Walnut (Juglans regia L.) plantlets were incubated during micropropagation in standard vessels (quasi confmed vessels) or in aerated vessels flushed with 360 or 20 000 cm^(C02) under irradiances of 70 (LI) and 250 (HI) pmol m"^ s'*. Plantlet morphology was strongly affected by the environment; leaf surface was increased, but shoot elongation and number of stems were reduced after increasing the irradiance of culture. Gross photosynthesis (Pq) capacity measured by using the •®02 isotope and mass-spectrometry techniques was increased by increasing photosynthetic photon flux (PPF) and CO2 concentration. Plantlets exhibited a potential for photorespiratory activity and Mehler-type reaction and a high rate of mitochondrial respiration in all vessel types and irradiances. When a long-term HI was applied, gas exchange rates (Pq and O2 uptake) were reduced in most of the vessel and PPF conditions, except in quasi confmed vessels. Under all the growth conditions, net photosynthetic rate (P^) was zero or slightly positive and the dry matter accumulation was very similar. Changes in O2 exchange, growth rate or enzyme activities linked to carbon fixation that were induced by changes in PFD and CO2 concentration showed that the photosynthetic characteristics of plantlets were typical for hetero-mixotrophic tissues.
The role of microtubules in the secretory processes in the tegument of adult trematode Fasciola hepatica L. is studied by estimating the effects of colchicine, a substance known to disrupt microtubules, on the number of T2 vesicles. Tissue slices of Ihe worm are incubated in Hedon-Fleig medium with or without 5 x 10'4M colchicine. The dynamics of the colchicine-provoked secretory block is examined by morphometry on samples processed for electron microscopy. T2 vesicles are estimated as a total number or separately within three levels (apical, sub-apical and central) of the distal tegument. The secretory block is demonstrated as reduction in the total number of T2 vesicles. The separate counting within three levels of the distal tegument demonstrates in control samples a trend of sub-apical condensation of T2 vesicles. This pattern of T2 distribution remains unchanged in colchicine-treated samples in spite of the reduction of the mean T2 counts within each of the levels examined. The data illustrate the role of microtubules in both the tegumental transport of secretory vesicles and the stratification of the organelles within the tegument.
Changes in various parameters of room temperature chlorophyll fluorescence were found during cold acciimation at 2.5 of Brassica napiis L, var. oleifera plants. At the beginning of cold stress the non-radiative energy dissipation was enhanced, and afterwards, the photochemistry was affected. After prolonged cold acciimation the fiill recovery of photochemical activity was observed when the plants had been transferred to higher temperature (20/15 oC) for several days.
Damage of molecules as a consequence of oxidative stress has been implicated in the pathogenes is of chronic diseases related to aging. Diet is a key environmental factor affecting the incidence of many chronic diseases. Antioxidant substances in diet enhance the DNA, lipid and protein protection by increasing the scavenging of free radicals. Products of oxidative damage of DNA (DNA strand breaks with oxidized purines or oxidized pyrimidines), lipids (conjugated dienes of fatty acids) and proteins (carbonyls) in relation to nutrition (vegetarian diet vs. non-vegetarian, traditional mixed diet) were measured in young women aged 20-30 years (46 vegetarians, 48 non-vegetarians) vs. older women aged 60-70 years (33 vegetarians, 34 non-vegetarians). In young subjects, no differences in values of oxidative damage as well as plasma values of antioxidative vitamins (C, β-carotene) were observed between vegetarian and non-vegetarian groups. In older vegetarian group significantly reduced values of DNA breaks with oxidized purines, DNA breaks with oxidized pyrimidines and lipid peroxidation and on the other hand, significantly increased plasma values of vitamin C and β-carotene were found compared to the respective non-vegetarian group. Significant age dependences of measured parameters (increase in all oxidative damage products and decrease in plasma vitamin concentrations in older women) were noted only in non-vegetarians. Vegetarian values of older women vs. young women were similar or non-significantly changed. Two greenhouse experiments were conducted in order to investigate the effects of different levels of water stress on gas exchange, chlorophyll fluorescence, chlorophyll content, antioxidant enzyme activities, lipid peroxidation, and yield of tomato plants (Solanum lycopersicum cv. Jinfen 2). Four levels of soil water content were used: control (75 to 80% of field water capacity), mild water stress (55 to 60%), moderate water stress (45 to 50%), and severe water stress (35 to 40%). The controlled irrigation was initiated from the third leaf stage until maturity. The results of
two-year trials indicated that the stomatal conductance, net photosynthetic rate, light-saturated photosynthetic rate, and saturation radiation decreased generally under all levels of water stress during all developmental stages, while compensation radiation and dark respiration rate increased generally. Water stress also declined maximum quantum yield of PSII photochemistry, electron transfer rate, and effective quantum yield of PSII photochemistry, while nonphotochemical quenching increased in all developmental stages. All levels of water stress also caused a marked reduction of chlorophyll a, chlorophyll b, and total chlorophyll content in all developmental stages, while activities of antioxidant enzymes, such as superoxide dismutase, peroxidase, and catalase, and lipid peroxidation increased., X. Sleep is regulated by complex biological systems and environmental influences, neither of which is fully clarified. This study demonstrates differential effects