Sensing and classification of drought stress levels are very important to agricultural production. In this work, rice drought stress levels were classified based on the commonly used chlorophyll a fluorescence (ChlF) parameter (Fv/Fm), feature data (induction features), and the whole OJIP induction (induction curve) by using a Support Vector Machine (SVM). The classification accuracies were compared with those obtained by the K-Nearest Neighbors (KNN) and the Ensemble model (Ensemble) correspondingly. The results show that the SVM can be used to classify drought stress levels of rice more accurately compared to the KNN and the Ensemble and the classification accuracy (86.7%) for the induction curve as input is higher than the accuracy (43.9%) with Fv/Fm as input and the accuracy (72.7%) with induction features as input. The results imply that the induction curve carries important information on plant physiology. This work provides a method of determining rice drought stress levels based on ChlF.
Growing evidence suggests that diabetes mellitus is associated with impairment of the intestinal barrier. However, it is not clear so far if the impairment of the intestinal barrier is a consequence of prolonged hyperglycemia or the consequence of external factors influencing the gut microbiota and intestinal mucosa integrity. Aim of the study was to perform an estimation of relationship between serological markers of impairment of the intestinal barrier: intestinal fatty acid-binding protein (I-FABP), cytokeratin 18 caspase-cleaved fragment (cCK-18), and soluble CD14 (sCD14) and markers of prolonged hyperglycemia, such as the duration of diabetes mellitus and glycated hemoglobin (HbA1c) via a correlation analysis in patients with diabetes mellitus. In 40 adult patients with type 1 diabetes mellitus and 30 adult patients with type 2 diabetes mellitus the estimation has been performed. Statistically significant positive correlation was found between cCK-18 and HbA1c (r=0.5047, p=0.0275) in patients with type 1 diabetes mellitus with fading insulitis (T1D). In patients with type 1 diabetes mellitus with ongoing insulitis (T1D/INS) and in patients with type 2 diabetes mellitus (T2D), no statistically significant positive correlations were found between serological markers of intestinal barrier impairment (I-FABP, cCK-18, sCD14) and duration of diabetes or levels of HbA1c. Similarly, in cumulative cohort of patients with T1D/INS and patients with T1D we revealed statistically positive correlation only between HbA1c and cCK-18 (r=0.3414, p=0.0311). Surprisingly, we found statistically significant negative correlation between the duration of diabetes mellitus and cCK-18 (r=-0.3050, p=0.0313) only in cumulative group of diabetic patients (T1D, T1D/INS, and T2D). Based on our results, we hypothesize that the actual condition of the intestinal barrier in diabetic patients is much more dependent on variable interactions between host genetic factors, gut microbiota, and environmental factors rather than effects of long-standing hyperglycemia (assessed by duration of diabetes mellitus or HbA1c).
Toxoplasmosis is a parasitic disease resulting from infection with the apicomplexan Toxoplasma gondii (Nicolle et Manceaux, 1908), one of the world's most common parasites in warm-blooded animals, including humans. Sources of infection can be exposed to infected cat faeces, mother-to-child transmission during pregnancy, and notably, the consumption of undercooked contaminated meat of intermediate hosts. In Thailand, water buffaloes are highly valued for rice cultivation, traditional culture and meat production. Like several other mammalian species, these animals play a role as reservoirs of T. gondii, thus representing a threat to human health. The seroprevalence of T. gondii infection in swamp buffaloes was examined in southern and northeastern Thailand. In total, serum samples of 721 water buffaloes were collected from seven provinces (Ubon Ratchathani, Roi Et, Si Sa Ket, Surin, Buri Ram, Sakon Nakhon, and Songkhla) and examined for the presence of T. gondii infection using commercial latex agglutination test kits (TOXOCHECK-MT, Eiken Chemical Co., Tokyo, Japan). Of the 721 animals analysed, 49 (6.8%) were positive for T. gondii. Songkhla province had the highest seroprevalence (14.7%) among the seven provinces covered in this survey. There was a potential risk to local citizens of T. gondii infection identified by the present study, notably in northeastern Thailand, where despite lower seroprevalence consuming raw buffalo meat salad should be restricted to avoid the risk of zoonotic infections.
