The lipid molecule, lysophosphatidylinositol (LPI), is hypothesis ed to form part of a novel lipid signalling system that involves the G protein- coupled receptor GPR55 and distinct in tracellular signalling cascades in endothelial cells. This work aimed to study the possible mechanisms involved in LPI -evoked cytosolic Ca 2+ mobilization in human brain microvascular endothelial cells. Changes in intracellular Ca 2+ concentrations were meas ured using cell population Ca 2+ assay. LPI evoked biphasic elevation of intracellular calcium concentration, a rapid phase and a sustained phase. The rapid phase was attenuated by the inhibitor of PLC (U 73122), inhibitor of IP 3 receptors, 2 -APB and the de pletor of endoplasmic reticulum Ca 2+ store, thapsigargin. The sustained phase, on the other hand, was enhanced by U 73122 and abolished by the RhoA kinase inhibitor, Y -27632. In conclusion, the Ca 2+ signal evoked by LPI is characterised by a rapid phase of Ca 2+ release from the endoplasmic reticulum, and requires activation of the PLC -IP 3 signalling pathway. The sustained phase mainly depends on RhoA kinase activation. LPI acts as novel lipid signalling molecule in endothelial cells, and elevation of cytosolic Ca 2+ triggered by it may present an important intracellular message required in gene expression and controlling of vascular tone., Y. Diatomite is a siliceous sedimentary rock, and it can be used in many industrial applications due to its high porosity, low density porous and permeable structure, chemical resistance, high purity, high specific surface area, absorptive capacity and to its absorbent properties. The best known Portuguese occurrences of diatomite are located in the regions of Óbidos and Rio Maior. The purpose of this research is to characterize the Rio Maior and Amieira diatomite and beneficiate this rock to be used in some potential applications locally or not. The studied diatomites present a fine texture; chemically the samples have a high SiO2 content, the principal minerlas are quartz and amorphous opal. The studied diatomites benefited with temperature and addition of NaCl (acting as accelerator of the crystallization process), with 5 % of NaCl being reduced the transformation temperature of the amorphous opal in opal C / CT at about 200 ºC. The diatomites from Rio Maior and Amieira in natural state has an enormous potential as absorbent, of heavy metals (e.g. in soil contamination) or dyes (textile industry). Catalytic test reaction of methylbutynol (MBOH) conversion was applied to investigation of natural clays and non-clay minerals from Jordanian (bentonite, kaolinite, diatomite, zeolit e) and Russian (palygorskite, kaolinite, hydromica) deposits as well as H-ZSM-5. Palygorskite and kaolinite containing samples have shown the highest catalytic activity. Conversionove r Jordanian clay minerals decreased in the order: zeolite > bentonite > red kaolinite > white kaolinite > diatomite that agreed to change of acidity, surface area of samples. Sufficient deactivation was observed for H-ZSM-5, kaolinite and hydromica due to adsorption of secondary product in micropores of raw minerals. Products of both acidic and basic pathways of test reaction were formed indicating the presence of acid and basic sites on clays surface, apart from acidic catalyst H-ZSM-5. Theyield ratio of acidic to basic products was above the unity for zeolite, bentonite, red kaolinite, and palygorskite; equal to unity for diatomite and hydromica, and less then unity for white kaolinite. It was observed for kaolinite containing sample that high activation temperature increased conversion of MBOH owing to fo rmation of additional basic sites as a result of mineral dehydroxylation. Let G be a finite group, and let N(G) be the set of conjugacy class sizes of G. By Thompson’s conjecture, if L is a finite non-abelian simple group, G is a finite group with a trivial center, and N(G) = N(L), then L and G are isomorphic. Recently, Chen et al. contributed interestingly to Thompson’s conjecture under a weak condition. They only used the group order and one or two special conjugacy class sizes of simple groups and characterized successfully sporadic simple groups (see Li’s PhD dissertation). In this article, we investigate validity of Thompson’s conjecture under a weak condition for the alternating groups of degrees p+1 and p+2, where p is a prime number. In this review we present immunohistochemical methods for visualization of capillaries and muscle fibres in thick muscle sections. Special attention is paid to the procedures that preserve good morphology. Applying confocal microscopy and virtual 3D stereological grids, or tracing of capillaries in virtual reality, length of capillaries within a muscle volume or length of capillaries adjacent to a muscle fibre per fibre length, fibre surface area