The impact of heat shock on minimising the activity of photosystem 2 (PS2) initiating high lipid peroxidation (POL) level and consequently changes in the enzymatic-antioxidant protective system was studied in seedlings of two Egyptian cultivars of barley (Giza 124 and 125). Heat doses (35 and 45 °C for 2, 4, 6, and 8 h) decreased chlorophyll (Chl) contents coupled with an increase in Chl a/b ratio, diminished Hill reaction activity, and quenched Chl a fluorescence emission spectra. These parameters reflect the disturbance of the structure, composition, and function of the photosynthetic apparatus as well as the activity of PS2. POL level, as dependent on the balance between pro- and anti-oxidant systems, was directly correlated with temperature, exposure time, and their interaction. Heat shock caused an increase in the electric conductivity of cell membrane, and malonyldialdehyde content (a peroxidation product) coupled with the disappearance of the polyunsaturated linolenic acid (C18:3), reflecting the peroxidation of membrane lipids which led to the loss of membrane selective permeability. Moreover, it induced distinct and significant changes in activities of antioxidant enzymes. Superoxide dismutase and peroxidase activities have been progressively enhanced by moderate and elevated heat doses, but the most elevated one (45 °C for 8 h) showed a decrease in activities of both enzymes. Winter wheat (Triticum aestivum L.) cultivars Yangmai 9 (water-logging tolerant) and Yumai 34 (water-logging sensitive) were subjected to water-logging (WL) from 7 d after anthesis to determine the responses of photosynthesis and anti-oxidative enzyme activities in flag leaf. At 15 d after treatment (DAT), net photosynthetic rate under WL was only 3.7 and 8.9 µmol(CO2) m-2 s-1 in Yumai 34 and Yangmai 9, respectively, which was much lower than in the control. Ratios of variable to maximum and variable to initial fluorescence, actual photosynthetic efficiency, and photochemical quenching were much lower, while initial fluorescence and non-photochemical quenching were much higher under WL than in control, indicating damage to photosystem 2. WL decreased activities of superoxide dismutase and catalase in both cultivars, and activity of peroxidase (POD) in Yumai 34, while POD activity in Yangmai 9 was mostly increased. Liriodendron tulipifera was exposed to gradually elevated ozone concentrations of 100-300 μg kg-1 in the naturally irradiated environment chamber. During 15 d of exposure to O3, net photosynthetic rate (PN) decreased and there was large difference between the control (C) and treatment with ozone (OT), while there was no significant difference in water use efficiency. Total chlorophyll content as well as the value of fluorescence parameter Fv/Fm decreased, while antioxidant enzyme activities related to ascorbate-glutathione cycle increased after 15 d of OT. Unchanged contents of ascorbate and glutathione indirectly suggest that the species hastened the antioxidant's oxidization/reduction cycle using enzymes instead of expanding their pool against oxidative stress. and S. With japonica rice 98-08, indica hybrids Shanyou 63, Gangyou 881, and X07S/Zihui 100, and sub-species hybrid Peiai 64S/9311 as materials, chlorophyll (Chl) content, Chl a fluorescence parameters, and membrane lipid peroxidation in flag leaf were measured at late developmental stages under natural conditions. Fv/Fm, qP, ΦPS2, and electron transport rate gradually decreased while qN increased conversely. Excessive photon energy led to the accumulation of active oxygen (O2-), H2O, malonyldialdehyde, and products of membrane lipid peroxidation, and resulted in reduced Chl content and early ageing subsequent to the photooxidation during flag leaf senescence. There was obvious diversification of these parameters among rice cultivars. In comparison with japonica cv. 98-08 (tolerant to photooxidation), Fv/Fm decreased in indica cv. Shanyou 63 (susceptible to photooxidation) with greater accumulation of active oxygen and a sharp drop in Chl content, which resulted in "yellowish" early ageing, and affected the filling and setting of rice grains. The mechanism for premature ageing in indica rice was related to irradiance and temperature at filling stages. On a sunny day at above 25 °C, the reaction centre of photosystem 2 (PS2) exhibited a dynamic change on reversible inactivation. Under the intense irradiance at noon, PS2 function in indica rice exhibited obvious down-regulation and photoinhibition. Under intense irradiance with lowered temperatures, PS2 resulted in photo-damage and early ageing, related to the degradation of PS2-D1 protein and the inhibition of endogenous protection systems such as the xanthophyll cycle and enzymes scavenging active oxygen. Hence for high-yield breeding, based on a good plant-type and utilising heterosis and tolerance of photooxidation, the selection of japonica rice or a sterile line with the japonica genotype as female is a strategy worthy of consideration. and Demao Jiao, Benhua Ji, Xia Li.
