1152. Anti-CD38 therapy with daratumumab for relapsed/refractory CD20-negative diffuse large B-Cell lymphoma

- Diffuse large B-cell lymphoma (DLBCL) is the most common and one of the most aggressive subtypes of non-Hodgkin’s lymphomas. Front-line therapy consists of chemotherapy in combination with anti-CD20 monoclonal antibody rituximab. Relapses after rituximab-based regimen have poor prognosis and call for new treatment options. Immunohistochemistry analysis of relapsed DLBCL often reveal CD20-negative lymphoma, which limits repeated use of rituximab in combination with salvage chemotherapy. CD38 is a surface antigen that binds to CD38, CD31/PECAM-1 and hyaluronic acid. CD38 is an important mediator of signal transmission from the microenvironment into the cell. Anti-CD38 monoclonal antibody daratumumab has been approved for the treatment of multiple myeloma. Expression of CD38 on the surface of DLBCL is highly variable (compared to strong expression on myeloma cells), but can be easily assessed by flow cytometry or immunohistochemistry. A patient-derived xenograft (PDX) model of CD20-negative, CD38-positive DLBCL derived from a patient with rituximab-refractory DLBCL was used for in vivo experiments. We demonstrated that daratumumab suppressed growth of subcutaneous PDX tumours significantly more effectively than rituximab. Analysis of tumours obtained from mice treated with daratumumab revealed down-regulation of surface CD38, suggesting endocytosis of CD38-daratumumab complexes. The results suggest a potential clinical use of daratumumab in combination with salvage chemotherapy in patients with relapses of CD20-negative DLBCL. In addition, daratumumab might potentially serve as a suitable antibody moiety for derivation of antibodydrug conjugates for the targeted delivery of toxic payloads to the lymphoma cells.

Empiricky overuje tézu, že mediálny marketing politických strán zneužíva diskriminované a marginalizované skupiny na dosahovanie straníckych cieľov. Prostredníctvom kritickej systemickej lingvistiky analyzuje významové štruktúry predvolebného bilbordu, čím odhaľuje intencie komunikácie politickej strany s verejnosťou. Postupmi kritickej diskurzívnej analýzy mediálnych textov o anti-rómskom bilborde autorky identifikujú diskurzívne pozície rozmanitých aktérov, prípadne ich argumentačné stratégie, pričom kriticky reflektujú najmä asymetriu ich mediálnej prezentácie. Popisujú Diskuze / 401 sa spôsoby, akými parlamentná strana SNS implicitne (prostredníctvom bilbordu) a explicitne (vo svojich vyjadreniach) znevažuje celú etnickú skupinu. Z analýzy vyplýva, že hoci médiá uvádzajú názory rôznych diskurzívnych aktérov, v textoch chýba prezentácia argumentov, ktoré by pomohli širokej verejnosti pochopiť, prečo dehonestácia tejto stigmatizovanej skupiny je neprijateľná. Politické zneužívanie „rómskeho problému“ novinári demaskovali, avšak nevysvetlili negatívne dôsledky falošnej etnizácie nezamestnanosti, chudoby a verejnej dehonestácie Rómov., The study deals critically with discursive activities of politics and newspapermen which serve as models for imitation of confirmation or on the contrary of opposing the negative stereotypes against Roma. The thesis that the media marketing of political parties misuses the discriminated and marginalized groups for achieving the party goals is empirically verified. The meaning structures of pre-election billboard are analyzed by the means of critical systemic linguistics and thus reveals the intentions of communication of political party with the public. The authors identify the discursive positions of diverse participants, alternatively their argumentation strategies, and critically reflect especially the asymmetry of their media presentation by procedures of critical discursive analysis of media texts about the anti-Roma billboard. The ways how the parliament party SNS implicitly (through the billboard) and explicitly (in its statements) discredit the whole ethnic group are described. From the analysis follows that although media present the opinions of different discourse participants, the texts are lacking the presentation of arguments that could help the wide public to understand why the dehonestation of this stigmatized group is unacceptable. - Associative memory (AM) is a very important part of the theory of neural networks. Although the Hebbian learning rule is always used to model the associative memory, it easily leads to spurious state because of the linear outer product method. In this work, nonlinear function constitution and dynamic synapses, against a spurious state for associative memory neural network are proposed. The model of the dynamic connection weight and the updating scheme of the states of neurons are presented. Nonlinear function constitution improves the conventional Hebbian learning rule to be a nonlinear outer product method. The simulation results show that both nonlinear function constitution and dynamic synapses can effectively enlarge the attractive basin. Comparing to the existing memory models, associative memory of neural network with nonlinear function constitution can both enlarge the attractive basin and increase the storage capacity. Owing to dynamic synapses, the attractive basin of the stored patterns is further enlarged, at the same time the attractive basin of the spurious state is diminished. But the storage capacity is decreased by using the dynamic synapses.

- Anti-suit injunctions are rather unknown in the civil law legal system of the Czech Repbulic. The policy reasons for the rejection of anti-suit injunctions in the Czech Republic are rooted in the constitutional right of every person to assert, through the legally prescribed procedure, their rights before an independent and impartial court or, in specified cases, before another body. In other words, every person may commence proceedings before a competent court, or respectively before any court or another body (e.g. like an arbitral tribunal, etc.), which is then exclusively empowered to rule on its jurisdiction. Therefore, there is also no distinction between anti-suit injunctions in domestic and international litigation. Czech law does not have any alternative procedural or substantive devices that may have similar functions as anti-suit injunctions related to arbitration. - The endothelin axis (endothelins and their receptors) is strongly involved in physiological and pathological processes. ET-1 plays a crucial role in particular in tumor diseases. Endothelin-1 receptors (ETA and ETB) are deregulated and overexpressed in several tumors such as melanoma and glioma. We studied the binding of 24 monoclonal antibodies directed against human ETB receptors (hETB) to different melanoma cell lines. Few of these mAbs bound to all the melanoma cell lines. One of them, rendomab B49, bound to ETB receptors expressed at the surface of human glioma stem cells. More recently, we produced new antibodies directed against human ETA receptor (hETA). Several antibodies have been isolated and have been screened on different tumoral cells lines. As for the mAbs directed against the hETB receptor only some of new antibodies directed against ETA receptor are capable to bind the human tumoral cell lines. Rendomab A63 directed against hETA is one of them. We report the specificity and binding properties of these mAbs and consider their potential use in diagnosis by an in vivo imaging approach., A.
