The role of neuronal nitric oxide synthase (nNOS) in the pathophysiology of epilepsy and seizures remains disputable. One of the reasons why results from the acute in vivo studies display controversies might be the effect on the regulation of cerebral blood flow (CBF) during pharmacologically induced alterations of NO system. We examined neurovascular coupling in the rat sensorimotor cortex in response to transcallosal stimulation under nNOS inhibition by 7-nitroindazole (7-NI). Adult Wistar rats were anesthetized with urethane and epidural silver EEG electrodes were implanted over sensorimotor cortices. Regional CBF was measured by Laser Doppler Flowmetry (LDF). We catheterized a common carotid artery to measure arterial blood pressure (BP). 7-NI did not significantly affect blood pressure and heart rate. Electrophysiological recordings of evoked potentials (EPs) revealed no effect on their ampl itude, rhythmic potentiation or depression of EPs. Transcallosal stimulation of the contralateral cortex induced a frequency dependent rise in CBF. Although 7-NI did not significantly affect basal CBF and cortical excitability, hemodynamic responses to the transcallosal stimulation were diminished implicating a role of nNOS in neurovascular coupling. We showed recently that increasing kidney epoxyeicosatrienoic acids (EETs) by blocking soluble epoxide hydrolase (sEH), an enzyme responsible for EETs degradation, retarded the development of renal dysfunction and progression of aorto-caval fistula(ACF)-induced congestive heart failure (CHF) in Ren-2 transgenic hypertensive rats (TGR). In that study the final survival rate of untreated ACF TGR was only 14 % but increased to 41 % after sEH blockade. Here we examined if sEH inhibition added to renin-angiotensin system (RAS) blockade would further enhance protection against ACF-induced CHF in TGR. The treatment regimens were started one week after ACF creation and the followup period was 50 weeks. RAS was blocked using angiotensinconverting enzyme inhibitor (ACEi, trandolapril, 6 mg/l) and sEH with an sEH inhibitor (sEHi, c-AUCB, 3 mg/l). Renal hemodynamics and excretory function were determined two weeks post-ACF, just before the onset of decompensated phase of CHF. 29 weeks post-ACF no untreated animal survived. ACEi treatment greatly improved the survival rate, to 84 % at the end of study. Surprisingly, combined treatment with ACEi and sEHi worsened the rate (53 %). Untreated ACF TGR exhibited marked impairment of renal function and the treatment with ACEi alone or combined with sEH inhibition did not prevent it. In conclusion, addition of sEHi to ACEi treatment does not provide better protection against CHF progression and does not increase the survival rate in ACF TGR: indeed, the rate decreases significantly. Ant-aphid mutualism is considered to be a beneficial association for the individuals concerned. The population and fitness of aphids affected by ant attendance and the outcome of this relationship affects the host plant of the aphid. The main hypothesis of the current study is that ant tending decreases aphid developmental time and/or increases reproduction per capita, which seriously reduces host plant fitness. The effect of attendance by the ant Tapinoma erraticum (Latreille, 1798) (Hymenoptera: Formicidae) on population growth and duration of different developmental stages of Aphis gossypii Glover (Hemiptera: Aphididae) were determined along with the consequences for the fitness of the host plant of the aphid, Vicia faba L., in greenhouse conditions. The initial aphid density was manipulated in order to study aphid performance due to density-dependent changes in ant attendance. The population growth rate of ant attended aphids was more than that of unattended aphids. However, the ratio of 1st-2nd nymphs to adults in aphid populations attended by ants was lower. The yields of bean plants on which the aphids were attended by ants were significantly greater than those of unattended plants. This study was designed to validate the measures of heart period variability for assessing of autonomic nervous system control in calves. Eight calves received an injection of either 0.5 mg/kg atenolol (sympathetic tone blockade), 0.2 mg/kg atropine sulfate (parasympathetic tone blockade), 0.5 mg/kg atenolol + 0.2 mg/kg atropine sulfate (double autonomic blockade) or saline. In the time-domain, we calculated the mean instantaneous heart rate (HR), mean of RR intervals (MeanRR), standard deviation of RR intervals (SDRR) and that of the difference between adjacent intervals (RMSSD). In the frequency-domain, the power of the spectral band 0-1 Hz (TPW), the power of the 0-0.15 Hz band (LF), that of the 0.15-1 Hz band (HF), and the LF/HF ratio were considered. The net vago-sympathetic effect (VSE) was calculated as the ratio of MeanRR in a defined situation to MeanRR during the double blockade. Atenolol injection had no effect on cardiac activity, whereas atropine induced large modifications which were moderated when atenolol was administered at the same time. VSE, HR, MeanRR and RMSSD were found to be valid indicators of the parasympathetic tone of calves because of large variations due to the drug and low individual variations. Energy transfer from resynthesized protochlorophyllide (PChlide) to chlorophyll (Chl) appeared after 5 h irradiation and additional redarkening of etiolated barley leaves. The transfer was more effective after a longer greening. Benzyladenine (BA) removed delay at the onset of the energy transfer and this process was effective even at very low Chl contents. BA enhanced Chl accumulation more strongly than the development of photosystems. The balance between the rate of Chl synthesis and that of its efflux from PChlide-containing site may determine the number of Chl molecules near PChlide, and thereby the probability of the energy transfer. At a high rate of Chl synthesis a part of its molecules remained near the PChlide synthesis site. Under various conditions of irradiation the slower rate of Chl accumulation corresponded to the more efficient energy transfer. Chl accumulation at PChlide forming site probably Controls the activity of the multienzyme systém of Chl biosynthesis by a feedback mechanism. The BA addition increased the number of EF-mtramembrane particles and the munber of enzyme systems of Chl biosynthesis to the same extent.