Wheat (Triticum aestivum L.) genotypes K-65 (salt tolerant) and HD 2329 (salt sensitive) were grown in pots under natural conditions and irrigated with NaCl solutions of electrical conductivity (ECe) 4.0, 6.0, and 8.0 dS m-1. Control plants were irrigated without saline water. Observations were made on the top most fully expanded leaf at tillering, anthesis, and grain filling stages. The net photosynthetic rate (PN), stomatal conductance (gs), and transpiration rate (E) were reduced with the addition of NaCl. The reduction was higher in HD 2329 than in K-65. Salinity enhanced leaf to air temperature gradient (ΔT) in both the genotypes. NaCl increased the activities of superoxide dismutase (SOD) and peroxidase (POX); the percent increment was higher in K-65. The sodium and potassium contents were higher in the roots and leaves of K-65 over HD 2329. Thus at cellular level K-65 has imparted salt tolerance by manipulating PN, E, gs, and K accumulation in leaves along with overproduction of antioxidative enzyme activities (SOD and POX). and N. With untransformed rice cv. Kitaake as control, the characteristics of carbon assimilation and photoprotection of a transgenic rice line over-expressing maize phosphoenolpyruvate carboxylase (PEPC) were investigated. The PEPC activity in untransformed rice was low, but the activity was stimulated under high irradiance or photoinhibitory condition. PEPC in untransformed rice contributed by about 5-10 % to photosynthesis, as shown by the application of the specific inhibitor 3,3-dichloro-2-(dihydroxyphosphinoylmethyl)propenoate (DCDP). When maize PEPC gene was introduced into rice, transgenic rice expressed high amount of maize PEPC protein and had high PEPC activity. Simultaneously, the activity of carbonic anhydrase (CA) transporting CO2 increased significantly. Thus the photosynthetic capacity increased greatly (50 %) under high CO2 supply. In CO2-free air, CO2 release in the leaf was less. In addition, PEPC transgenic rice was more tolerant to photoinhibition. Treating by NaF, an inhibitor of phosphatase, showed that in transgenic rice more phosphorylated light-harvesting chlorophyll a/b-binding complexes (LHC) moved to photosystem 1 (PS1) protecting thus PS2 from photo-damage. Simultaneously, the introduction of maize PEPC gene could activate or induce activities of the key enzymes scavenging active oxygen, such as superoxide dismutase (SOD) and peroxidase (POD). Hence higher PS2 photochemical efficiency and lower superoxygen anion (O2.-) generation and malonyldiadehyde (MDA) content under photoinhibition could improve protection from photo-oxidation. and D. Physiological responses of two wheat (Triticum aestivum L.) genotypes (salt-tolerant DK961 and salt-sensitive JN17) to increased salt concentrations (50, 100, 150 mM NaCl: NaCl50, NaCl100, NaCl150) were studied. Photosynthetic capacity, irradiance response curves, contents of soluble sugars, proteins, and chlorophyll (Chl), K+/Na+ ratio, and activities of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase) in flag leaves were measured on 7 d after anthesis. In control (NaCl0) plants, non-significant (p>0.05) differences were found in gas exchange and saturation irradiance (SI) between salt-tolerant (ST) and salt-sensitive (SS) wheat genotypes. However, we found higher soluble sugar and protein contents, K+/Na+ ratio, and antioxidant enzyme activities, but lower Chl content and yield in ST wheat. Salinity stresses remarkably increased soluble sugar and protein contents and the antioxidant activities, but decreased K+/Na+ ratio, Chl contents, SI, photosynthetic capacities, and yield, the extent being considerably larger in JN17 than DK961. Although the soluble sugar and protein contents and the antioxidant activities of JN17 elevated more evidently under salt stresses, those variables never reached the high levels of DK961. The antioxidant enzyme activities of SS wheat increased in NaCl50 and NaCl100, but decreased rapidly when the NaCl concentration reached 150 mM. Thus the ST wheat could maintain higher grain yield than the SS one by remaining higher osmoregulation and antioxidative abilities, which led to higher photosynthetic capacity. A crop legume Vigna unguiculata L. (Walp.) and a wild legume Crotalaria juncea L. were evaluated for their relative responses to the oxidative stress injury induced by various doses of UV-B radiation (UV-B, 280-315 nm; 0, 1.0, 1.4, 4.7, and 6.0 kJ m-2 d-1). A dose-dependent damage in lipid peroxidation was determined as an index of membrane injury caused by UV-B. The impact was significantly higher in V. unguiculata than in C. juncea. The specific activities of superoxide dismutase, ascorbate peroxidase, monodehydroascorbate reductase, and dehydroascorbate reductase increased directly proportional to UV-B doses. However, the activities of these enzymes were significantly higher in V. unguiculata than in C. juncea indicating that V. unguiculata was inflicted with more severe oxidative stress injury under UV-B. In C. juncea the glutathione reductase and ascorbate oxidase activities were 35 and 40 % greater than in V. unguiculata, respectively. Further, the non-enzymatic antioxidants ascorbate and glutathione, and their reduced/oxidizes ratios in C. juncea were much greater than V. unguiculata indicating C. juncea has an inherently greater antioxidative potential than V. unguiculata. Thus C. juncea is better adapted to oxidative stress than V. unguiculata by means of efficient cellular antioxidant mechanisms helping to combat the photooxidative stress injury elicited by UV-B.