Nocturnal pollinators such as moths have received less attention than diurnal insects. To elucidate whether nocturnal moths are important pollinators, we observed both the diurnal and nocturnal visitors to the flowers of Uncaria rhynchophylla (Rubiaceae) in a warm-temperate forest in central Japan. The diurnal visitors included various taxonomic groups (e.g., bees, hoverflies and butterflies). The nocturnal visitors were exclusively moths (Geometridae, Erebidae, Noctuidae and Crambidae). Pollen grains of U. rhynchophylla were attached to both diurnal and nocturnal visitors. Although diurnal flower visitors carried pollen grains of other plant species, nocturnal moths did not carry heterospecific pollen grains. Following their first appearance, the invasive fishes Pseudorasbora parva and Perccottus glenii have been in Lithuania for several decades. However, until recently, information relating to their distribution and secondary spread was limited. For this reason, suitable habitats for these fish species were surveyed for their presence across the entire country. Additionally, all previously reported records on the presence of these species were summarized. Results revealed P. glenii to be widely distributed within the country with abundant populations in habitats suitable for the species. The recent distribution of P. parva is restricted to only a few water bodies. It was shown that both species are associated with human mediated transfer, while no natural dispersal of these invasive species was observed. The results of this study suggest that the invasion of Lithuanian inland waters by P. parva and P. glenii is still ongoing, and their occurrence in numerous water bodies, which are still devoid of these species, now seems probable. Demonstrated vectors of P. parva and P. glenii introductions in Lithuania highlight the importance of controlling and screening human activities related to aquaculture, recreational angling and the ornamental fish trade in order to restrict further P. glenii and P. parva expansion in this region.
Extracellular DNA (ecDNA) activates immune cells and is involved in the pathogenesis of diseases associated with inflammation such as sepsis, rheumatoid arthritis or metabolic syndrome. DNA can be cleaved by deoxyribonucleases (DNases), some of which are secreted out of cells. The aim of this experiment was to describe plasma DNase activity in relation to extracellular DNA in adult rats, to analyse potential sex differences and to prove whether they are related to endogenous testosterone. Adult Lewis rats (n=28) of both sexes were included in the experiment. Male rats were gonadectomized or sham-operated and compared to intact female rats. Plasma ecDNA and DNase activity were measured using fluorometry and single radial enzyme diffusion assay, respectively. Concentrations of nuclear ecDNA and mitochondrial ecDNA were determined using real-time PCR. Females had 60% higher plasma DNase activity than males (p=0.03). Gonadectomy did not affect plasma DNase aktivity in males. Neither the concentration of total ecDNA, nor nuclear or mitochondrial DNA in plasma differed between the groups. No significant correlations between DNase aktivity and ecDNA were found. From previous studies on mice, it was expected, that male rats will have higher DNase activity. In contrast, our study in rats showed the opposite sex difference. This sex difference seems not to be caused by endogenous testosterone. Interestingly, no sex differences were observed in plasma ecDNA suggesting a complex or missing association between plasma ecDNA and DNase. The observed sex difference in plasma DNase aktivity should be taken into account in animal models of ecDNAassociated diseases.
The sex steroid hormones (SSHs) such as testosterone, estradiol, progesterone, and their metabolites have important organizational and activational impacts on the brain during critical periods of brain development and in adulthood. A variety of slow and rapid mechanisms mediate both organizational and activational processes via intracellular or membrane receptors for SSHs. Physiological concentrations and distribution of SSHs in the brain result in normal brain development. Nevertheless, dysregulation of hormonal equilibrium may result in several mood disorders, including depressive disorders, later in adolescence or adulthood. Gender differences in cognitive abilities, emotions as well as the 2-3 times higher prevalence of depressive disorders in females, were already described. This implies that SSHs may play a role in the development of depressive disorders. In this review, we discuss preclinical and clinical studies linked to SSHs and development of depressive disorders. Our secondary aim includes a review of up-to-date knowledge about molecular mechanisms in the pathogenesis of depressive disorders. Understanding these molecular mechanisms might lead to significant treatment adjustments for patients with depressive disorders and to an amelioration of clinical outcomes for these patients. Nevertheless, the impact of SSHs on the brain in the context of the development of depressive disorders, progression, and treatment responsiveness is complex in nature, and depends upon several factors in concert such as gender, age, comorbidities, and general health conditions.