The aim of this study was to investigate nitric oxide (NO) production and L-NAME-sensitive component of endothelium-dependent vasorelaxation in adult normotensive Wistar-Kyoto rats (WKY), borderline hypertensive rats (BHR) and spontaneously hypertensive rats (SHR). Blood pressure (BP) of WKY, BHR and SHR (determined by tailcuff) was 111±3, 140±4 and 184±6 mm Hg, respectively. NO synthase activity (determined by conversion of [3H]-Larginine) was significantly higher in the aorta of BHR and SHR vs. WKY and in the left ventricle of SHR vs. both BHR and WKY. L-NAME-sensitive component of endothelium-dependent relaxation was investigated in the preconstricted femoral arteries using the wire myograph during isometric conditions as a difference between acetylcholine-induced relaxation before and after acute NG-nitro-L-arginine methyl ester pre-treatment (L-NAME, 10-5 mol/l). Acetylcholineinduced vasorelaxation of SHR was significantly greater than that in WKY. L-NAME-sensitive component of vasorelaxation in WKY, BHR and SHR was 20±3 %, 29±4 % (p<0.05 vs. WKY) and 37±3 % (p<0.05 vs. BHR), respectively. There was a significant positive correlation between BP and L-NAME-sensitive component of relaxation of the femoral artery. In conclusion, results suggest the absence of endothelial dysfunction in the femoral artery of adult borderline and spontaneously hypertensive rats and gradual elevation of L-NAME-sensitive component of vasorelaxation with increasing blood pressure., A. The cadmium treatment of dark-grown leaves and isolated etioplast inner membranes of wheat resulted in a decrease of the amount of the 657 nm emitting (77 K fluorescence) protochlorophyllide (PChlide) form, a simultaneous increase of the 633 nm form and the appearance of a 641.5 nm emitting form. This effect did not occur if excess NADPH was added to the isolated membranes: these samples showed spectral properties identical to those of non-treated (control) samples. Inhibition of the PChlide phototransformation was observed in the cadmium-treated leaves and membranes, the irradiation resulted in the appearance of a smáli amount of chlorophyllide (Chlide) with characteristic emission band at 678 nm. If excess NADPH was added, the inhibition did not occur and flash irradiation resulted in formation of the 694 nm Chlide form similarly as in control plants.
Chlorophyll content, photosynthetíc oxygen evolution and the activities of ATP- sulphurylase (ATP-s) (E.C. 2.7,7.4.), OAS-sulphydrylase (OAS-s) (E.C, 4.2.99.8.), nitráte reductase (NR) (E.C. 1.6.6.1.) and glutamine synthetase (GS) (E.C. 6.3.1.2), were determined in leaves of Zea mays L. Dekalb cv. Sponsor plants grown in the presence of 0, 10, 100, 250 pM Cd in order to evaluate the effect of this metal on sulphate and nitráte assimilation pathway. Cd induced a slight decrease of photosynthetíc oxygen evolution (-21 % of the control at 250 pM), whereas the enzyme activities were differently influenced: OAS-s and GS increased up to 40 and 25 % of the control, respectively, at 250 pM Cd; NR showed a 20 % stimulation at 100 pM Cd, and ATP-s was slightly inhibited. The results stress the importance of experimental conditions adopted on the response of enzyme activities to Cd and suggest that the observed increases of enzyme activities are related to a defence mechanism.
Chronic continuous light exposure leads to melatonin deficiency along with complex neurohumoral activation resulting in hypertension development in rats. The aim of this study was to show, whether continuous light in duces fibrotic rebuilding of the aorta and whether the treatment with melatonin or angiotensin converting enzyme inhibitor captopril can prevent these potential alterations. In a six-week experiment, 3-month-old Wistar rats were divided into 4 groups (t en per group): controls, rats exposed to continuous light, exposed to continuous light plus treated with captopril (100 mg/kg/24 h) and exposed to continuous light plus treated with melatonin (10 mg/kg/24 h). Systolic blood pressure (SBP) and collagen type I and III in the media of thoracic aorta were me asured. Continuous light induced hypertension and fibrotic rebuilding of the aorta in terms of enhancement of collagen I and III concentration in the aortic media. Both captopril and melatonin prevented SBP rise and reduced collagen III concentration in the aorta. However, only melatonin reduced collagen I and the sum of collagen I and III in the aortic tissue. We conclude that in continuous light-induced hypertension, administration of melatonin, along with SBP reduction, decreases collagen I and III concentration in the aorta. It is suggested that antifibrotic effect of melatonin may reduce the stiffness of the aorta and small arteries and beneficially influence the nature of the pulse wave and peripheral vascular resistance., K.
