Alkaline stress is important abiotic stress that restricts the growth and physiological activity of sorghum (Sorghum bicolor L. Moench). We aimed to investigate the effects of alkaline stress on alkali-tolerant SX44B and alkali-sensitive 262B sorghum inbred lines. The results showed that alkaline stress decreased the content of chlorophyll, activity of photosystem II, net photosynthetic rate, and destroyed chloroplast morphology. These changes were less pronounced in SX44B, possibly owing to its higher antioxidant enzyme activity and nonphotochemical quenching. Alkaline stress decreased water content, transpiration rate, and stomatal conductance while increasing the leaf temperature, with the effect being more pronounced in 262B. A significant correlation was observed between leaf-air temperature difference (ΔT) and relative water content and gas-exchange parameters, especially in 262B. Therefore, ΔT is an effective indicator for monitoring changes in sorghum leaves under alkaline stress and evaluating the alkali tolerance of different sorghum germplasm.
Excessive cadmium (Cd) content in soil leads to a number of phytotoxic effects and challenges agricultural production. Aim of this study was to investigate different responses of two maize inbreds and their hybrid to an elevated Cd content in soil by measuring photosynthetic and biochemical activity and to identify a Cd tolerance mechanism. Antioxidant statusrelated parameters varied significantly between inbreds and treatments. Dry mass increased in both inbreds, but remained unchanged in hybrid. After the Cd treatment, parameters of chlorophyll a fluorescence varied between inbreds and hybrid performance was similar to inbred B84. We concluded that inbred B84 is Cd-sensitive compared to Os6-2, which did not appear to be negatively affected by Cd treatment at this growth stage studied. We suspect that due to a dilution effect in the hybrid, there was no or very weak Cd stress detected by biochemical parameters, although stress was detected by chlorophyll a fluorescence., M. The effect of drought stress on energy dissipation and antioxidant enzyme system in two sweet sorghum inbred lines (M-81E and Roma) was investigated. Results showed that the germination indicator increased more in M-81E than that in Roma under rehydration. Under drought stress, both the maximal photochemical efficiency of PSII (Fv/Fm) and oxidoreductive activity (ΔI/I0) of Roma decreased more than those in M-81E. Relative to Fv/Fm, the ΔI/I0 decreased markedly, which indicated that PSI was more sensitive to drought stress than PSII. Increases in the reduction state of QA (1-qp), nonphotochemical quenching (NPQ) and minimal fluorescence yield of the dark-adapted state (F0) were greater in Roma than those in M-81E; meanwhile, the H2O2 content was lower in M-81E than that in Roma. Our results suggested that the photoinhibition might be related to the accumulation of reactive oxygen species (ROS). The hypothesis that application of exogenous glycine betaine (GBEX) may attenuate the effects of mild water deficit in leaf gas exchange and lipid peroxidation in Carapa guianensis was examined. For this reason, 110-d old plants were sprayed with 0, 25, and 50 mM GBEX and then subjected to two watering regimes. In the first, irrigation was continuously performed to maintain the soil near to field capacity (watered plants). In the second, irrigation was withheld and water deficit resulted from progressive evapotranspiration (water-stressed plants). Treatment comparisons were assessed when predawn leaflet water potential (Ψpd) of stressed plants reached -1.28 ± 0.34 MPa. Regardless of the watering regime, significant (P<0.05) increases in foliar glycine betaine (GBLeaf) concentration were observed in response to increasing GBEX; however, such increases were more expressive in stressed plants. The net photosynthetic rate, stomatal conductance to water vapor, and intercellular to ambient CO2 concentration ratio were significantly lower in water-stressed plants independently of GBEX concentration sprayed on leaves. The application of 25 and 50 mM GBEX caused significant (P<0.05) increases in ascorbate peroxidase (APX) activity in stressed plants, while significant (P<0.05) increases in catalase activity was observed just in the stressed plants treated with 50 mM GBEX. Malondialdehyde concentrations did not differ between watered and stressed plants regardless of GBEX concentration. In conclusion, C. guianensis was able to incorporate GBEX through their leaves and the resulting increases in GBLeaf attenuated lipid peroxidation in stressed plants through positive modulation of APX and CAT activities., F. Melatonin mediates many physiological processes in animals and plants. To examine the potential roles of melatonin in salinity tolerance, we investigated the effects of exogenous melatonin on growth and antioxidant system in cucumber under 200 mM NaCl stress conditions. The results showed that the melatonin-treated plants significantly increased growth mass and antioxidant protection. Under salinity stress, the addition of melatonin effectively alleviated the decrease in the net photosynthetic rate, the maximum quantum efficiency of PSII, and the total chlorophyll content. Our data also suggested that melatonin and the resistance of plants exhibited a concentration effect. The application of 50-150 μM melatonin significantly improved the photosynthetic capacity. Additionally, the pretreatment with melatonin reduced the oxidative damage under salinity stress by scavenging directly H2O2 or enhancing activity of antioxidant enzymes (including superoxide dismutase, peroxidase, catalase, ascorbate peroxidase) and concentrations of antioxidants (ascorbic acid and glutathione). The effects of foliar spraying of the dithiocarbamate zineb on two cultivars of tomato grown in the field in a site with high ozone concentrations were studied by means of biomass assessment, antioxidant enzyme assays, lipid peroxidation, and chlorophyll fluorescence measurements. Zineb prevented the peroxidation of membrane lipids and decreased the activity of scavenging enzymes, which suggests that plants sprayed with zineb are subjected to lower oxidative stress than