The present study focused on the evolution of the karyotype in 21 taxa of the genus Isophya, which was done by mapping the location on the chromosomes of ribosomal RNA (rRNA) coding genes using fluorescence in situ hybridization (FISH) with an 18S rDNA probe and using silver staining (AgNO3) to evaluate the activity of major rDNA clusters. Since the chromosome number and sex determination do not vary in this genus, the above markers were used in a detailed comparison of the cytogenetic features of species of Isophya. The species analyzed were placed into three groups based on the location of rDNA on their chromosomes: (1) rDNA-FISH signals only on the two long pairs of autosomes, (2) rDNA-FISH signals on one long and one short pair of autosomes, and (3) rDNA-FISH signals on three to five different sized pairs of autosomes. These groupings partly correspond to the morphological groupings proposed in earlier studies. One long pair of autosomes frequently carried rDNA in all the Isophya species and probably is a plesiomorphic character for these taxa. The cytogenetic mapping revealed great variability among Isophya species in the chromosomal location of major rDNA clusters. Our results suggest that the observed variation in the number of rDNA clusters can be an important species-group specific phylogenetic marker. Analysis of 18S rDNA hybridization signals showed that the evolutionary dynamics of rDNA in this genus is remarkably high and accompanied by changes in the structure of chromosomes bearing rDNA at an inter- and intra-specific level. a1_There are few studies on the morphology and meiosis in the testicles of Heteroptera, but are extremely important, especially for the family Pentatomidae, because in some species in this family meiosis in the testicular lobes results in the production of non-fertile spermatozoa. With the aim of improving the level of understanding of this phenomenon the morphology of the testes and spermatogenesis in 10 species of Pentatomidae were recorded and compared. All of them have testes covered by a pigmented peritoneal sheath. In some species the internal membrane or just inside the peritoneal sheath is pigmented. The pigmentation of both membranes varied. The pigmentation of both was either yellow, or the internal membrane was yellowish and the external one reddish. When the membrane is pigmented, the colour is red or yellow. The number of lobes varied from 3 to 7, with intermediate numbers of 4 and 6. The size and diameter of the lobes are similar in all the species studied, except Proxys albopunculatus, in which the diameter of the third and fifth is smaller than that of the other lobes. The behaviour of the cells during spermatogenesis was the same in all lobes of most species, except in P. albopunctulatus, in which the harlequin lobe is absent and the cells in lobes 4 and 6 exhibited characteristics different from those of cells in lobes 1-3 and 5. Chlorocoris complanatus and Loxa deducta (both Pentatominae) have a harlequin lobe (lobe 5). The chromosome complements recorded were: 2n = 12 (10A + XY) in Dichelops melacanthus and Edessa collaris, 2n = 14 (12A + XY) in C. complanatus, Edessa meditabunda, Ladeaschistus sp., Loxa deducta, P. albopunctulatus, Piezodorus guildinii and Thyanta perditor and 2n = 16 (14A + XY) in Edessa affinis. This investigation addresses the interaction of insulin (INS) and glucocorticoid (GC) signaling in the hepatic regulation of tryptophan oxygenase (TO) enzyme activity in the rat. Male Wistar rats (200-250 g b.w) received an injection of the different doses of INS (10, 25, 50, 70 and 100 μg/200 g b.w., i.p.) and were used for experiments 3 h and 18 h after INS administration. This study shows that maximum of TO activity was found at dose of 50 μg of INS with peak increases observed at 3 h and 18 h after injection of INS, while INS had no effect on TO activity in adrenalectomized rats. The analysis of INS effects on glucocorticoid receptor-complex (GC/GR complex) stability shows that complexes from INS-treated rats are less stable than those from control animals. In addition, INS-stimulated stability of glucocorticoid receptor (GR) protein was significantly increased from the controls. Furthermore, the results show that GC/GR complexes from