We studied the effect of flooding on a carabid community inhabiting grassland in a large river valley (W Poland). We used pitfall-traps to catch beetles from April to November 1999–2001. Some of the samples collected were preceded by floods during the collecting period, which enabled us to evaluate the effects of flooding on species composition and abundance. We collected 17,722 individuals belonging to 108 species. The number of species and individuals per sample differed between plots and showed a nonlinear decrease over time, from spring to autumn. Carabids were more abundant in samples collected after floods than in the control samples. In contrast, the expected cumulative number of species as a function of the number of individuals collected was lower in samples collected after floods than in the control samples. In the case of the most abundant species the species-specific responses in terms of the numbers caught after flooding differed, with those of (e.g. Agonum micans) increasing and those of (Amara plebeja, Epaphius secalis) decreasing. This study shows that floods filter the community with the result that some species increase in abundance but the overall species richness decreases. The aim of the study was to determine the effect of selective dopaminergic agents [(±)-SKF-81297 (Di agonist), R(-)-2,10,ll-trihydroxy-N-propyl-noraporphine (D2 agonist), pergolid (D agonist), R( + )-SCH-23390 (Di antagonist), S(—)-eticlopride (D2 antagonist) and cis-(Z)-flupenthixol (D antagonist)] on cell-mediated immune response in vivo and in vitro and to verify the presence of dopamine receptors on murine splenocytes. The tested dopaminergic compounds exhibited a pronounced inhibitory effect on T-dependent immunity. They suppressed alloantigen-induced immune response in vivo and in vitro, IL-2 production was also markedly reduced. No substantial difference was found between the effect of dopamine agonists and antagonists or among ligands of subtypes of dopamine receptors. The effect of dopaminergic agents in vitro indicates a direct interaction with immunocompetent cells at the peripheral level. As the binding studies did not confirm the presence of dopamine receptors on splenocytes, the immunosuppressive efficacy of dopaminergic agents does not seem to be mediated via specific dopamine receptors.
Infection and tumors provoke substantial changes accompanied with the disbalance of many neuroendocrine factors which in their summarizing effects influence the life span of animals. Our previous results showed enhanced mortality after one injection of morphine in association with Friend leukaemia virus infection. The aim of this study was to examine the effects of some other opioids (pethidine and pentazocine) and an acetylcholine esterase inhibitor neostigmine on the survival of animals under two conditions: (1) Friend leukaemia virus infection which mostly depressed immune functions, and (2) Toxoplasma gondii infection which in general enhanced the immune status. In contrast to our previous observation with morphine, the mortality induced by single doses of pethidine (150 mg/kg) or pentazocine (50-75 mg/kg) was unchanged during the Friend leukaemia virus infection. A single injection of neostigmine (0.42 or 0.56 mg/kg) was significantly more lethal in DBA-2 mice infected with Friend leukaemia virus. Neostigmine in doses of 0.33 and 0.4 mg/kg caused death in 46 % and 57 %, respectively, of animals infected with Toxoplasma gondii which was significantly higher in comparison with only 8 % and 12.5 % in control groups. Pethidine (150 mg/kg) killed 70 % of Toxoplasma gondii infected animals and even 90 % of non-infected mice. Thus, the Friend leukaemia virus and Toxoplasma gondii infections increased toxicity only of some drugs which may, at least partly, be associated with altered immune status during infection and involvement of the cholinergic system.
a1_Renal transplantation is associated with a large number of risk factors that can have an influe nce on early renal graft function (ERGF). One of these factors could be the increasing number of obese kidney donors. The mechanisms of reduced ERGF in obese kidney donors are still poorly understood. To that end, w e compared ERGF in recipients with body mass index (BMI), perivascular fat and plasma inflammation markers of live kidney donors. We hypothesi zed that the BMI of donors would negatively correlate with an average increase of glomerular filtration rate (GFR ) and that it would also be associated wi th increased perivascular and plasma inflammation markers in the first seven days after transplantation. Between January 2013 and December 2014, some 58 living kidney transplantation pairs were included in the study. Donor and recipient demographic data, preoperative BMI, blood C -reactive protein (CRP) and adiponectin levels, perivascular adipose tissue (PAT) samples and recipient blood creatinine levels were analy zed. The median CRP of donors was 0.68 mg/l (max: 8.66 mg/l, min: 0.33 mg/l), the median of M1 macrophages (CD14+CD16+) in one gram of PAT was 5940 (max: 41 100, min: 248) and the median of adiponectin was 411 930 pg/ml (max: 14 217 000, min: 167 300) in plasma. We did not find any association between early renal graft function and the percentage o f M1 macrophages in donor perirenal adipose tissue (p=0.83, r=0.03, n=58), adiponectin (p=0.65, r=0.06, n=58) or CRP (p=0.16, r=0.2, n=58) in plasma. The obesity level of donors, expressed as BMI, did not correlate with early renal graft function in the first seven days after transplantation. In the field, the blister beetle Mylabris phalerata Pallas (Coleoptera: Meloidae) undergoes larval diapause in the ground, which lasts for nearly six months. The effect of the soil environment on this diapause was examined. Final instar larvae kept at temperatures of >= 26°C do not enter diapause and continued to develop regardless of the soil water content and photoperiod. Below 25°C the final instar larvae entered diapause regardless of soil water content and photoperiod. The early stages, particularly L2, appeared to be more important for diapause induction than the later stages. However, the other instars were also sensitive. Temperature, rather then photoperiod was the main factor influencing pupal duration.
