The Academy of Sciences of the Czech Republic (AS CR) established a new public research institution, the Institute of Biotechnology AS CR, v. v. i. The primary ambition of this new institute is to develop cutting-edge basic and oriented research on topics opening for diagnostic and therapeutic applicatons in human medicine. This investigation addresses the interaction of insulin (INS) and glucocorticoid (GC) signaling in the hepatic regulation of tryptophan oxygenase (TO) enzyme activity in the rat. Male Wistar rats (200-250 g b.w) received an injection of the different doses of INS (10, 25, 50, 70 and 100 μg/200 g b.w., i.p.) and were used for experiments 3 h and 18 h after INS administration. This study shows that maximum of TO activity was found at dose of 50 μg of INS with peak increases observed at 3 h and 18 h after injection of INS, while INS had no effect on TO activity in adrenalectomized rats. The analysis of INS effects on glucocorticoid receptor-complex (GC/GR complex) stability shows that complexes from INS-treated rats are less stable than those from control animals. In addition, INS-stimulated stability of glucocorticoid receptor (GR) protein was significantly increased from the controls. Furthermore, the results show that GC/GR complexes from INS-treated rats could be activated and accumulated at higher rate in cell nuclei of control animals. Patch clamp method developed more than 30 years ago is widely used for investigation of cellular excitability manifested as transmembrane ionic current and/or generation of action potentials. This technique could be applied to measurement of ionic currents flowing through individual (single) ion channels or through the whole assembly of ion channels expressed in the whole cell. Whole cell configuration is more common for measurement of ion currents and the only one enabling measurement of action potentials. This method allows detailed analysis of mechanisms and structural determinants of voltagedependent gating of ion channels as well as regulation of channel activity by intracellular signaling pathways and pharmacological agents., M. The purpose of this Summer School, which took place in Liblice 1 to 7 September 2008, was to introduce the students and post-docs to updated procedures used in the analyses of molecular data-sets applied to problem-solving in conservation genetics. This was done by bringing together geneticists statisticians and empirical conservation geneticists who presented theoretical lectures and led practical computer work. and Josef Bryja, Pavel Munclinger.
22nd Annual Mammalian Genome Conference took place in the new edifice of the Institute of Molecular Chemistry AS CR, v. v. i., in Prague. The symposium was aimed at dissiminating research news on the topic mammalian genomes, epigenomes and phenotypes. It began with Bioinformatics speakers folowed by a Student Satellite Meeting and it continued with a number of sessions. and Jiří Forejt.
