutathione S-transferase, the crucial enzyme of glutathione detoxification system. It summarizes findings associated with their unique mode of reproduction, challenging species delimitation and insufficient understanding of the biology and systematics of these fishes in the past. Molekulární genetika vstoupila na antropologické kolbiště koncem 60. let, ale teprve přímá analýza archaické DNA (aDNA) z fosilních pozůstatků od 80. let umožnila přesnější vhled do evoluce našeho druhu ve středním a mladém pleistocénu. Navzdory různým „Jurským parkům“ totiž DNA po smrti organismu rychle degraduje a časové okno její možné analýzy je poměrně omezené. Navíc jsou vzorky kontaminovány DNA okolních organismů. Nicméně velice záhy se pozornost paleogenetiků soustředila na naše příbuzné - neandertálce. Analýza jejich genomu ukázala, že ~2 % jejich DNA se vyskytuje v genomu anatomicky moderního člověka s výjimkou subsaharské Afriky a celkový rozsah tohoto přenosu může dosahovat až 20 %. Křížení s neandertálci lidem zřejmě umožnilo snadnější adaptaci na chladnější podmínky eurasijského kontinentu, současně však přineslo i výskyt některých chorob. Překvapení přinesla sekvence aDNA izolovaná z článku prstu nalezeného v Denisově jeskyni na Altaji. Ukázalo se, že tento jedinec patřil k neznámému druhu odlišnému jak od moderních lidí, tak i od neandertálců. I tito hominini přispěli až 6 % svojí DNA do genomu některých současných populací člověka (JV Asie, Oceánie). Podle nejnovějších poznatků byl tok genů mezi homininy středního a mladého paleolitu poměrně složitý, byla např. detekována příměs neandertálské DNA v genomu denisovců, kteří navíc získali další sekvence od dalšího, blíže neurčeného druhu hominina. The objective of this prospective double-blind study was to determine whether postoperative residual paralysis (PORP) after pancuronium or vecuronium results in hypoxemia and hypercapnia in the immediate admission period to the recovery ward. Eighty-three consecutive surgical patients received balanced or intravenous anesthesia with pancuronium for operations lasting longer than one hour or vecuronium for those lasting less than 60 min, both combined with neostigmine at the end of anesthesia. Standard clinical criteria assessed neuromuscular function intraoperatively. Postoperatively, we determined neuromuscular function (acceleromyography with supramaximal train-of-four (TOF) stimulation of the ulnar nerve, and a 5-s head lift) and pulmonary function (pulse oximetry: SpO2, and blood gas analysis: SaO2, PaCO2). We defined PORP as a TOF-ratio 70 %, hypoxemia as a postoperative SpO2³ 5 % below the pre-anesthestic level together with a postoperative SaO293 %, and hypercapnia as a PaCO2³ 46 mm Hg. Among the 49 pancuronium and 27 vecuronium patients studied, the PORP rates were 20 % in the pancuronium group and 7 % in the vecuronium group (p>0.05). Hypoxemia and hypercapnia occurred more often in pancuronium patients with PORP than in those without PORP namely 60 % vs. 10% (p<0.05) and 30 % vs. 8 % (p>0.05), respectively. Jedním ze zdrojů informací o pravěku je mitochondriální DNA (mtDNA). Díky mutacím, které v její molekule v průběhu času zcela přirozeně vznikají, lze rekonstruovat fylogenetický strom a datovat vznik jednotlivých větví i jejich další vývoj v jednotlivých oblastech světa. První evropské větve lidské mtDNA fylogeneze jsou zřejmě starší více než 50 tis. let a pocházejí s největší pravděpodobností z Předního východu. Díky mtDNA bylo rovněž zjištěno, jak se na složení evropského genofondu projevilo poslední maximum doby ledové před 20 tis. lety, kdy se kvůli ochlazení lidé stáhli do jihoevropských refugií a po oteplení před 15 tis. lety severnější oblasti znovu kolonizovali. Také se předpokládalo, že se vznikem zemědělství před 10 tis. lety proběhla další větší migrace z Předního východu. Ve střední Evropě je ale námi zjištěný příspěvek ,,neolitických“ mtDNA linií poměrně malý, takže se zemědělství v této oblasti šířilo spíše akulturací původních obyvatel, tedy přenosem kulturních dovedností. Zbarvení hraje v životě zvířat zásadní roli a je často pod přísným dohledem přírodního a/nebo pohlavního výběru. Abychom mohli objektivně zhodnotit jeho vliv na životní strategie zvířat, je zaprvé důležité barvu správně měřit a zadruhé nahlížet na ni zrakovým aparátem zainteresovaného pozorovatele. Obě tyto metody popisujeme na příkladu ptáků, konkrétně hodnocení mimeze parazitických vajec., Colouration plays a very important role in the life of animals and its evolution is often under natural and/or sexual selection. For objective assessment of the influence of colouration on animal life strategies the colour should be measured properly and evaluated from the perspective of the studied species.