controls. The beneficial effects of zineb protection is the utilization of a larger fraction of absorbed radiant energy in photosynthesis and a larger fruit yield in plants of both cultivars. and Á. a1_We compared the interactive effects of temperature and light intensity on growth, photosynthetic performance, and antioxidant enzyme activity in Zizania latifolia Turcz. plants in this study. Plants were grown under field (average air temperature 9.6-25°C and average light intensity 177-375 W m-2) or greenhouse (20-32°C and 106-225 W m-2) conditions from the spring to the early summer. The results indicated that greenhouse-grown plants (GGP) had significantly higher plant height, leaf length, and leaf width, but lower leaf thickness and total shoot mass per cluster compared with field-grown plants (FGP). Tiller emergence was almost completely suppressed in GGP. Significantly higher chlorophyll (Chl) content and lower Chl a/b ratio were observed in GGP than in FGP. From 4 to 8 weeks after treatment (WAT), net photosynthetic rate (PN) was significantly lower in FGP than in GGP. However, from 9 to 12 WAT, PN was lower in GGP, accompanied by a decrease in stomatal conductance (gs) and electron transport rate (ETR) compared with FGP. Suppressed PN in GGP under high temperature combined with low light was also indicated by photosynthetic photon flux density (PPFD) response curve and its diurnal fluctuation 10 WAT. Meanwhile, ETR in GGP was also lower than in FGP according to the ETR - photosynthetically active radiation (PAR) curve. The results also revealed that GGP had a lower light saturation point (LSP) and a higher light compensation point (LCP). From 4 to 8 WAT, effective quantum yield of PSII photochemistry (ΦPSII), photochemical quenching (qP), and ETR were slightly lower in FGP than in GGP. The activities of ascorbate peroxidase (APX), guaiacol peroxidase (POD), glutathione reductase (GR), superoxide dismutase (SOD), and malondialdehyde (MDA) content were significantly higher from 4 to 8 WAT, but lower from 10 to 12 WAT in FGP., a2_However, catalase (CAT) activity was significantly lower in FGP from 4 to 8 WAT. b1_We previously demonstrated in rats that noninvasive delayed limb ischemic preconditioning (LIPC) induced by three cycles of 5-min occlusion and 5-min reperfusion of the left hind limb per day for three days confers the sa me cardioprotective effect as local ischemic preconditioning of the heart, but the mechanism has not been studied in depth. The aim of this project was to test the hypothesis that delayed LIPC enhances myocardial antioxidative ability during ischemia-reperfusion by a mitochondrial KATP channel (mito KATP)-dependent mechanism. Rats were randomized to five gr oups: ischemia-reperfusion (IR)- control group, myocardial ischemic preconditioning (MIPC) group, LIPC group, IR-5HD group and LIPC-5HD group. The MIPC group underwent local ischemic precondi tioning induced by three cycles of 5-min occlusion and 5-min reperfusion of the left anterior descending coronary arteries. The LIPC and LIPC-5HD groups underwent LIPC induced by three cycles of 5-min occlusion and 5-min reperfusion of the left hind limb using a modified blood pressure aerocyst per day for three days. All rats were subjected to myocardial ischemia-reperfu sion injury. The IR-5HD and LIPC-5HD groups received the mito KATP channel blocker 5-hydroxydecanoate Na (5-HD) before and during the myocardial ischemia-reperfusion injury. Compar ed with the IR-control group, both the LIPC and MIPC groups showed an amelioration of ventricular arrhythmia, reduced my ocardial infarct size, increased activities of total superoxide dismutase, manganese-superoxide dismutase (Mn-SOD) and glutathione peroxidase, increased expression of Mn-SOD mRNA and decreased xanthine oxidase activity and malondialdehyde concentration. These beneficial effects of LIPC were prevented by 5-HD. Nicosulfuron is a post-emergence herbicide used for weed control in fields of maize (Zea mays L.). We used a pair of nearly isogenic inbred lines, SN509-R (nicosulfuron-resistant) and SN509-S (nicosulfuron-sensitive), to study the effect of nicosulfuron on waxy maize seedling. After the nicosulfuron treatment, net photosynthetic rate, stomatal conductance, transpiration rate, leaf maximum photochemical efficiency of PSII, photochemical quenching of chlorophyll fluorescence, and the actual photochemical efficiency of PSII were significantly lower in SN509-S than those of SN509-R, contrary to intercellular CO2 concentration, stomatal limitation, and nonphotochemical quenching. Compared to SN509-R, antioxidant enzyme activities in SN509-S decreased significantly in response to the nicosulfuron treatment, while SN509-S exhibited an increased malondialdehyde content, which was associated with lower antioxidant enzyme activities. Glyphosate herbicide caused oxidative stress and exhibited negative effects on photosynthesis and gas exchange of peanut [Arachis hypogaea L. cv. Giza (G) 5 and 6] leaves. We demonstrated that glyphosate caused various morphological symptoms, such as chlorosis, yellowing, and appearance of curly edges in leaves treated with high doses of herbicide in both cultivars; however, the G5 cultivar was more sensitive and showed severer symptoms. Glyphosate lowered photosynthesis and reduced contents of pigments and proteins as well as free amino acids in both cultivars. The gas-exchange parameters, such as photosynthetic (P N) and transpiration rate (E), were highly altered by the glyphosate application. For example, P N and E were reduced by 65 and 61%, respectively, in G5 treated with high dose of glyphosate compared with control. Antioxidant enzymes, such as peroxidase, catalase, ascorbate peroxidase, and superoxide dismutase were induced by both low and high concentrations in the glyphosate-treated leaves. Moreover, the level of lipid peroxidation, indicated by a malondialdehyde content, as well as the hydrogen peroxide content increased in the glyphosate-treated leaves. However, an increase in total antioxidant activity was detected in leaves and this reflected changes in the antioxidant status and accumulation of antioxidants as a defense mechanism against glyphosate toxicity in peanut., D.