or fibre volume can be evaluated by an unbiased approach. Moreover, 3D models of capillaries and muscle fibres can be produced. Comparison of the developed methods with counting capillary profiles from 2D sections is discussed and the reader is warned that counting capillary profiles from 2D sections can underestimate the capillary length by as much as 75 percent. Application of the described 3D methodology is illustrated by the anatomical remodelling of capillarity during acute denervation and early reinnervation in the ra t soleus and extensor digitorum longus muscles., I. The complete mitochondrial genome (mitogenome) of Spilarctia robusta (Lepidoptera: Noctuoidea: Erebidae) was sequenced and analyzed. The circular mitogenome is made up of 15,447 base pairs (bp). It contains a set of 37 genes, with the gene complement and order similar to that of other lepidopterans. The 12 protein coding genes (PCGs) have a typical mitochondrial start codon (ATN codons), whereas cytochrome c oxidase subunit 1 (cox1) gene utilizes unusually the CAG codon as documented for other lepidopteran mitogenomes. Four of the 13 PCGs have incomplete termination codons, the cox1, nad4 and nad6 with a single T, but cox2 has TA. It comprises six major intergenic spacers, with the exception of the A+T-rich region, spanning at least 10 bp in the mitogenome. The nucleotide composition of the genome is greatly A+T biased (81.09%), with a negative AT skewness (-0.007), indicating the presence of fewer As than Ts, similar to other Noctuoidea. The A+T-rich region is 343 bp long, and contains some conserved regions, including an "ATAGA" motif followed by a 19 bp poly-T stretch, a microsatellite-like (AT)9 and a poly-A element, a characteristic shared with other lepidopteran mitogenomes. Phylogenetic analysis, based on 13 PCGs using Maximum likelihood methods revealed that S. robusta belongs to the superfamily Noctuoidea., Yu Sun, Sen Tian, Cen Qian, Yu-Xuan Sun, Muhammad N. Chemical stability and aggregation activity of assembly pheromone of argasid ticks and its synthetic purine analogues were studied during long-lasting storage at room temperature as the potential components of attractant/acaricide mixtures. Guanine spherules from dry excreta of Omithodoros moubata Murray, 1877 were very stable in their chemical composition and did not suffer from purine degradation. However, an intensive purine conversion into uric acid occurred in samples of the fluid of the excreta of Argas persicus (Oken, 1818) and to a lesser extent also in an artificial mixture of synthetic guanine, xanthine and hypoxanthine in saline. The presence of bacteria Bacillus sp. and the moulds Talaromyces flavus (syn. Pénicillium dangeardii) and Aspergillus carbonarius, isolated from some samples, might explain the enzymic degradation of purines. The suspension of guanine hydrochloride in saline or in saline with ethanol (1 : 1 v/v) as a potential acaricide solvent, and with diatomaceous earth as the pheromone carrier, was very stable and no guanine degradation occurred. This proved to be suitable for potential use mixed with acaricides for tick control. The assembly of A. persicus males, on most of the substrates tested, was very high up to day 77 of experiment but decreased significantly on day 114-119 in samples of synthetic analogue of assembly pheromone variant 1 in which the absolute amount of guanine in solutions strongly decreased or disappeared completely.
Nitric oxide (NO) plays a crucial role not only in regulation of blood pressure but also in maintenance of cardiac autonomic tone and its deficiency induced hypertension is accompanied by cardiac autonomic dysfunction. However, underlying mechanisms are not clearly defined. We hypothesized that sympathetic activation mediates hemodynamic and cardiac autonomic changes consequent to deficient NO synthesis. We used chemical sympathectomy by 6-hydroxydopamine to examine the influence of sympathetic innervation on baroreflex sensitivity (BRS) and heart rate variability (HRV) of chronic NG-nitro-L-arginine methyl ester (L-NAME) treated adult Wistar rats. BRS was determined from heart rate responses to changes in systolic arterial pressure achieved by intravenous administration of phenylephrine and sodium nitroprusside. Time and frequency domain measures of HRV were calculated from 5-min electrocardiogram recordings. Chronic L-NAME administration (50 mg/kg per day for 7 days orally through gavage) in control rats produced significant elevation of blood pressure, tachycardia, attenuation of BRS for bradycardia and tachycardia reflex and fall in time as well as frequency domain parameters of HRV. Sympathectomy completely abolished the pressor as well as tachycardic effect of chronic L-NAME. In addition, BRS and HRV improved after removal of sympathetic influence in chronic L-NAME treated rats.