INS-treated rats could be activated and accumulated at higher rate in cell nuclei of control animals. Under optimal conditions, most of the light energy is used to drive electron transport. However, when the light energy exceeds the capacity of photosynthesis, the overall photosynthetic efficiency drops down. The present study investigated the effects of high light on rice photooxidation-prone mutant 812HS, characterized by a mutation of leaf photooxidation 1 gene, and its wild type 812S under field conditions. Our results showed no significant difference between 812HS and 812S before exposure to high sunlight. However, during exposure to high light, shoot tips of 812HS turned yellow and their chlorophyll (Chl) content decreased. Transmission electron microscopy showed that photooxidation resulted in significant damage of chloroplast ultrastructure. It was confirmed also by inhibited photophosphorylation and reduced ATP content. The decreased coupling factor of ATP, Ca2+-ATPase and Mg2+-ATPase activities also verified these results. Further, significantly enhanced activities of antioxidative enzymes were observed during photooxidation. Malondialdehyde, hydrogen peroxide, and the superoxide generation rates also increased. Chl a fluorescence analysis found that the performance index and maximum quantum yield of PSII declined on August 4, 20 days after high-light treatment. Net photosynthetic rate also decreased and substomatal CO2 concentration increased in 812HS at the same time. In conclusion, our findings indicated that excessive energy triggered the production of toxic reactive oxygen species and promoted lipid peroxidation in 812HS plants, causing severe damage to cell membranes, degradation of photosynthetic pigments and proteins, and ultimately inhibition of photosynthesis., J. The aim of our study was to investigate the underlying molecular mechanisms of exogenously supplied trehalose affecting wheat photosynthesis under heat stress. The amount of ATP synthase (ATPase), oxygen-evolving enhancer protein (OEE), PsbP, Rubisco, chloroplast fructose-bisphosphate aldolase (FBPA), and ferredoxin-NADP(H) oxidoreductase (FNR) were downregulated, while PSI reaction center subunits were upregulated under heat stress. However, in the trehalose-pretreated groups, the amount of FNR, cytochrome b6f complex, PSI reaction center subunits, ATPase, FBPA, and Rubisco were upregulated under normal growth conditions and heat stress. Besides, during the recovery period, the upregulation in CAB, PsbP, OEE2, and ATPase suggested that trehalose pretreatment might help to the recovery of PSII and PSI. In the present work, we have characterized the chromosomes of 13 Cassidinae beetles, belonging to four tribes, the broad aim being to increase the cytogenetic data and establish the mechanisms involved in chromosome evolution of this subfamily, which appear to be conserved karyotypically, i.e. 2n = 16 + Xyp. The analysis of mitotic and meiotic cells revealed a high diversity of diploid numbers (2n = 18, 2n = 22, 2n = 26, 2n = 32, 2n = 36, 2n = 40, 2n = 42), and the presence of sex chromosome system of the Xyp type in most species, with the exception of two representatives that exhibited Xyr and XY systems. C-banding showed constitutive heterochromatin predominantly localized in the pericentromeric region of the chromosomes, but differences regarding the number of chromosomes with positive C-bands, intensity of the blocks, and presence of additional bands in autosomes and/or sex chromosomes were observed among the species investigated. Our data revealed that the karyotype 2n = 16 + Xyp does not occur in all 13 tribes of the Cassidinae characterized cytogenetically, seeming to be only a shared feature among the species of the Cassidini. Variations in the C-band pattern, mainly in closely related species, suggest that the interspecific karyotype diversification occurred as a result of changes in the quantity and distribution of constitutive heterochromatin. The occurrence of the Xyp sex chromosome system in the tribe Mesomphaliini, which showed the highest diversity of simple and multiple systems among the coleopteran as a whole, reinforces the view that derived systems originated by chromosome rearrangements involving the Xyp