of partial sleep deprivation (SD) on sleep architecture and psychomotor vigilance task (PVT) performance using two different protocols (sequentially) that each restricted daily sleep to 3 hours in healthy adult men. The protocols differed only in the period of sleep restriction; in one, sleep was restricted to a 3-hour block from 12:00 AM to 3:00 AM, and in the other, sleep was restricted to a block from 3:00 AM to 6:00 AM. Subjects in the earlier sleep restriction period showed a significantly lower percentage of rapid-eye-movement (REM) sleep after 4 days (17.0 vs. 25.7 %) and a longer latency to the onset of REM sleep (L-REM) after 1 day (78.8 vs. 45.5 min) than they did in the later sleep restriction period. Reaction times on PVT performance were also better (i.e. shorter) in the earlier SR period on day 4 (249.8 vs. 272 ms). These data support the view that earlier-night sleep may be more beneficial for daytime vigilance than later-night sleep. The study also showed that cumulative declines in daytime vigilance resulted from loss of total sleep time, rather than from specific stages, and underscored the reversibility of SR effects with greater amounts of sleep., H. Drought stress has multiple effects on the photosynthetic apparatus. Herein, we aimed to study the effect of drought stress on fluorescence characteristics of PSII in leaves of Plectranthus scutellarioides and explore potentially underlying mechanisms. Plants of P. scutellarioides were grown in a greenhouse and subjected to drought (DS, drought-stressed) or daily irrigation (control group). Leaf chlorophyll (Chl) index and induction kinetics curves of Chl a fluorescence and the JIP-test were used to evaluate effects of drought lasting for 20 d. Our results showed that both the leaf and soil relative water content decreased with increasing treatment duration. The leaf Chl index was reduced to half in the DS plants compared with the control group after 20 d. The minimal fluorescence in the DS plants was higher than that in the control plants after 10 d of the treatment. Maximum photochemical efficiency and lateral reactivity decreased with increasing treatment duration in the DS plants. With the continuing treatment, values of absorption flux per reaction center (RC), trapped energy flux per RC, dissipated energy flux per RC, and electron transport flux per RC increased in the earlier stage in the DS plants, while obviously decreased at the later stage of the treatment. The present study aimed to determine effects of drought stress on Lycium ruthenicum Murr. seedlings. Our results showed that mild drought stress was beneficial to growth of L. ruthenicum seedlings. Their height, basal diameter, crown, leaf number, stem dry mass, leaf and root dry mass increased gradually when the soil water content declined from 34.7 to 21.2%. However, with further decrease of the soil water content, the growth of L. ruthenicum seedlings was limited. After 28 d of treatment, the seedlings were apparently vulnerable to drought stress, which resulted in significant leaf shedding and slow growth. However, growth was restored after rehydration. Drought treatments led to a decrease in contents of chlorophyll (Chl) a, b, and Chl (a+b) and increase in the Chl a/b ratio. After rewatering, the Chl content recovered to the content of the control plants. Under drought stress, minimal fluorescence and nonphotochemical quenching coefficient increased, thereby indicating that L. ruthenicum seedlings could protect PSII reaction centres from damage. Maximum fluorescence, maximum quantum yield, actual quantum yield of PSII photochemistry, and photochemical quenching decreased, which suggested that drought stress impacted the openness of PSII reaction centres. A comparison of these responses might help identify the drought tolerance mechanisms of L. ruthenicum. Effects of ectopic pacing on left ventricular repolarization were studied in six anesthetized open-c hest chickens. In each animal, unipolar electrograms were acquired from as many as 98 sites with 14 plunge needles (seven transmural locations between epicardium and endocardium in each needle). Activation-recovery intervals (ARIs), corrected to the cycle length, were used for estimating repolarization. At baseline, the nonuniform ARI distribution in the left ventricle resulted in the apicobasal differences being greater than the transmural gradient. Nonuniform ARI prolongation caused by ectopic pacing resulted in decreasing the transmural repolarization gradient and increasing the differences in th e apex-to-base direction. The basal, but not apical transmural differences contributed to the total left ventricular transmural gradient. The total left ventricular apicobasal gradient was contribute d by the apicobasal differences in mid-myocardial and subendoc ardial layers more than in subepicardial ones. Thus, in in situ chicken hearts, the transmural and apicobasal ARI gradients exist within the left ventricle with the shortest ARIs in the basal subepicardium and the longest ARIs in the subendocardium of th e apical and middle parts of the left ventricle. Ascorbic acid (Asc) is a major plant antioxidant. L-galactono-1,4-lactone dehydrogenase (GLDH) is an enzyme that catalyzes the last step of Asc biosynthesis in higher plants. Effects of endogenous Asc on resistance to high-temperature stress were studied by using GLDH-overexpressed (GO-2) and GLDH-suppressed transgenic rice (GI-2) as experimental materials. After high-temperature treatment, the maximal quantum yield of PSII was significantly lower in GI-2, and higher in GO-2 compared to wild type rice. The content of reactive oxygen species (ROS) was the highest in GI-2. The higher Asc content resulted in lower lipid peroxidation in GO-2. The contents of chlorophyll, soluble proteins, and Rubisco large and small subunit were positively correlated to the Asc content. These results show that the higher Asc content reduced the accumulation of ROS and maintained the function of rice leaves.