AIMS: Authors studied potential side effects of fetal calf serum (FCS) in cultivation media on human dental pulp stem cells (DPSC) during long-term cultivation. METHODS: Two lines of DPSC obtained healthy donors (male 22 years, female 23 years) were used. Both lines were cultivated under standard cultivation conditions in four different media containing 10% or 2% FCS and substituted with growth factors. During long-term cultivation proliferation ability, karyotype and phenotype of DPSC were measured. RESULTS: Both lines of DPSC cultivated in a media containing 2% FCS and ITS supplement showed the highest number of population doublings. On the other hand the proliferation rate of DPSC cultivated in a media with 2% FCS without ITS supplement was slowest. Proliferation rate of DPSC cultivated in 10% FCS media with or without FGF-2 was comparable. DPSC cultivated in a media with 10% FCS showed a significantly higher amount of chromosomal aberrations. These chromosomal aberrations do not seem to be clonal but surprisingly we found large amounts of tetraploid cells in the 9th passage in both media containing 10% FCS. CONCLUSIONS: Our study proved that cultivation of DPSC in media containing higher concentration of FCS has critical side effects on cell chromosomal stability. and J. Suchánek, TS. Kleplová, M. Increased and prolonged postprandial lipemia has been identified as a risk factor of cardiovascular disease. However, there is no consensus on how to test postprandial lipemia, especia lly with respect to the composition of an experimental meal. To address this question of how glucose, when added to a fat load, affects the selected parameters of postprandial lipemia, we carried out a study in 30 healthy male volunteers. Men consumed an experimental meal containing either 75 g of fat + 25 g of glucose (F+G meal) or 75 g of fat (F meal) in a control experiment. Blood was taken before the meal and at selected time points within the following 8 h. Glucose, when added to a fat load, induced an increase of glycemia and insulinemia and, surprisingly, a 20 % reduction in the response of both total and active glucagon -like peptide -1 (GLP -1) concentration. The addition of glucose did not affect the magnitude of postprandial triglyceridemia and TRL -C and TRL -TG concentrations but stimulated a faster response of chylomicrons to the test meal, evaluated by changes in apolipoprotein B -48 concentrations. The addition of glucose induced the physiological response of insulin and the lower response of GLP -1 to the test meal during the early postprandial phase, but had no effect on changes of TRL -cholesterol and TRL -TG within 8 h after the meal., K. Experiments were conducted to study the effect of goldthioglucose (GTG) upon the processes associated with lipid peroxidation. The glucose-6-phosphate dehydrogenase activity (G6PD; E.C.1.1.1.49) in red blood cells (RBC) and the amount of malonaldehyde precursors (MDA) per gram of brain, liver and kidney were determined. Adult mice received i.p. injections for three consecutive days of either saline (controls) or GTG dissolved in saline, in a dose of 0.10 mg.g-1 or 0.15 mg.g-1 b.w. In mice receiving higher dose of GTG the G6PD activity was significantly increased (349.38± 17.46 mU.10-9 RBC compared to 258.2± 14.46 mU.10-9 RBC in control animals). The content of MDA precursors rose significantly from 4.8± 0.81 m mol.g-1 of the liver in controls to 8.12± 1.41 m mol.g -1 and 7.88± 0.51 m mol.g-1 and from 18.71± 1.01 m mol.g-1 of the kidneys in controls to 24.25± 1.25 m mol.g-1 and 24.88± 1.7 m mol.g-1 respectively. The GTG-induced higher levels of MDA precursors and increased G6PD activity in RBC corresponds to the rise in lipid peroxidation and its participation in producing the lesions after experimental and therapeutic use of gold-containing substances seems possible., D. Group scan sampling and focal animal sampling was used to evaluate the relationship between the vigilance and group size of Mongolian gazelle (
Procapra gutturosa) from July to August 2012 around Lake Dalai, Inner Mongolia, China. We recorded 348 groups, including 107 mixed groups, 54 male groups, and 187 female groups with juveniles. The total number of individuals observed was 2026, including 4 solitary males, and the group average size was 5.76 individuals. The index of the models for curve fitting revealed that the power model Y= 2291.40–0.821 (R2 = 0.846, P = 0.013, Y
= vigilance distance and X = group size) was the best-fit model to represent
the relationship between vigilance distance and group size. The cubic model
Y = 108.1 – 16.95X + 1.09X2 – 0.023X3 (R2 = 0.826, P = 0.019, Y = vigilance rate and X = group size) was the best-fit model to represent the relationship between vigilance rate and group size. While, the logarithmic model Y =35.84 – 6.26lnX (R2 = 0.792, P = 0.024, Y = vigilance time and X = group size) was the best-fit model to represent the relationship between individual vigilance time and group size. Results indicate that vigilance rate, individual vigilance
time, and vigilance distance of Mongolian gazelle decreased significantly with increasing group size. Our research provides data on Mongolian gazelles’ vigilance and which will further benefit gazelle conservation.
The average soil temperature was significantly lower in plots covered with hay. Hay mulching, however, did not increase significantly the total number of soil micro-arthropods collected using a soil pin trap and a soil sampler. Significant increases in the number of individuals was recorded only for certain groups. 64% of all the arthropods collected using soil pin traps were collected in hay-covered plots and 36% in control plots. This increase was statistically significant for the orders Entomobryomorpha and Poduromorpha of the subclass Collembola and surface-dwelling (epigeic) Coleoptera. For the samples collected using the soil sampler, 57% of the specimens were collected from hay-covered plots and 43% from control plots. As for the pin traps this was reflected in differences in the numbers of the orders Entomobrhyomorpha and Poduromorpha of the subclass Collembola and Pauropoda, collected in the treated and control plots.