The expression of sexually dimorphic phenotypes from a shared genome between males and females is a longstanding puzzle in evolutionary biology. Increasingly, research has made use of transcriptomic technology to examine the molecular basis of sexual dimorphism through gene expression studies, but even this level of detail misses the metabolic processes that ultimately link gene expression with the whole organism phenotype. We use metabolic profiling in Drosophila melanogaster to complete this missing step, with a view to examining variation in male and female metabolic profiles, or metabolomes, throughout development. We show that the metabolome varies considerably throughout larval, pupal and adult stages. We also find significant sexual dimorphism in the metabolome, although only in pupae and adults, and the extent of dimorphism increases throughout development. We compare this to transcriptomic data from the same population and find that the general pattern of increasing sex differences throughout development is mirrored in RNA expression. We discuss our results in terms of the usefulness of metabolic profiling in linking genotype and phenotype to more fully understand the basis of sexually dimorphic phenotypes., Fiona C. Výzkumy dlouhodobě poukazují na celou řadu zdravotních nerovností a rozdílů v kvalitě života lidí, jejichž hlavním rozlišujícím znakem je jejich sexuální orientace, sexuální sebe-identifikace či obecně sexualita. Tato rozmanitost ovšem bývá jen zřídka běžně zařazována mezi demografické otázky, a tak se aktuálně potýkáme nejen s nedostupností důležitých dat, ale také s celou řadou nejasností, které s "měřením sexuality" v rámci výzkumných šetření souvisí. Tento článek si klade čtyři vzájemně propojené cíle. Nejprve přispět k diskusi, která by typic-kou nepřítomnost položek zjišťujících sexualitu respondentů v rámci relevantních studií identifikovala jako významnou bariéru omezující dostupnost informací o zdraví neheterosexuálních lidí. Následně vyjasnit klíčové terminologické problémy, které se pojí zejména s neustále se vyvíjející sexuálně-identitní terminologií. Dále se zaměřuje na diskusi metodologických, ale také kontextuálních úskalí, která se dotýkají problematiky dotazování v oblasti sexuality a přispívají k různým typům výběrových zkreslení. V závěrečné části je diskutována populární otázka "reprezentativity" dostupných výsledků výzkumných šetření a s využitím aktuálních českých a zahraničních reprezentativních šetření zodpovězena otázka, zda stále platí, že neheterosexuální lidé tvoří 4 % populace., Research has long pointed to a number of health inequalities and differences in the quality of life of people, whose main distinguishing feature is their sexual orientation, sexual self-identification, or sexuality in general. However, this diversity is rarely included among demographic questions, so we currently face not only the unavailability of essential data but also many ambiguities associated with the "measurement of sexuality" in research. This article has four interconnected primary goals. First, to contribute to the discussion, which may identify the absence of items determining the sexuality of respondents in relevant studies as a significant barrier limiting the availability of information about the health of non-heterosexual people. Further, it clarifies key terminological issues, which are mainly related to the continually evolving sexual-identity terminology. Third, it focuses on the discussion of methodological but also contextual pitfalls, which touch on the issue of measuring in the field of sexuality and con-tribute to various types of selection bias. In the final part, the question of "representativeness" of available research results is discussed and using current Czech and foreign representative surveys, the question of whether it is still true that non-heterosexual people make up 4% of the population is addressed., Michal Pitoňák., and Obsahuje bibliografické odkazy
Glucagon-like-peptide 2 (GLP-2) is an endogenous enteroendocrine physiological trophic peptide. Glepaglutide is a novel long-acting GLP-2 analog under development for the treatment of patients with Short Bowel Syndrome (SBS). The objective of this work was to compare the small intestinal trophic effects in both genders following short (1 week) versus long-term (26-39 weeks) GLP-2 treatment in Wistar rats and Beagle dogs. Following both short- and long-term treatment with glepaglutide, a significant dose-dependent intestinotrophic effect was seen in both genders and species. At all doses increased length and weight of the small intestine as well as macroscopic thickening and villous hypertrophy were noted in all segments of the small intestine, without any differences between genders. The findings were still present following a 6-week recovery period, indicating long-acting intestinotrophic effects of glepaglutide. These studies demonstrate that a long-acting GLP-2 analog (glepaglutide) has a fast onset and long duration of intestinotrophic action with similar profile in both genders and species (rat and dog).