ancestral system., Amália T. The objective of this work was to evaluate the relative trap efficiency of the three modified traps baited with 1-octen-3-ol as attractant, Box, Nzi and Canopy, for horse fly (Tabanidae) survey. Nine traps (three traps per trap type) were tested in 3 × 3 Latin square designs during summer 2011 in riparian oak-ash forests in the Danube floodplain of Croatia. Overall, the traps caught 1,986 specimens of 11 horse flies species during the study period. The most abundant species caught was Tabanus bromius (58%), followed by Tabanus tergestinus (21%), Tabanus maculicornis (16%), Tabanus sudeticus (2%), Atylotus loewianus (1%) and Tabanus autumnalis (1%). Both the Box traps and the Nzi traps had significantly greater catch efficiencies than the Canopy traps (βbox = 2.47, X2 = 65.48, df = 1, P < 0.001 and βnzi = 1.54, X2 = 25.12, df = 1, P < 0.001, respectively). Nzi traps had a catch rate 4.65 (95% CI: 2.55, 8.48) times greater than Canopy traps and Box traps had a catch rate 11.77 (95% CI: 6.48, 21.39) times greater than Canopy traps. A field study was conducted with the aim to elucidate photosynthetic responses of five emmer hulled wheat (Triticum turgidum ssp. dicoccum) accessions to 30 (N-limited) and 100 kg(N) ha-1
(N-sufficient) conditions at control and drought stress (irrigation after 30-40% and 60-70% depletion of available soil water, respectively). Chlorophyll (Chl) a and Chl b concentrations of the emmer wheats remained unchanged but net photosynthetic rate and dry mass increased and decreased, respectively, when received a sufficient amount of N. Smaller drought-induced decreases in Chl concentration, membrane stability index, and dry mass were concomitant to a greater decrease in intercellular CO2 concentration of emmer compared to the durum (Triticum turgidum) and bread wheats (Triticum aestivum). The lack of negative effect of insufficient N on Chl concentration and dry mass of emmer wheat suggests that this type of wheat possesses an obvious potential for organic farming., M. Vaghar, P. The endothelium of different organs displays a remarkable heterogeneity, although it presents many common functional and morphological features. However, despite our knowledge of heterogeneity among endothelial cells from different sites, the differences between brain microvascular endothelial cells (BMEC) and coronary microvascular endothelial cells (CMEC) are poorly defined. The aim of this study was to investigate whether BMEC are distinct from CMEC at the protein level. Using the proteomic approach, we comparatively analyzed the proteome of cultured BMEC and CMEC. We reproducibly separated over 2000 polypeptides by using two-dimensional electrophoresis (2-DE) at pH range of 3-10. Using PDQuest software to process the 2-DE gel images, forty-seven protein spots were differentially expressed in the two-endothelial cells. Of these, thirty-five proteins are highly expressed in BMEC, whereas twelve proteins are highly expressed in CMEC. Fifteen proteins in BMEC and seven proteins in CMEC were identified with high confidence by matrix-associated laser desorption ionization time-of-flight mass spectrometer (MALDI-TOF-MS). Our data suggested that BMEC and CMEC were different in several aspects including cytokine and growth-related molecules, stress-related proteins, metabolic enzymes, signal transduction proteins and others. The aim of study was to gain a deeper knowledge about local and systemic changes in photosynthetic processes and sugar production of pepper infected by Obuda pepper virus (ObPV) and Pepper mild mottle virus (PMMoV). PSII efficiency, reflectance, and gas exchange were measured 48 and/or 72 h after inoculation (hpi). Sugar accumulation was checked 72 hpi and 20 d after inoculation (as a systemic response). Inoculation of leaves with ObPV led to appearance of hypersensitive necrotic lesions (incompatible interaction), while PMMoV caused no visible symptoms (compatible interaction). ObPV (but not PMMoV) lowered Fv/Fm (from 0.827 to 0.148 at 72 hpi). Net photosynthesis decreased in ObPV-infected leaves. In ObPV-inoculated leaves, the accumulation of glucose, fructose, and glucose-6-phosphate was accompanied with lowered sucrose, maltoheptose, nystose, and trehalose contents.
